Urinary exosomal microRNA-145-5p and microRNA-27a-3p act as noninvasive diagnostic biomarkers for diabetic kidney disease

BACKGROUND Diabetic kidney disease(DKD),characterized by increased urinary microalbumin levels and decreased renal function,is the primary cause of end-stage renal di-sease.Its pathological mechanisms are complicated and multifactorial;Therefore,sensitive and specific biomarkers are needed.Urinary exosome originate from diverse renal cells in nephron segments and partially mirror the pathological changes in the kidney.The microRNAs(miRNAs)in urinary exosome are remark-ably stable and highly tissue-specific for the kidney.METHODS Type 2 diabetic mellitus(T2DM)patients were recruited from the Second Hospital of Hebei Medical University and were divided into two groups:DM,diabetic pa-tients without albuminuria[urinary albumin to creatinine ratio(UACR)hain reaction.The correlation between exosomal miRNAs and the clinical in-dexes was evaluated.The diagnostic values of exosomal miR-145-5p and miR-27a-3p in DKD were determined using receiver operating characteristic(ROC)analysis.Biological functions of miR-145-5p were investigated by performing RESULTS Urinary exosomal expression of miR-145-5p and miR-27a-3p was more upregulated in the DKD group than in the DM group(miR-145-5p:4.54±1.45 vs 1.95±0.93,P<0.001;miR-27a-3p:2.33±0.79 vs 1.71±0.76,P<0.05)and the NC group(miR-145-5p:4.54±1.45 vs 1.55±0.83,P<0.001;miR-27a-3p:2.33±0.79 vs 1.10±0.51,P<0.001).The exosomal miR-145-5p and miR-27a-3p positively correlated with albuminuria and serum creatinine and negatively correlated with the estimated glomerular filtration rate.miR-27a-3p was also closely related to blood glucose,gly-cosylated hemoglobin A1c,and low-density lipoprotein cholesterol.ROC analysis revealed that miR-145-5p had a better area under the curve of 0.88[95%confidence interval(CI):0.784-0.985,P<0.0001]in diagnosing DKD than miR-27a-3p with

Effect of pre-existing shear bands on mechanical properties and serration behaviors in bulk metallic glasses

Pre-existing（multiple）shear bands were introduced into the ductile Zr56Co28Al16 and Zr65Ni10Cu15Al10bulk metallic glasses（BMGs）through the lateral-deformation,respectively.It was found that the pre-exiting shear bands can further enhance the compressive plasticity of ductile BMGs.According to the serration analysis on the plastic deformation of the as-cast and the pre-deformed samples,the serration events in the stress-strain curves during deformation display a self-organized critical（SOC）behavior.Compared with the as-cast BMGs,a larger power-law scaling exponent calculated based on serrated flow behaviors becomes larger for the pre-deformed BMGs,implying that the shear banding stability of BMGs is effectively enhanced.This should be caused by the pronounced interactions of shear bands during plastic deformation for the pre-deformed BMGs.However,by introducing a large amount of multiple shear bands into the glassy matrix,it also becomes easier for shear bands to propagate along the pre-existing shear bands,leading to a lower cut-off elastic energy density for the pre-deformed BMGs.More multiple shear bands with stronger interactions for the pre-deformed BMGs could provide a larger chance to activate the shear-band cracking but less local elastic energies are remained for the subsequent crack-linking.

Improving mathematical model of burden distribution and correcting chute angle to cope with fluctuation of stock line

Accurate evaluations of the burden distribution are of critical importance to stabilize the operation of blast furnace.The mathematical model and discrete element method(DEM)are two attractive methods for predicting burden distribution.Based on DEM,the initial velocities of the pellet,sinter,and coke were calculated,and the velocity attenuations of the above three particles between the burden and the chute were analyzed.The initial velocity and velocity attenuation were applied to a mathematical model for improving the accuracy.Additionally,based on the improved model,a scheme for rectifying the chute angles was proposed to address the fluctuation of the stock line and maintain a stable burden distribution.The validity of the scheme was confirmed via a stable burden distribution under different stock lines.The mathematical model has been successfully applied to evaluate the online burden distribution and cope with the fluctuation of the stock line.

