Digital design and manufacturing have been under pinned by digital modeling, simulation, and automation controls for decades. Under the new market requirement of mass customized products and services, the advancements in artificial intelligence （AI）, smart technology, virtual reality （VR）, big data, digital twin, robotics and human-centered design are becoming driving forces for the development of future digital design and manufacturing. This special issue focuses on the future digital design and manufacturing especially under the Industry 4.0 framework and beyond. This editorial introduces the papers in this special issue, which linked to the International Workshop on Digital Design and Manufacturing Technologies - Embracing Industry 4.0 and Beyond at Northumbria University in Newcastle, UK, held on 12-13 April 2016. In the Part I of the issue [1], there are 13 papers published in 2016, Vol- ume 29, No 6 of the Chinese Journal of Mechanical Engineering （this journal）.

As the overall population ages, driving-related accidents and injuries, associated with elderly drivers, have risen. Existing research about elderly drivers mainly focuses on factual data collection and analysis, indicating the elderly＇s growing fatal accident rates and their different behaviours compared to younger drivers. However, few research has focused on design-led practical solutions to mitigate the elderly＇s growing fatal accidents, by consid- ering their usability and body conditions, afflicting the elderly, such as decreased vision, hearing, and reaction times. In this paper, first, current worldwide situations on growing fatal accident rates for elderly drivers is reviewed and the key impact factors are identified and discussed with regarding to usability and design trend in the automotive technology for elderly. Second, existing smart vehicle technology-based solutions to promote safe driving are explored and their pros and cons are discussed and anal- ysed. Most of solutions are not created by people with driving difficulties, which are caused by health problems most commonly afflicting the elderly. Thirdly, diverse design-led research activities are taken, such as a survey, observation, and interviews to gain new understanding of what kinds of driving problems elderly drivers have and demonstrate how new system concepts could be developed for the elderly＇s benefits. Finally, it is found that the elderly＇s low vision and late reaction are main factors causing their driving difficulties. Based on this finding, usable vehicle system design ideas have been proposed, by utilising facial expression sensing technology as a solution. The proposed solutions would ensure reducing both the elderly＇s driving problems and high fatal accident rates and provide a more enjoyable driving environment for the elderly population.

Objective:To study the pharmacological mechanism of Guanxin Ⅱ formula(冠心Ⅱ号方,GXⅡ)for treatment of coronary heart disease(CHD).Methods:A network pharmacology-based method was utilized.First candidate compounds,targets of GXⅡ were collected using Pharm Mapper,BATMAN-TCM,Drug Bank and Swiss Target Prediction,and targets on CHD were mined from Gene Cards,Dis Genet,Drug Bank and GEO.Afterwards,the big hub compounds and targets were chosen in the candidate compounds-direct therapeutic targets on the CHD(C-T)network and the direct therapeutic targets on the CHD(T-D)network.Furthermore,the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis were performed to identify the enriched terms.Finally,a molecular docking simulation strategy was adopted to verify the binding capacity between the big hub compounds and big hub targets on CHD.Results:First,114 candidate compounds were selected with the following criteria:OB 30%,DL 0.18,and HL 4 h.Then,1,035 targets of GXⅡ were gathered,while 928 targets on CHD were collected.Afterwards,196 common targets of compound targets and therapeutic targets on CHD were defined as direct therapeutic targets acting on CHD.In addition,the contribution index(CI)in the C-T network was calculated,and 4 centrality properties,including degree,betweenness,closeness and coreness,in the T-D network,4 big hub compounds,and 6 big hub targets were eventually chosen.Furthermore,the GO and KEGG analysis indicated that GXⅡ acted on CHD by regulating the reactive oxygen species metabolism,steroid metabolism,lipid metabolism,inflammatory response,proliferation,differentiation and apoptosis.The docking results manifested excellent binding capacity between the 4 big hub compounds and 6 big hub targets on CHD.Conclusion:This network pharmacology-based exploration revealed that GXⅡ might prevent and inhibit the primary pathological processes of CHD.

