Caspase-3 mRNA expression in rats with apoptosis of retinal photoreceptor cells

BACKGROUND： Apoptosis of retinal photoreceptor cells is a commonly pathological characteristic of various eye diseases, while caspase-3 is an important regulating gene and plays a key role in apoptosis. OBJECTIVE： To measure the expression of caspase-3 mRNA in rats with apoptosis of retinal photoreceptor cells by using real-time fluorescent quantitative polymerase chain reaction and compare with those of the normal rats. DESIGN： Randomized controlled animal study. SETTING： Zhongshan Ophthalmological Center of Sun Yat-sen University. MATERIALS： A total of 36 female SD rats of 50 days old and clean grade and weighing （150±10） g were provided by Experimental Animal Center of Northern Area of Sun Yat-sen University. All rats were randomly divided into normal control group （n =6） and N-methyl-N-nitrosourea （MNU） group （n =30）, and they were observed at 12 hours, 1, 2, 3 and 5 days after model establishment, with 6 rats at each time point. METHODS： The experiment was carried out at Zhongshan Ophthalmological Center, Key Laboratory of Ophthalmology by State Ministry of Education from March to December 2004. Rats in the normal control group were intraperitoneally injected with saline and rats in the MNU group were intraperitoneally injected with 40 mg/kg MNU. And then, retinal photoreceptor injured models were established. At 12 hours, 1, 2, 3 and 5 days after model establishment, the rats were sacrificed for enucleating right eyeballs, isolating retina immediately and extracting total RNA. The expression of caspase-3 mRNA in retina was measured with real-time fluorescent quantitative polymerase chain reaction. MAIN OUTCOME MEASURES： Expression of caspase-3 mRNA in retina of rats in the two groups. RESULTS： A total of 36 SD rats were involved in the final analysis. The expressions of caspase-3 mRNA in the rat retina of both groups at the five time points （12 hours, 1, 2, 3 and 5 days） after model establishment were 1.52×105, 18.35×105, 25.14×105, 29.25×105, 13.72×105 and 12.24×105, respectively. Efficacy of Carbamazepine Combined with Botulinum Toxin A in the Treatment of Blepharospasm and Hemifacial Spasm

Purpose:To observe the efficacy of the combined treatment of carbamazepine and botulinum toxin A for blepharospasm and hemifacial spasm. Methods:Fifty-eight patients with either blepharospasm or hemifacial spasm were randomly divided into treatment and control groups. In the treatment group, 30 patients were administered with local intramuscular injections of botulinum toxin A and oral carbamazepine 100 mg/time,3 times/day for 60 days. Twenty-eight subjects in the control group under-went local intramuscular injections of botulinum toxin A only. Results:After combined treatment, the complete remission rate was 90%, which was significantly higher than that of the of the control group (67.9% , Plinum toxin A had increased efficacy in the treatment of blepharospasm or hemifacial spasm, but had no significant effect in terms of the duration of the therapeutic effect.

目的基于术前磁共振小肠成像(MRE)构建列线图模型预测克罗恩病(CD)患者首次肠切除术后早期吻合口复发(EAR)风险的价值。方法回顾性纳入术前行MRE和初次肠切除术的CD患者,分为EAR组(18例)和无EAR组(12例)。EAR组包括内镜下Rutgeerts评分≥I_(2)或术后12个月内需要切除吻合口的患者。分析患者术前人口学特征、实验室检查、切除肠段及其邻近肠系膜的MRE参数、组织学分析和术后药物治疗数据共38个参数。采用LASSO回归分析多因素二分类logistic回归筛选出与EAR相关的独立危险因素,构建列线图预测模型并评估其预测性能。结果 MRE上的梳状征和肠系膜爬行脂肪指数为CD术后发生早期吻合口复发的独立因素,一致性指数为0.882 (95%CI:0.764~1)。校准图显示了实际观测和预测早期吻合口复发概率之间的强相关性。结论基于术前MRE构建的列线图模型可能是一种预测CD患者术后早期吻合口复发的潜在工具,有利于CD患者肠切除术后个体化管理。为EAR高风险患者术后早期个体化药物辅助治疗方案的制定提供参考。

Differentiation of human epidermis-derived mesenchymal stem cell-like pluripotent cells into neural-like cells in culture and after transplantation