Interpretation and characterization of rate of penetration intelligent prediction model

Accurate prediction of the rate of penetration(ROP)is significant for drilling optimization.While the intelligent ROP prediction model based on fully connected neural networks(FNN)outperforms traditional ROP equations and machine learning algorithms,its lack of interpretability undermines its credibility.This study proposes a novel interpretation and characterization method for the FNN ROP prediction model using the Rectified Linear Unit(ReLU)activation function.By leveraging the derivative of the ReLU function,the FNN function calculation process is transformed into vector operations.The FNN model is linearly characterized through further simplification,enabling its interpretation and analysis.The proposed method is applied in ROP prediction scenarios using drilling data from three vertical wells in the Tarim Oilfield.The results demonstrate that the FNN ROP prediction model with ReLU as the activation function performs exceptionally well.The relative activation frequency curve of hidden layer neurons aids in analyzing the overfitting of the FNN ROP model and determining drilling data similarity.In the well sections with similar drilling data,averaging the weight parameters enables linear characterization of the FNN ROP prediction model,leading to the establishment of a corresponding linear representation equation.Furthermore,the quantitative analysis of each feature's influence on ROP facilitates the proposal of drilling parameter optimization schemes for the current well section.The established linear characterization equation exhibits high precision,strong stability,and adaptability through the application and validation across multiple well sections.

Transparent micropatterned conductive films based on highlyordered nanowire network

Transparent conductive films that are based on nanowire networks are essential to construct flexible,wearable,and even stretchable electronics.However,large-scale precise micropatterning,especially with regard to the controllability of the organizing orientation of nanowires,is a critical challenge.Herein,we proposed a liquid film rupture self-assembly approach for manufacturing transparent conductive films with microstructure arrays based on a highly ordered nanowire network.The large-scale microstructure conductive films were fabricated through air-liquid interface self-assembly and liquid film rupture self-assembly.Six typical micropattern morphologies,including square,hexagon,circle,serpentine,etc.,were prepared to reveal the universal applicability of the proposed approach.The homogeneity and controllability of this approach were verified for multiple assemblies.With the assembly cycles increasing,the optical transmittance decreases slightly.In addition,theoretical model analysis is carried out,and the analytical formula of the speed of the film moving with the surface tension and the density of the liquid film is presented.Finally,the feasibility of this approach for piezoresistive strain sensors is verified.This fabrication approach demonstrated a cost-effective and efficient method for precisely arranging nanowires,which is useful in transparent and wearable applications.

Highly Transparent Fire-resistant Coatings with Intumescent Three-source Integration

Wood,a readily available and sustainable natural resource,has found widespread use in construction and furniture.However,its inherent flammability poses a potential fire risk.Although intumescent fire-retardant coatings effectively mitigate this risk,achieving high transparency in such coatings presents a significant challenge.In our approach,we employed a cross-linked network of phytic acid anion and N-[3-(trimethoxysilyl)propyl]-N,N,N-trimethylammonium cation to create a transparent"three-in-one"intumescent coating.The collaborative P/N/Si flame-retardant effect markedly improved the intumescent char-forming capability,preventing the wood from rapid decomposition.This resulted in a substantial reduction in heat release(13.9%decrease in THR)and an increased limiting oxygen index(LOI)value of 35.5%.Crucially,the high transparency of the coating ensured minimal impact on the wood's appearance,allowing the natural wood grains to remain clearly visible.This innovative approach provides a straightforward method for developing transparent intumescent flame-retardant coatings suitable for wooden substrates.The potential applications extend to preserving ancient buildings and heritage conservation efforts.

Species profile and reactivity of volatile organic compounds emission in solvent uses,industry activities and from vehicular tunnels

A survey was conducted of the volatile organic compounds(VOCs)released from sources of solvent use,industry activities and vehicle emissions in Guiyang,a capital city of China.Samples were collected by canisters and analyzed by GC-MS-FID.The species profiles of VOCs emitted from sources were obtained.Results showed that xylenes,ethylbenzene,acetone and dichloromethane were the characteristics species for painting,2-propanol and ethyl acetate for printing,α-pinene for solid wood furniture manufacturing,and 2-butanone for biscuit baking.These characteristics species could be as tracers for the sources respectively.In most of samples from the solvent use,the benzene/toluene(B/T)ratio was less than 0.3,indicating that the ratio could be as the indicator for tracing the solvent use related sources.The results also suggested that the toluene/xylene(T/X)ratio be as the indicator to distinguish the VOCs sources of painting(2).Aromatics contributed the most to ozone formation potential(OFP)of most painting and non-paper printing sources,and oxygen-containing VOCs(OVOCs)were major species contributing to OFP of the sources from food production and paper printing.The OFP of the VOCs emissions from vehicle in tunnels and from other manufactures were dominated by both aromatics and alkenes.Theα-pinene could explain 56.94%and 32.54%of total OFP of the VOCs sources from filing cabinet and solid wood furniture manufacturing,which was rarely been involved in previous studies of VOCs source profiles,indicating that the species of concern for VOCs sources are still insufficient at present.

