Calcitriol Suppressed Isoproterenol-induced Proliferation of Cardiac Fibroblasts via Integrinβ3/FAK/Akt Pathway

Objective Cardiac fibroblasts(CFs)proliferation and extracellular matrix deposition are important features of cardiac fibrosis.Various studies have indicated that vitamin D displays an anti-fibrotic property in chronic heart diseases.This study explored the role of vitamin D in the growth of CFs via an integrin signaling pathway.Methods MTT and 5-ethynyl-2′-deoxyuridine assays were performed to determine cell viability.Western blotting was performed to detect the expression of proliferating cell nuclear antigen(PCNA)and integrin signaling pathway.The fibronectin was observed by EliSA.Immunohistochemical staining was employed to evaluate the expression of integrinβ3.Results The PCNA expression in the CFs was enhanced after isoproterenol(ISO)stimulation accompanied by an elevated expression of integrin beta-3(β3).The blockade of the integrinβ3 with a specific integrinβ3 antibody reduced the PCNA expression induced by the ISO.Decreasing the integrinβ3 by siRNA reduced the ISO-triggered phosphorylation of FAK and Akt.Both the FAK inhibitor and Akt inhibitor suppressed the PCNA expression induced by the ISO in the CFs.Calcitriol(CAL),an active form of vitamin D,attenuated the ISO-induced CFs proliferation by downregulating the integrinβ3 expression,and phosphorylation of FAK and Akt.Moreover,CAL reduced the increased levels of fibronectin and hydroxyproline in the CFs culture medium triggered by the ISO.The administration of calcitriol decreased the integrinβ3 expression in the ISO-induced myocardial injury model.Conclusion These findings revealed a novel role for CAL in suppressing the CFs growth by the downregulation of the integrinβ3/FAK/Akt pathway.

Magnetic behaviors and magnetocaloric effects in rare earth high-entropy amorphous/nanocrystalline alloys

Rare earth high-entropy alloys(RE-HEAs)exhibit great potential to be applied as refrigerants due to their good comprehensive magnetocaloric properties.In this work,octary GdTbDyHoErTmCoAl and GdTbDyHoErTmCoNi RE-HEAs with amorphous/nanocrystalline structure exhibiting comparable magnetocaloric effect were synthesized.Both RE-HEAs show a second-order magnetic phase transition in the temperature range of hydrogen liquefaction.Due to the complex magnetic interactions,a spin glasslike behavior at low temperatures is observed in the RE-HEAs.A superior magnetocaloric effect is obtained in the nanocrystalline GdTbDyHoErTmCoNi high-entropy alloy that is multiphase attributed to a stronger magnetic exchange interaction when compared with the other that exhibits single amorphous structure.Despite heterogeneous microstructure,homogeneous chemical distributions are observed in the partially crystallized high-entropy alloy.In addition,the magnetocaloric effect and magnetic transition behavior of rare earth medium-and high-entropy alloys,including the RE-HEAs in this study,are summarized and discussed.The results in this work provide a helpful guide for the design of RE-HEAs for hydrogen liquefaction applications with excellent magnetocaloric effects.

Improved damage tolerance and oxidation resistance of (Ti_(0.2)Zr_(0.2)Hf_(0.2)Nb_(0.2)Ta_(0.2))B_(2)–SiC by introducing chopped carbon fibers

High-entropy diborides(HEBs)are considered as promising high-temperature structure materials owing to their high melting point and excellent thermal stability.However,the intrinsic brittleness is the main obstacle that seriously limits their practical applications.To overcome with this obstacle,carbon fibers(Cf)with outstanding mechanical properties are used in the present work as a first attempt to improve the damage tolerance of HEBs.The as-prepared C_(f)/(Ti_(0.2)Zr_(0.2)Hf_(0.2)Nb_(0.2)Ta_(0.2))B_(2)–SiC composite(C_(f)/HEB–SiC)shows high relative density(97.9%)and good mechanical properties with flexural strength of 411±3 MPa and fracture toughness of 6.15±0.11 MPa·m^(1/2).More importantly,the damage tolerance parameter(Dt)has increased from 0.10 m^(1/2) for HEB–SiC to 0.29 m^(1/2) for C_(f)/HEB–SiC.Through microstructural analysis and Vickers indentation of the composite,the toughening mechanisms are disclosed.The carbon fibers coated with carbon coatings demonstrate unique capacity for prolonging the crack propagation path,which promotes the reliability of the composite effectively.Moreover,the C_(f)/(Ti_(0.2)Zr_(0.2)Hf_(0.2)Nb_(0.2)Ta_(0.2))B_(2)–SiC composite also exhibits good static oxidation resistance in the temperature range of 1100–1500℃in air due to the formation of the protective oxide layer constituting of multicomponent oxides(Zr)HfTiO4 and(Zr)Hf_(6)Ta_(2)O_(17) embedded in a continuous SiO_(2) glass.These results are promising,and this primary work can be used as a reference to the synthesis of C_(f)/HEBs for thermal protection materials under hightemperature serving conditions.

