Hereditary severe insulin resistance syndrome:Pathogenesis,pathophysiology,and clinical management

Severe insulin resistance has been linked to some of the most globally prevalent disorders,such as diabetes mellitus,nonalcoholic fatty liver disease,polycystic ovarian syndrome,and hypertension.Hereditary severe insulin resistance syndrome(H-SIRS)is a rare disorder classified into four principal categories:primary insulin receptor defects,lipodystrophies,complex syndromes,and obesity-related H-SIRS.Genes such as INSR,AKT2,TBC1D4,AGPAT2,BSCL2,CAV1,PTRF,LMna,PPARG,PLIN1,CIDEC,LIPE,PCYT1A,MC4R,LEP,POMC,SH2B1,RECQL2,RECQL3,ALMS1,PCNT,ZMPSTE24,PIK3R1,and POLD1 have been linked to H-SIRS.Its clinical features include insulin resistance,hyperglycemia,hyperandrogenism,severe dyslipidemia,fatty liver,abnormal topography of adipose tissue,and low serum leptin and adiponectin levels.Diagnosis of H-SIRS is based on the presence of typical clinical features associated with the various H-SIRS forms and the identification of mutations in H-SIRS-linked genes by genetic testing.Diet therapy,insulin sensitization,exogenous insulin therapy,and leptin replacement therapy have widely been adopted to manage H-SIRS.The rarity of H-SIRS,its highly variable clinical presentation,refusal to be tested for genetic mutations by patients’family members who are not severely sick,unavailability of genetic testing,and testing expenses contribute to the delayed or underdiagnoses of H-SIRS.Early diagnosis facilitates early management of the condition,which results in improved glycemic control and delayed onset of diabetes and other complications related to severe insulin resistance.The use of updated genetic sequencing technologies is recommended,and long-term studies are required for genotype–phenotype differentiation and formulation of diagnostic and treatment protocols.

The Adjustment Analysis Method of the Active Surface Antenna Based on Convolutional Neural Network

Active surface technique is one of the key technologies to ensure the reflector accuracy of the millimeter/submillimeter wave large reflector antenna. The antenna is complex, large-scale, and high-precision equipment,and its active surfaces are affected by various factors that are difficult to comprehensively deal with. In this paper,based on the advantage of the deep learning method that can be improved through data learning, we propose the active adjustment value analysis method of large reflector antenna based on deep learning. This method constructs a neural network model for antenna active adjustment analysis in view of the fact that a large reflector antenna consists of multiple panels spliced together. Based on the constraint that a single actuator has to support multiple panels(usually 4), an autonomously learned neural network emphasis layer module is designed to enhance the adaptability of the active adjustment neural network model. The classical 8-meter antenna is used as a case study,the actuators have a mean adjustment error of 0.00252 mm, and the corresponding antenna surface error is0.00523 mm. This active adjustment result shows the effectiveness of the method in this paper.

Anti-osteosarcoma trimodal synergistic therapy using NiFe-LDH and MXene nanocomposite for enhanced biocompatibility and efficacy

Osteosarcoma is usually resistant to immunotherapy and,thus primarily relies on surgical resection and high-dosage chemotherapy.Unfortunately,less invasive or toxic therapies such as photothermal therapy(PTT)and chemodynamic therapy(CDT)generally failed to show satisfactory outcomes.Adequate multimodal therapies with proper safety profiles may provide better solutions for osteosarcoma.Herein,a simple nanocomposite that synergistically combines CDT,PTT,and chemotherapy for osteosarcoma treatment was fabricated.In this composite,small 2D Ni Fe-LDH flakes were processed into 3D hollow nanospheres via template methods to encapsulate 5-Fluorouracil(5-FU)with high loading capacity.The nanospheres were then adsorbed onto larger 2D Ti3C2MXene monolayers and finally shielded by bovine serum albumin(BSA)to form 5-FU@Ni Fe-LDH/Ti3C2/BSA nanoplatforms(5NiTiB).Both in vitro and in vivo data demonstrated that the 5-FU induced chemotherapy,Ni Fe-LDH driven chemodynamic effects,and MXene-based photothermal killing collectively exhibited a synergistic“all-in-one”anti-tumor effect.5NiTiB improved tumor suppression rate fromnanotherapeutic platform achieved higher therapeutic efficacy with a lower agent dose,thereby minimizing side effects.Moreover,the composite is simple to produce,enabling the fine-tuning of dosages to suit different requirements.Thus,the platform is versatile and efficient,with potential for further development.

