Risk Factors of Depression Screened by Two-Sample Mendelian Randomization Analysis:A Systematic Review

Objective Recombinase-aided polymerase chain reaction(RAP)is a sensitive,single-tube,two-stage nucleic acid amplification *** study aimed to develop an assay that can be used for the early diagnosis of three types of bacteremia caused by Staphylococcus aureus(SA),Pseudomonas aeruginosa(PA),and Acinetobacter baumannii(AB)in the bloodstream based on recombinant human mannanbinding lectin protein(M1 protein)-conjugated magnetic bead(M1 bead)enrichment of pathogens combined with *** Recombinant plasmids were used to evaluate the assay *** blood influenza bacteria were used for the specific *** and clinical plasma samples were enriched with M1 beads and then subjected to multiple recombinase-aided PCR(M-RAP)and quantitative PCR(qPCR)*** analysis was used to evaluate the consistency between the two *** The M-RAP method had sensitivity rates of 1,10,and 1 copies/μL for the detection of SA,PA,and AB plasmids,respectively,without cross-reaction to other bacterial *** M-RAP assay obtained results for<10 CFU/mL pathogens in the blood within 4 h,with higher sensitivity than qPCR.M-RAP and qPCR for SA,PA,and AB yielded Kappa values of 0.839,0.815,and 0.856,respectively(P<0.05).Conclusion An M-RAP assay for SA,PA,and AB in blood samples utilizing M1 bead enrichment has been developed and can be potentially used for the early detection of bacteremia.

目的探讨T淋巴细胞在人乳头瘤病毒(HPV)感染小鼠模型生殖道黏膜上皮细胞免疫反应中的作用。方法采用随机数字法将C57BL/6小鼠分为对照组、空载组、模型A组、模型B组,各12只,构建HPV16感染小鼠模型,进行小鼠生殖道分泌物、外周血、黏膜组织中炎性细胞分类计数;观察生殖道黏膜上皮病理状态;流式细胞仪检测外周血T淋巴细胞激活水平;实时荧光定量聚合酶链反应(RT-PCR)检测黏膜组织CD_4~+T淋巴细胞转录因子T盒子21转录因子(T-bet)、GATA结合蛋白3(GATA3)、维A酸孤儿核受体γt(ROR-γt)、叉头样转录因子P3(FOXP3),以及炎性因子白细胞介素(IL)-6、IL-10、肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)、干扰素-γ(IFN-γ)的mRNA转录水平、蛋白相对表达水平。结果与对照组、空载组比较,模型A组、模型B组生殖道分泌物、外周血、黏膜组织中的炎性细胞数量高,外周血中CD_(3)^(+)T淋巴细胞亚群激活水平降低,CD_(4)^(+)、CD_(8)^(+)T淋巴细胞亚群激活水平升高,CD_(4)^(+)/CD_(8)^(+)比值降低,黏膜组织中CD_(4)^(+)T细胞亚群转录因子T-bet、GATA3、RORγt、FOXP3的mRNA转录水平高,以及T细胞亚群特异细胞因子IFN-γ、IL-4、IL-17A、FOXP3表达水平高,黏膜组织中炎性因子IL-6、TNF-α、IL-10、TGF-β、IFN-γ的mRNA转录水平、蛋白相对表达水平高,且模型B组高于模型A组(P<0.05)。结论 T淋巴细胞亚群通过不同的激活水平参与机体对HPV感染的免疫反应,其激活水平随HPV拷贝数而改变;其中CD_(4)^(+)在免疫应答的调节和效应中起着重要作用,通过调节炎性细胞因子的分泌达到抗病毒的作用。

