Identification of C9-C11 unsaturated aldehydes as prediction markers of growth and feed intake for non-ruminant animals fed oxidized soybean oil

Background: The benefits of using the oxidized oils from rendering and recycling as an economic source of lipids and energy in animal feed always coexist with the concerns that diverse degradation products in these oxidized oils can negatively affect animal health and performance. Therefore, the quality markers that predict growth performance could be useful when feeding oxidized oils to non-ruminants. However, the correlations between growth performance and chemical profiles of oxidized oils have not been well examined. In this study, six thermally oxidized soybean oils(OSOs) with a wide range of quality measures were prepared under different processing temperatures and processing durations, including 45 °C-336 h;67.5 °C-168 h;90 °C-84 h;135 °C-42 h;180 °C-21 h;and225 °C-10.5 h. Broilers and nursery pigs were randomly assigned to diets containing either unheated control soybean oil or one of six OSOs. Animal performance was determined by measuring body weight gain, feed intake,and gain to feed ratio. The chemical profiles of OSOs were first evaluated by common indicative tests, including peroxide value, thiobarbituric acid reactive substances, p-anisidine value, free fatty acids, oxidized fatty acids,unsaponifiable matter, insoluble impurities, and moisture, and then analyzed by the liquid chromatography-mass spectrometry-based chemometric ***: Among common quality indicators, p-anisidine value(An V), which reflects the level of carbonyl compounds,had the greatest inverse correlation with the growth performance of both broilers and pigs, followed by free fatty acids and oxidized fatty acids. The reintroduction of fire to landscapes where it was once common is considered a priority to restore historical forest dynamics,including reducing tree density and decreasing levels of woody biomass on the forest ***,reintroducing fire causes tree mortality that can have unintended ecological outcomes related to woody biomass,with potential impacts to fuel accumulation,carbon sequestration,subsequent fire severity,and forest *** this study,we examine the interplay between fire and carbon dynamics by asking how reintroduced fire impacts fuel accumulation,carbon sequestration,and subsequent fire severity *** pre-fire,and continuing 6 years post-fire,we tracked all live,dead,and fallen trees≥1 cm in diameter and mapped all pieces of deadwood(downed woody debris)originating from tree boles≥10 cm diameter and≥1 m in length in 25.6 ha of an Abies concolor/Pinus lambertiana forest in the central Sierra Nevada,California,*** also tracked surface fuels along 2240 m of planar transects pre-fire,immediately post-fire,and 6 years *** years after moderate-severity fire,deadwood≥10 cm diameter was 73 mg ha^(−1),comprised of 32 mg ha^(−1) that persisted through fire and 41 mg ha^(−1) of newly fallen wood(compared to 72 mg ha^(−1) pre-fire).Woody surface fuel loading was spatially heterogeneous,with mass varying almost four orders of magnitude at the scale of 20 m×20 m quadrats(minimum,0.1 mg ha^(−1);mean,73 mg ha^(−1);maximum,497 mg ha^(−1)).Wood from large-diameter trees(≥60 cm diameter)comprised 57%of surface fuel in 2019,but was 75%of snag biomass,indicating high contributions to current and future fuel *** of fire does not consume all large-diameter fuel and generates high levels of surface fuels≥10 cm diameter within 6 *** fires are needed to reduce surface fuel loading.

Intron splicing in eukaryotic organisms requires the interactions of five snRNAs and numerous different proteins in the spliceosome. Although the molecular mechanism behind splicing has been well studied, relatively little is known about regulation of expression for these splicing factor proteins. One of these proteins is the evolutionarily-conserved Drosophila RNP-4F splicing assembly factor. This protein is transcribed from a single gene into two developmentally regulated mRNAs that differ in their 5’-UTR structure. In the longer isoform, known to be abundant in the developing fly central nervous system, a conserved retained intron which folds into a stem-loop has been implicated in expression control of the mRNA. Here, we describe construction and utilization of several new rnp-4f gene expression study vectors using a GFP reporter in the ΦC31 system. The results confirm our previous observation that presence of the regulatory stem-loop enhances RNP-4F protein expression. However, in that study, the enhancement factor protein was not identified. We show here that overexpression of the RNP-4F transgene compared to the control results in additional translation, as indicated by the GFP reporter in the fluorescent images. These results are interpreted to show that RNP-4F protein acts back on its own mRNA 5’-UTR regulatory region via a feedback pathway to enhance protein synthesis in the developing fly central nervous system. A model is proposed to explain the molecular mechanism behind rnp-4f gene expression control.

The two ubiquitous, outside the retina, G protein-coupled receptor(GPCR)adapter proteins, β-arrestin-1 and-2(also known as arrestin-2 and-3,respectively), have three major functions in cells: GPCR desensitization, i.e.,receptor decoupling from G-proteins; GPCR internalization via clathrin-coated pits; and signal transduction independently of or in parallel to G-proteins. Bothβ-arrestins are expressed in the heart and regulate a large number of cardiac GPCRs. The latter constitute the single most commonly targeted receptor class by Food and Drug Administration-approved cardiovascular drugs, with about onethird of all currently used in the clinic medications affecting GPCR *** β-arrestin-1 and-2 play important roles in signaling and function of several GPCRs, in particular of adrenergic receptors and angiotensin II type 1 receptors,in cardiac myocytes, they have been a major focus of cardiac biology research in recent years. Perhaps the most significant realization coming out of their studies is that these two GPCR adapter proteins, initially thought of as functionally interchangeable, actually exert diametrically opposite effects in the mammalian myocardium. Specifically, the most abundant of the two β-arrestin-1 exerts overall detrimental effects on the heart, such as negative inotropy and promotion of adverse remodeling post-myocardial infarction(mI). In contrast, β-arrestin-2 is overall beneficial for the myocardium, as it has anti-apoptotic and antiinflammatory effects that result in attenuation of post-mI adverse remodeling,while promoting cardiac contractile function. Thus, design of novel cardiac GPCRligands that preferentially activate β-arrestin-2 over β-arrestin-1 has the potential of generating novel cardiovascular therapeutics for heart failure and other heart diseases.
