The development of single-cell subclones,which can rapidly switch from dormant to dominant subclones,occur in the natural pathophysiology of multiple myeloma(MM)but is often"pressed"by the standard treatment of *** emerging subclones present a challenge,providing reservoirs for chemoresistant *** advancement is required to track MM subclonal changes,as understanding MM's mechanism of evolution at the cellular level can prompt the development of new targeted ways of treating this *** methods to study the evolution of subclones in MM rely on technologies capable of phenotypically and genotypically characterizing plasma cells,which include immunohistochemistry,flow cytometry,or ***,all of these technologies may be limited by the sensitivity for picking up rare *** contrast,more incisive methods such as RNA sequencing,comparative genomic hybridization,or whole-genome sequencing are not yet commonly used in clinical *** we introduce the epidemiological diagnosis and prognosis of MM and review current methods for evaluating MM subclone evolution,such as minimal residual disease/multiparametric flow cytometry/next-generation sequencing,and their respective advantages and *** addition,we propose our new single-cell method of evaluation to understand MM's mechanism of evolution at the molecular and cellular level and to prompt the development of new targeted ways of treating this disease,which has a broad prospect.

Spatiotemporal switching signals for cancer stem cell activation in pediatric origins of adulthood cancer:Towards a watch-and-wait lifetime strategy for cancer treatment

Pediatric origin of cancer stem cell hypothesis holds great promise and potential in adult cancer treatment, however; the road to innovation is full of obstacles as there are plenty of questions left unanswered. First, the key question is to characterize the nature of such stem cells (concept). Second, the quantitative imaging of pediatric stem cells should be implemented(technology). Conceptually, pediatric stem cell origins of adult cancer are based on the notion that plasticity in early life developmental programming evolves local environments to cancer. Technologically, such imaging in children is lacking as all imaging is designed for adult patients. We postulate that the need for quantitative imaging to measure space-time changes of plasticity in early life developmental programming in children may trigger research and development of the imaging technology. Such quantitative imaging of pediatric origin of adulthood cancer will help develop a spatiotemporal monitoring system to determine cancer initiation and progression. Clinical validation of such speculative hypothesis-that cancer originates in a pediatric environment-will help implement a wait-andwatch strategy for cancer treatment.

Dynamic response of mountain tunnel,bridge,and embankment along the Sichuan-Tibet transportation corridor to active fault based on model tests

The Sichuan-Tibet transportation corridor is prone to numerous active faults and frequent strong *** extensive studies have individually explored the effect of active faults and strong earthquakes on different engineering structures,their combined effect remains *** research employed multiple physical model tests to investigate the dynamic response of various engineering structures,including tunnels,bridges,and embankments,under the simultaneous influence of cumulative earthquakes and stick-slip misalignment of an active *** prototype selected for this study was the Kanding No.2 tunnel,which crosses the Yunongxi fault zone within the Sichuan-Tibet transportation *** results demonstrated that the tunnel,bridge,and embankment exhibited amplification in response to the input seismic wave,with the amplification effect gradually decreasing as the input peak ground acceleration(PGA)*** PGAs of different engineering structures were weakened by the fault rupture ***,the misalignment of the active fault may decrease the overall stiffness of the engineering structure,leading to more severe damage,with a small contribution from seismic ***,the seismic vibration effect might be enlarged with the height of the engineering structure,and the tunnel is supposed to have a smaller PGA and lower dynamic earth pressure compared to bridges and embankments in strong earthquake zones crossing active *** findings contribute valuable insights for evaluating the dynamic response of various engineering structures crossing an active fault and provide an experimental reference for secure engineering design in the challenging conditions of the Sichuan-Tibet transportation corridor.

BACKGROUND Radiation induces rapid bone loss and enhances bone resorption and adipogenesis, leading to an increased risk of bone fracture. However, the mechanisms of radiation-induced osteoporosis are not fully *** To investigate the role of CR6-interacting factor-1(Crif1) in osteoclastogenesis after radiation and its possible *** C57 BL/6 mice were exposed to Co-60 gamma rays and received 5 Gy of wholebody sublethal irradiation at a rate of 0.69 Gy/min. For in vitro study, mouse bone marrow mesenchymal stem/stromal cells(BM-MSCs) were irradiated with Co-60 at a single dose of 9 Gy. For osteoclast induction, monocyte-macrophage RAW264.7 cells were cocultured with mouse BM-MSCs for 7 d. Clus Pro and Inter Pro Surf were used to investigate the interaction interface in Crif1 and protein kinase cyclic adenosine monophosphate(c AMP)-activited catalytic subunit alpha complex. Virtual screening using 462608 compounds from the life Chemicals database around His120 of Crif1 was carried out using the program Autodock_vina. A tetrazolium salt(WST-8) assay was carried out to study the toxicity of compounds to different cells, including human BM-MSCs, mouse BMMSCs, and Vero *** Crif1 expression increased in bone marrow cells after radiation in *** of Crif1 in mouse BM-MSCs and radiation exposure could increase RANKL secretion and promote osteoclastogenesis in vitro. Deletion of Crif1 in BM-MSCs could reduce both adipogenesis and RANKL expression,resulting in the inhibition of osteoclastogenesis. The influence of different types of roots on the soil is complex and still remains *** in-situ extrusion tests were conducted on two types of root systems,namely fibrous and tap root system,for three plants,Eleusine indica,Potentilla anserine,and Artemisia argyi,according to the classification in Botany,and the thrust-displacement curves and failure patterns of different samples were analysed by comparison to fill the aforementioned *** reveal that the roots can reduce the characteristics of soil brittleness and enhance its capability to resist large deformation,and different root types contribute different effects to the strain-hardening behavior of the root-soil *** contribution of the fibrous root system to strength is limited,whilst the tap root system substantially enhances strength and *** of failure patterns show that fibrous and tap root systems affect soil solidification and surface cracking ***,the effect of the tap root system depends on the composition of lateral and tap roots:long and rich lateral roots are effective for resisting the creation of cracks,but thick tap roots with few and thin lateral roots may lead to several surface cracks.