Derivation of Expanded Isospectral-Nonisospectral Integrable Hierarchies via the Column-vector Loop Algebra

A scheme for generating nonisospectral integrable hierarchies is introduced.Based on the method,we deduce a nonisospectral hierarchy of soliton equations by considering a linear spectral problem.It follows that the corresponding expanded isospectral and nonisospectral integrable hierarchies are deduced based on a 6 dimensional complex linear space ■.By reducing these integrable hierarchies,we obtain the expanded isospectral and nonisospectral derivative nonlinear Schr?dinger equation.By using the trace identity,the biHamiltonian structure of these two hierarchies are also obtained.Moreover,some symmetries and conserved quantities of the resulting hierarchy are discussed.

Fangji Fuling Decoction Alleviates Sepsis by Blocking MAPK14/FOXO3A Signaling Pathway

Objective:To examine the therapeutic effect of Fangji Fuling Decoction(FFD) on sepsis through network pharmacological analysis combined with in vitro and in vivo experiments.Methods:A sepsis mouse model was constructed through intraperitoneal injection of 20 mg/kg lipopolysaccharide(LPS).RAW264.7 cells were stimulated by 250 ng/m L LPS to establish an in vitro cell model.Network pharmacology analysis identified the key molecular pathway associated with FFD in sepsis.Through ectopic expression and depletion experiments,the effect of FFD on multiple organ damage in septic mice,as well as on cell proliferation and apoptosis in relation to the mitogen-activated protein kinase 14/Forkhead Box O 3A(MAPK14/FOXO3A) signaling pathway,was analyzed.Results:FFD reduced organ damage and inflammation in LPS-induced septic mice and suppressed LPS-induced macrophage apoptosis and inflammation in vitro(P<0.05).Network pharmacology analysis showed that FFD could regulate the MAPK14/FOXO signaling pathway during sepsis.As confirmed by in vitro cell experiments,FFD inhibited the MAPK14 signaling pathway or FOXO3A expression to relieve LPS-induced macrophage apoptosis and inflammation(P<0.05).Furthermore,FFD inhibited the MAPK14/FOXO3A signaling pathway to inhibit LPS-induced macrophage apoptosis in the lung tissue of septic mice(P<0.05).Conclusion:FFD could ameliorate the LPS-induced inflammatory response in septic mice by inhibiting the MAPK14/FOXO3A signaling pathway.

Sustained release of vascular endothelial growth factor A and basic fibroblast growth factor from nanofiber membranes reduces oxygen/glucose deprivation-induced injury to neurovascular units

Upregulation of vascular endothelial growth factor A/basic fibroblast growth factor(VEGFA/b FGF)expression in the penumbra of cerebral ischemia can increase vascular volume,reduce lesion volume,and enhance neural cell proliferation and differentiation,thereby exerting neuroprotective effects.However,the beneficial effects of endogenous VEGFA/b FGF are limited as their expression is only transiently increased.In this study,we generated multilayered nanofiber membranes loaded with VEGFA/b FGF using layer-by-layer self-assembly and electrospinning techniques.We found that a membrane containing 10 layers had an ideal ultrastructure and could efficiently and stably release growth factors for more than 1 month.This 10-layered nanofiber membrane promoted brain microvascular endothelial cell tube formation and proliferation,inhibited neuronal apoptosis,upregulated the expression of tight junction proteins,and improved the viability of various cellular components of neurovascular units under conditions of oxygen/glucose deprivation.Furthermore,this nanofiber membrane decreased the expression of Janus kinase-2/signal transducer and activator of transcription-3(JAK2/STAT3),Bax/Bcl-2,and cleaved caspase-3.Therefore,this nanofiber membrane exhibits a neuroprotective effect on oxygen/glucose-deprived neurovascular units by inhibiting the JAK2/STAT3 pathway.
