Theories and experiments on dirty superconductors are complex but important in terms of both theoretical fundamentals and practical *** activities are even more challenging when magnetic fields are present because the field distribution,electron density of states,and superconducting pairing potentials become ***,we present tunneling microspectroscopic experiments on NbC single crystals and demonstrate that NbC is a homogeneous dirty *** applying magnetic fields to the samples,we found that the zero-energy local density of states and the pairing energy gap followed the explicit scaling relation proposed by de Gennes for homogeneous dirty superconductors in high magnetic *** significantly,our experimental findings indicate that the validity of the scaling relation extends to magnetic field strengths far below the upper critical field,calling for a new nonperturbative understanding of this fundamental property in dirty *** the practical side,we used the observed scaling relation to derive a simple and straightforward experimental scheme for estimating the superconducting coherence length of a dirty superconductor in magnetic fields.

Spondylocarpotarsal syndrome(SCT) is a rare musculoskeletal disorder characterized by short stature and vertebral, carpal, and tarsal fusions resulting from biallelic nonsense mutations in the gene encoding filamin B(FLNB). Utilizing a FLNB knockout mouse,we showed that the vertebral fusions in SCT evolved from intervertebral disc(IVD) degeneration and ossification of the annulus fibrosus(AF), eventually leading to full trabecular bone formation. This resulted from alterations in the TGFβ/BMP signaling pathway that included increased canonical TGFβ and noncanonical BMP signaling. In this study, the role of FLNB in the TGFβ/BMP pathway was elucidated using in vitro, in vivo, and ex vivo treatment methodologies. The data demonstrated that FLNB interacts with inhibitory Smads 6 and 7(i-Smads) to regulate TGFβ/BMP signaling and that loss of FLNB produces increased TGFβ receptor activity and decreased Smad 1 ubiquitination. Through the use of small molecule inhibitors in an ex vivo spine model, TGFβ/BMP signaling was modulated to design a targeted treatment for SCT and disc degeneration. Inhibition of canonical and noncanonical TGFβ/BMP pathway activity restored Flnb-/-IVD morphology. These most effective improvements resulted from specific inhibition of TGFβand p38 signaling activation. FLNB acts as a bridge for TGFβ/BMP signaling crosstalk through i-Smads and is key for the critical balance in TGFβ/BMP signaling that maintains the IVD. These findings further our understanding of IVD biology and reveal new molecular targets for disc degeneration as well as congenital vertebral fusion disorders.

Background Ciliopathies are a group of diseases associated with abnormal structure or function of primary cilia. Ciliopathies include polycystic kidney disease （PKD）, a pathology associated with vascular hypertension. We previously showed that cilia length regulates cilia function, and cilia function is required for nitric oxide （NO） biosynthesis in endothelial cells. Because patients with PKD show abnormal sensory cilia function, the aim of our current study was to search for a targeted therapy focused on primary cilia, which we refer to as ＇cilio- therapy＇. Methods and Results In the present studies, our in vitro analyses refined fenoldopam as an equipotent and more specific dopa- minergic agonist to regulate cilia length and function. Our in vivo studies indicated that fenoldopam increased cilia length and serum NO thereby reducing blood pressure in a PKD mouse model. Our crossover, multicenter, double-blind and placebo-controlled clinical study further indicated that cilia-targeting therapy showed an overall reduction in mean arterial pressure in PKD patients. Conclusions Overall, our studies provide the first evidence of ciliotherapy as an innovative intervention in patients with abnormal primary cilia.

The stability against various environmental stresses of the curcumin-loaded secondary and tertiary emulsions that was emulsified by whey protein isolate(WPI)and coated by chitosan(ChI),carboxymethyl konjac glucomannan(CMKGM),or their combination through layer-by-layer assembly was ***,the multilayered emulsions were destabilized in high Na Cl concentrations or medium p h that could interrupt the electrostatic interaction between the three polyelectrolytes or deprotonate ChI,indicating that electrostatic interaction played an important role in the stability of *** with the primary emulsion that was solely stabilized by WPI,extra coating with ChI and CMKGM generally increased the stability of the emulsion against repeated freezing-thawing,improved the retention of curcumin against heating,UV irradiation,and long-term storage,and the effects were more remarkable in the tertiary emulsion with CMKGM locating in the outmost *** CMKGM has shown the colon-targeted delivery potency,the multilayered emulsions assembled by layer-by-layer deposition,especially the tertiary emulsion,could be used as an effective carrier for the targeted delivery of curcumin.

During the deformation of Mg alloys,{10-12} extension twin often contributes to the formation of basal texture but rarely assists the nucleation of recrystallization,i.e.,effective grain refinement,therefore it seems to make against the improvement of formability and mechanical *** this work,{10-12} extension twin has been creatively utilized as a preference nucleation site for static recrystallization(SRX),achieving grain refinement and orientation randomization in a Mg-Gd-Y alloy using multi-directional impact forging(MDIF)and subsequent annealing *** of {10-12} extension twin on SRX behavior has been investigated by annealing treatment at 450℃ using quasi-in-situ optical microscopy(OM)and quasi-in-situ electron back-scattering diffraction(EBSD).The microstructural evolution during annealing shows that several SRX gains can nucleate from the grain boundary of untwinned grains,but they only have few influences on the final microstructure due to their limited volume faction and sluggish *** contrast,a large number of SRX gains can initiate from {10-12} extension twin and grow up without the confine of twin ***,they consume their parent grains and make the main contribution to grain *** should be attributed to those pinned {10-12} twin boundary,by interacting with various dislocation slips during the MDIF process,which can operate like grain boundary,store enough strain energy,and promote the nucleation of SRX during *** the other hand,SRX grains usually keep initial random orientation and further randomize the forging texture during annealing treatment.
