A practical guide for fast implementation of SNARE-mediated liposome fusion

Soluble N-ethylmaleimide-sensitive factor attachment protein receptor(SNAER) family proteins are the engines of most intra-cellular and exocytotic membrane fusion pathways(Jahn and Scheller 2006).Over the past two decades, in-vitro liposome fusion has been proven to be a powerful tool to reconstruct physiological SNARE-mediated membrane fusion processes(Liu et al. 2017). The reconstitution of the membrane fusion process not only provides direct evidence of the capability of the cognate SNARE complex in driving membrane fusion but also allows researchers to study the functional mechanisms of regulatory proteins in related pathways(Wickner and Rizo 2017). Heretofore, a variety of delicate methods for in-vitro SNARE-mediated liposome fusion have been established(Bao et al. 2018;Diao et al. 2012; Duzgunes 2003; Gong et al. 2015; Heo et al. 2021; Kiessling et al. 2015; Kreye et al.2008; Kyoung et al. 2013; Liu et al. 2017; Scott et al. 2003). Although technological advances have made reconstitution more physiologically relevant, increasingly elaborate experimental procedures, instruments, and data processing algorithms nevertheless hinder the non-experts from setting up basic SNARE-mediated liposome fusion assays. Here, we describe a low-cost, timesaving, and easy-to-handle protocol to set up a foundational in-vitro SNARE-mediated liposome fusion assay based on our previous publications(Liu et al. 2023; Wang and Ma 2022). The protocol can be readily adapted to assess various types of SNARE-mediated membrane fusion and the actions of fusion regulators by using appropriate alternative additives(e.g., proteins, macromolecules, chemicals, etc.). The total time required for one round of the assay is typically two days and could be extremely compressed into one day.

Detection of Pb in Tieguanyin tea and ash by laser-induced breakdown spectroscopy

In this work, laser-induced breakdown spectroscopy(LIBS) was applied for the detection of Pb in Tieguanyin tea and ash. Firstly, the Tieguanyin tea and ash containing Pb were prepared, and the difference of intensities of Pb I spectral lines before and after the ashing treatment was studied. It was found that the intensities of Pb I lines increased by 30 times and the standard deviation of background signal decreased by 41% after the ashing treatment. Therefore, the enrichment of Pb element by ashing treatment was used to detect Pb in tea with high sensitivity. Then, the calibration curve of Pb was established using spectral lines without self-absorption, and the determination coefficient(R^(2)) for the linear fitting of calibration curve was 0.979 9. Finally, it was found that the limit of detection of Pb was 233.8 ppb. Compared with the results of other works which detect Pb directly, the enrichment of Pb by ashing treatment improved the detection sensitivity of Pb by about 200 times. In addition, this method can be applied to the high sensitivity detection of other heavy metals, such as Cr, Cd, Hg, etc in plants, Chinese herbal medicine, flour, rice, coal and other solid materials.

Quasi-2D weak lensing cosmological constraints using the PDF-SYM method

Cosmic shear statistics,such as the two-point correlation function(2PCF),can be evaluated with the PDF-SYM method instead of the traditional weighted-sum approach.It makes use of the full PDF information of the shear estimators,and does not require weightings on the shear estimators,which can in principle introduce additional systematic biases.This work presents our constraints on S8and?mfrom the shear-shear correlations using the PDF-SYM method.The data we use is from the z-band images of the Dark Energy Camera Legacy Survey(DECa LS),which covers about 10000 deg2with more than 100 million galaxies.The shear catalog is produced by the Fourier Quad method,and well tested on the real data itself with the field-distortion effect.Our main approach is called quasi-2D as we do use the photo-z information of each individual galaxy,but without dividing the galaxies into redshift bins.We mainly use galaxy pairs within the redshift interval between 0.2 and 1.3,and the angular range from 4.7 to 180 arcmin.Our analysis yields S8=0.762±0.026 and?m=0.234±0.075,with the baryon effects and the intrinsic alignments included.The results are robust against redshift uncertainties.We check the consistency of our results by deriving the cosmological constraints from auto-correlations ofγ1andγ2separately,and find that they are consistent with each other,but the constraints from theγ1component are much weaker than that fromγ2.It implies a much worse data quality ofγ1,which is likely due to additional shear uncertainties caused by CCD electronics(according to the survey strategy of DECa LS).We also perform a pure 2D analysis,which gives S8=0.81_(-0.04)~(+0.03)and?m=0.25_(-0.05)~(+0.06).Our findings demonstrate the potential of the PDF-SYM method for precision cosmology.

Nanomedicine delivery technology plays an important role in modern medicine and has shown good therapeutic effects in scientific research.Polysaccharides have the characteristics of wide sources,excellent biocompatibility,and non-toxicity.In addition,there are multifunctional groups on the main chain of polysaccharides,which can be surface-modified or functionalized to have targeting ability through specific sugar parts.Amphiphilic polysaccharide micelles with good biocompatibility,degradability,high safety,easy structural modification,and special core-shell structure are regarded as ideal carriers for nanomedicines.Therefore,this review is focused on the hydrophobic modification designs of polysaccharides,the preparation methods and characteristics of micelles,and the applications of amphiphilic polysaccharide micelles in the field of biomedicine.It is expected to provide some ideas and inspiration for the design of polysaccharide drug carriers.

