Non-human primates(NHPs)are increasingly used in preclinical trials to test the safety and efficacy of biotech-nology therapies.Nonetheless,given the ethical issues and costs associated with this model,it would be highly advantageous to use NHP cellular models in clinical studies.However,developing and maintaining the naive state of primate pluripotent stem cells(PSCs)remains difficult as does in vivo detection of PSCs,thus limiting biotech-nology application in the cynomolgus monkey.Here,we report a chemically defined,xeno-free culture system for culturing and deriving monkey PSCs in vitro.The cells display global gene expression and genome-wide hypometh-ylation patterns distinct from monkey-primed cells.We also found expression of signaling pathways components that may increase the potential for chimera formation.Crucially for biomedical applications,we were also able to integrate bioluminescent reporter genes into monkey PsCs and track them in chimeric embryos in vivo and in vitro.The engineered cells retained embryonic and extra-embryonic developmental potential.Meanwhile,we generated a chimeric monkey carrying bioluminescent cells,which were able to track chimeric cells for more than 2 years in living animals.Our study could have broad utility in primate stem cell engineering and in utilizing chimeric monkey models forclinical studies.

Alterations of soil aggregates and intra-aggregate organic carbon fractions after soil conversion from paddy soils to upland soils:Distribution,mineralization and driving mechanism

Investigating the impacts of soil conversion on soil organic carbon(OC) content and its fractions within soil aggregates is essential for defining better strategies to improve soil structure and OC sequestration in terrestrial ecosystems. However, the consequences of soil conversion from paddy soil to upland soil for soil aggregates and intra-aggregate OC pools are poorly understood. Therefore, the objective of this study was to quantify the effects of soil conversion on soil aggregate and intra-aggregate OC pool distributions. Four typical rice-producing areas were chosen in North and South China, paired soil samples(upland soil converted from paddy soil more than ten years ago vs. adjacent paddy soil) were collected(0–20 cm) with three replicates in each area. A set of core parameters(OC preservation capacity, aggregate carbon(C) turnover, and biological activity index) were evaluated to assess the responses of intra-aggregate OC turnover to soil conversion. Results showed that soil conversion from paddy soil to upland soil significantly improved the formation of macro-aggregates and increased aggregate stability. It also notably decreased soil intra-aggregate OC pools, including easily oxidized OCa(EOCa), particulate OCa(POCa), and mineral-bound(MOCa) OC, and the sensitivity of aggregate-associated OC pools to soil conversion followed the order: EOCa(average reduction of 21.1%) > MOCa(average reduction of 15.4%) > POCa(average reduction of 14.8%). The potentially mineralizable C(C_(0)) was significantly higher in upland soil than in paddy soil, but the corresponding decay constant(k) was lower in upland soil than in paddy soil. Random forest model and partial correlation analysis showed that EOCa and pH were the important nutrient and physicochemical factors impacting k of C mineralization in paddy soil,while MOCa and C-related enzyme(β-D-cellobiohydrolase) were identified as the key factors in upland soil. A 3D-printed microhemispherical shell resonator with electrostatic tuning for a Coriolis vibratory gyroscope

The emergence of microhemispherical resonant gyroscopes,which integrate the advantages of exceptional stability and long lifetime with miniaturization,has afforded new possibilities for the development of whole-angle gyroscopes.However,existing methods used for manufacturing microhemispherical resonant gyroscopes based on MEMS technology face the primary drawback of intricate and costly processing.Here,we report the design,fabrication,and characterization of the first 3D-printable microhemispherical shell resonator for a Coriolis vibrating gyroscope.We remarkably achieve fabrication in just two steps bypassing the dozen or so steps required in traditional micromachining.By utilizing the intricate shaping capability and ultrahigh precision offered by projection microstereolithography,we fabricate 3D high-aspect-ratio resonant structures and controllable capacitive air gaps,both of which are extremely difficult to obtain via MEMS technology.In addition,the resonance frequency of the fabricated resonators can be tuned by electrostatic forces,and the fabricated resonators exhibit a higher quality factor in air than do typical MEMS microhemispherical resonators.This work demonstrates the feasibility of rapidly batch-manufacturing microhemispherical shell resonators,paving the way for the development of microhemispherical resonator gyroscopes for portable inertial navigation.Moreover,this particular design concept could be further applied to increase uptake of resonator tools in the MEMS community.