Design Change Model for Effective Scheduling Change Propagation Paths

Changes in requirements may result in the increasing of product development project cost and lead time, therefore, it is important to understand how require- ment changes propagate in the design of complex product systems and be able to select best options to guide design. Currently, a most approach for design change is lack of take the multi-disciplinary coupling relationships and the number of parameters into account integrally. A new design change model is presented to systematically analyze and search change propagation paths. Firstly, a PDS-Be- havior-Structure-based design change model is established to describe requirement changes causing the design change propagation in behavior and structure domains. Secondly, a multi-disciplinary oriented behavior matrix is utilized to support change propagation analysis of complex product systems, and the interaction relationships of the matrix elements are used to obtain an initial set of change paths. Finally, a rough set-based propagation space reducing tool is developed to assist in narrowing change propagation paths by computing the importance of the design change parameters. The proposed new design change model and its associated tools have been demonstrated by the scheduling change propagation paths of high speed train＇s bogie to show its feasibility and effectiveness. This model is not only supportive to response quickly to diversified market requirements, but also helpful to satisfy customer require- ments and reduce product development lead time. The proposed new design change model can be applied in a wide range of engineering systems design with improved efficiency.

Upregulation of erythropoietin and erythropoietin receptor in castration-resistant progression of prostate cancer

Hypoxia-induced erythropoietin signaling plays an important role in tumor growth and invasion.In the present study,we investigated the contribution of erythropoietin signaling pathway to castration-resistant prostate cancer and the development of a neuroendocrine phenotype.Immunohistochemical staining showed that the erythropoietin and erythropoietin receptor scores in castration-resistant prostate cancer and androgen-dependent prostate cancer were 7.55 versus 4.5 and 7.45 versus 5.9,respectively(P<0.001).Furthermore,a cell proliferation assay was conducted,and the differential expression of erythropoietin and erythropoietin receptor in LNCaP cells and hypoxia-induced LNCaP cells was evaluated using western blot and quantitative real-time PCR.The proliferation capacity of hypoxia-induced LNCaP cells was similar in cultures of both fetal bovine serum and charcoal-stripped fetal bovine serum,suggesting that LNCaP cells acquired hypoxia-induced androgen-independent growth.After 2 weeks of hypoxic culture,LNCaP cells showed a neuroendocrine cell change and increased expression of neuron-specific enolase,erythropoietin,and erythropoietin receptor;knockdown of erythropoietin receptor reversed the hypoxia-induced upregulation of neuron-specific enolase in the LNCaP cells.In conclusion,the concurrent upregulation of erythropoietin and erythropoietin receptor in castration-resistant prostate cancer suggests that the erythropoietin/erythropoietin receptor autocrine loop plays an important role in the progression of castration resistance and is responsible for the development of a neuroendocrine phenotype.

Effects of MgO and TiO_2 on the viscous behaviors and phase compositions of titanium-bearing slag

The effects of MgO and TiO_2 on the viscosity, activation energy for viscous flow, and break-point temperature of titanium-bearing slag were studied. The correlation between viscosity and slag structure was analyzed by Fourier transform infrared（FTIR） spectroscopy. Subsequently, main phases in the slag and their content changes were investigated by X-ray diffraction and Factsage 6.4 software package. The results show that the viscosity decreases when the MgO content increases from 10.00wt% to 14.00wt%. Moreover, the break-point temperature increases, and the activation energy for viscous flow initially increases and subsequently decreases. In addition, with increasing TiO_2 content from 5.00wt% to 9.00wt%, the viscosity decreases, and the break-point temperature and activation energy for viscous flow initially decrease and subsequently increase. FTIR analyses reveal that the polymerization degree of complex viscous units in titanium-bearing slag decreases with increasing MgO and TiO_2 contents. The mechanism of viscosity variation was elucidated. The basic phase in experimental slags is melilite. Besides, as the MgO content increases, the amount of magnesia–alumina spinel in the slag increases. Similarly, the sum of pyroxene and perovskite phases in the slag increases with increasing TiO_2 content.

