Multiple sclerosis(MS)is a neuroinflammatory demyelinating disease,mediated by pathogenic T helper 17(Th17)cells.However,the therapeutic effect is accompa-nied by the fluctuation of the proportion and function of Th17 cells,which prompted us to find the key regulator of Th17 differentiation in MS.Here,we demonstrated that the triggering receptor expressed on myeloid cells 2(TREM-2),a modulator of pattern recognition receptors on innate immune cells,was highly expressed on pathogenic CD4-positive T lymphocyte(CD4^(+)T)cells in both patients with MS and experimental autoimmune encephalomyelitis(EAE)mouse models.Conditional knockout of Trem-2 in CD4^(+)T cells significantly alleviated the disease activity and reduced Th17 cell infiltration,activation,differentiation,and inflammatory cytokine production and secretion in EAE mice.Furthermore,with Trem-2 knockout in vivo experiments and in vitro inhibitor assays,the TREM-2/zeta-chain associated protein kinase 70(ZAP70)/signal transducer and activator of transcription 3(STAT3)signal axis was essential for Th17 activation and differentiation in EAE progression.In conclusion,TREM-2 is a key regulator of pathogenic Th17 in EAE mice,and this sheds new light on the potential of this therapeutic target for MS.

Effect of Heat Treatment on Microstructure and Mechanical Behavior of Cu-Bearing 316L Stainless Steel Produced by Selective Laser Melting

Cu-bearing stainless steels(SSs)with high strength,excellent plasticity,and effective antimicrobial properties hold significant potential for applications in the marine industry.In this study,Cu-bearing SSs with copper ranging from 0 to 6.0 wt%were successfully prepared using selective laser melting(SLM)technology.For the Cu-bearing SSs with different copper contents,the effect of heat treatment on the microstructural and mechanical behaviors was studied systematically.Microstructural observations revealed that the subgrain size of Cu-bearing SSs increased with heat treatment at 500℃ and 700℃ for 6 h.Furthermore,the tensile strength and elongation increased after the heat treatment temperature due to the combined effect of dislocations,twins,andε-Cu precipitated phases.Notably,after heat treatment at 700℃,the SLM4.5Cu sample exhibited an abnormal rise in tensile strength and elongation.This finding suggests that the diffusion strengthening caused byε-Cu precipitates exceeded the stacking fault energy.Consequently,the tensile strength and elongation reached 693.32 MPa and 56.94%,respectively.This work provides an efficient approach for preparing Cu-bearing SSs with exceptional strength and plasticity.

To achieve high-performance compressor cascades at low Reynolds number(Re),it is important to organize the boundary layer transition and separation processes efficiently and reasonably.In this study,the airfoil is focused on at a 5% blade height at the root of the orthogonal blade in the downflow passage of the high-load booster stage.The bionics modeling design is carried out for the leading edge of the original blade cascade;the response characteristics of laminar transition and separation to blades with different leading edge shapes at low Reynolds numbers are studied by using large eddy simulations combined with Omega vortex identification.The findings of this study demonstrate that bionic leading edge modeling can significantly improve the aerodynamic performance of blades at low Reynolds numbers.The blades effectively suppress the formation of separation bubbles at low Reynolds numbers and weaken or even eliminate large-scale flow separation at the trailing edge.In addition,the blades can weaken the vortex intensity on the blade surface,reduce the areas of high-velocity fluctuations,and minimize aerodynamic losses caused by turbulence dissipation.These results should serve as a valuable reference for the aerodynamic design and flow control of the high-load booster stage blade at low Re.

Mg-based amorphous alloys exhibit efficient catalytic performance and excellent biocompatibility with a promising application probability,specifically in the field of azo dye wastewater degradation.However,the problems like difficulty in preparation and poor cycling stability need to be solved.At present,Mg-based amorphous alloys applied in wastewater degradation are available in powder and ribbon.The amorphous alloy powder fabricated by ball milling has a high specific surface area,and its reactivity is thousands of times better than that of gas atomized alloy powder.But the development is limited due to the high energy consumption,difficult and costly process of powder recycling.The single roller melt-spinning method is a new manufacturing process of amorphous alloy ribbon.Compared to amorphous powder,the specific surface area of amorphous ribbon is relatively lower,therefore,it is necessary to carry out surface modification to enhance it.Dealloying is a way that can form a pore structure on the surface of the amorphous alloys,increasing the specific surface area and providing more reactive sites,which all contribute to the catalytic performance.Exploring the optimal conditions for Mg-based amorphous alloys in wastewater degradation by adjusting amorphous alloy composition,choosing suitable method to preparation and surface modification,reducing cost,expanding the pH range will advance the steps to put Mg-based amorphous alloys in industrial environments into practice.

