Genetic characteristics of a patient with multiple primary cancers:A case report

BACKGROUND Two or multiple primary malignant neoplasms(MPMNs)rarely occur in the same *** has been reported that MPMNs are easily misdiagnosed as the recurrence or metastasis of malignancies in clinical practice,affecting the choice of treatment for the patients,thereby resulting in the delay of optimal *** generation sequencing(NGS)can be used to distinguish between multiple primary lung cancers and intrapulmonary metastasis,and may distinguish the origin of tumours in different sites of the *** suMMARY We report the case of 66-year-old woman who suffered from different malignant neoplasms in the rectum and esophageal and gastrointestinal *** first neoplasm rectal adenocarcinoma was diagnosed and removed in *** second and third lesions were diagnosed with esophageal squamous-cell carcinoma(ESCC)and gastrointestinal stromal tumour(GIST),respectively,in *** whole exome sequencing was performed on the tissue specimens of rectal carcinoma,esophageal cancer,GIST,and white blood cells to investigate the relationship between malignancies at different timeframe and determine whether the ESCC and GIST evolved from the rectal *** including v-Ki-ras2-Kirsten rat sarcoma viral oncogene homolog,adenomatosis polyposis coli,and mothers against decapentaplegic homolog 4 were detected in rectal adenocarcinoma sample,mast/stem cell growth factor receptor was detected in GIST tissue,and lysine methyltransferase 2D was detected in ESCC ***,ESCC and GIST were not genetically evolved from rectal adenocarcinoma,and this patient did not have a trunk driven *** NGS is an effective tool to study clonal evolution of tumours and distinguish between MPMNs and intrapulmonary metastasis.

surrogate modeling for unsaturated infiltration via the physics and equality-constrained artificial neural networks

Machine learning(ML)provides a new surrogate method for investigating groundwater flow dynamics in unsaturated *** pure data-driven methods(*** neural network,DNN)can provide rapid predictions,but they do require sufficient on-site data for accurate training,and lack interpretability to the physical processes within the *** this paper,we provide a physics and equalityconstrained artificial neural network(PECANN),to derive unsaturated infiltration solutions with a small amount of initial and boundary *** takes the physics-informed neural network(PINN)as a foundation,encodes the unsaturated infiltration physical laws(*** equation,RE)into the loss function,and uses the augmented Lagrangian method to constrain the learning process of the solutions of RE by adding stronger penalty for the initial and boundary *** unsaturated infiltration cases are designed to test the training performance of PECANN,***-dimensional(1D)steady-state unsaturated infiltration,1D transient-state infiltration,two-dimensional(2D)transient-state infiltration,and 1D coupled unsaturated infiltration and *** predicted results of PECANN are compared with the finite difference solutions or analytical *** results indicate that PECANN can accurately capture the variations of pressure head during the unsaturated infiltration,and present higher precision and robustness than DNN and *** is also revealed that PECANN can achieve the same accuracy as the finite difference method with fewer initial and boundary training ***,we investigate the effect of the hyperparameters of PECANN on solving RE *** provides an effective tool for simulating unsaturated infiltration.

Hepatocellular carcinoma(HCC),a prevalent solid carcinoma of significant concern,is an aggressive and often fatal disease with increasing global incidence rates and poor therapeutic *** etiology and pathological progression of non-alcoholic steatohepatitis(NASH)-related HCC is multifactorial and ***,no single animal model can accurately mimic the full NASH-related HCC pathological progression,posing considerable challenges to transition and mechanistic ***,a novel conditional inducible wild-type human HRAS overexpressed mouse model(HRAS-HCC)was established,demonstrating 100%morbidity and mortality within approximately one month under normal dietary and lifestyle *** symptoms of HCC such as ascites,thrombus,internal hemorrhage,jaundice,and lung metastasis were successfully replicated in ***-depth pathological features of NASH-related HCC were demonstrated by pathological staining,biochemical analyses,and typical marker gene *** murine anti-PD-1 and sorafenib treatment effectively prolonged mouse survival,further confirming the accuracy and reliability of the *** on protein-protein interaction(PPI)network and RNA sequencing analyses,we speculated that overexpression of HRAS may initiate the THBS1-COL4A3 axis to induce NASH with severe fibrosis,with subsequent progression to ***,our study successfully duplicated natural sequential progression in a single murine model over a very short period,providing an accurate and reliable preclinical tool for therapeutic evaluations targeting the NASH to HCC continuum.

