Ferritic/martensitic(F/M)steel is widely used as a structural material in thermal and nuclear power ***,it is susceptible to intergranular damage,which is a critical issue,under service *** this study,to improve the resistance to intergranular damage of F/M steel,a thermomechanical process(TMP)was employed to achieve a grain boundary engineering(GBE)microstructure in F/M steel *** TMP,including cold-rolling thickness reduction of 6%,9%,and 12%,followed by austenitization at 1323 K for 40 min and tempering at 1053 K for 45 min,was applied to the as-received(AR)P92 *** prior austenite grain(PAG)size,prior austenite grain boundary character distribution(GBCD),and connectivity of prior austenite grain boundaries(PAGBs)were *** to the AR specimen,the PAG size did not change *** fraction of coincident site lattice boundaries(CSLBs,3≤Σ≤29)and Σ3^(n) boundaries along PAGBs decreased with increasing reduction ratio because the recrystallization fraction increased with increasing reduction *** PAGB connectivity of the 6%deformed specimen slightly deteriorated compared with that of the AR ***,potentiodynamic polarization studies revealed that the intergranular damage resistance of the studied steel could be improved by increasing the fraction of CSLBs along the PAGBs,indicating that the TMP,which involves low deformation,could enhance the intergranular damage resistance.

视网膜神经节细胞(RGCs)是视网膜视觉信号输出到大脑的终极神经元,可参与成像视觉(IFV)(图像形成)和非成像视觉(NIFV)(非图像形成)。视觉处理系统除了传递图像的视觉信息外,传入的光信号对人的生理活动和行为也会产生影响,称为NIFV。NIFV较少依赖于传统光感受器细胞产生的信号,而是由视网膜上一类特殊的视网膜感光神经节细胞(ipRGCs)来完成。ipRGCs是RGCs中一类能表达感光黑视蛋白的细胞,其轴突投射至特定核团,参与调控多种NIFV行为,从基础生理调节(如心率和瞳孔大小)到更复杂的行为调节(如昼夜节律),甚至更高层次的认知过程(如焦虑等情绪)。NIFV环路是对光的重要反应,ipRGCs在NIFV环路中起着至关重要的作用。本文就NIFV环路对生理活动和行为的调控作用进行综述,归纳ipRGCs投射核团与NIFV功能的关系,以期为临床医生提供更加全面的视觉认识。

shen-Jing as a Chinese Medicine Concept Might Be a Counterpart of Stem Cells in Regenerative Medicine

As the epitome of the modern regenerative medicine, stem cells were proposed in the basic sense no more than 200 years ago. However, the concept of "stem cells" existed long before the modern medical description. The hypothesis that all things, including our sentient body, were generated from a small origin was shared between Western and Chinese people. The ancient Chinese philosophers considered Jing(also known as essence) as the origin of life. In Chinese medicine(CM), Jing is mainly stored in Kidney(shen) and the so-called shen-Jing(Kidney essence). Here, we propose that shen-Jing is the CM term used to express the meaning of "origin and regeneration". This theoretical discovery has at least two applications. First, the actions underlying causing shen-Jing deficiency, such as excess sexual intercourse, chronic diseases, and aging, might damage the function of stem cells. Second, a large number of Chinese herbs with shen-Jing-nourishing efficacy had been proven to affect stem cell proliferation and differentiation. Therefore, if shen-Jing in CM is equivalent with stem cells in regenerative medicine, higher effective modulators for regulating stem-cell behaviors from Kidney-tonifying herbs would be expected.

T-A cloning takes advantage of the unpaired adenosyl residue added to the 3＇ terminus of amplified DNAs by Taq and other thermostable DNA polymerase and uses a Ilnearlzed plasmld vector with a protruding 3＇ thymldylate residue at each of Its 3＇ termini to clone polymerase chain reaction （PCR）-derived DNA fragments. It Is a simple, reliable, and efficient Ilgatlon-dependent cloning method for PCR products, but the drawback of variable cloning efficiency occurs during application. In the present work, the relationship between variable T-A cloning efficiency and the different 5＇ end nucleotlde base of primers used In PCR amplification was studied. The results showed that different cloning efficiency was obtained with different primer pairs containing A, T, C and G at the 5＇ terminus respectively. The data shows that when the 5＇ end base of primer pair was adenosyl, more white colonies could be obtained In cloning the corresponding PCR product In comparison with other bases. And the least white colonies were formed when using the primer pair with 5＇ cytldylate end. The gluanylate end primers resulted In almost the same cloning efficiency In the white colonies amount as the thymldylate end primer did, and this efficiency was much lower than that of adenosyl end primers. This presumably is a consequence of variability In 3＇dA addition to PCR products mediated by Taq polymerase. Our results offer instructions for primer design for researchers who choose T-A cloning to clone PCR products.

