The Effect of Bovine IFN-α on the Immune Response in Guinea Pigs Vaccinated with DNA Vaccine of Foot-and-Mouth Disease Virus

In this study, we constructed recombinant plasmid pcDNA3.1/P12X3C3D including P1, 2A, 3C, 3D and part of 2B gene of FMDV and pcDNA3.1/IFN containing the gene encoding bovine IFN-α. We inoculated the DNA vaccine pcDNA3.1/P12X3C3D with or without pcDNA3.1/IFN to evaluate the effi- ciency of this DNA vaccine and the immunogenicity of DNA vaccine enhanced by the co-delivery with pcDNA3.1/IFN. After two times of vaccination with DNA vaccine, all of guinea pigs were challenged with 103 ID50 FMDV type O. Anti-FMDV antibody levels were detected by ELISA and T lymphocyte proliferation response was tested by MTT assay. The result shows that guinea pigs inoculated by pcDNA3.1/P12X3C3D alone or with pcDNA3.1/IFN generated specific antibodies and induced an FMDV-specific T lymphocyte proliferation response. FMDV challenge tests showed that one in four guinea pigs immunized by pcDNA3.1/ P12X3C3D with pcDNA3.1/IFN was protected from the FMDV serotype O infection. This result indicated that the efficiency of the DNA vaccine was enhanced by co-delivery with pcDNA3.1/IFN. However, the protection rate was considerably lower than that immunized with conventional FMD vaccine.

Effects of Soil Water Content on Cotton Root Growth and Distribution Under Mulched Drip Irrigation

The relation between soil water content and the growth of cotton root was studied for the scheme of field water and cotton yield under mulched drip irrigation. Based on the field experiments, three treatments of soil water content were conducted with 90%, 75%θf, and 60%θf （θfis field water capacity）. Cotton roots and root-shoot ratio were studied with digging method, and the soil moisture was observed with TDR （time domain reflector）, and cotton yield was measured. The results indicated that the growth of cotton root accorded with Logistic growth curve in the three treatments, the cotton root grew quickly and its weight was very high under 75%θf because of the suitable soil water condition, while grew slowly and its weight was lower under 90%θf due to water moisture beyond the suitable condition, and the root weight was in between under 60%θf For the three water treatments, the cotton root weight decreased with soil depth, and decreased more significantly in deeper soil layer with the soil moisture increasing. And the ratio of cotton root weight in 0-30 cm soil layer to the total root weight was the highest under 75%θf. The cotton root system was distributed mainly in the soil of narrow row and wide row mulched with plastic film, and little in the soil outside plastic film. The weight of cotton root was the highest in the soil of narrow row or wide row mulched with plastic film under 75%θf. Root-shoot ratio decreased with the soil moisture increasing. The soil water content affected cotton yields, and cotton yield was the highest under 75%θf. The higher soil moisture level is unfavorable to the growth of cotton root system and yield of cotton under mulched drip irrigation.

To investigate the relationship between 27 bp repeat polymorphism in intron 4 in the endothelial nitric oxide synthase (eNOS4) gene and essential hypertension in the Kazakh Chinese population, 151 patients with essential hypertension and 138 healthy people were selected from the Boertonggu countryside of Shawan region in the Xinjiang Uygur Autonomous Region of China in 2006. The polymorphism of eNOS in the two groups was detected with polymerase chain reaction assays and the genotype frequencies in each group were calculated following the Hardy-Weinberg law. Four and five tandem 27 bp repeats were designated as "a" and "b", respectively. It was found that the frequencies of b/b, b/a and a/a genotypes of the eNOS4 gene were 84.06%, 15.22% and 0.72% in the control group, and 81.46%, 15.89% and 2.65% in the hypertension group, respectively. The frequencies of gene "b" and "a" were 91.67% and 8.33% in the control group and 89.40% and 10.60% in the hypertension group, respectively. It was found that plasma eNOS activity was not associated with genotypes and alleles of eNOS gene. Plasma eNOS activity in the hypertension group was significantly decreased compared with the control group (P<0.01). The results suggest that eNOS4 gene polymorphisms are unlikely to be the major genetic susceptibility factors for essential hypertension in the Xinjiang Kazakh population. However, a positive association between plasma eNOS activity and essential hypertension has been revealed.

