The rice PLATZ protein SHORT GRAIN6 determines grain size by regulating spikelet hull cell division

Grain size is a major determinant of cereal grain yields;however,the relevant regulatory mechanisms controlling this trait have not been fully elucidated.The rice(Oryza sativa)mutant short grain6(sg6)was identified based on its reduced grain length and weight.Here,we functionally characterized the role of SG6 in determining grain size through the regulation of spikelet hull cell division.SG6 encodes a previously uncharacterized plant ATrich sequence and zinc-binding(PLATZ)protein that is ubiquitously localized throughout the cell and is preferentially expressed in the early developing panicles but not in the endosperm.The overexpression of SG6 resulted in significantly larger and heavier grains,as well as increased plant heights,which is consistent with its elevated spikelet hull cell division rate.Yeast two-hybrid analyses revealed that SG6 interacts with the core cell cycle machinery DP protein and several other putative cell division regulators,consistent with our transcriptomic analysis,which showed that SG6 activates the expression of many DNA replication and cell-cycle-related genes.These results confirm the crucial role of SG6 in determining grain size by regulating spikelet hull cell division and provide clues for understanding the functions of PLATZ family proteins and the network regulating cereal grain size.

CRISPR Primer Designer:Design primers for knockout and chromosome imaging CRISPR-Cas system

The clustered regularly interspaced short palin- dromic repeats （CRiSPR）-associated system enables biologists to edit genomes precisely and provides a powerful tool for perturbing endogenous gene regulation, modulation of epigenetic markers, and genome architecture. However, there are concerns about the specificity of the system, especially the usages of knocking out a gene. Previous designing tools either were mostly built-in websites or ran as command-line programs, and none of them ran locally and acquired a user- friendly interface. In addition, with the development of CRISPR-derived systems, such as chromosome imaging, there were still no tools helping users to generate specific end-user spacers. We herein present researchers to design primers CRISPR Primer Designer for for CRISPR applications. Theprogram has a user-frfendly interface, can analyze the BLAST results by using multiple parameters, score for each candidate spacer, and generate the primers when using a certain plasmid. In addition, CRISPR Primer Designer runs locally and can be used to search spacer clusters, and exports primers for the CRiSPR-Cas system-based chromosome imaging system.

Deficiency of mitochondrial outer membrane protein 64 confers rice resistance to both piercing-sucking and chewing insects in rice

The brown planthopper(BPH)and striped stem borer(SSB)are the most devastating insect pests in rice(Oryza sativa)producing areas.Screening for endogenous resistant genes is the most practical strategy for rice insect-resistance breeding.Forty-five mutants showing high re-sistance against BPH were identified in a rice T-DNA in-sertion population(11,000 putative homozygous lines)after 4 years of large-scale field BPH-resistance phenotype screening.Detailed analysis showed that deficiency of rice mitochondrial outer membrane protein 64(OM64)gene resulted in increased resistance to BPH.Mitochondrial outer membrane protein 64 protein is located in the outer mitochondrial membrane by subcellular localization and its deficiency constitutively activated hydrogen peroxide(H2O2)signaling,which stimulated antibiosis and tolerance to BPH.The om64 mutant also showed enhanced resist-ance to SSB,a chewing insect,which was due to promotion of Jasmonic acid biosynthesis and related responses.Im-portantly,om64 plants presented no significant changes in rice yield-related characters.This study confirmed OM64 as a negative regulator of rice herbivore resistance through regulating H2O2 production.Mitochondrial outer membrane protein 64 is a potentially efficient candidate to improve BPH and SSB resistance through gene deletion.Why the om64 mutant was resistant to both piercing-sucking and chewing insects via a gene deficiency in mitochondria is discussed.

Cis-12-Oxo-Phytodienoic Acid Stimulates Rice Defense Response to a Piercing-Sucking Insect

The brown planthopper （BPH, Nilaparvata lugens） is a destructive, monophagous, piercing-sucking insect pest of rice. Previous studies indicated that jasmonic acid （JA） positively regulates rice defense against chewing insect pests but negatively regulates it against the piercing-sucking insect of BPH. We here demonstrated that overexpression of allene oxide cyclase （AOC） but not OPR3 （cis-12-oxo-phytodienoic acid （OPDA） reductase 3, an enzyme adjacent to AOC in the JA synthetic pathway） significantly increased rice resistance to BPH, mainly by reducing the feeding activity and survival rate. Further anal- ysis revealed that plant response to BPH under AOC overexpression was independent of the JA pathway and that significantly higher OPDA levels stimulated rice resistance to BPH. Microarray analysis identified multiple candidate resistance-related genes underAOCoverexpression. OPDA treatment stimulated the resistance of radish seedlings to green peach aphid Myzus persicae, another piercing-sucking insect. These results imply that rice resistance to chewing insects and to sucking insects can be enhanced simultaneously through AOC-mediated increases of JA and OPDA and provide direct evidence of the potential application of OPDA in stimulating plant defense responses to piercing-sucking insect pests in agriculture.

