a.M:To exa.ine the influence of ghrelin on the regenera.ive potentia. of ga.trointestina.(GI)***:Da.a.e to GI epithelium wa. induced in mice by two intra.enous injections of doxorubicin(10 a.d 6 mg/kg).Some of the doxorubicin-trea.ed mice received a.continuous subcuta.eous infusion of ghrelin(1.25μg/h)for 10 d via.impla.ted mini-osmotic *** la.el dividing stem cells in the S-pha.e of the cell cycle,a.l mice received a.single intra.eritonea. injection of 5'-bromo-2'-deoxyuridine(BrdU)one hour before *** stoma.h a.ong with the duodenum were then removed a.d processed for histologica. exa.ina.ion a.d immunohistochemistry using a.ti-BrdU ***:The results showed dra.a.ic da.a.e to the GI epithelium 3 d a.ter a.ministra.ion of chemothera.y which bega. to recover by da. *** ghrelintrea.ed mice,a.tenua.ion of GI mucosa. da.a.e wa. evident in the tissues exa.ined *** a.a.ysis showed a. increa.e in the number of BrdUla.eled cells a.d a. a.tera.ion in their distribution a.ong the epithelia. lining in response to da.a.e by *** mice trea.ed with both doxorubicin a.d ghrelin,the number of BrdUla.eled cells wa. reduced when compa.ed with mice trea.ed with doxorubicin ***:The present study suggests tha. ghrelin enha.ces the regenera.ive potentia. of the GI epithelium in doxorubicintrea.ed mice,a. lea.t in pa.t,by modula.ing cell prolifera.ion.
