Frequencies and mechanisms of pesticide resistance in Tetranychus urticae field populations in China

The two-spotted spider mite Tetranychus urticate is an important agricultural pest worldwide.It is extremely polyphagous and has developed resistance to many pesticides.Here,we assessed the pesticide resistance of seven field populations of T.urticae in China,their target site mutations and the activities of their detoxification enzymes.The results showed that abamectin and the traditional pesticides pyridaben,profenofos and bifenthrin had higher resistance or lower toxicity than more recently developed pesticides including chlorfenapyr,spinetoram,cyflumetofen,cyenopyrafen,bifenazate and B-azolemiteacrylic.The frequency of point mutations related to abamectin resistance,G314D in the glutamate-gated chloride channel 1(GluCl1)and G326E in GluC13,ranged 47%-70%and 0%-97%,respectively.The frequency of point mutations in A1215D and F1538I of the voltage-gated sodium channel gene(VGSC),which may increase resistance to pyrethroids,ranged 88%-100%and 10%-100%,respectively.For target sites related to organophosphate resistance,mutation frequencies ranged 25%-92%for G119S and 0%-23%for A201S in the acetycholinesterase gene(Ace).Mutation G126S in the bifenazate resistance-related cytochrome b gene(Cytb)was observed in three of the seven T.urticae populations.Higher activities of detoxification enzymes(P450,GST,CarEs and UGTs)were observed in two T.urticae populations,with significant difference in the XY-SX population.These results provide useful information on the status of pesticide resistance of T.urticae in China and suggest that T.urticae field populations may have multiple resistance mechanisms.

Accurate and efficient methods for identifying and tracking each animal in a group are needed to study complex behaviors and social interactions.Traditional tracking methods(e.g.,marking each animal with dye or surgically implanting microchips)can be invasive and may have an impact on the social behavior being measured.To overcome these shortcomings,video-based methods for tracking unmarked animals,such as fruit flies and zebrafish,have been developed.However,tracking individual mice in a group remains a challenging problem because of their flexible body and complicated interaction patterns.In this study,we report the development of a multi-object tracker for mice that uses the Faster region-based convolutional neural network(R-CNN)deep learning algorithm with geometric transformations in combination with multi-camera/multi-image fusion technology.The system successfully tracked every individual in groups of unmarked mice and was applied to investigate chasing behavior.The proposed system constitutes a step forward in the noninvasive tracking of individual mice engaged in social behavior.

Cancer profiles in China and comparisons with the USA:a comprehensive analysis in the incidence,mortality,survival,staging,and attribution to risk factors

China faces a disproportionate cancer burden to the population size and is undergoing a transition in the cancer spectrum.We extracted data in five aspects of cancer incidence,mortality,survival,staging distributions,and attribution to risk factors in China,the USA and worldwide from open-source databases.We conducted a comprehensive secondary analysis of cancer profiles in China in the above aspects,and compared cancer statistics between China and the USA.A total of 4,546,400 new cancer cases and 2,992,600 deaths occurred in China in 2020,accounting for 25.1%and 30.2%of global cases,respectively.Lifestyle-related cancers including lung cancer,colorectal cancer,and breast cancer showed an upward trend and have been the leading cancer types in China.41.6%of new cancer cases and 49.3%of cancer deaths occurred in digestive-system cancers in China,and the cancers of esophagus,nasopharynx,liver,and stomach in China accounted for over 40%of global cases.Infection-related cancers showed the highest population-attributable fractions among Chinese adults,and most cancers could be attributed to behavioral and metabolic factors.The proportions of stage I for most cancer types were much higher in the USA than in China,except for esophageal cancer(78.2%vs.41.1%).The 5-year relative survival rates in China have improved substantially during 2000–2014,whereas survival for most cancer types in the USA was significantly higher than in China,except for upper gastrointestinal cancers.Our findings suggest that although substantial progress has been made in cancer control,especially in digestive system cancers in China,there was still a considerable disparity in cancer burden between China and the USA.More robust policies on risk factors and standardized screening practices are urgently warranted to curb the cancer growth and improve the prognosis for cancer patients.

Cancer statistics for young adults aged 20 to 49 years in China from 2000 to 2017:a population-based registry study

An increasing cancer incidence among adults younger than 50 years has been reported for several types of cancer in multiple countries.We aimed to report cancer profiles and trends among young adults in China.Data from the China Cancer Registry Annual Report were used to estimate incidence and mortality among young adults(ages 20–49 years)in China in 2017,and an age-period-cohort model was employed to estimate the average annual percent change(AAPC)in incidence and mortality from 2000 to 2017.All 25 cancer types were grouped into obesity-or overweight-associated cancers(12 cancer types)and additional cancers(13 cancer types).In 2017,there were 681,178new cases and 214,591 cancer deaths among young adults in China.Among young adults,the most common cancers were thyroid,breast,cervical,liver,lung,and colorectal cancer,and the leading causes of cancer deaths were liver,lung,cervical,stomach,breast,and colorectal cancer.From 2000 to 2017,the cancer incidence increased for all cancers combined among young adults,with the highest AAPC(1.46%)for adults aged 20–24 years,while cancer mortality decreased,with the highest AAPC(-1.63%)for those aged 35–39 years.In conclusion,the cancer incidence in China has increased among young adults,while cancer mortality has decreased for nearly all ages.Cancer control measures,such as obesity control and appropriate screening,may contribute to reducing the increasing cancer burden among young adults.

