N^(6)-methyladenosine(m^(6)A)RNA modification is widely perceived as the most abundant and common modification in transcripts.This modification is dynamically regulated by specific m^(6)A“writers”,“erasers”and“readers”and is reportedly involved in the occurrence and development of many diseases.Since m^(6)A RNA modification was discovered in the 1970s,with the progress of relevant research technologies,an increasing number of functions of m^(6)A have been reported,and a preliminary understanding of m^(6)A has been obtained.In this review,we summarize the mechanisms through which m^(6)A RNA modification is regulated from the perspectives of expression,posttranslational modification and protein interaction.In addition,we also summarize how external and internal environmental factors affect m^(6)A RNA modification and its functions in tumors.The mechanisms through which m^(6)A methylases,m^(6)A demethylases and m^(6)A-binding proteins are regulated are complicated and have not been fully elucidated.Therefore,we hope to promote further research in this field by summarizing these mechanisms and look forward to the future application of m^(6)A in tumors.

B cells are generally considered to be positive regulators of the immune response because of their capability to produce antibodies, including autoantibodies. The production of antibodies facilitates optimal CD4＋ T-cell activation because B cells serve as antigen-presenting cells and exert other modulatory functions in immune responses. However, certain B cells can also negatively regulate the immune response by producing regulatory cytokines and directly interacting with pathogenic T cells via cell-to-cell contact. These types of B Cells are defined as regulatory B （Breg） cells. The regulatory function of Breg cells has been demonstrated in mouse models of inflammation, cancer, transplantation, and particularly in autoimmunity. In this review, we focus on the recent advances that lead to the understanding of the development and function of Breg cells and the implications of B cells in human autoimmune diseases.

福建福海创石油化工有限公司将含有高浓度无机盐的除盐水再生废水引至精对苯二甲酸(PTA)厂区废水处理装置,把高浓度的无机盐均匀稀释,补充至PTA废水上流式厌氧污泥床(UASB)厌氧反应器,作为厌氧微生物微量元素营养剂添加。装置运行4个月后,颗粒污泥占厌氧污泥总量的60%以上,厌氧反应器化学需氧量(COD)降低率提升5百分点以上,说明除盐水再生废水对厌氧污泥颗粒化有明显促进作用。

Peptide:N-glycanase has been thought to be responsible for proteasome-dependent degradationof misfolded glycoproteins translocated from the endoplasmic reticulum (ER) to the cytosol.Therefore,theenzyme was supposed to be able to distinguish between native and non-native glycoproteins.In the presentstudy,a recombinant,yeast peptide:N-glycanase,Png lp, was expressed in Escherichia coli as inclusionbodies and was purified,refolded and characterized.The results showed that the recombinant enzymehas a broad pH range adaptation,from pH 4.0 to pH 10.0,and has an optimum temperature of 30 ℃.This enzyme is a zinc metalloenzyme.Its activity was abolished with the addition of EDTA and notrestored by adding metal ions.Furthermore,the deglycosylation efficiency of recombinant Pnglpfrom E.coli was investigated with respect to the substrate conformation in vitro.When ribonuclease B(RNase B) was denatured at 60-65 ℃ or by 40-60 mM dithiothreitol, indicated by its obvious structuralchange and sharpest activity change,its deglycosylation by Pnglp was most prominent.The deglycosylationefficiency of RNase B by Pnglp was found to be related to its structural conformation and enzymaticactivity.

Recent studies have identified olfactory ecto-mesenchymal stem cells （OE-MSCs） as a new type of resident stem cell in the olfactory lamina propria. However, it remains unclear whether OE-MSCs possess any immunoregulatory functions. In this study, we found that mouse OE-MSCs expressed higher transforming growth factor-beta and interleukin-10 levels than bone marrow-derived MSCs. In culture, OE-MSCs exerted their immunosuppressive capacity via directly suppressing effector T-cell proliferation and increasing regulatory T （Treg） cell expansion. In mice with collagen-induced arthritis, adoptive transfer of OE-MSCs markedly suppressed arthritis onset and disease severity, which was accompanied by increased Treg cells and reduced Th1/Th17 cell responses in vivo. Taken together, our findings identified a novel function of OE-MSCs in regulating T-cell responses, indicating that OE-MSCs may represent a new cell therapy for the treatment of rheumatoid arthritis and other autoimmune diseases.