Synthesis of Long-Chain Oligomeric Donor and Acceptors via Direct Arylation for Organic Solar Cells

The rapid synthesis of structurally complicated electron donors&acceptors still remains a major challenge in organic solar cells(OSC).In this work,we developed a highly efficient strategy to access long-chain oligomeric donor and acceptors for OSC applications.A series of cyclopentadithiophene(CPDT)and benzothiadiazole(BT)-basedπ-conjugated oligomers,i.e.,three oligomeric acceptors(BTDT)n-IC(n=1—3)and one long-chain oligomeric donor(BTDT)4-RD,are facilely synthesized by an atom-and step-economical,and labor-saving direct C—H arylation(DACH)reaction(i.e.,C—H/C—Br cross coupling).Note that(BTDT)4-RD involving five CPDT,four BT and two rhodamine(RD)building blocks is the longest oligomeric donor in the fullerene-free OSC devices ever reported.The dependence of the structure-property-performance correlation of(BTDT)n-IC(n=1—3)and(BTDT)4-RD on theπ-conjugation lengths is thoroughly investigated by opto-electrochemical measurements,bulk heterojunction(BHJ)OSC devices and microscopies.The(BTDT)1-IC:PBDB-T and(BTDT)4-RD:Y6 BHJs achieve power conversion efficiencies of 9.14%and 4.51%,respectively.Our findings demonstrate that DACH reaction is a powerful tool to tune the opto-electronic properties and device performances by regulating the lengths ofπ-conjugated oligomers with varied numbers of repeating units.

Modulation of starch synthesis in Arabidopsis via phytochrome B-mediated light signal

Starch is a major storage carbohydrate in plants and is critical in crop yield and quality.Starch synthesis is intricately regulated by internal metabolic processes and external environmental cues;however,the precise molecular mechanisms governing this process remain largely unknown.In this study,we revealed that high red to far-red(high R:FR)light significantly induces the synthesis of leaf starch and the expression of synthesis-related genes,whereas low R:FR light suppress these processes.Arabidopsis phytochrome B(phyB),the primary R and FR photoreceptor,was identified as a critical positive regulator in this process.Downstream of phyB,basic leucine zipper transcription factor ELONGATED HYPOCOTYL5(HY5)was found to enhance starch synthesis,whereas the basic helix-loop-helix transcription factors PHYTOCHROME INTERACTING FACTORs(PIF3,PIF4,and PIF5)inhibit starch synthesis in Arabidopsis leaves.Notably,HY5 and PIFs directly compete for binding to a shared G-box cis-element in the promoter region of genes encoding starch synthases GBSS,SS3,and SS4,which leads to antagonistic regulation of their expression and,consequently,starch synthesis.Our findings highlight the vital role of phyB in enhancing starch synthesis by stabilizing HY5 and facilitating PIFs degradation under high R:FR light conditions.Conversely,under low R:FR light,PIFs predominantly inhibit starch synthesis.This study provides insight into the physiological and molecular functions of phyB and its downstream transcription factors HY5 and PIFs in starch synthesis regulation,shedding light on the regulatory mechanism by which plants synchronize dynamic light signals with metabolic cues to module starch synthesis.

