Understanding endometriosis from an immunomicroenvironmental perspective

Endometriosis,a heterogeneous,inflammatory,and estrogen-dependent gynecological disease defined by the presence and growth of endometrial tissues outside the lining of the uterus,affects approximately 5-10%of reproductive-age women,causing chronic pelvic pain and reduced fertility.Although the etiology of endometriosis is still elusive,emerging evidence supports the idea that immune dysregulation can promote the survival and growth of retrograde endometrial debris.Peritoneal macrophages and natural killer(NK)cells exhibit deficient cytotoxicity in the endometriotic microenvironment,leading to inefficient eradication of refluxed endometrial fragments.In addition,the imbalance of T-cell subtypes results in aberrant cytokine production and chronic inflammation,which contribute to endometriosis development.Although it remains uncertain whether immune dysregulation represents an initial cause or merely a secondary enhancer of endometriosis,therapies targeting altered immune pathways exhibit satisfactory effects in preventing disease onset and progression.Here,we summarize the phenotypic and functional alterations of immune cells in the endometriotic microenvironment,focusing on their interactions with microbiota and endocrine and nervous systems,and how these interactions contribute to the etiology and symptomology of endometriosis.

The cubic or third-power(TP)nonlinear energy sink(NES)has been proven to be an effective method for vibration suppression,owing to the occurrence of targeted energy transfer(TET).However,TET is unable to be triggered by the low initial energy input,and thus the TP NES would get failed under low-amplitude vibration.To resolve this issue,a new type of NES with fractional nonlinearity,e.g.,one-third-power(OTP)nonlinearity,is proposed.The dynamic behaviors of a linear oscillator(LO)with an OTP NES are investigated numerically,and then both the TET feature and the vibration attenuation performance are evaluated.Moreover,an analogy circuit is established,and the circuit simulations are carried out to verify the design concept of the OTP NES.It is found that the threshold for TET of the OTP NES is two orders of magnitude smaller than that of the TP NES.The parametric analysis shows that a heavier mass or a lower stiffness coefficient of the NES is beneficial to the occurrence of TET in the OTP NES system.Additionally,significant energy transfer is usually accompanied with efficient energy dissipation.Consequently,the OTP NES can realize TET under low initial input energy,which should be a promising approach for micro-vibration suppression.

Chemical proteomics:terra incognita for novel drug target profiling

The growing demand for new therapeutic strategies in the medical and pharmaceutic fields has resulted in a pressing need for novel druggable targets.Paradoxically,however,the targets of certain drugs that are already widely used in clinical practice have largely not been annotated.Because the pharmacologic effects of a drug can only be appreciated when its interactions with cellular components are clearly delineated,an integrated deconvolution of drug-target interactions for each drug is necessary.The emerging field of chemical proteomics represents a powerful mass spectrometry(MS)-based affinity chromatography approach for identifying proteome-wide small molecule-protein interactions and mapping these interactions to signaling and metabolic pathways.This technique could comprehensively characterize drug targets,profile the toxicity of known drugs,and identify possible off-target activities.With the use of this technique,candidate drug molecules could be optimized,and predictable side effects might consequently be avoided.Herein,we provide a holistic overview of the major chemical proteomic approaches and highlight recent advances in this area as well as its potential applications in drug discovery.

Down-regulation of argininosuccinate lyase induces hepatoma cell apoptosis through activating Bax signaling pathway

Argininosuccinate lyase(ASL)plays an important role in the hepatic urea cycle,and can catalyze the reversible reaction of argininosuccinate to arginine and fumarate.However,the function of ASL in hepatocellular carcinoma(HCC)is not fully understood.In this study,we found that ASL expression was frequently upregulated in HCC tissues and HCC cell lines.Knock down of ASL inhibited cell proliferation and induced apoptosis in HCC cells.Mechanistic studies revealed the BCL2-associated X protein(Bax)signaling pathway which determines cancer cell apoptosis was regulated by ASL.Moreover,the depletion of Bax restored the inhibition of cell growth and reduced apoptosis initiated by ASL silencing.Together,the study demonstrated that ASL regulated HCC cell growth and apoptosis by modulating Bax signaling.Thus,the therapeutic targeting of ASL may offer options for HCC treatment.

DC breakdown characteristics of XLPE/BNNS nanocomposites considering BN nanosheet concentration,space charge and temperature

Breakdown failure in insulation material is one of the key problems that threaten the safe operation of high-voltage direct current cable.In this work,the effect of boron nitride nanosheets(BNNSs)concentration,space charge and temperature on DC breakdown strength have been explored.Cross-linked polyethylene(XLPE)/BNNS nanocomposites were prepared by the melt blending method,and the basic characteristics of nanoparticles and composite were characterised.The experimental results indicate that DC breakdown strength of nanocomposite can be effectively improved when a small amount of BN nanosheet is doped into the matrix.The breakdown strength of the sample reaches the maximum value of 407.52 kV/mm when BNNS content is 0.5 wt%,which is about 33%higher than that of pure XLPE.Further,the effect of space charge on the breakdown of nanocomposites has been studied by pre-injecting charges.For the samples with different BNNS contents,all the breakdown strength present ascending trend when the polarity of the applied voltage is the same as that of the pre-injected charges.Besides,it can be found that the breakdown strength of the XLPE/BNNSs composite decreases significantly at 50°C,which is due to more charge accumulation at 50°C.It reaches 2.06×10^(−8)C which increases by about 2.2 times than the room temperature.