Density functional theory study on the structural and electronic properties of Ag-adsorbed (MgO)_n clusters

Equilibrium geometries, charge distributions, stabilities and electronic properties of the Ag-adsorbed (MgO)n (n = 1-8) clusters have been investigated by density functional theory (DFT) with generalized gradient approximation (GGA) for exchange-correlation functional. The results show that hollow site is energetically preferred for n≥4, and the incoming Ag atoms tend to cluster on the existing Ag cluster. The Mulliken populations indicate that the interaction between the Ag atom and Magnesia clusters is mainly induced by a weak atomic polarization. The adsorbed Ag atom only causes charge redistributions of the atoms near itself. The effect of the adsorbed Ag atom on the bonding natures and structural features of Magnesia clusters is minor. Furthermore, the investigations on the first energy difference, fragmentation energies and electron affinities show that the Ag(MgO)4 and Ag(MgO)6 are the most stable among studied clusters.

Modulatory effect of substance P on GABA-activated currents from rat dorsal root ganglion

AIM: To explore the modulatory effect of substance P (SP) on GABA-activated current of dorsal root ganglion(DRG) neurons in rat. METHODS: The whole-cell patch-clamp technique was used to record SP- and GABA-activated currents in neurons freshly dissociated from rat DRG neurons. Drugs were applied by rapid solutionexchange. RESULTS: Application of SP (28/41, 68.5 %) and GABA (36/41, 88.2 %) could induce concentration-dependent inward current in some cells. SP-(10 mol/L) and GABA (100 mol/L)-activated inward currents were(24483) pA (n=9) and (1.80.5) nA (n=13), respectively. The majority of GABA-activated current had obviousthree processes, the peak value (Ip), the steady state (Iss) and the desensitization (Id). The desensitization of GABA-activated current was a biphasic process, including fast and slow desensitization. However, pre-application of SP(0.0011 mol/L) could inhibit the GABA-activated inward current which was identified to be GABAA receptor-mediated current. The inhibitory effects were concentration-dependent. The inhibitory effect of SP on the peakvalue of GABA-activated current was more than the steady state of GABA-activated current. The inhibition ofGABA-activated current by SP (0.1 mol/L) was related to the time after application of SP, the inhibition of GABA-activated currents by SP reached the peak at about 4 min (49.8 %7.2 %, n=7, P<0.01) and took about 12 min toget a full recovery. The inhibition of GABA-activated currents by SP was almost completely removed after block-ade of PKC by H-7 with the re-patch clamp. CONCLUSION: Pre-application of SP exerts a more strong inhibitoryeffect on the peak value of GABA-activated current than the steady state of GABA-activated current.

To evaluate the characteristics of chicken interleukin-18 (ChIL-18) in different forms in vitro,the ChIL-18 full-length gene (ChIL-18-F) and the ChIL-18 presumed mature protein gene (ChIL-18-M) werecloned and inserted into the eukaryotic expression vector pCI,to construct recombinant pCI-ChIL-18-Fand *** recombinant plasmids were then transferred into chicken splenic lymphocytes(CSLs).Western blot showed that ChIL-18-F,with a molecular weight of 23.0 kDa,was produced in CSLstransfected by pCI-ChlL-18-F;ChIL-18-M,with a molecular weight of 19.5 kDa,was produced in CSLstransfected by *** nitric oxide (NO) level in the transfected CSLs and the culture mediumat different time points was further examined under confocal microscopy using *** results showed that both pCI-ChIL-18-F and pCI-ChIL-18-M groups showed significantincrease in intracellular and extracellular NO production compared with pCI transfected control *** suggest that both ChIL-18-F and ChIL-18-M could stimulate NO secretion in *** characterizethe intracellular distribution of ChIL-18,ChIL-18-F and ChIL-18-M were each fused to the enhanced greenfluorescent protein gene,and expressed in Vero *** results showed that the ChIL-18-F tended to themembranous region in Vero cells,while ChIL-18-M did *** indicates that the N-terminal 27 amino acidpeptide helped ChIL-18 target to Veto cell membranes.

Empirical Study on the Relationship between Farmers' Human Capital Investment and Rural Poverty

Literatures about the relationship between human capital investment and rural poverty are *** to the time-series data from 1990 to 2007,VAR model and variance decomposition research are used to study the relationship between household human capital investment and rural *** shows that there is long-run equilibrium relationship between household human capital investment and rural *** investment and health investment have significant impacts on the alleviation of rural poverty;while migration investment does not have significant impact on the alleviation of rural *** the factors causing poverty fluctuations,educational investment has greater impacts on poverty fluctuations than health investment in the short run,but health investment has greater impacts on poverty fluctuations than educational investment in the long *** on this,related countermeasures are put forward,such as further increasing the investment in education and health,implementing the compulsory education and social security system,consolidating the rural cooperative medical care,improving the retirement pension system in rural areas,and perfecting the training and technical extension system for rural population under the poverty line.