Flower-like CuS/γ-Fe_(2)O_(3) van der Waals heterostructures with highefficient electromagnetic wave absorption

The escalating electromagnetic(EM)pollution issues and the demand to elevate military stealth technology make it imperative to develop cost-effective and high-performance electromagnetic wave(EMW)absorbing materials.In this paper,the flower-like CuS/γ-Fe_(2)O_(3) van der Waals(vdW)heterostructures have been synthesized via a facile two-step solvothermal approach.The flower-like CuS skeleton increases the attenuation path of EMW while reducing the material density.Different contents ofγ-Fe_(2)O_(3) nanoparticles anchor between the flower-like CuS nanosheets to constitute a heterogeneous structure,which enables dielectric and magnetic loss synergistically to optimize impedance matching and remarkably improve the EMW absorption performance.The minimum reflection loss(RLmin)is-49.36 dB with a thickness of only 1.6 mm and the effective absorption bandwidth(EAB)reaches 4.64 GHz(13.36–18 GHz).By adjusting the thickness of the absorber,the EAB can cover 96%of the GHz band.Notably,the superior absorption of-61.53 dB at middle frequency band can be obtained by adjusting the amount of Fe_(2)O_(3) addition.In this study,the adjustment of EM parameters and the optimization of impedance matching have been achieved by constructing a novel vdW heterogeneous structure,which provides fresh ideas and references for the design of high-performance EMW absorbing materials.

Unveiling the performance of a novel alkalizing bacterium Enterobacter sp.LYX-2 in immobilization of available Cd

A novel alkalizing strain Enterobacter sp.LYX-2 that could resist 400 mg/L Cd was isolated from Cd-contaminated soil,which immobilized 96.05%Cd^(2+)from medium.Cd distribution analysis demonstrated that more than half of the Cd^(2+)was converted into extracellular precipitated Cd through mobilization of the alkali-producing mechanism by the strain LYX-2,achieving the high immobilization efficiency of Cd^(2+).Biosorption experiments revealed that strain LYX-2 had superior biosorption capacity of 48.28 mg/g for Cd.Pot experiments with Brassica rapa L.were performed with and without strain LYX-2.Compared to control,15.92%bioavailable Cd was converted to non-bioavailable Cd and Cd content in aboveground vegetables was decreased by 37.10%with addition of strain LYX-2.Available Cd was mainly immobilized through extracellular precipitation,cell-surface biosorption and intracellular accumulation of strain LYX-2,which was investigated through Cd distribution,Scanning Electron Microscope and Energy-Dispersive X-ray Spectroscopy(SEM-EDS),Fourier Transform Infrared Spectroscopy(FT-IR),X-ray Photoelectron Spectroscopy(XPS)and Transmission Electron Microscopy(TEM)analysis.In addition,the application of strain LYX-2 significantly promoted the growth of vegetables about 2.4-fold.Above results indicated that highly Cd-resistant alkalizing strain LYX-2,as a novel microbial passivator,had excellent ability and reuse value to achieve the remediation of Cd-contaminated soil coupled with safe production of vegetables simultaneously.

Comparative analysis of the microbiome of sympatric wintering Bean Geese,Domestic Ducks,humans,and soil at Shengjin Lake of China reveals potential public risk to human health