Analysis of ancient Rongcharong landslide dam failure events in the Suwalong reach of the upper reaches of the Jinsha River

Active tectonic movements and geological disasters frequently occur in the upper reaches of the Jinsha River， increasing the likelihood of landslides obstructing the river. Taking the ancient Rongcharong landslide dam failure events in the Suwalong reach as an example， this paper first analyzes the accuracy and applicability of the commonly used methods in calculating the peak flow of the dam failure， such as the empirical formula， the numerical method based on the physical mechanism， and the computational fluid dynamics (CFD) method. Then， the peak flood flow of the Rongcharong-dammed lake when it overflows the dam is determined to be 28393～64272m3/s. At the same time， the failure process of landslide dam due to flood erosion was elucidated using the CFD method， which can be divided into three stages: gradual erosion in the initial stage， rapid development in the middle stage， and gradual expansion in the final stage. Finally， the factors that affect the peak flow of floods are analyzed， and suggestions for emergency treatment of landslide dams are put forward. The findings of this research can serve as a valuable reference for disaster prevention and mitigation strategies to adapt to the increasing frequency of landslide-induced river blockages.

目的检测CRIF1在骨肉瘤组织、癌旁组织及骨肉瘤细胞系中的表达情况,探讨CRIF1在骨肉瘤细胞放射耐受中的作用及其分子机制,探索抑制CRIF1在裸鼠移植瘤模型放疗增敏中的作用,以及以CRIF1为靶点在骨肉瘤的放疗增敏中应用前景。材料和方法收集新鲜骨肉瘤组织标本和石蜡包埋骨肉瘤组织标本,免疫组化、免疫荧光、RT-PCR以及Western blotting检测CRIF1的表达水平并进行生存率分析。检测骨肉瘤细胞系中CRIF1的表达水平。建立CRIF1稳定低表达的U2OS细胞系,检测CRIF1低表达对对细胞周期、细胞增殖、DNA复制、DNA修复、线粒体结构及膜电位的影响。通过免疫荧光、免疫共沉淀、Western blotting等方法检查CRIF1与CDK2之间的相互作用,以及该作用对放射后CDK2的表达水平、磷酸化状态、亚细胞分布和关键上下游调控分子表达水平的影响。建立CRIF1低表达的裸鼠移植瘤模型,探索其放疗敏感性。结果骨肉瘤肿瘤组织高表达CRIF1,其表达水平与患者生存率呈负相关。CRIF1在骨肉瘤细胞系中高表达,抑制CRIF1的表达可导致G1/S检查点失活、DNA修复延滞以及线粒体结构功能受损,从而降低骨肉瘤细胞的放射抗性。CRIF1与CDK2存在相互结合作用,放射后二者在细胞核中同步聚集。抑制CRIF1可诱导CDK2放射后入核转运受阻,进而影响CDK2的Thr14和Thr160位点磷酸化水平。抑制CRIF1可导致放射后未完全修复DSBs的细胞通过G1期检查点进入S期,EdU和γH2AX双阳性细胞比率显著增加;还可延滞NBS1 Ser432位点的磷酸化增强,γH2AX的阳性数明显高于对照细胞;最终通过影响G1/S期检查点激活和DNA修复引起细胞凋亡。动物实验证实,抑制CRIF1并联合放疗可显著增加TUNEL阳性细胞数量,导致肿瘤消退。结论体内外研究结果均显示,CRIF1与骨肉瘤的放射抗性密切相关。CRIF1介导的放射后CDK2入核转运和Thr14/Thr160磷酸化可通过促进G1/S期检查点激活和DNA修复参与骨肉瘤的放疗抵抗。该研究有望为寻找放射增敏靶分子,克服骨肉瘤的放疗耐受性提供了新思路。

Twisting immune responses for allogeneic stem cell therapy