This paper concerns the linearization problem on rational maps of degree d≥2 and polynomials of degree d>2 from the perspective of non-linearizability.The authors introduce a set l_(∞) of irrational numbers and show that if α∈l_(∞),then any rational map is not linearizable and has infinitely many cycles in every neighborhood of the fixed point with multiplier λ=e^(2πiα),Adding more constraints to cubic polynomials,they discuss the above problems by polynomial-like maps.For the family of polynomials,with the help of Yoccoz's method,they obtain its maximum dimension of the set in which the polynomials are non-linearizable.

Physiological and pathological functions of circular RNAs in the nervous system

Circular RNAs(circRNAs)are a class of covalently closed single-stranded RNAs that are expressed during the development of specific cells and tissues.CircRNAs play crucial roles in physiological and pathological processes by sponging microRNAs,modulating gene transcription,controlling the activity of certain RNA-binding proteins,and producing functional peptides.A key focus of research at present is the functionality of circRNAs in the nervous system and several advances have emerged over the last 2 years.However,the precise role of circRNAs in the nervous system has yet to be comprehensively reviewed.In this review,we first summarize the recently described roles of circRNAs in brain development,maturity,and aging.Then,we focus on the involvement of circRNAs in various diseases of the central nervous system,such as brain cancer,chronic neurodegenerative diseases,acute injuries of the nervous system,and neuropathic pain.A better understanding of the functionality of circRNAs will help us to develop potential diagnostic,prognostic,and therapeutic strategies to treat diseases of the nervous system.

Vibration Control of A Flexible Marine Riser System Subject to Input Dead Zone and Extraneous Disturbances

An observer-based adaptive backstepping boundary control is proposed for vibration control of flexible offshore riser systems with unknown nonlinear input dead zone and uncertain environmental disturbances.The control algorithm can update the control law online through real-time data to make the controller adapt to the environment and improve the control precision.Specifically,based on the adaptive backstepping framework,virtual control laws and Lyapunov functions are designed for each subsystem.Three direction interference observers are designed to track the timevarying boundary disturbance.On this basis,the inverse of the dead zone and linear state transformation are used to compensate for the original system and eliminate the adverse effects of the dead zone.In addition,the stability of the closed-loop system is proven by Lyapunov stability theory.All the system states are bounded,and the vibration offset of the riser converges to a small area of the initial position.Finally,four examples of flexible marine risers are simulated in MATLAB to verify the effectiveness of the proposed controller.

Colorectal cancer(CRC)is one of the most common cancers around the world,and it is one of the leading causes of cancer-related death.1 Although anti-EGFR therapy and immune checkpoint inhibitor(ICl)therapy are becoming more and more important for colorectal therapy,however,clinical outcomes of current treatments for metastatic CRC,especially with ICls,have been shown to be affected by the status of KRAS.Therefore,KRAS mutation status detection has become a very important diagnostic factor for managing metastatic CRC patients.

Iridoids modulate inflammation in diabetic kidney disease: A review

In recent years,preclinical research on diabetic kidney disease (DKD) has surged to the forefront of scientific and clinical attention.DKD has become a pervasive complication of type 2 diabetes.Given the complexity of its etiology and pathological mechanisms,current interventions,including drugs,dietary modifications,exercise,hypoglycemic treatments and lipid-lowering methods,often fall short in achieving desired therapeutic outcomes.Iridoids,primarily derived from the potent components of traditional herbs,have been the subject of long-standing research.Preclinical data suggest that iridoids possess notable renal protective properties;however,there has been no summary of the research on their efficacy in the management and treatment of DKD.This article consolidates findings from in vivo and in vitro research on iridoids in the context of DKD and highlights their shared anti-inflammatory activities in treating this condition.Additionally,it explores how certain iridoid components modify their chemical structures through the regulation of intestinal flora,potentially bolstering their therapeutic effects.This review provides a focused examination of the mechanisms through which iridoids may prevent or treat DKD,offering valuable insights for future research endeavors.

Rice grain shape,which is closely related to seed development and is determined by multiple factors,is an important trait affects both production and its economic value. In order to investigate the molecular mechanism controlling grain shape,a mutant population with japonica rice Zhonghua11 background was generated by EMS(ethyl methane sulfonate) mutagenesis. Various mutants with alternation of grain shape were identified and slender grain 1(slgr1),a mutant with significant increase of grain length was selected for further analysis. The grain length of slgr1 is 8% longer than that of wild-type,but significant alternation was found in neither width nor thickness of grain. Genetic analysis revealed that the increased grain length was controlled by a single recessive gene,which was further mapped by Mut Map method. Five candidate genes with amino acids change of SLGR1 were identified and will be further analyzed. Detailed results will be presented.