The gut microbiota of migratory waterbirds is affected by various complex factors,including cross-species transmission,which increases the risk of pathogen spreading among sympatric animals and poses a potential public health risk to humans.In this study,we investigated the microbial communities of wintering Bean Geese(Anser fabalis),Domestic Ducks(A.platyrhynchos domesticus),humans,and soil using high-throughput sequencing of the 16S rRna gene region in Shengjin Lake,China.In total,6,046,677 clean reads were obtained,representing 41,119 operational taxonomic units(OTUs)across the four groups.The dominant microbial phyla were the Proteobacteria,Firmicutes,Bacteroidota,and Actinobacteriota.The Sorensen similarity index and alpha and beta diversity results showed that the gut microbial communities of Bean Geese and Domestic Ducks were more similar to those of the other pairs.Network analysis revealed that Faecalibacterium prausnitzii,Pseudomonas fragi,and Bradyrhizobium elkanii were hubs of the three major modules.Fourteen common microbiomes were iden-tified in Bean Geese,Domestic Ducks,humans,and soil in Shengjin Lake.A total of 96 potential pathogens were identified among the four groups,with 20 specific potentially pathogenic microbiomes found in the gut of Bean Geese.Some of these pathogens are responsible for significant financial losses in the poultry industry and pose risks to human health.Klebsiella pneumoniae,Morganella morganii,Escherichia coli,and Ralstonia insidiosa are potential core pathogens found in the four groups at Shengjin Lake that can cause diseases in humans and an-imals and facilitate cross-species transmission through various media.Therefore,humans are at risk of con-tracting these pathogens from migratory birds because of their frequent contact with domestic poultry.However,further studies are required to explore the potential pathogenic species and transmission pathways among sympatric wintering Bean Geese,Domestic Ducks,humans,and soil.

Crystal engineering regulation achieving inverse temperature symmetry breaking ferroelasticity in a cationic displacement type hybrid perovskite system

Ferroelastic hybrid perovskite materials have been revealed the significance in the applications of switches,sensors,actuators,etc.However,it remains a challenge to design high-temperature ferroelastic to meet the requirements for the practical applications.Herein,we reported an one-dimensional organicinorganic hybrid perovskites(OIHP)(3-methylpyrazolium)CdCl_(3)(3-MBCC),which possesses a mmmF2/m ferroelastic phase transition at 263 K.Moreover,utilizing crystal engineering,we replace-CH_(3) with-NH_(2) and-H,which increases the intermolecular force between organic cations and inorganic frameworks.The phase transition temperature of(3-aminopyrazolium)CdCl_(3)(3-ABCC),and(pyrazolium)CdCl_(3)(BCC)increased by 73 K and 10 K,respectively.Particularly,BCC undergoes an unconventional inverse temperature symmetry breaking(ISTB)ferroelastic phase transition around 273 K.Differently,it transforms from a high symmetry low-temperature paraelastic phase(point group 2/m)to a low symmetry high-temperature ferroelastic phase(point group ī)originating from the rare mechanism of displacement of organic cations phase transition.It means that crystal BCC retains in ferroelastic phase above 273 K until melting point(446 K).Furthermore,characteristic ferroelastic domain patterns on crystal BCC are confirmed with polarized optical microscopy.Our study enriches the molecular mechanism of ferroelastics in the family of organic-inorganic hybrids and opens up a new avenue for exploring high-temperature ferroic materials.

A Knowledge-enhanced Two-stage Generative Framework for Medical Dialogue Information Extraction

This paper focuses on term-status pair extraction from medical dialogues(MD-TSPE),which is essential in diagnosis dia-logue systems and the automatic scribe of electronic medical records(EMRs).In the past few years,works on MD-TSPE have attracted increasing research attention,especially after the remarkable progress made by generative methods.However,these generative methods output a whole sequence consisting of term-status pairs in one stage and ignore integrating prior knowledge,which demands a deeper un-derstanding to model the relationship between terms and infer the status of each term.This paper presents a knowledge-enhanced two-stage generative framework(KTGF)to address the above challenges.Using task-specific prompts,we employ a single model to com-plete the MD-TSPE through two phases in a unified generative form:We generate all terms the first and then generate the status of each generated term.In this way,the relationship between terms can be learned more effectively from the sequence containing only terms in the first phase,and our designed knowledge-enhanced prompt in the second phase can leverage the category and status candidates of the generated term for status generation.Furthermore,our proposed special status"not mentioned"makes more terms available and en-riches the training data in the second phase,which is critical in the low-resource setting.The experiments on the Chunyu and CMDD datasets show that the proposed method achieves superior results compared to the state-of-the-art models in the full training and low-re-sourcesettings.