Bradysia cellarum Frey (Diptera: Sciaridae) is an important subterranean pestand is especially damaging to Chinese chive. An effective and moreenvironmentally safe method than pesticides is needed for its control. Theefficacy of B. cellarum control, growth of Chinese chive and soil microbialdiversity were investigated after uae of soil solarization to exterminate thisinsect pest. The results show that on the first day after soil solarization 100%control of B. cellarum was achieved. Growth of Chinese chive was lower insolarized plots than in control plots over the first 10 days after treatment. Chivegrowth in solarized plots increased subsequently to match that in the controlplots. Moreover, the soil microbial community diversity in the treatment groupdecreased initially before gradually recovering. In addition, the abundance ofbeneficial microorganisms in the genus Bacillus and the phyla Proteobacteria,Chloroflexi and Firmicutes increased significantly. Soil solarization is thereforepractical and worthy of promotion in Chinese chive-growing regions.

It is of great significance to study the effects of desert plants on soil enzyme activities and soil organic carbon(SOC)for maintaining the stability of the desert ecosystem.In this study,we studied the responses of soil enzyme activities and SOC fractions(particulate organic carbon(POC)and mineral-associated organic carbon(MAOC))to five typical desert plant communities(Convolvulus tragacanthoides,Ephedra rhytidosperma,Stipa breviflora,Stipa tianschanica var.gobica,and Salsola laricifolia communities)in the proluvial fan in the eastern foothills of the Helan Mountain in Ningxia Hui Autonomous Region,China.We recorded the plant community information mainly including the plant coverage and herb and shrub species,and obtained the aboveground biomass and plant species diversity through sample surveys in late July 2023.Soil samples were also collected at depths of 0–10 cm(topsoil)and 10–20 cm(subsoil)to determine the soil physicochemical properties and enzyme activities.The results showed that the plant coverage and aboveground biomass of S.laricifolia community were significantly higher than those of C.tragacanthoides,S.breviflora,and S.tianschanica var.gobica communities(P<0.05).Soil enzyme activities varied among different plant communities.In the topsoil,the enzyme activities of alkaline phosphatase(ALP)andβ-1,4-glucosidas(βG)were significantly higher in E.rhytidosperma and S.tianschanica var.gobica communities than in other plant communities(P<0.05).The topsoil had higher POC and MAOC contents than the subsoil.Specifically,the content of POC in the topsoil was 18.17%–42.73%higher than that in the subsoil.The structural equation model(SEM)indicated that plant species diversity,soil pH,and soil water content(SWC)were the main factors influencing POC and MAOC.The soil pH inhibited the formation of POC and promoted the formation of MAOC.Conversely,SWC stimulated POC production and hindered MAOC formation.Our study aimed to gain insight into the effects of desert plant communities on soil enzyme activities and SOC fra

Objective:To establish the quality standards for the preparation of Xiao’er Qingre enema in hospitals.Method:Thin-layer chromatography(TLC)was used to identify Radix Isatidis and Glycyrrhizae in the prescription.The content of(R,S)-Goitrin was determined by high-performance liquid chromatography(HPLC).Results:In TLC identification,there was no interference between the negative controls of Radix Isatidis and glycyrrhizae,and the spots were well-separated.The HPLC results of(R,S)-Goitrin showed a good linear relationship between the peak area and concentration within the range of 0.476-95.2μg·mL-1(r=0.9999).Conclusion:The TLC and HPLC methods established in this experiment are simple,reproducible,and specific,making them suitable for the quality control Xiao’er Qingre enema preparation in hospitals.

Gastrointestinal stromal tumors(GISTs)are the most common type of soft tissue sarcoma in the gastrointestinal tract.Most GISTs have been attributed to activated gain-of-function mutations in either KIT or platelet-derived growth factor receptorα,making these molecular features essential targets for therapeutic interventions.Although surgery is the standard treatment for localized GISTs,patients often experience relapse and disease progression even after surgery.In recent years,targeted therapy has significantly improved the prognosis of patients with advanced GISTs.Imatinib mesylate,a KIT inhibitor,is the first-line treatment for advanced GISTs and has revolutionized the treatment of this disease.However,drug resistance remains a major issue with imatinib treatment,as a significant majority of patients become resistant to imatinib either after initiation or after 2–3 years of treatment.Consequently,novel tyrosine kinase inhibitors such as sunitinib,regorafenib,ripretinib,and avapritinib have been introduced to address drug resistance.Immunotherapy has emerged as a potential approach for the treatment of advanced GISTs.This review comprehensively summarizes the pathogenesis of GISTs and the development of targeted therapies and immunotherapies,provides an overview of the emergence of drug resistance in advanced GISTs,and discusses the challenges and prospects associated with the treatment of GISTs.