Study on preparation,characterization and application of mixed-colorants thermochromic microcapsules

In order to expand the color-change range and make the discoloration temperature suitable for the daily textiles,a new formulation of mixed-colorants thermochromic core material is designed,in which crystal violet lactone(CVL)is the basic color former,bisphenol A(BPA)is the developer,especially the solvent-based dye and dodecyl dedecanoate are used as the extended dye and the solvent,respectively.Scanning electron microscope(SEM),differential scanning calorimeter(DSC)and thermogravimetric analyzer(TGA)were applied to study the morphology,encapsulation rate,non-isothermal crystallization behavior and thermal stability of the mixed-colorant thermochromic microcapsules(MCTMs)prepared by in-situ polymerization.The results show that MCTMs have good sphericity,uniform particle size,good thermal stability and the encapsulation rate can reach 86.73%.Under the non-isothermal conditions,the encapsulation of formaldehyde-melamine resin wall material can slightly reduce the crystallization rate of the solvent,but does not affect the relative crystallinity,thermal properties and discoloration sensitivity of the core materials.With the variety of ambient temperatures in the range of 20–35°C,the printed cotton fabrics with MCTMs could change color reversibly between different tones with good reliability and durability,and exhibit more gorgeous colors than the common thermochromic materials.

Objective:Real-word data on long-acting luteinizing hormone-releasing hormone(LHRH)agonists in Chinese patients with prostate cancer are limited.This study aimed to determine the real-world effectiveness and safety of the LHRH agonist,goserelin,particularly the long-acting 10.8-mg depot formulation,and the follow-up patterns among Chinese prostate cancer patients.Methods:This was a multicenter,prospective,observational study in hormone treatment-na?ve patients with localized or locally advanced prostate cancer who were prescribed goserelin 10.8-mg depot every 12 weeks or 3.6-mg depot every 4 weeks with or without an anti-androgen.The patients had follow-up evaluations for 26 weeks.The primary outcome was the effectiveness of goserelin in reducing serum testosterone and prostate-specific antigen(PSA)levels.The secondary outcomes included testosterone and PSA levels,attainment of chemical castration(serum testosterone<50 ng/d L),and goserelin safety.The exploratory outcome was the monitoring pattern for serum testosterone and PSA.All analyses were descriptive.Results:Between September 2017 and December 2019,a total of 294 eligible patients received≥1 dose of goserelin;287 patients(97.6%)were treated with goserelin 10.8-mg depot.At week 24±2,the changes from baseline[standard deviation(95%confidence interval)]in serum testosterone(n=99)and PSA(n=131)were-401.0 ng/d L[308.4 ng/d L(-462.5,-339.5 ng/d L)]and-35.4 ng/m L[104.4 ng/m L(-53.5,-17.4 ng/m L)],respectively.Of 112 evaluable patients,100(90.2%)achieved a serum testosterone level<50 ng/d L.Treatment-emergent adverse events(TEAEs)and severe TEAEs occurred in 37.1%and 10.2%of patients,respectively.The mean testing frequency(standard deviation)was 1.6(1.5)for testosterone and 2.2(1.6)for PSA.Conclusions:Goserelin 10.8-mg depot effectively achieved and maintained castration and was well-tolerated in Chinese patients with localized and locally advanced prostate cancer.

Inhibition of proliferation,migration,and invasiveness of bladder cancer cells through SAPCD2 knockdown

Introduction:Bladder cancer(BC)has a high incidence and mortality rate worldwide.Suppressor anaphasepromoting complex domain containing 2(SAPCDC2)is over-expressed in a variety of tumors.Objectives:This study investigated the effects of SAPCD2 knockdown on BC cells.Methods:T24 and UMUC3 cell models and the xenografted BC tumor model with SAPCD2 knockdown were established to observe the malignant phenotype of BC cells by cell counting kit-8 assay,colony formation test,wound healing,and Transwell assay,mRNA and proteins expressions were measured with quantitative real-time polymerase chain reaction,western blotting,and tissue immunohistochemistry.Lithium chloride agonist on the Wnt/β-catenin pathway was used to clarify the molecular mechanism of SAPCD2 knockdown.Results:SAPCD2 expression was significantly higher in BC cell lines than in SVHUC-1 cells.SAPCD2 knockdown inhibited viability and cloning,hindered the G0/G1 phase of the cell cycle in UMUC3 and T24 cells,and decreased the migration and invasiveness of BC cells.SAPCD2 knockdown inhibited expression levels of cyclin D1,cyclin B1,N-cadherin,vimentin,Snail,β-catenin,c-Myc,and cyclin-dependent kinase 4,while the P21 and E-cadherin were raised by SAPCD2 knockdown.Furthermore,lithium chloride reversed the effects of SAPCD2 knockdown on the expression levels of the above proteins in UMUC3 and T24 cells.In vivo,SAPCD2 knockdown inhibited the volume,weight,and expression of Ki-67 andβ-catenin in tumors and increased the E-cadherin expression.Conclusion:SAPCD2 knockdown inhibits the malignant phenotype of BC via a pathway involvingβ-catenin.