Ex vivo liver resection and auto-transplantation as an alternative for the treatment of liver malignancies: Progress and challenges

Hepatectomy is still the major curative treatment for patients with liver malignancies.However,it is still a big challenge to remove the tumors in the central posterior area,especially if their location involves the retrohepatic inferior vena cava and hepatic veins.Ex vivo liver resection and auto-transplantation(ELRA),a hybrid technique of the traditional liver resection and transplantation,has brought new hope to these patients and therefore becomes a valid alternative to liver transplantation.Due to its technical difficulty,ELRA is still concentrated in a few hepatobiliary centers that have experienced surgeons in both liver resection and liver transplantation.The efficacy and safety of this technique has already been demonstrated in the treatment of benign liver diseases,especially in the advanced alveolar echinococcosis.Recently,the application of ELRA for liver malignances has gained more attention.However,standardization of clinical practice norms and international consensus are still lacking.The prognostic impact in these oncologic patients also needs further evaluation.In this review,we summarized the principles and recent progresses on ELRA.

Diverse interactions of five core type Ⅲ effectors from Ralstonia solanacearum with plants

Ralstonia solanacearum is a widespread plant bacterial pathogen that can launch a range of type Ⅲ effectors(T3Es)to cause disease.In this study,we isolate a pathogenic R.solanacearum strain named P380 from tomato rhizosphere.Five out of 12 core T3Es of strain P380 are introduced into Pseudomonas syringae DC3000D36E separately to determine their functions in interacting with plants.DC3000D36E that harbors each effector suppresses FliC-triggered Pti5 and ACRE31 expression,ROS burst,and callose deposition.RipAE,RipU,and RipW elicit cell death as well as upregulate the MAPK cascades in Nicotiana benthamiana.The derivatives RipC1^(△DDXDX(T/V))and RipW^(△DDKXXQ)but not RipAE^(K310R) fail to suppress ROS burst.Moreover,RipAE^(K310R) and RipW^(△DDKXXQ) retain the cell death elicitation ability.RipAE and RipW are associated with salicylic acid and jasmonic acid pathways,respectively.RipAE and RipAQ significantly promote the propagation of DC3000D36E in plants.The five core T3Es localize in diverse subcellular organelles of nucleus,plasma membrane,endoplasmic reticulum,and Golgi network.The suppressor of G2 allele of Skp1 is required for RipAE but not RipU-triggered cell death in N.benthamiana.These results indicate that the core T3Es in R.solanacearum play diverse roles in plantpathogen interactions.

Highly adaptive and broadband triboelectric energy harvester with stretching silicone rubber strip for variable harmonic frequency vibration

An enormous number of wireless sensing nodes(WSNs)are of great significance for the Internet of Things(IoT).It is tremendously prospective to realize the in-situ power supply of WSNs by harvesting unutilized mechanical vibration energy.A harmonic silicone rubber triboelectric nanogenerator(HSR-TENG)is developed focusing on ubiquitous constant working frequency machinery.The unique design of the strip serving as a flexible resonator realizes both soft contact and high and broadband output.The significant factors influencing the 1^(st)-order vibration mode of the strip are developed for realizing the harmonic frequency adaptation to external vibration.The surface treatment of the strip improves the output performance of HSR-TENG by 49.1%as well as eliminates the adhesion effect.The HSR-TENG is able to achieve a voltage output bandwidth of 19 Hz under a vibration strength of 3.0,showing its broadband capability.The peak power density of 153.9 W/m^(3)is achieved and 12×0.5 W light-emitting diodes(LEDs)are successfully illuminated by the HSR-TENG.It can continuously power a temperature sensor by harvesting the actual compressor vibration energy.In brief,the HSR-TENG provides a promising way for constant frequency vibration energy harvesting,so as to achieve in-situ power supply for the WSNs in the vicinity.

Lactate is present at a high level in the microenvironment of mammalian preimplantation embryos in vivo and in vitro. However, its role in preimplantation development is unclear. Here, we report that lactate is highly enriched in the nuclei of early embryos when major zygotic genome activation(ZGA) occurs in humans and mice. The inhibition of its production and uptake results in developmental arrest at the 2-cell stage, major ZGA failure, and loss of lactate-derived H3K18lac, which could be rescued by the addition of Lac-Co A and recapitulated by overexpression of H3K18R mutation. By profiling the landscape of H3K18lac during mouse preimplantation development, we show that H3K18lac is enriched on the promoter regions of most major ZGA genes and correlates with their expressions. In humans, H3K18lac is also enriched in ZGA markers and temporally concomitant with their expressions. Taken together, we profile the landscapes of H3K18lac in mouse and human preimplantation embryos, and demonstrate the important role for H3K18lac in major ZGA, showing that a conserved metabolic mechanism underlies preimplantation development of mammalian embryos.