宫颈病变与阴道灌洗液炎性因子的关系及宫颈癌变列线图预测模型构建

目的分析不同宫颈病变患者阴道灌洗液辅助性T(Th)细胞与调节性T(Treg)细胞细胞因子差异性,并建立宫颈癌变列线图预测模型。方法选取2022年1月一2022年12月于邢台市人民医院就诊的高危型人乳头瘤病毒(HR-HPV)16感染患者200例为研究对象,根据宫颈癌变情况分为宫颈癌组35例和非宫颈癌组165例,其中非宫颈癌组根据宫颈病变程度分为无病变组52例、低级别鳞状上皮内病变(LSIL)组58例、高级别鳞状上皮内病变(HSIL)组55例。另外选取同期健康体检者100名为对照组。比较各组受试者阴道灌洗液Th与Treg细胞因子表达水平情况。应用R软件建立预测宫颈癌变的列线图模型,采用受试者操作特征(ROC)曲线分析该模型预测宫颈癌变的效能。结果5组受试者阴道灌洗液肿瘤坏死因子-α(TNF-α)、转化生长因子-β(TGF-β)、干扰素(IFN-)、白细胞介素(IL)-2、IL-6、IL-10、IL-17表达水平差异有统计学意义(P<0.05),且随着疾病进展,IL 6、IL-10、IL-17、TGF-β呈增加趋势,而TNF-α、IFN-γ、IL-2呈降低趋势(P<0.05)。宫颈癌组IL-6、IL-10、IL-17、TGF-β高于非宫颈癌组,TNF-α、IFN-γ、IL-2水平低于非宫颈癌组(P<0.05)。ROC曲线分析显示,列线图模型预测宫颈癌变的曲线下面积(AUC)为0.818(95%CI0.736~0.900),敏感度、特异度分别为77.14%、78.79%。结论阴道灌洗液Th与Treg细胞因子与宫颈癌变密切关联,以各细胞因子建立的列线图模型具有良好的宫颈癌变风险预测效能。

Follicular fluid levels of prostaglandin E2 and the effect of prostaglandin E2 on steroidogenesis in granulosa-lutein cells in women with moderate and severe endometriosis undergoing in vitro fertilization and embryo transfer

Background The mechanisms of endometriosis with infertility have not been fully studied. The present study aimed to assess the follicular fluid （FF） levels of prostaglandin E2 （PGE2）, which plays a critical role within the ovary, and to investigate the effect of PGE2 on steroidogenesis in granulosa-lutein cells （GLCs） from women with and without endometriosis. Methods Thirty-three women with laparoscopically documented endometriosis and 40 controls undergoing in vitro fertilization （IVF） were studied. We assayed the concentrations of PGE2 in FF, the production of E2 and progesterone in FF and in culture medium, and the expression of steroidogenic acute regulatory protein （STAR） and CYP19A1 in GLCs with the intervention of PGE2. Results PGE2 and progesterone concentrations were increased and displayed positive correlation in endometriotic FE PGE2 induced the expression of STAR and the production of progesterone in GLCs from women with endometriosis, and the expression of STAR and the production of progesterone were increased in GLCs from women with endometriosis. However, there were no significant effects of PGE2 on promoting the production of E2 or the expression of CYP19A1 in GLCs. Moreover, the production of E2 and the expression of CYP19A1 in GLCs from women with endometriosis were significantly decreased compared to the controls. Conclusions PGE2 concentrations are increased in endometriotic FF, along with concomitant increases in progesterone and STAR. In contrast, the E2 and CYP19A1 are decreased in GLCs, which may delay the development of the follicles and cause an imbalance in the follicular steroid hormone levels. These changes may have close relationship with endometriosis-associated infertility.

Factors Influencing the Number of Rabies Cases in Children in China

To understand the epidemic situation and factors influencing rabies cases in children in China, we obtained an overview of the current epidemic based on individual data of rabies cases in children and a descriptive analysis was carried on the prevalence and related factors. The results showed that the rabies cases in children accounted for 21.3% of the total number of rabies cases in China, 97.0% of these cases occurred in rural areas, they were mainly caused by dogs （81.5%）, and were primarily level Ill exposure （47.7%）. More than half of the cases were not treated with wound care, vaccination rate was extremely low （15.7%）, and only 5.9% of cases were injected with antibodies. Furthermore, 25.4% of cases adopted incorrect treatments such as extruding bleed and wound closure, cases vaccinated with 5 injections acco- unted for only 22.5%. In conclusion, the prevalence of rabies cases in children in China remains a serious concern, the number and immune status of dogs in rural areas, and knowledge of rabies by risk populations should be considered in future rabies prevention and control programs.

Preparation and Initial Application of a Monoclonal Antibody Specific for a Newly Discovered Conserved Linear Epitope of Rabies Virus Nucleoprotein

Objective To prepare monoclonal antibodies against a newly discovered and conserved linear epitope of Rabies virus nucleoprotein and to use them in a rabies diagnostic test. Methods Synthetic peptide containing the epitope was used as immunogen to prepare hybridoma cell lines by classical hybridoma technology. Anti-peptide monoclonal antibodies produced in ascites of inoculated Balb/c mice were labeled with fluorescein isothiocyanate （FITC） after purification and used in fluorescent antibody test （FAT）. Results Two positive hybridoma cell lines, RVNP-mAbl-CL and RVNP-mAb2-CL, were obtained. RVNP- mAbl-CL produced a higher concentration of monoclonal antibody RVNP-mAbl in Balb/c ascites. FITC-labeled RVNP-mAbl showed correct results on certain Rabies virus-positive canine brain tissue samples and cells of a small subclone of baby hamster kidney 21 cell line （BSR）. Conclusion FITC-labeled RVNP-mAbl has potential application for laboratory diagnosis of rabies

