Analysis of Occurrence and Mixed Infection of Sugarcane Bacilliform Virus Disease in Hainan Sugarcane-growing Area

[Objectives]The paper was to explore the occurrence and mixed infection of sugarcane bacilliform virus disease in Hainan sugarcane-growing area.[Methods]A total of 348 sugarcane leaf samples were collected from 7 sugarcane-growing areas in Hainan *** detection of sugarcane bacilliform virus(SCBV)was carried out by PCR using specific primers.[Results]SCBV was detected in 244 out of 348 sugarcane samples,with an average detection rate of 70.11%.The highest detection rate was 76.66%in the Danzhou sugarcane-growing area,while the lowest was 57.14%in the Baisha sugarcane-growing *** SCBV-positive samples were subjected to testing for SCYLV,SCSMV,SrMV,and SCMV,*** results indicated that 106 out of 244 positive samples exhibited a single infection with SCBV,while 138 samples exhibited mixed infections with SCBV and other sugarcane *** proportion of mixed infections among the SCBV-positive samples was as high as 56.56%.Among the various types of mixed infections,two-virus and three-virus mixed infections were the most prevalent.[Conclusions]SCBV has emerged as a significant threat to the secure production of sugarcane in the Hainan sugarcane-growing *** presents an explosive infection in the Hainan sugarcane-growing region and frequently combines with other sugarcane viruses to infect *** findings of this study will provide a theoretical foundation for the prevention and control of sugarcane bacilliform virus disease.

特发性耳鸣因其机制尚不明确,缺乏特效的治疗方法,是耳鼻喉科常见难题之一。近年来,随着功能磁共振技术快速发展,越来越多的研究认为,耳鸣的病理机制不仅涉及听觉系统,更是听觉、默认、执行控制、突显及注意网络等多个重叠大脑网络之间动态交互的结果。我们综述耳鸣相关大脑功能网络的功能磁共振成像研究进展,为进一步探索耳鸣的病理机制夯实基础,以期为临床治疗特发性耳鸣提供新的作用靶点及干预措施。

采用文献计量和知识图谱方法分析近10年火针临床随机对照试验(RCT)文献的研究热点、前沿与趋势。检索中国期刊全文数据库(CNKI)、万方数据知识服务平台(Wanfang)、维普资讯中文期刊服务平台(VIP)、中国生物医学文献数据库(SinoMed)、PubMed、Web of Science(WOS)共6个中英文数据库中有关火针的RCT研究文献,采用CiteSpaceⅤ6.1.R6及VOSviewerⅤ1.6.18软件进行合作网络、关键词共现、关键词聚类、关键词时间线、关键词突现等分析,并绘制可视化知识图谱。共纳入1 973篇中文文献、3篇英文文献;发文量前3位的机构分别为广州中医药大学、黑龙江中医药大学及首都医科大学附属北京中医医院;火针多与针刺、拔罐及放血疗法联合应用治疗痤疮、白癜风、腰椎间盘突出症、带状疱疹、中风后遗症、面神经麻痹、膝关节骨关节炎等;研究前沿包括火针与其他疗法的联合应用、临床机制研究及疗效评价指标研究。未来应拓展优势病种,优化研究设计,加强团队之间的合作,深入开展高水平的临床研究。

中风软瘫以中风后患者身体运动功能异常,肌肉张力下降为主要表现,是中风后较为常见的病理状态,软瘫时间越长预后越差。中医理论认为,中风主因“虚、风、火、痰、瘀、气”而发病,当邪气搏结、阻于经络则使气血凝滞、经筋损伤、肌肉痿废,导致中风软瘫,因此需立以通经柔筋为其主要治法。火针治疗中风软瘫具有通经温阳、柔筋止痛的作用,可温阳散寒、充养经脉经气、松解筋脉结节、推动气血运行、濡养四肢百骸。火针疗法可促进神经通路的恢复,加强局部新陈代谢,改善局部肌张力,从而恢复肢体功能。火针治疗中风后软瘫的高质量临床研究、选穴规律、规范化操作手法等有待进一步深化。

Isolation,Screening and Identification of Antagonistic Actinomycetes against Ustilago scitaminea Syd

[Objectives] This study was conducted to obtain actinomycetes strains having antagonistic effect on Ustilago scitaminea Syd.[Methods] At first, actinomycetes strains were isolated from 22 soil samples in Hainan sugarcane regions. Then, antagonistic actinomycetes against U. scitaminea were screened by confrontation culture. Finally, the taxonomic status of antagonistic actinomycetes was determined using 16S rDNA.[Results] From the 22 samples, 984 actinomycetes strains were isolated. From all the isolated strains, 23 antagonistic actinomycetes strains were obtained through primary screening, and one strains with better antagonistic effect was then obtained through secondary screening, and designated FAS. 16S rDNA identification showed that strain FAS shared 99% sequence similarity with Streptomyces cealestis US24. A phylogenetic tree was built with MAGE 7.0 software, and the results showed that strain FAS had the shortest genetic distance with S. caelestis US24. Therefore, the actinomycetes FAS was determined as S. caelestis .[Conclusions] This study provides a new biocontrol method for the biological control of sugarcane smut, thereby ensuring sustainable development of sugarcane industry and sugar industry.

