·AIM: To explore the potential mechanism of molecular hydrogen in the regulation of miRNA expression and signal-modulating activities. ·METHODS: Retinal microglia cells were activated by Lipopolysaccharides (LPS) and then treated with hydrogen -saturated medium or normal medium without hydrogen. qRT -PCR was used to detect the expression difference in miR-9, miR-21 and miR-199 between these two groups. Moreover, the expression of LPS -induced signaling proteins, including Myd88, IKK -β, NF -kB, and PDCD4, were detected by Western blotting. ·RESULTS: The results demonstrated a marked downregulation of miR -9 and miR -21 and up -regulation of miR-199 by hydrogen treatment; the expression of Myd88 and IKK-β was decreased after hydrogen treatment, whereas PDCD4 was increased, and there was no significant change in NF-kB expression. · CONCLUSION: The results in the present study indicate that miR -9, miR -199 and miR -21 play an important role in the anti -inflammatory regulation of LPS -activated microglia cells by molecular hydrogen, which will help to explain the protective mechanism of molecular hydrogen against inflammatory injury.·

AIM：To develop a new method to produce recombinant reprogramming proteins,c Myc,Klf4,Oct4,and Sox2,in soluble format with low cost for the generation of induced pluripotent stem cells（i PSCs）.METHODS：A short polypeptide sequence derived from the HIV trans-activator of transcription protein（TAT） and the nucleus localization signal（NLS） polypeptide were fused to the N terminus of the reprogramming proteins and they were constructed into p Cold-SUMO vector which can extremely improve the solubility of recombinant proteins.Then these vector plasmids were transformed into E.coli BL21（DE3） Chaperone competent cells for amplification.The solubility of these recombinant proteins was determined by SDS-PAGE and Coomassie brilliant blue staining.The recombinant proteins were purified by NiNTA resin and identified by Western blot.The transduction of these proteins into HEK 293 T cells were evaluated by immunofluorescence staining.RESULTS：These four reprogramming proteins could be produced in soluble format in p Cold-SUMO expression vector system with the assistance of chaperone proteins in bacteria.The proteins were purified successfully with a purity of over 70% with a relative high transduction rate into 293 cells.CONCLUSION：The results in the present study indicate the four important reprogramming proteins,c Myc,Klf4,Oct4,and Sox2,can be produced in soluble format in bacteria with low cost.Our new method thus might be expected to greatly contribute to the future study of i PSCs.

Optimization problems are prevalent in various fields,and the gradient-based gradient descent algorithm is a widely adopted optimization method.However,in classical computing,computing the numerical gradient for a function with variables necessitates at least d+1 function evaluations,resulting in a computational complexity of O(d).As the number of variables increases,the classical gradient estimation methods require substantial resources,ultimately surpassing the capabilities of classical computers.Fortunately,leveraging the principles of superposition and entanglement in quantum mechanics,quantum computers can achieve genuine parallel computing,leading to exponential acceleration over classical algorithms in some cases.In this paper,we propose a novel quantum-based gradient calculation method that requires only a single oracle calculation to obtain the numerical gradient result for a multivariate function.The complexity of this algorithm is just O(1).Building upon this approach,we successfully implemented the quantum gradient descent algorithm and applied it to the variational quantum eigensolver(VQE),creating a pure quantum variational optimization algorithm.Compared with classical gradient-based optimization algorithm,this quantum optimization algorithm has remarkable complexity advantages,providing an efficient solution to optimization problems.The proposed quantum-based method shows promise in enhancing the performance of optimization algorithms,highlighting the potential of quantum computing in this field.

Here,we characterize the temporal and spatial dynamics of forest community structure and species diversity in a subtropical evergreen broad-leaved forest in China.We found that community structure in this forest changed over a 15-year period.Specifically,renewal and death of common species was large,with the renewal of individuals mainly concentrated within a few populations,especially those of Aidia canthioides and Cryptocarya concinna.The numbers of individual deaths for common species were concentrated in the small and mid-diameter level.The spatial distribution of community species diversity fluctuated in each monitoring period,showing a more dispersed diversity after the 15-year study period,and the coefficient of variation on quadrats increased.In 2010,the death and renewal of the community and the spatial variation of species diversity were different compared to other survey years.Extreme weather may have affected species regeneration and community stability in our subtropical monsoon evergreen broad-leaved forests.Our findings suggest that strengthening the monitoring and management of the forest community will help better understand the long-and short-term causes of dynamic fluctuations of community structure and species diversity,and reveal the factors that drive changes in community structure.