Small incision lenticule extraction(SMILE)has become one of the mainstream refractive surgeries in recent years,with satisfactory efficacy,safety,and predictability.SMILE-derived refractive lenticule,the byproduct of the surgery,holds great potential in clinical practice given its easy access and good biocompatibility.Numerous studies have been published to describe its applications in refractive correction,corneal ectasia diseases,and corneal defects.The feasibility and safety were validated in both animal models and clinical studies.Moreover,the preservation method is also crucial for its further promotion and application.Novel techniques are also evaluated and applied in lenticule preservation.We covered the recent advances in the preservation of corneal stromal lenticules and their clinical reuse in this review.

Efficient reduction of hexavalent chromium with microscale Fe/Cu bimetals:Efficiency and the role of Cu

Microscale zero valent iron(mFe^(0))is one of the most potential water pollution remediation materials,but the effective utilization ability of electrons released by mFe^(0)in the reduction of hexavalent chromium(Cr(VI))is not satisfactory.Here,we find the microscale iron-copper(m Fe/Cu)bimetals coated with copper on the surface of mFe^(0)can significantly improve the effective utilization of electrons released by mFe^(0).Electrochemical analysis displays that copper plating on the surface of m Fe/Cu can promote the release the electrons from mFe^(0)and reduce the impedance of mFe^(0).Spin-polarized density functional theory(DFT)calculation reveals that Cu on the surface of m Fe/Cu bimetals promotes the release of electrons from mFe^(0)and reduces the adsorption energy of Fe to Cr.As the electron transporter,moreover,Cu can always attract Cr to the hollow position near itself of the Fe surface,which could promote the effective utilization of electrons released by Fe.Effective utilization ability of electrons in m Fe/Cu system is 12.5 times higher than that in mFe^(0)system.Our findings provide another basis for the efficient reduction of Cr(VI)by m Fe/Cu bimetals,which could promote the application and popularization of m Fe/Cu bimetals.

Impeding the combination of astrocytic ASCT2 and NLRP3 by talniflumate alleviates neuroinflammation in experimental models of Parkinson's disease

Alanine-serine-cysteine transporter 2(ASCT2)is reported to participate in the progression of tumors and metabolic diseases.It is also considered to play a crucial role in the glutamate-glutamine shuttle of neuroglial network.However,it remains unclear the involvement of ASCT2 in neurological diseases such as Parkinson’s disease(PD).In this study,we demonstrated that high expression of ASCT2 in the plasma samples of PD patients and the midbrain of MPTP mouse models is positively correlated with dyskinesia.We further illustrated that ASCT2 expressed in astrocytes rather than neurons significantly upregulated in response to either MPP+or LPS/ATP challenge.Genetic ablation of astrocytic ASCT2 alleviated the neuroinflammation and rescued dopaminergic(DA)neuron damage in PD models in vitro and in vivo.Notably,the binding of ASCT2 to NLRP3 aggravates astrocytic inflammasometriggered neuroinflammation.Then a panel of 2513 FDA-approved drugs were performed via virtual molecular screening based on the target ASCT2 and we succeed in getting the drug talniflumate.It is validated talniflumate impedes astrocytic inflammation and prevents degeneration of DA neurons in PD models.Collectively,these findings reveal the role of astrocytic ASCT2 in the pathogenesis of PD,broaden the therapeutic strategy and provide a promising candidate drug for PD treatment.

Viruses replicate and proliferate in host cells while continuously adjusting to and modulating the host environment.They encode a wide spectrum of multifunctional proteins,which interplay with and modify proteins in host cells.Viral genomes were chronologically the first to be sequenced.However,the corresponding viral proteomes,the alterations of host proteomes upon viral infection,and the dynamic nature of proteins,such as post-translational modifications,enzymatic cleavage,and activation or destruction by proteolysis,remain largely unknown.Emerging high-throughput techniques,in particular quantitative or semi-quantitative mass spectrometry-based proteomics analysis of viral and cellular proteomes,have been applied to define viruses and their interactions with their hosts.Here,we review the major areas of viral proteomics,including virion proteomics,structural proteomics,viral protein interactomics,and changes to the host cell proteome upon viral infection.

宫颈癌患者病死率位居女性恶性肿瘤前列,严重威胁女性的生命健康。近年来,得益于靶向治疗和免疫治疗研究的发展,尤其是抗血管生成药物贝伐单抗的使用,宫颈癌患者的生存时间得到了显著延长,然而仍有部分患者疗效不佳。肿瘤分子分型从分子层面对宫颈癌进行亚型划分,可进一步区分靶向治疗及免疫治疗适应人群,有望提升临床治疗效果。现针对宫颈癌靶向治疗及免疫治疗的基础研究、临床试验及其分子分型最新进展进行系统综述。