Objective:Extreme gradient boosting(XGBoost)was used to predict the 7^(th)day efficacy of the acupoint application(AP)of Chinese herbs(Xiao Zhong Zhi Tong Tie)in patients with diarrhea.Materials and Methods:We consecutively collected medical records of patients with diarrhea nationwide on the Chun Bo Wan Xiang cloud platform from August 22 to November 5,2020.Demographic and clinical data and the fecal properties were included in this study.We established the XGBoost model to predict the 7^(th)day efficacy of AP in patients with diarrhea.The XGBoost model was evaluated using the area under the receiver operating characteristic(ROC)curve(AUC).We next compared the performance of XGBoost with that of artificial neural network(ANN),ANN+boosting,ANN+bagging,and support vector machine(SVM).Results:The XGBoost model provided a prediction accuracy of 84.86%(95%confidence interval=82.74%to 86.81%)and the ROC curve analysis showed an AUC of 0.81.The top-three variables with the highest importance are age,duration of diarrhea,and region(North).Our study revealed that XGBoost was not superior to ANN,ANN+boosting,ANN+bagging,and SVM.Conclusions:The established XGBoost model for predicting the 7^(th)day efficacy of AP in patients with diarrhea exhibited good accuracy and precision,which can be used for efficacy prediction.

Lactic acid bacteria （LAB） are widely used in food industries. Correct identification and safety evaluation of these bacteria at the species even strain level should take considerations into account. In this study, the LAB were recovered from yoghurt and characterized phenotypically and genetically. Fifty-two isolates of LAB from 31 yoghurt samples were cultured and grouped into 6 species including Luctobucillus bulguricus （24 isolates）, Streptococcus thermophilus （15 isolates）, L. ucidophilus （7 isolates）, L. porucusei/cusei （3 isolates）, L. delbrueckii （2 isolates）, and L. fermentum （1 isolate）, based on their Gram-staining, colony morphology and biochemical properties.

Erectile dysf un ction and penile shri nkage are the comm on complications after radical prostatectomy.Penile rehabilitati on is widely applied after the surgery.Vacuum therapy is one of the three penile rehabilitation methods used in the clinical setting,but its mechanism is not well known.This study was designed to investigate whether vacuum erectile device(VED)can prevent corporeal veno-occlusive dysfunction and penile shrinkage in the bilateral cavernous nerve crush(BCNC)rat model.Adult male Sprague-Dawley rats were randomly assigned into three groups:sham group,BCNC group,and BCNC+VED group.After 4 weeks,penile length and intracavernosal pressure(ICP)were measured,and then the middle part of the penis was harvested after dynamic inf usion caver no sometry to complete the followi ng items:smooth muscle/collage n ratios and collagenⅠ/Ⅲratios;ultramicrostructure of the tunica albuginea,endothelial cell,and smooth muscle cell;and the expression of calponin・l and osteopontin.The penile shortening,peak ICP and ICP drop rate after alprostadil injection were significantly improved with vacuum therapy after 4-week treatme nt.Compared with BCNC group,VED sign ifica ntly in creased smooth muscle/collagen ratios,decreased collagenⅠ/Ⅲratios,and preserved the ultramicrostructure of the tunica albuginea,endothelial cell,and smooth muscle cell.The data also showed that an imals exposed to VED could partially reverse the expression of calp onin・l and osteop ontin induced by BCNC.In conclusion,vacuum therapy is effective to prevent penile shrinkage and veno-occlusive dysfunction in penile rehabilitation,which may be associated with well-preserved structure and function of the tunica albuginea,endothelial cell,and smooth muscle cell.