Soil salinization is an essential environmental stressor,threatening agricultural yield and ecological security worldwide.Saline soils accumulate excessive soluble salts which are detrimental to most plants by limiting plant growth and productivity.It is of great necessity for plants to efficiently deal with the adverse effects caused by salt stress for survival and successful reproduction.Multiple determinants of salt tolerance have been identified in plants,and the cellular and physiological mechanisms of plant salt response and adaption have been intensely characterized.Plants respond to salt stress signals and rapidly initiate signaling pathways to re-establish cellular homeostasis with adjusted growth and cellular metabolism.This review summarizes the advances in salt stress perception,signaling,and response in plants.A better under-standing of plant salt resistance will contribute to improving crop performance under saline conditions using multiple engineering approaches.The rhizosphere microbiome-mediated plant salt tolerance as well as chemical priming for enhanced plant salt resistance are also discussed in this review.

Two-Staged Method for Ice Channel Identification Based on Image Segmentation and Corner Point Regression

Identification of the ice channel is the basic technology for developing intelligent ships in ice-covered waters,which is important to ensure the safety and economy of navigation.In the Arctic,merchant ships with low ice class often navigate in channels opened up by icebreakers.Navigation in the ice channel often depends on good maneuverability skills and abundant experience from the captain to a large extent.The ship may get stuck if steered into ice fields off the channel.Under this circumstance,it is very important to study how to identify the boundary lines of ice channels with a reliable method.In this paper,a two-staged ice channel identification method is developed based on image segmentation and corner point regression.The first stage employs the image segmentation method to extract channel regions.In the second stage,an intelligent corner regression network is proposed to extract the channel boundary lines from the channel region.A non-intelligent angle-based filtering and clustering method is proposed and compared with corner point regression network.The training and evaluation of the segmentation method and corner regression network are carried out on the synthetic and real ice channel dataset.The evaluation results show that the accuracy of the method using the corner point regression network in the second stage is achieved as high as 73.33%on the synthetic ice channel dataset and 70.66%on the real ice channel dataset,and the processing speed can reach up to 14.58frames per second.

Circular RNAs(circRNAs)are a class of covalently closed single-stranded RNAs that are expressed during the development of specific cells and tissues.CircRNAs play crucial roles in physiological and pathological processes by sponging microRNAs,modulating gene transcription,controlling the activity of certain RNA-binding proteins,and producing functional peptides.A key focus of research at present is the functionality of circRNAs in the nervous system and several advances have emerged over the last 2 years.However,the precise role of circRNAs in the nervous system has yet to be comprehensively reviewed.In this review,we first summarize the recently described roles of circRNAs in brain development,maturity,and aging.Then,we focus on the involvement of circRNAs in various diseases of the central nervous system,such as brain cancer,chronic neurodegenerative diseases,acute injuries of the nervous system,and neuropathic pain.A better understanding of the functionality of circRNAs will help us to develop potential diagnostic,prognostic,and therapeutic strategies to treat diseases of the nervous system.

Chitin hydrogel has been recognized as a promising material for various biomedical applications because of its biocompatibility and biodegradability.However,the fabrication of strong chitin hydrogel remains a big challenge because of the insolubility of chitin in many solvents and the reduced chain length of chitin regenerated from solutions.We herein introduce the fabrication of chitin hydrogel with biomimetic structure through the chemical transformation of chitosan,which is a water-soluble deacetylated derivative of chitin.The reacetylation of the amino group in chitosan endows the obtained chitin hydrogel with outstanding resistance to swelling,degradation,extreme temperature and pH conditions,and organic solvents.The chitin hydrogel has excellent mechanical properties while retaining a high water content(more than 95 wt.%).It also shows excellent antifouling performance that it resists the adhesion of proteins,bacteria,blood,and cells.Moreover,as the initial chitosan solution can be feasibly frozen and templated by ice crystals,the chitin hydrogel structure can be either nacre-like or wood-like depending on the freezing method of the precursory chitosan solution.Owing to these anisotropic structures,such chitin hydrogel can exhibit anisotropic mechanics and mass transfer capabilities.The current work provides a rational strategy to fabricate chitin hydrogels and paves the way for its practical applications as a superior biomedical material.

Audio mixing is a crucial part of music production.For analyzing or recreating audio mixing,it is of great importance to conduct research on estimating mixing parameters used to create mixdowns from music recordings,i.e.,audio mixing inversion.However,approaches of audio mixing inversion are rarely explored.A method of estimating mixing parameters from raw tracks and a stereo mixdown via embodied self-supervised learning is presented.In this work,several commonly used audio effects including gain,pan,equalization,reverb,and compression,are taken into consideration.This method is able to learn an inference neural network that takes a stereo mixdown and the raw audio sources as input and estimate mixing parameters used to create the mixdown by iteratively sampling and training.During the sampling step,the inference network predicts a set of mixing parameters,which is sampled and fed to an audio-processing framework to generate audio data for the training step.During the training step,the same network used in the sampling step is optimized with the sampled data generated from the sampling step.This method is able to explicitly model the mixing process in an interpretable way instead of using a black-box neural network model.A set of objective measures are used for evaluation.The experimental results show that this method has better performance than current state-of-the-art methods.
