Largemouth bass(Micropterus salmoides) is an economically important fish species in North America, Europe, and China. Various genetic improvement programs and domestication processes have modified its genome sequence through selective pressure, leaving nucleotide signals that can be detected at the genomic level. In this study,we sequenced 149 largemouth bass fish, including protospecies(imported from the US) and improved breeds(four domestic breeding populations from China). We detected genomic regions harboring certain genes associated with improved traits, which may be useful molecular markers for practical domestication, breeding, and selection. Subsequent analyses of genetic diversity and population structure revealed that the improved breeds have undergone more rigorous genetic changes. Through selective signal analysis, we identified hundreds of putative selective sweep regions in each largemouth bass line. Interestingly, we predicted 103 putative candidate genes potentially subjected to selection,including several associated with growth(psst1 and grb10), early development(klf9, sp4, and sp8), and immune traits(pkn2, sept2, bcl6, and ripk2). These candidate genes represent potential genomic landmarks that could be used to improve important traits of biological and commercial interest. In summary, this study provides a genome-wide map of genetic variations and selection footprints in largemouth bass, which may benefit genetic studies and accelerate genetic improvement of this economically important fish.

This study was performed to evaluate prostate-specific antigen-age volume （PSA-AV） scores in predicting prostate cancer （PCa） in a Chinese biopsy population. A total of 2355 men who underwent initial prostate biopsy from January 2006 to November 2015 in Huashan Hospital were recruited in the current study. The PSA-AV scores were calculated and assessed together with PSA and PSA density （PSAD） retrospectively. Among 2133 patients included in the analysis, 947 （44.4%） were diagnosed with PCa. The mean age, PSA, and positive rates of digital rectal examination result and transrectal ultrasound result were statistically higher in men diagnosed with PCa （all P 〈 0.05）. The values of area under the receiver operating characteristic curves （AUCs） of PSAD and PSA-AV were 0.864 and 0.851, respectively, in predicting PCa in the entire population, both performed better than PSA （AUC = 0.805; P 〈 0.05）. The superiority of PSAD and PSA-AV was more obvious in subgroup with PSA ranging from 2.0 ng ml^-1 to 20.0 ng ml^-1. A PSA-AV score of 400 had a sensitivity and specificity of 93.7% and 40.0%, respectively. In conclusion, the PSA-AV score performed equally with PSAD and was better than PSA in predicting PCa. This indicated that PSA-AV score could be a useful tool for predicting PCa in Chinese population.

An accelerator-based Boron Neutron Capture Therapy(AB-BNCT)experimental facility called D-BNCT01 has been recently completed and is currently able to generate a high-intensity neutron beam for BNCTrelated research.In this study,we perform several experiments involving water phantoms to validate the Monte Carlo simulation results and analyze the neutron beam characteristics.According to our measurements,D-BNCT01 can generate a neutron flux about 1.2×10^(8)n/cm^(2)/s at the beam port using a 5 kW proton beam.Our results also show that the thermal neutron flux depth distribution inside the water phantom is in good agreement with simulations.We conclude that D-BNCT01 may be effectively employed for BNCT research.

Although glucorticosteroids(GCs)are the standard first-line therapy for acute graft-versus-host disease(a Gv HD),nearly 50%of a Gv HD patients have no response to GCs.The role of T cell metabolism in murine a Gv HD was recently reported.However,whether GCs and metabolism regulators could cooperatively suppress T cell alloreactivity and ameliorate a Gv HD remains to be elucidated.Increased glycolysis,characterized by elevated 6-phosphofructo-2-kinase/fructose-2,6-biphosphatase 3(PFKFB3),and higher rates of glucose consumption and lactate production were found in Tcells from a Gv HD patients.Genetic upregulation of PFKFB3 induced T cell proliferation and differentiation into proinflammatory cells.In a humanized mouse model,PFKFB3-overexpressing or PFKFB3-silenced T cells aggravated or prevented a Gv HD,respectively.Importantly,our integrated data from patient samples in vitro,in a humanized xenogeneic murine model of a Gv HD and graft-versus-leukaemia(GVL)demonstrate that GCs combined with a glycolysis inhibitor could cooperatively reduce the alloreactivity of T cells and ameliorate a Gv HD without loss of GVL effects.Together,the current study indicated that glycolysis is critical for T cell activation and induction of human a Gv HD.Therefore,the regulation of glycolysis offers a potential pathogenesis-oriented therapeutic strategy for a Gv HD patients.GCs combined with glycolysis inhibitors promises to be a novel first-line combination therapy for a Gv HD patients.