An online detection technology must be developed for realizing the real-time control of friction stir welding.In this study,the three-dimensional force exerted on a material during friction stir welding was collected synchronously and the relationship between the forces and welding quality was investigated.The results indicated that the fluctuation period of the traverse force was equal to that of the lateral force during the stable welding stage.The phase difference between two horizontal forces wasπ/2.The values of the horizontal forces increased with welding speed,whereas their amplitudes remained the same.The proposed force model showed that the traverse and lateral forces conformed to an elliptical curve,and this result was consistent with the behavior of the measured data.The variational mode decomposition was used to process the plunge force.The intrinsic mode function that represented the real fluctuation in the plunge force varied at the same frequency as the spindle rotational speed.When tunnel defects occurred,the fluctuation period features were consistent with those obtained during normal welding,whereas the ratio parameter defined in this study increased significantly.

A Perturbation Analysis of Low-Rank Matrix Recovery by Schatten p-Minimization

A number of previous papers have studied the problem of recovering low-rank matrices with noise, further combining the noisy and perturbed cases, we propose a nonconvex Schatten p-norm minimization method to deal with the recovery of fully perturbed low-rank matrices. By utilizing the p-null space property (p-NSP) and the p-restricted isometry property (p-RIP) of the matrix, sufficient conditions to ensure that the stable and accurate reconstruction for low-rank matrix in the case of full perturbation are derived, and two upper bound recovery error estimation ns are given. These estimations are characterized by two vital aspects, one involving the best r-approximation error and the other concerning the overall noise. Specifically, this paper obtains two new error upper bounds based on the fact that p-RIP and p-NSP are able to recover accurately and stably low-rank matrix, and to some extent improve the conditions corresponding to RIP.

单晶碳化硅(SiC)的高脆性、高硬性和强化学惰性是制约第三代半导体超精密抛光发展的关键,实现衬底高效率、超光滑表面的加工具有挑战性。对于单晶SiC的化学机械抛光(CMP),分别从材料去除和工艺优化两个维度出发,阐述了CMP SiC的影响因素和规律,指出了该方法的不足。介绍了光催化、超声振动、电场、等离子体、磁流变、表面预处理等辅助CMP抛光方法,分析了复合增效抛光的去除机理和优势。通过对比发现,辅助能场的介入有助于改善SiC表面质量,并能获得较好的加工效果,然而,复合抛光技术涉及的能场复杂,多能场作用下的材料去除机制和工艺参数匹配仍需进行深入研究。最后,对未来单晶SiC超精密加工的研究给出了建议,并进行了展望。

Intelligent diagnostic model for pterygium by combining attention mechanism and MobileNetV2

AIM:To evaluate the application of an intelligent diagnostic model for pterygium.METHODS:For intelligent diagnosis of pterygium,the attention mechanisms—SENet,ECANet,CBAM,and Self-Attention—were fused with the lightweight MobileNetV2 model structure to construct a tri-classification model.The study used 1220 images of three types of anterior ocular segments of the pterygium provided by the Eye Hospital of Nanjing Medical University.Conventional classification models—VGG16,ResNet50,MobileNetV2,and EfficientNetB7—were trained on the same dataset for comparison.To evaluate model performance in terms of accuracy,Kappa value,test time,sensitivity,specificity,the area under curve(AUC),and visual heat map,470 test images of the anterior segment of the pterygium were used.RESULTS:The accuracy of the MobileNetV2+Self-Attention model with 281 MB in model size was 92.77%,and the Kappa value of the model was 88.92%.The testing time using the model was 9ms/image in the server and 138ms/image in the local computer.The sensitivity,specificity,and AUC for the diagnosis of pterygium using normal anterior segment images were 99.47%,100%,and 100%,respectively;using anterior segment images in the observation period were 88.30%,95.32%,and 96.70%,respectively;and using the anterior segment images in the surgery period were 88.18%,94.44%,and 97.30%,respectively.CONCLUSION:The developed model is lightweight and can be used not only for detection but also for assessing the severity of pterygium.
