NUMB is an evolutionarily conserved protein that plays an important role in cell adhesion,migration,polarity,and cell fate *** has also been shown to play a role in the pathogenesis of certain cancers,although it remains controversial whether NUMB functions as an oncoprotein or tumor ***,we show that NUMB binds to anaplastic lymphoma kinase(ALK),a receptor tyrosine kinase aberrantly activated in several forms of cancer,and this interaction regulates the endocytosis and activity of ***,the function of the NUMB-ALK interaction is *** both p66-NUMB and p72-NUMB isoforms are capable of mediating the endocytosis of ALK,the former directs ALK to the lysosomal degradation pathway,thus decreasing the overall ALK level and the downstream MAP kinase *** contrast,the p72-NUMB isoform promotes ALK recycling back to the plasma membrane,thereby maintaining the kinase in its active *** work sheds light on the controversial role of different isoforms of NUMB in tumorigenesis and provides mechanistic insight into ALK regulation.

Pancreatoscopy plays a significant role in the diagnosis and treatment of pancreatic ***,the risk of pancreatoscopy is remarkably greater than that of other endoscopic procedures,such as gastroscopy and bronchoscopy,owing to its severe *** comparison,virtual pancreatoscopy(VP)has shown notable ***,because of the low resolution of current computed tomography(CT)technology and the small diameter of the pancreatic duct,VP has limited clinical *** this study,an optimal path algorithm and super-resolution technique are investigated for the development of an open-source software platform for VP based on 3D *** proposed segmentation of the pancreatic duct from the abdominal CT images reached an average Dice coefficient of 0.85 with a standard deviation of *** to the excellent segmentation performance,a fly-through visualization of both the inside and outside of the duct was successfully reconstructed,thereby demonstrating the feasibility of *** addition,a quantitative analysis of the wall thickness and topology of the duct provides more insight into pancreatic diseases than a fly-through *** entire VP system developed in this study is available at https://***/gaoyi/***.

Patterns of in-hospital mortality and bleeding complications following PCI for very elderly patients： insights from the Dartmouth Dynamic Registry

Background Very elderly patients （age 〉 85 years） are a rapidly increasing segment of the population. As a group, they experience high rates of in-hospital mortality and bleeding complications following percutaneous coronary intervention （PCI）. However, the relationship between bleeding and mortality in the very elderly is unknown. Methods Retrospective review was performed on 17,378 consecutive PCI procedures from 2000 to 2015 at Dartmouth-Hitchcock Medical Center. Incidence of bleeding during the index PCI admission （bleeding requiring transfusion, access site hematoma 〉 5 cm, pseudoaneurysm, and retroperitoneal bleed） and in-hospital mortality were reported for four age groups （〈 65 years, 65-74 years, 75-84 years, and ≥ 85 years）. The mortality of patients who suffered bleeding complications and those who did not was calculated and multivariate analysis was performed for in-hospital mortality. Lastly, known predictors of bleeding were compared between patients age 〈 85 years and age ≥85 years. Results Of 17,378 patients studied, 1019 （5.9%） experienced bleeding and 369 （2.1%） died in-hospital following PCI. Incidence of bleeding and in-hospital mortality increased monotonically with increasing age （mortality： 0.94%, 2.27%, 4.24% and 4.58%; bleeding： 3.96%, 6.62%, 10.68% and 13.99% for ages 〈 65, 65-4, 75-84 and ≥ 85 years, respectively）. On multivariate analysis, bleeding was associated with increased mortality for all age groups except patients age ≥85 years [odds ratio （95% CI）： age 〈 65 years, 3.65 （1.99-6.74）; age 65-74 years, 2.83 （1.62-4.94）; age 75-84 years, 3.86 （2.56-5.82）, age ≥ 85 years 1.39 （0.49-3.95）]. Conclusions Bleeding and mortality following PCI increase with increasing age. For the very elderly, despite high rates of bleeding, bleeding is no longer predictive of in-hospital mortality following PCI.

Ultrasound (US) has become one of the most commonly performed imaging modalities in clinical practice. It is a rapidly evolving technology with certain advantages and with unique challenges that include low imaging quality and high variability. From the perspective of image analysis, it is essential to develop advanced automatic US image analysis methods to assist in US diagnosis and/or to make such assessment more objective and accurate. Deep learning has recently emerged as the leading machine learning tool in various research fields, and especially in general imaging analysis and computer vision. Deep learning also shows huge potential for various automatic US image analysis tasks. This review first briefly introduces several popular deep learning architectures, and then summarizes and thoroughly discusses their applications in various specific tasks in US image analysis, such as classification, detection, and segmentation. Finally, the open challenges and potential trends of the future application of deep learning in medical US image analysis are discussed.