OBJECTIVE: In this study, we investigated the effects of ethanol extract of Hupiruisu Fang(HPRS), a formula of Traditional Chinese Medicine, on HCT116 cell line.METHODS: MKN45, A549, Hela and HCT116 cells were treated with ethanol extract of HPRS alone or the extract plus 5-Fluorouracil(5-FU) for 48 h, and then the cell viabilities were measured using CCK-8Kit. The early apoptosis rate and total apoptosis rate in both HCT116 cells were evaluated by flow cytometry. The m RNA levels of apoptosis-related genes including caspase-3, caspase-8, Bcl2 and Baxwere detected by real-time quantitative polymerase chain reaction. Lastly, the protein activities and expressions of those apoptosis related genes were observed for further verifying the pro-apoptosis of the extract of HPRS.RESULTS: Ethanol extract of HPRS could significantly induce apoptosis in HCT116 cell line. Synergistic analysis revealed that the extract exhibited a significant effect upon 5-FU-associated cytotoxicity in the cell line.CONCLUSION: The ethanol extract of HPRS plus5-FU might have the potential to improve the treatment of colorectal cancer.

Different sensitivities to rocuronium of the neuromuscular junctions innervated by normal/damaged facial nerves and somatic nerve in rats： the role of the presynaptic acetylcholine quantal release

Background Muscles present different responses to muscle relaxants, a mechanism of importance in surgeries requiring facial nerve evoked electromyography under general anaesthesia. The non-depolarizing muscle relaxants have multiple reaction formats in the neuromuscular junction, in which pre-synaptic quantal release of acetylcholine was one of the important mechanisms. This study was to compare the pre-synaptic quantal release of acetylcholine from the neuromuscular junctions innervated by normal/damaged facial nerves and somatic nerve under the effect of rocuronium in rats in vitro. Methods Acute right-sided facial nerve injury was induced by nerve crush axotomies. Both sided facial nerve connected orbicularis oris strips and tibial nerve connected gastrocnemius strips were isolated to measure endplate potentials （EPP） and miniature endplate potentials （MEPP） using an intracellular microelectrode gauge under different rocuronium concentrations. Then, the pre-synaptic quantal releases of acetylcholine were calculated by the ratios of the EPPs and the MEPPs, and compared among the damaged or normal facial nerve innervated orbicularis oris and tibial nerve innervated gastrocnemius. Results The EPP/MEPP ratios of the three neuromuscular junctions decreased in a dose dependent manner with the increase of the rocuronium concentration. With the concentrations of rocuronium being 5 pg/ml, 7.5 IJg/ml and 10 pg/ml, the decrease of the EPP/MEPP ratio in the damaged facial nerve group was greater than that in the normal facial nerve group. The decrease in the somatic nerve group was the biggest, with significant differences. Conclusions Rocuronium presented different levels of inhibition on the pre-synaptic quantal release of acetylcholine in the three groups of neuromuscular junctions. The levels of the inhibition showed the following sequence： somatic nerve 〉 damaged facial nerve 〉 normal facial nerve. The difference may be one of the reasons causing the different sensitivities to rocuronium among the muscles innervated

Understanding the X-ray spectrum of anomalous X-ray pulsars and soft gamma-ray repeaters

Hard X-rays above 10 keV are detected from several anomalous X-ray pul- sars （AXPs） and soft gamma-ray repeaters （SGRs）, and different models have been proposed to explain the physical origin within the frame of either a magnetar model or a fallback disk system. Using data from Suzaku and INTEGRAL, we study the soft and hard X-ray spectra of four AXPs/SGRs： 1RXS J 170849-400910, 1E 1547.0- 5408, SGR 1806-20 and SGR 0501＋4516. It is found that the spectra could be well reproduced by the bulk-motion Comptonization （BMC） process as was first suggested by Triimper et al., showing that the accretion scenario could be compatible with X- ray emission from AXPs/SGRs. Simulated results from the Hard X-ray Modulation Telescope using the BMC model show that the spectra would have discrepancies from the power-law, especially the cutoff at -200 keV. Thus future observations will allow researchers to distinguish different models of the hard X-ray emission and will help us understand the nature of AXPs/SGRs.

Ductal breast carcinoma metastasized to the rectum: A case report and review of the literature

BACKGROUND Gastrointestinal(GI)metastasis from breast cancer(BC)is rarely encountered in clinical practice.Nonspecific symptoms and long intervals make early diagnosis difficult.Therefore,increased awareness of GI metastasis secondary to BC and a deep understanding of the clinical and pathological features,and intervention for GI metastasis are fundamental to avoid delay in correct diagnosis and management.CASE SUMMARY The present report discusses the case of a Chinese female patient aged 36 years.The patient presented with difficult defecation along with bloody stools and hypogastralgia.In 2015,she had undergone right modified radical mastectomy and axillary lymph node dissection in another hospital to treat the infiltrating ductal breast carcinoma pT1N1M0.The presenting symptoms were investigated by colonoscopy,which indicated a circumferential stricture in the lower rectum at 3 cm from the anal edge.Further investigation with positron emission tomography-computed tomography revealed an uptake of fluorodeoxyglucose within the distal rectum as well as in the left acetabulum.The samples from laparoscopic exploration were biopsied,which revealed metastases of BC.Immunohistochemical analysis of the tumor confirmed that the patient had rectal metastasis of infiltrating ductal BC.CONCLUSION Rectal metastasis should be considered when patients with a history of BC present with changed bowel habits.