Immunoproteasome activation in immune cells is involved in the modulation of immune responses.Increasing evidence indicates that proteasome inhibitors show beneficial effects in treating autoimmune diseases,but it remains unclear whether proteasome inhibition is an effective approach for suppressing autoimmune development in Sjo?gren’s syndrome(SS).Our previous work has demonstrated a critical role for Th17 cells in the development of experimental SS(ESS)in mice.In this study,we detected high levels of low-molecular-weight protein 7(LMP7),a key subunit of the immunoproteasome,in Th17 cells from ESS mice.Moreover,treatment with bortezomib(BTZ),a proteasome inhibitor,markedly suppressed Th17 differentiation in both murine and human naive T cells in culture.Furthermore,ESS mice treated with BTZ displayed significantly higher saliva flow rates and a reduction in tissue destruction in the salivary glands compared with vehicle-treated ESS mice.Notably,BTZ-treated ESS mice showed markedly decreased Th17 cells,germinal center B cells and plasma cells in the peripheral lymphoid organs.In addition,adoptively transferred wild type naive CD4+T cells rapidly differentiated into Th17 cells and induced salivary dysfunction in IL-17-deficient mice immunized for ESS induction.However,BTZ treatment profoundly suppressed the donor T-cell-derived Th17 response and ameliorated the reduction in salivary secretion in IL-17-deficient recipient mice.Taken together,our findings demonstrate that proteasome inhibition can effectively ameliorate ESS by suppressing the Th17 response,which may contribute to the development of a novel therapeutic strategy for the treatment of SS.

Extensive studies have demonstrated that mesenchymal stem cells(MSCs)are multipotent mesoderm-derived stromal cells that can differentiate into a variety of cell types,including adipocytes,osteoblasts,chondrocytes,myocytes and neuronal cells.1 MSCs are found in numerous organs and tissues,including bone marrow,heart,lung,muscle,peripheral blood,adipose tissue,cartilage,synovium,dental pulp,tonsil,umbilical cord,placenta,thymus and olfactory mucosa.1 MSCs have been shown to possess potent immunosuppressive functions and tissue repair capacities,which have facilitated the clinical applications of MSCs in treating a diverse range of disorders involving angiogenesis and fibrosis,including rheumatic diseases and graft-versus-host diseases.2,3 Here,we provide a brief commentary on newly emerging evidence of the immunoregulatory function and potential application of ecto-mesenchymal stem cells.

目的探讨STUB1基因变异所致常染色体隐性脊髓小脑共济失调16型患者的临床特点进而提高临床医生对该病的认识。方法收集山东大学齐鲁医院2022年5月确诊的1例常染色体隐性脊髓小脑共济失调16型患者的临床资料、辅助检查和基因检测结果,同时结合相关文献复习,对该类疾病的临床及遗传学特点进行总结。结果先证者为35岁男性,临床表现为步态不稳和构音障碍。头颅磁共振检查可见小脑萎缩。二代测序发现患者STUB1基因存在c.322dupG(p.Glu108Glyfs*4)和c.433A>C(p.Lys145Gln)复合杂合突变(参考转录本NM005861.4)。其中c.322dupG(p.Glu108Glyfs*4)为新突变。家系验证结果显示2个突变分别来自先证者表型正常的父母。通过文献复习检索到既往共12篇外文文献报道的32例常染色体隐性遗传脊髓小脑共济失调16型患者,未检索到相关中文文献报道。总结该病的主要临床表现为共济失调、构音障碍和腱反射亢进,还可伴有眼球震颤、痉挛状态、动作性震颤和肌阵挛等症状。头颅磁共振检查主要表现为小脑萎缩。结论STUB1基因变异所致常染色体隐性脊髓小脑共济失调16型在中国较罕见,以小脑共济失调为主要临床表现,影像学检查可见明显的小脑萎缩。基因检测有助于明确诊断。

Glucocorticoid-induced tumor-necrosis factor receptor(GITR)and its ligand,GITRL,play significant roles in regulating immune responses.It is clear that human soluble GITRL(hsGITRL)transduces signal activity through multiple oligomerization states.To develop human soluble trimeric GITRL protein as a potential therapeutic target,we explored the link of the isoleucine-zipper(ILZ)motif to the N-terminus of the human soluble GITRL with two leucine sequences.hsGITRL,with the ILZ motif(ILZ-hsGITRL),was firstly expressed in Escherichia coli,which exhibited a predominant trimer when identified by Sephadex G-100 filtration and non-reducing SDS–polyacrylamide gel electrophoresis(SDS-PAGE).The significantly higher biological activity of the ILZ-hsGITRL compared with hsGITRL was confirmed by CD41 T proliferation,interferon-c(IFN-c)secretion and binding activity assay.To reveal and compare the underlying mechanisms,the level of extracellular signal-regulated kinase-1/2(ERK1/2)phosphorylation was examined,indicating that ILZ-hsGITRL induced more persistent and stronger ERK1/2 activation than hsGITRL.In conclusion,the incorporation of an ILZ motif could markedly improve the costimulation of hsGITRL.