Rapid and simultaneous detection of human hepatitis B virus and hepatitis C virus antibodies based on a protein chip assay using nano-gold immunological amplification and silver staining method

A sensitive method for detection of Hepatitis A virus (HAV) by utilizing gold-DNA probe on an array was developed. Amino- modified oligodeoxynucleotides at the 5’ position were arrayed on an activated glass surface to function as capture probes. Sandwich hybridization occurred among capture probes, the HAV amplicon, and gold nanoparticlesupported oligonucleotide probes. After a silver enhancement step, signals were detected by a standard flatbed scanner or just by naked eyes. As little as 100 fM of HAV amplicon could be detected on the array. Therefore, the array technology is an alternative to be applied in detection of HAV due to its low-cost and high-sensitivity.

Background: Viral hepatitis due to hepatitis B virus and hepatitis C virus are major public health problems all over the world. Traditional detection methods including polymerase chain reaction (PCR)-based assays and enzyme-linked immunosorbent assays (EliSA) are expensive and timeconsuming. In our assay, a protein chip assay using Nano-gold Immunological Amplification and Silver Staining (NIASS) method was applied to detect HBV and HCV antibodies rapidly and simultaneously.

The interactions between bovine serum albumin(BSA) and gold nanoparticles(AuNPs) ,and the conformational changes of BSA induced by this interaction,were investigated by UV-visible absorption spectroscopy,fluorescence spectroscopy,and Fourier transform infrared in combination with attenuated total reflection spectroscopy(ATR-FTIR) .The critical adsorption density for preventing AuNP aggregation in 0.1 mol/L phosphate buffered saline(pH 7.2) was 23 BSA molecules per gold particle or 3.8×1012 BSA molecules/*** bound to the AuNPs with high affinity(binding constant Ks=7.59×108 L/mol) ,and the intrin-sic fluorescence of BSA was quenched by the AuNPs in accordance with the static quenching *** fluorescence spectroscopy and ATR-FTIR showed that AuNPs induced conformational changes in BSA,which resulted in it becoming less compact and increased the polarity of the microenvironment around the tryptophan residue Trp-212.

Starch is the major component in maize kernels,providing a stable carbohydrate source for humans and livestock as well as raw material for the biofuel *** maize kernel starch content will help meet industry demands and has the potential to increase overall *** developed a pair of maize near-isogenic lines（NILs） with different alleles for a starch quantitative trait locus on chromosome 3（q HS3）, resulting in different kernel starch content. To investigate the candidate genes for q HS3 and elucidate their effects on starch metabolism, RNA-Seq was performed for the developing kernels of the NILs at 14 and 21 d after pollination（DAP）. Analysis of genomic and transcriptomic data identified 76 genes with nonsynonymous single nucleotide polymorphisms and 384 differentially expressed genes（DEGs） in the in trogressed fragment, including a hexokinase gene, Zm HXK3 a, which catalyzes the conversion of glucose to glucose-6-phosphate and may play a key role instarch metabolism. The expression pattern of all DEGs in starch metabolism shows that altered expression of the candidate genes for q HS3 promoted starch synthesis,with positive consequences for kernel starch content. These results expand the current understanding of starch biosynthesis and accumulation in maize kernels and provide potential candidate genes to increase starch content.

Experimental data shows that the binding of human prolactin(hPRL)to human prolactin receptor(hPRLr-ECD)is strongly pH-dependent,while the binding of the same receptor to human growth hormone(hGH)is *** we carry in silico analysis of the molecular effects causing such a difference and reveal the role of individual amino *** is shown that the computational modeling correctly predicts experimentally determined pKa’s of histidine residues in an unbound state in the majority of the cases and the pH-dependence of the binding free *** analysis carried in conjunction with calculated pH-dependence of the binding revealed that the main reason for pH-dependence of the binding of hPRL-hPRLr-ECD is a number of salt-bridges across the interface of the complex,while no salt-bridges are formed in the ***,most of the salt-bridges involve histidine residues and this is the reason for the pH-dependence across a physiological range of *** analysis not only revealed the molecular mechanism of the pH-dependence of the hPRL-hPRLr-ECD,but also provided critical insight into the underlying physicchemical mechanism.