OBJECTIVE: To explore whether the regulation of matrix metalloproteinase 9(MMP-9)/tissue inhibitors of MMPs(TIMPs) gene expression through histone acetylation is a possible mechanism by which electroacupuncture(EA)protects blood-brain barrier(BBB) integrity in a middle cerebral artery occlusion(MCAO) rat model.METHODS: Male Sprague-Dawley rats were divided into four groups: the sham group, the MCAO group, the MCAO + EA(MEA) group, and the MCAO + EA + HAT inhibitor(HATi) group. The MCAO model was generated by blocking the middle cerebral artery. EA was applied to Baihui(GV20). Samples were collected 1 or 3 d after reperfusion. Neurological function scores and Evans blue extravasation were employed to evaluate the poststroke injury. The effect of EA on MMP-9/TIMPs gene expression was assessed by real-time fluorescence quantitative polymerase chain reaction(RT-qPCR) and chromatin immunoprecipitation(ChIP). RESULTS: Our results showed that EA treatment prominently improved neurological function and ameliorated BBB disruption. The RT-qPCR assay showed that EA reduced the expression of MMP-9 and promoted TIMP-2 mRNA expression, but HATi reversed these effects of EA. In addition, ChIP results revealed that EA decreased the enrichment of H3K9ace/H3K27ace at MMP-9 promoters and notably stimulated the recruitment of H3K9ace/H3K27ace at TIMP-2 promoter. CONCLUSION: EA treatment at Baihui(GV20) regulates the transcription of MMP-9 and TIMP-2 through histone acetylation modification in the acute stage of stroke, which preserves the structural integrity of the BBB in MCAO rats. These findings suggested that the histone acetylation-mediated transcriptional activity of target genes may be a crucial mechanism of EA treatment in stroke.

对口帮扶作为一种横向转移支付方式,在缓解地区发展不平衡等方面取得了突出成就。文章基于2010—2020年地级市层面异地投资子公司数量的面板数据,以2016年《关于进一步加强东西部扶贫协作工作的指导意见》为政策背景,运用双重差分法探讨了对口帮扶对企业异地投资的政策效应。研究发现,两地建立对口帮扶结对关系显著促进了帮扶地对被帮扶地进行跨区域投资,且不存在地理距离远近和产权性质不同的异质性影响,但受到市场化水平和方言异质性的影响。进一步分析发现,对口帮扶政策在两城市行政等级不同组合下呈现出不同的影响效应。最后,从帮扶双方角度分别探究该政策对异地投资的调节效应。本文研究结论对新时期开展对口帮扶政策的研究创新提供了新的经验证据。

厘清政府间垂直管理关系,是优化省以下财政体制的重要前提。本文从市县距离这一区位特征出发,基于中国地级市政府驻地迁移改革,构建双重差分模型,识别政府邻近与县域经济发展之间的因果关系。研究发现,政府邻近具有显著的经济效应,并且从动态趋势上看,这一经济效应是持续的而非短期的。机制检验表明,公共投资的增长是政府邻近优势能够实现的重要途径,不仅信息优势提高了市政府对邻近县域投资的力度,与市政府邻近也激励了县域自身公共投资的动力。异质性分析表明,在初始距离近、财政分权程度高以及财政压力小的地区影响更为显著,并且这一经济效应主要体现在第二产业增长上。进一步研究发现,政府邻近对资本及劳动力的带动作用是负向的,对县域经济效率的影响也不明显。但是,当地方制度环境改善时,政府邻近对县域经济效率的带动作用会逐步显现。本文研究将有利于深入理解上下级政府间经济互动的地理诱因,对进一步促进县域经济高质量发展的政策实践具有借鉴意义。