Skin is the largest organ of the human body and a possible source of stem cells for research and cell-based therapy. We have isolated a population of mesenchymal stem cell-like pluripotent cells from human epidermis, termed human (h) EMSCPCs. This preliminary study tested if these hEMSCPCs can be induced to differentiate into neural-like cells. Human EMSCPCs were first cultured for four to seven days in a serum-free neural stem cell (NSC) medium for pre-induction. During pre-induction, hEMSCPCs coalesced into dense spheres that resembled neural rosettes. In the presence of a conditioned differentiation medium, pre-induced cells took on the morphological characteristics of neural cells, including slender projections with inflated or claw-like ends that contacted the soma or projections of other cells as revealed by confocal microscopy. Moreover, these differentiating cells expressed the neural-specific markers β-III tubulin, MAP2, GFAP, and synapsin I as evidenced by immunocytochemistry. Both pre-induced hEMSCPCs and uninduced hEMSCPCs were labeled with CM-DiI and transplanted into the vitreous cavities of nude mice. Transplanted cells were examined four weeks later in frozen eyeball sections by immunofluorescence staining, which demonstrated superior retinal migration and neural differentiation of pre-induced cells. Our study is the first to demonstrate that hEMSCPCs possess the capacity to differentiate into neural-like cells, suggesting potential uses for the treatment of retinal diseases such as age-related macular degeneration.

如何精细遥感监测复杂的不同类型农田作物种植情况,是智慧农业农村领域实现农耕面积调查与农作物估产的关键。目前的高分辨率影像的作物种植像素级语义分割中,深度卷积神经网络难以兼顾空间多尺度全局特征和局部细节特征,从而导致各类农田地块之间边界轮廓模糊和同类农田区域内部完整性不高等问题。针对这些不足,本文提出了一种耦合DeepLabv3+和Transformer编码器的双分支并行特征融合网络FDTNet,以实现农作物种植类型的精细遥感监测。首先,在FDTNet中并行嵌入DeepLabv3+和Transformer分别捕获农田影像的局部特征和全局特征;其次,应用耦合注意力融合模块CAFM有效融合两者的特征;然后,在解码器阶段应用卷积注意力模块CBAM增强卷积层有效特征的权重;最后,采用渐进式多层特征融合策略将编码器和解码器中的有效特征全面融合并输出特征图,以实现晚稻、中稻、藕田、菜地和大棚的高精度分类识别。为了验证FDTNet网络模型在高分辨率作物分类应用的有效性,本文选择不同高分辨率的Yuhu数据集和Zhejiang数据集验证,mIoU分别达到74.7%和81.4%。相比于已有的UNet、DeepLabv3、DeepLabv3+、ResT和Res-Swin等深度学习方法,FDTNet的mIoU可分别高2.2%和3.6%。结果表明,FDTNet在纹理单一、大样本量,以及纹理多样、小样本量的两类农田场景中同时表现出优于对比方法的性能,具有较全面的多类别农作物有效特征提取能力。

Coherent Transfer of Excitation in a Nanomechanical Artificial Lattice

We realize a coherent transfer of mechanical excitation in a finely controlled artificial nanomechanical lattice.We also realize strong dynamic coupling between adjacent high-Q mechanical resonators,via modulated dielectric force at the frequency difference between them.An excitation transfer across a lattice consisting of 7 nanobeams is observed by applying a design sequence of switching for couplings,with the final effective population reaching 0.94.This work not only demonstrates the ability to fully control an artificial lattice but also provides an efficient platform for studying complicated dynamics in one-dimensional systems.

Among several dark matter candidates,bosonic ultra-light(sub-meV)dark matter is well motivated because it could couple to the Standard Model and induce new forces.Previous MICROSCOPE and E¨ot–Wash torsion experiments have achieved high accuracy in the sub-1 Hz region.However,at higher frequencies there is still a lack of relevant experimental research.We propose an experimental scheme based on the diamagnetic levitated micromechanical oscillator,one of the most sensitive sensors for acceleration sensitivity below the kilohertz scale.In order to improve the measurement range,we utilize a sensor whose resonance frequencyω0 could be adjusted from 0.1 Hz to 100 Hz.The limits of the coupling constant gB-Lare improved by more than 10 times compared to previous reports,and it may be possible to achieve higher accuracy by using the array of sensors in the future.

Fluidics is one of the most historic subjects that are well-established over centuries on the macroscopic scale.In recent years,fluid detection using a number of micro/nano scale devices has been achieved.However,the interaction of microfluid and solid devices on micro/nano-meter scale still lacks in-depth research.We demonstrate a practical nanomechanical detector for microfluidics via a string resonator with high Q-factor,suspended over a hole.This device is placed under a jet nozzle with several microns of diameter,and the interaction between the micro-gas flow and the resonator is observed by monitoring the variation of the fundamental frequency and the quality factor.Moreover,we manage to measure the fluctuations of the micro-gas flow on the nanomechanical resonator by means of stochastic resonance.This work manifests a potential platform for detecting dynamical fluid behaviors at microscopic scale for novel fluid physics.