Objective: This study evaluated the safety and efficacy of an anti-epidermal growth factor receptor(EGFR) antibody(SCT200) and an anti-programmed cell death 1(PD-1) antibody(SCT-I10A) as third-line or subsequent therapies in patients with rat sarcoma viral oncogene(RAS)/v-raf murine sarcoma viral oncogene homolog B(BRAF) wild-type(wt) metastatic colorectal cancer(mCRC).Methods: We conducted a multicenter, open-label, phase Ib clinical trial. Patients with histologically confirmed RAS/BRAF wt m CRC with more than two lines of treatment were enrolled and treated with SCT-I10A and SCT200. The primary endpoints were the objective response rate(ORR) and safety. The secondary endpoints included disease control rate(DCR), progression-free survival(PFS), and overall survival(OS).Results: Twenty-one patients were enrolled in the study through January 28, 2023. The ORR was 28.57% and the DCR was 85.71%(18/21). The median PFS and OS were 4.14 and 12.84 months, respectively. The treatment-related adverse events(TRAEs) were tolerable. Moreover, compared with the monotherapy cohort from our previous phase I study evaluating SCT200 for RAS/BRAF wt m CRC in a third-line setting, no significant improvements in PFS and OS were observed in the combination ***: SCT200 combined with SCT-I10A demonstrated promising efficacy in previously treated RAS/BRAF wt m CRC patients with an acceptable safety profile. Further head-to-head studies with larger sample sizes are needed to validate whether the efficacy and safety of combined anti-EGFR and anti-PD-1 therapy are superior to anti-EGFR monotherapy in the third-line setting.(Registration No. NCT04229537).

目的观察新型光敏蛋白PsCatCh2.0转染至视网膜色素变性模型rd1小鼠视网膜1年后视网膜神经节细胞(RGC)对光反应、视网膜炎症及细胞凋亡情况。方法雄性rd1小鼠24只随机均分为rd1实验组、rd1对照组,各12只。rd1实验组小鼠鼻侧角巩膜缘下1 mm处玻璃体腔注射重组腺相关病毒(rAAV)2/2-巨细胞病毒启动子(CMV)-PsCatCh2.0-增强型绿色荧光蛋白(EGFP)1.5μl,2周后颞侧角巩膜缘下1 mm处再次注射相同剂量的重组病毒。注射后1年,用膜片钳技术记录表达PsCatCh2.0的RGC对光反应;行免疫荧光染色评估PsCatCh2.0在视网膜中的表达情况;行视网膜铺片染色评估重组病毒的转染效率,并计数RGC;使用苏木精-伊红染色评估内层视网膜厚度;采用蛋白免疫印迹法检测视网膜中核因子(NF)-κB p65的蛋白表达;采用实时荧光定量聚合酶链反应检测各组小鼠视网膜中肿瘤坏死因子(TNF)-α、白细胞介素(IL)-6、B淋巴细胞瘤-2相关X蛋白(Bax)mRNA相对表达量;采用原位末端标记法观察各组小鼠视网膜细胞凋亡情况。组间比较采用单因素方差分析。结果注射重组病毒后1年,表达PsCatCh2.0的RGC产生的电流约为30 pA。PsCatCh2.0-EGFP与RGC呈共定位表达;少量与无长突细胞共定位,几乎不与水平细胞、双极细胞共定位。rd1实验组、rd1对照组小鼠RGC密度、内层视网膜厚度比较,差异均无统计学意义(F=14.35、0.05,P>0.05)。rd1实验组小鼠视网膜NF-κB p65蛋白表达量以及TNF-α、IL-6 mRNA表达均低于rd1对照组,差异均有统计学意义(F=4.61、5.91、5.78,P<0.05)。rd1实验组小鼠视网膜中呈红色荧光的凋亡细胞数量少于rd1对照组,Bax mRNA表达低于rd1对照组,差异有统计学意义(F=7.52,P<0.01)。结论玻璃体腔注射rAAV2/2-CMV-PsCatCh2.0-EGFP后1年,表达PsCatCh2.0的RGC仍可产生光电流;长期转染表达PsCatCh2.0对RGC无明显细胞毒性作用,也不增加视网膜的炎性反应。

目的探讨轻度创伤性颅脑损伤患者(mTBI)谵妄的发生率及危险因素。方法前瞻性纳入2021年1月—2023年12月在广西壮族自治区人民医院急诊科住院的107例根据《颅脑创伤临床救治指南》标准诊断为mTBI的患者,患者入院后72 h内每24h采用意识模糊评估法中文修正版(CAM-CR)进行谵妄评估,同时收集患者年龄、性别、合并疾病、入院血清钾、血清钠、血肌酐、血清白蛋白、入院24 h匹兹堡睡眠质量指数(PSQI)等临床指标。根据患者有无谵妄发生分为谵妄组和正常组,比较两组患者的相关临床指标,采用单因素分析和多因素Logistic回归分析筛选患者发生谵妄的危险因素。结果 107例mTBI患者中有41例在住院期间发生谵妄,发生率为38.32%(95%CI:22%~47.7%),谵妄组患者年龄、合并冠心病、高血压病的比例和入院24 h PSQI指数、WBC、CRP数值高于正常组,血清钠、血清钾、血清白蛋白数值低于正常组,两组比较差异有统计学意义(P均<0.05)。Logistic多因素回归分析提示年龄(OR=1.208,95%CI:1.089~1.341)、血清钠(OR=0.758,95%CI:0.624~0.920)、血清白蛋白(OR=0.755,95%CI:0.586~0.971)、入院24 h PSQI指数(OR=2.725,95%CI:1.237~6.002)是mTBI患者住院期间发生谵妄的独立影响因素(P均<0.05)。结论年龄增加及PSQI指数增加与mTBI患者住院期间发生谵妄风险增加显著相关,血清钠浓度和血清白蛋白水平增加与mTBI患者住院期间发生谵妄风险降低显著相关。