Optimization of Streptomyces caelestis Culture Conditions and Preliminarily Determination of Its Antifungal Components

Streptomyces caelestis FAS is an actinomyces strain isolated with Ustilago scitaminea as an indicator. With GAUZE s medium as the basic medium, carbon source and nitrogen source in the medium and ventilation volume were optimized, and the antifungal substances produced by S. caelestis FAS were preliminarily determined through organic solvent extraction and ammonium sulfate precipitation. The results showed that the antifungal components produced by S. caelestis FAS were proteins, and the optimal culture conditions were using corn flour as carbon source and KNO 3 as nitrogen source and culturing in 250 ml triangular flask added with 70 ml of culture medium. The crude protein obtained under these culture conditions had the best antifungal effect with an inhibition zone diameter of 25 mm. This study will provide a scientific basis for further study on the antifungal mechanism and application of S. caelestis FAS.

Study on Wide Antifungal Property of Bacillus subtilis HAS

Bacillus subtilis HAS is a biocontrol strain with better inhibitory effect on Sporisorium scitaminea Sydow. In order to further understand its action range,with fungal pathogens in sugarcane production as targets,confrontation tests and tests of inhibitory effect of secondary metabolites on growth of pathogens were carried out. The results showed that B. subtilis HAS not only had better inhibitory effect on S. Scitaminea,but also inhibited the growth of various fungal pathogens in sugarcane production. The inhibitory effect was the best on Ceratocystis adiposum,Thielaviopsis paradoxa,Fusarium moniliforme and Colletotrichum fuleatum,which was reflected by the radius of inhibitory zone over 10 mm. The inhibitory rates of the secondary metabolites were about 70%. It indicates that B. subtilis HAS has wide antifungal property,providing reference for further application of this strain.

Identification of A Strain HN-1 against Banana Wilt Disease and Determination of Its Antagonism

[Objective] The paper was to identify strain HN-1 against banana wilt disease and to determine its antagonism. [Method] The strain HN-1 was ob- tained from the soil in fields heavily infected by Fusarium oxysporum f. sp. cubense （FOC）. Antagonism of the strain against F. oxysporum was tested via dual-cul- ture and inhibition test on spore germination. [Result] HN-1 effectively inhibited mycelial growth and spore germination of F. oxysporum, Strain HN-1 was identi- fied as BrevibaciUus brevis according to its characteristics in morphology, physiology and biochemistry and its 16S rDNA sequence. The strain showed high inhibition effect on 15 species of fungal pathogens in the dual-culture trials with fungal pathogens. [ Conclusion] The study provides theoretical basis for application of strain HN-1 in agricultural fields.

Colonization Ability of Bacillus subtilis HAS in Sugarcane

Bacillus subtilis HAS had good control effects on sugarcane smut in the field. Colonization dynamics of B. subtilis HAS were studied through biotics marker and pot culture, to understand the bio-control mechanism of the strain and provide experimental basis for commercialization. The results showed that *** HAS had strong colonization ability, which could be colonized in sugarcane root, stem, leaf and rhizosphere for long time, but the colonization levels were different. The colonization density of *** HAS remained 103 CFU/g soil in the rhizosphere and 102 CFU/g （fresh weight） in the root after inoculation of 60 d. The colonization in the rhizosphere decreased first, then increased and decreased again, and finally stabilized. The colonization in the root first increased then decreased. Meantime, colonization status in stems and leaves showed that the colonization density in stems and leaves was 10 CFU/g （fresh weight）, indicating that the colonization level was not high in stem and leaf.

对甘蔗Southern杂交体系的优化,旨为转基因甘蔗Southern杂交鉴定分析提供参考。以转基因甘蔗为材料,就探针不同标记方法的比较、甘蔗基因组DNA的提取、基因组DNA酶切量、酶切时间及杂交过程等方面,对地高辛标记的Southern杂交技术进行了优化研究。结果表明,改良的CTAB法提取的甘蔗DNA能满足后期实验的要求;PCR法标记探针的效率较随机引物法标记探针的效率高,更适合用于Southern杂交;40μg的DNA在400μL酶切体系中,酶切10 h可获得良好的酶切效果;杂交温度40℃,杂交18 h,可获得清晰的杂交条带。