Propagation Properties of Shock Waves in Polyurethane Foam based on Atomistic Simulations

Porous materials are widely used in the field of protection because of their excellent energy absorption characteristics.In this work,a series of polyurethane microscopic models are established and the effect of porosity on the shock waves is studied with classical molecular dynamics simulations.Firstly,shock Hugoniot relations for different porosities are obtained,which compare well with the experimental data.The pores collapse and form local stress wave,which results in the complex multi-wave structure of the shock wave.The microstructure analysis shows that the local stress increases and the local velocity decreases gradually during the process of pore collapse to complete compaction.Finally,it leads to stress relaxation and velocity homogenization.The shock stress peaks can be fitted with two exponential functions,and the amplitude of attenuation coefficient decreases with the increase of density.Besides,the pore collapse under shock or non-shock are discussed by the entropy increase rate of the system.The energy is dissipated mainly through the multiple interactions of the waves under shock.The energy is dissipated mainly by the friction between atoms under non-shock.

Recent progress in functional modification and crosslinking of bioprosthetic heart valves

Valvular heart disease(VHD),clinically manifested as stenosis and regurgitation of native heart valve,is one of the most prevalent cardiovascular diseases with high mortality.Heart valve replacement surgery has been recognized as golden standard for the treatment of VHD.Owing to the clinical application of transcatheter heart valve replacement technic and the excellent hemodynamic performance of bioprosthetic heart valves(BHVs),implantation of BHVs has been increasing over recent years and gradually became the preferred choice for the treatment of VHD.However,BHVs might fail within 10-15 years due to structural valvular degeneration(SVD),which was greatly associated with drawbacks of glutaraldehyde crosslinked BHVs,including cytotoxicity,calcification,component degradation,mechanical failure,thrombosis and immune response.To prolong the service life of BHVs,much effort has been devoted to overcoming the drawbacks of BHVs and reducing the risk of SVD.In this review,we summarized and analyzed the research and progress on:(i)modification strategies based on glutaraldehyde crosslinked BHVs and(ii)nonglutaraldehyde crosslinking strategies for BHVs.

For a quasi-split Satake diagram,we define a modified q-Weyl algebra,and show that there is an algebra homomorphism between it and the corresponding■quantum group.In other words,we provide a differential operator approach to■quantum groups.Meanwhile,the oscillator representations of■quantum groups are obtained.The crystal basis of the irreducible subrepresentations of these oscillator representations are constructed.

Adaptive and augmented active anomaly detection on dynamic network traffic streams

Active anomaly detection queries labels of sampled instances and uses them to incrementally update the detection model,and has been widely adopted in detecting network attacks.However,existing methods cannot achieve desirable performance on dynamic network traffic streams because(1)their query strategies cannot sample informative instances to make the detection model adapt to the evolving stream and(2)their model updating relies on limited query instances only and fails to leverage the enormous unlabeled instances on streams.To address these issues,we propose an active tree based model,adaptive and augmented active prior-knowledge forest(A3PF),for anomaly detection on network trafic streams.A prior-knowledge forest is constructed using prior knowledge of network attacks to find feature subspaces that better distinguish network anomalies from normal traffic.On one hand,to make the model adapt to the evolving stream,a novel adaptive query strategy is designed to sample informative instances from two aspects:the changes in dynamic data distribution and the uncertainty of anomalies.On the other hand,based on the similarity of instances in the neighborhood,we devise an augmented update method to generate pseudo labels for the unlabeled neighbors of query instances,which enables usage of the enormous unlabeled instances during model updating.Extensive experiments on two benchmarks,CIC-IDS2017 and UNSW-NB15,demonstrate that A3PF achieves significant improvements over previous active methods in terms of the area under the receiver operating characteristic curve(AUC-ROC)(20.9%and 21.5%)and the area under the precision-recall curve(AUC-PR)(44.6%and 64.1%).