Thermodynamic property of sandwich cylindrical shell structure with metallic wire mesh:Numerical modeling and experimental analysis

As a new addition to lightweight composite structures,the sandwich cylindrical shell with a metallic wire mesh core has emerged as a promising solution for thermodynamic performance analysis at elevated *** intricate interwoven cellular formations within the metallic wire mesh pose difficulties for thermo-mechanical modeling and property ***,the constitutive models employed to characterize hysteresis phenomena were presented,comprising isotropic elasticity,Bergstrom-Boyce model,Ogden hyper-elasticity,and parameter identification through mechanical examinations at varying ***,the finite element modeling of cylindrical shell structures was determined for modal and steady-state dynamic ***,the experimental procedures were carried out,including the preparation of the sandwich cylindrical shell and the dynamic testing *** first-order natural frequency of the cylindrical shell structure is close to the resonance frequency of the dynamic test results,with a maximum error of 6.5%,demonstrating the accuracy of the simulation *** compared to the solid-core cylindrical shell,the average insertion loss of the sandwich cylindrical shell structure within the frequency range of 10–1000 Hz at room temperature is up to 11.09 ***,at elevated temperatures,the average insertion loss of the sandwich cylindrical shell decreases but fluctuates as the temperature changes.

Generalized Embedding Machines for Recommender Systems

Factorization machine (FM) is an effective model for feature-based recommendation that utilizes inner products to capture second-order feature interactions. However, one of the major drawbacks of FM is that it cannot capture complex high-order interaction signals. A common solution is to change the interaction function, such as stacking deep neural networks on the top level of FM. In this work, we propose an alternative approach to model high-order interaction signals at the embedding level, namely generalized embedding machine (GEM). The embedding used in GEM encodes not only the information from the feature itself but also the information from other correlated features. Under such a situation, the embedding becomes high-order. Then we can incorporate GEM with FM and even its advanced variants to perform feature interactions. More specifically, in this paper, we utilize graph convolution networks (GCN) to generate high-order embeddings. We integrate GEM with several FM-based models and conduct extensive experiments on two real-world datasets. The results demonstrate significant improvement of GEM over the corresponding baselines.

Highly selective electrosynthesis of 3,4-dihydroisoquinoline accompanied with hydrogen production over three-dimensional hollow CoNi-based microarray electrocatalysts

Electrocatalytic oxidation reaction of biomass-based derivatives is an excellent candidate to replace water oxidation for obtaining both value-added products and hydrogen(H_(2)),but the exploration of competent electrocatalysts is still highly ***,two new types of three-dimensional self-supported hollow microarrays containing CoNi layered double hydroxide(CoNi-LDH)and N-doped carbon nanosheets decorated with CoNi alloyed nanoparticles(CoNi-NC)on carbon cloth(CC)are prepared,which are further used as efficient electrocatalysts for tetrahydroisoquinoline(THIQ)electrooxidation and hydrogen evolution reaction(HER),*** demonstrate that the Co-modulated electronic environment for Ni(II)/Ni(Ⅲ)redox-looping in CoNi-LDH is the main factor to boost the selectivity of 3,4-dihydroisoquinoline(DHIQ)for the indirect electrooxidation process of *** functional theory(DFT)calculations reveal that the Ni(Ⅲ)/Co(Ⅲ)dual sites of CoNi-LDH exhibit enhanced adsorption for THIQ but poorer adsorption for DHIQ compared to pure Co(Ⅲ)or Ni(Ⅲ).Therefore,the Ni(Ⅲ)/Co(Ⅲ)dual sites can effectively inhibit the peroxidation of DHIQ to isoquinoline(IQ)over CoNi-LDH,thus improving the selectivity of DHIQ to nearly 100%,much higher than that of its pure Ni ***,CC@CoNi-NC can deliver high HER activity with low overpotential(40 mV@10 mA·cm^(-2))and high exchange current density(3.08 mA·cm^(-2)).Impressively,the assembled flow-cell device with CC@CoNi-LDH anode and CC@CoNi-NC cathode only requires low cell voltage and electricity consumption of 1.6 V and 3.50 kWh per cubic meter of H_(2)(@25 mA·cm^(-2)).