P-and SV-wave dispersion and attenuation in saturated microcracked porous rock with aligned penny-shaped fractures

P-and SV-wave dispersion and attenuation have been extensively investigated in saturated poroelastic media with aligned ***,there are few existing models that incorporate the multiple wave attenuation mechanisms from the microscopic scale to the macroscopic ***,in this work,we developed a unified model to incorporate the wave attenuation mechanisms at different scales,which includes the microscopic squirt flow between the microcracks and pores,the mesoscopic wave-induced fluid flow between fractures and background(FB-WIFF),and the macroscopic Biot's global flow and elastic scattering(ES)from the *** Tang's modified Biot's theory and the mixed-boundary conditions,we derived the exact frequency-dependent solutions of the scattering problem for a single penny-shaped fracture with oblique incident P-and *** then developed theoretical models for a set of aligned fractures and randomly oriented fractures using the Foldy *** results indicated that microcrack squirt flow considerably influences the dispersion and attenuation of P-and SV-wave *** coupling effects of microcrack squirt flow with the FB-WIFF and ES of fractures cause much higher velocity dispersion and attenuation for P waves than for SV *** oriented fractures substantially reduce the attenuation caused by the FB-WIFF and ES,particularly for the ES attenuation of SV *** a comparison with existing models in the limiting cases and previous experimental measurements,we validated our model.

Improving antioxidant activities of water-soluble lignin-carbohydrate complex isolated from wheat stalk through prolonging ball-milling pretreatment and homogeneous extraction

Water-soluble lignin-carbohydrate complex(LCC)rich in polysaccharides exhibits benign in vitro antioxidant activities and distinguishes high biocompatibility from lignin-rich LCC and ***,the antioxidant activity of water-soluble LCCs remains to be improved and its structure-antioxidant relationship is still ***,structurally diversified water-soluble LCCs were isolated under different ball-milling pretreatment durations(4,6,8 h),extraction pathways(ho-mogeneous and heterogeneous),and isolation routines(water extracts and residues after water extraction).Their structures were characterized by wet chemistry,chromatography and *** activities were evaluated by ferric reducing antioxidant power and 1,1-diphenyl-2-picrylhydrazyl radicals scavenging rate(RDPPH).Results show that altering ball-milling duration and isolation procedures cause varied structures and antioxidant activities of the water-soluble ***,prolonging ball-milling duration to 8 hours and homogeneous extrac-tion can enhance their antioxidant activity through releasing more phenolic structures and pro-moting the extraction of high-molecular-weight LCCs via reducing mass-transfer resistance,*** a result,the RDPPH of water-soluble LCCs reaches up to 97.35%,which is associated with the arabinan content with statistical significance(P<0.05).This study provides new insights into the structure-antioxidation relationship of herbaceous LCC as potential antioxidants.

BACKGROUND Multiple endocrine neoplasias(MENs)are a group of hereditary diseases invol-ving multiple endocrine glands,and their prevalence is *** type 1(MEN1)has diverse clinical manifestations,mainly involving the parathyroid glands,gastrointestinal tract,pancreas and pituitary gland,making it easy to miss the clinical *** suMMARY We present the case of a patient in whom MEN1 was detected early.A middle-aged male with recurrent abdominal pain and diarrhea was admitted to the *** tests at admission revealed hypercalcemia and hypophosphatemia,and emission computed tomography of the parathyroid glands revealed a hy-perfunctioning parathyroid *** findings suggested a duodenal bulge and *** endoscopy revealed a hypoechoic lesion in the duodenal *** blood tests revealed elevated levels of serum *** was performed,and pathological analysis of the surgical specimens revealed a parathyroid adenoma after parathyroidectomy and a neuroendocrine tumor after duodenal *** time from onset to the definitive diagnosis of MEN1 was only approximately 1 *** For patients who present with gastrointestinal symptoms accompanied by hyper-calcemia and hypophosphatemia,clinicians need to be alert to the possibility of MEN1.

Anti-OX40 Antibody Combined with HBc VLPs Delays Tumor Growth in a Mouse Colon Cancer Model

Background:Hepatocellular carcinoma(HCC)is a serious complication of hepatic vena cava Budd-Chiari syndrome(HVC-BCS)that significantly reduces the survival time of *** study aimed to analyze the prognostic factors influencing the survival of HVC-BCS patients with HCC and to develop a prognostic scoring ***:The clinical and follow-up data of 64 HVC-BCS patients with HCC who received invasive treatment at the First Affiliated Hospital of Zhengzhou University between January 2015 and December 2019 were retrospectively ***-Meier curves and log-rank tests were used to analyze the survival curve of patients and the difference in prognoses between the *** and multivariate Cox regression analyses were performed to analyze the influence of biochemical,tumor,and etiological characteristics on the total survival time of patients,and a new prognostic scoring system was developed according to the regression coefficients of the independent predictors in the statistical *** prediction efficiency was evaluated using the time-dependent receiver operating characteristics curve and concordance ***:Multivariate analysis showed that serum albumin level7 cm(HR=8.623,95%CI:3.771-19.715,Psurvival.A prognostic scoring system was developed according to the above-mentioned independent predictors,and patients were classified into grades A,B,C and *** differences in survival were found among the four ***:This study successfully developed a prognostic scoring system for HVC-BCS patients with HCC,which is helpful for clinical evaluation of patient prognosis.