FeNi Prussian blue analogues on highly graphitized carbon nanosheets as efficient glucose sensors

Meeting the continuous glucose monitoring requirements of individuals necessitates the research and development of sensors with high sensitivity and stability.In this study,a straightforward strategy was proposed for synthesizing ultra-thin oxygen-rich graphitized carbon nanosheets（denoted as GCS-O）.These nanosheets are obtained by calcining a topologically two-dimensional indium-based coordination polymer.Subsequently,the growth of FeNi Prussian blue analogue（PBA） on GCS-O effectively introduces active sites and increases the nitrogen content within the carbonaceous matrix.The resulting FeNi-PBA/GCS-O composite exhibits excellent glucose sensing performance with a broad linear range of 1 to 1300μmol·L-1.Meanwhile,it also achieves a high sensitivity of 2496 μA·mmol-1·L·cm-2,a limit of detection of 100nmol·L-1（S/N=3）,and commendable long-term durability.The relatively simple synthesis process,exceptional sensitivity,and satisfactory electrochemical sensing performance of FeNi-PBA/GCS-O open up new directions for biosensor applications.

Prediction of cell-cell communication patierns of dorsal root ganglion cells: single-cell RNA sequencing data analysis

Dorsal root ganglion neurons transmit peripheral somatic information to the central nervous system, and dorsal root ganglion neuron excitability affects pain perception. Dorsal root ganglion stimulation is a new approach for managing pain sensation. Knowledge of the cell-cell communication among dorsal root ganglion cells may help in the development of new pain and itch management strategies. Here, we used the single-cell RNA-sequencing(scRNA-seq) database to investigate intercellular communication networks among dorsal root ganglion cells. We collected scRNA-seq data from six samples from three studies, yielding data on a total of 17,766 cells. Based on genetic profiles, we identified satellite glial cells, Schwann cells, neurons, vascular endothelial cells, immune cells, fibroblasts, and vascular smooth muscle cells. Further analysis revealed that eight types of dorsal root ganglion neurons mediated proprioceptive, itch, touch, mechanical, heat, and cold sensations. Moreover, we predicted several distinct forms of intercellular communication among dorsal root ganglion cells, including cell-cell contact, secreted signals, extracellular matrix, and neurotransmitter-mediated signals. The data mining predicted that Mrgpra3-positive neurons robustly express the genes encoding the adenosine Adora2b(A2B) receptor and glial cell line-derived neurotrophic factor family receptor alpha 1(GFRα-1). Our immunohistochemistry results confirmed the coexpression of the A2B receptor and GFRα-1. Intrathecal injection of the A2B receptor antagonist PSB-603 effectively prevented histamine-induced scratching behaviour in a dose-dependent manner. Our results demonstrate the involvement of the A2B receptor in the modulation of itch sensation. Furthermore, our findings provide insight into dorsal root ganglion cell-cell communication patterns and mechanisms. Our results should contribute to the development of new strategies for the regulation of dorsal root ganglion excitability.

For a polymer/polymer dismissible blend with two crystallizable components,the crystallization behavior of different components and the reciprocal influences between different crystals are interesting and important,but did not investigate in detail.In this study,the L-poly(lactic acid)/polypropylene(PLLA/PP)blends with different weight ratios were prepared by melt mixing and the crystallization behavior of the blends were investigated.Results showed that the crystalline structures of PLLA and PP were not altered by the composition.For the crystallization of PLLA,both the diffusion of chain segments and crystallization rate were enhanced under the existence of PP crystals.For the crystallization of PP,its crystallization rate was depressed under the existence of amorphous PLLA molecular chains.When the PP crystallized from the existence of PLLA crystals,although the diffusion rate of PP was reduced by PLLA crystals,the nucleation positions were obviously enhanced,which accelerated the formation of PP crystals.This investigation would supply more basic data for the application of PLLA/PP blend.
