Intron-Targeted Intron-Exon Splice Conjunction (IT-ISJ) Marker and Its Application in Construction of Upland Cotton linkage Map

To develop a new DNA maker, which could be used in genetic diversity analysis and genetic map construction in plants, IT-ISJ （intron targeted intron-exon splice junction） primer combinations, which were designed according to the intronexon splice junction conserved sequences, were used to construct cotton genetic linkage map in the present study. 49 out of 704 IT-ISJ primer combinations showed polymorphism between upland cotton high quality cultivar Yumian 1 and multiple dominant gene line T586, and the polymorphic primer combinations accounted for 7.0% of total primer combinations. 49 IT-ISJ primer combinations were used to genotype 270 F2：7 recombinant inbred lines developed from （Yumian 1 × T586） F2, and 58 IT-ISJ loci were obtained. 58 IT-ISJ, together with 150 SSR and 8 morphological loci, were used to conduct linkage analysis, and a linkage map including 22 linkage groups and 113 loci （49 IT-ISJ, 62 SSR, and 2 morphological loci） was constructed. The linkage map covered 714.5 cM with an average interval of 6.3 cM between two markers, accounting for 16.1% of cotton genome. The present study demonstrated that the polymorphism of IT-ISJ marker is high, and it could be effectively applied in plant genetic map construction.

The safety of risers in hang-off states is a vital challenge in offshore oil and gas engineering.A new hang-off system installed on top of risers is proposed for improving the security of *** approach leads to a challenging problem:coupling the dynamics of risers with a new hang-off system combined with multiple structures and complex *** accurately analyze the dynamic responses of the coupled system,a coupled dynamic model is established based on the Euler-Bernoulli beam-column theory and penalty function method.A comprehensive analysis method is proposed for coupled dynamic analysis by combining the finite element method and the Newmarkβ*** analysis program is also developed in MATLAB for dynamic *** simulation results show that the dynamic performances of the risers at the top part are significantly improved by the new hang-off system,especially the novel design,which includes the centralizer and articulation *** bending moment and lateral deformation of the risers at the top part decrease,while the hang-off joint experiences a great bending moment at the bottom of the lateral restraint area which requires particular attention in design and *** platform navigation speed range under the safety limits of risers expands with the new hang-off system in use.

Dear Editor,We report a rare case of endogenous endophthalmitis caused by Candida albicans in a patient with chronic renal failure concomitant hemodialysis.A 71-year-old man with a history of hypertension and 2-year chronic renal failure on hemodialysis via a native arteriovenous fistula(AVF)was presented to the local hospital due to fever 3wk *** pathogens were detected through blood culture on admission,and his fever gradually subsided with intravenous ceftriaxone *** a week of fever,he experienced progressive blurred vision in both eyes,accompanied by pain and *** magnetic resonance imaging(MRI)of the brain revealed *** had not undergone any previous eye surgeries or trauma.

To ensure the long-term safety and stability of bridge pile foundations in permafrost regions,it is necessary to investigate the rheological effects on the pile tip and pile side bearing *** creep characteristics of the pile-frozen soil interface are critical for determining the long-term stability of permafrost pile *** study utilized a self-developed large stress-controlled shear apparatus to investigate the shear creep characteristics of the frozen silt-concrete interface,and examined the influence of freezing temperatures(−1,−2,and−5°C),contact surface roughness(0,0.60,0.75,and 1.15 mm),normal stress(50,100,and 150 kPa),and shear stress on the creep characteristics of the contact *** incorporating the contact surface’s creep behavior and development trends,we established a creep constitutive model for the frozen silt-concrete interface based on the Nishihara model,introducing nonlinear elements and a damage *** results revealed significant creep effects on the frozen silt-concrete interface under constant load,with creep displacement at approximately 2-15 times the instantaneous displacement and a failure creep displacement ranging from 6 to 8 *** different experimental conditions,the creep characteristics of the frozen silt-concrete interface varied.A larger roughness,lower freezing temperatures,and higher normal stresses resulted in a longer sample attenuation creep time,a lower steady-state creep rate,higher long-term creep strength,and stronger creep *** upon the Nishihara model,we considered the influence of shear stress and time on the viscoelastic viscosity coefficient and introduced a damage factor to the *** improved model effectively described the entire creep process of the frozen silt-concrete *** results provide theoretical support for the interaction between pile and soil in permafrost regions.
