PROTECTION OF CARBON MONOXIDE INHALATION ON LIPOPOLY-SACCHARIDE-INDUCED MULTIPLE ORGAN INJURY IN RATS

Objective To observe the protection of carbon monoxide （CO） inhalation on lipopolysaccharide （LPS）-induced rat multiple organ injury. Methods Sprague-Dawley rats with multiple organ injury induced by 5 mg/kg LPS intravenous injection were exposed to room air or 2.5 × 10 ^-4 （V/V） CO for 3 hours. The lung and intestine tissues of rats were harvested to measure the expression of heme oxygenase-1 （ HO-1 ） with reverse transcription-polymerase chain reaction, the levels of pulmonary tumor necrosis factor-or （ TNF-α）, interleukin-6 （ IL-6）, and intestinal platelet activator factor （ PAF）, intercellular adhesion molecule-1 （ICAM-1） with enzyme-linked immunosorbent assay, the content of maleic dialdehyde （MDA） and the activity of myeloperoxidase （MPO） with chemical method, the cell apoptosis rate with flow cytometry, and the pathological changes with light microscope. Results CO inhalation obviously up-regulated the expression of HO-1 in lung （5.43 ± 0. 92） and intestine （6. 29 ± 1.56） in LPS ＋ CO group compared with （ 3.08 ± 0. 82） and （ 3.97 ± 1.16 ） in LPS group （ both P 〈 0. 05 ）. The levels of TNF-ot, IL-6 in lung and PAF, ICAM-1 in intestine ofLPS ＋ CO group were 0. 91 ±0. 25,0. 64 ±0.05, 1. 19 ± 0. 52, and 1.83 ±0. 35 pg/mg, respectively, significantly lower than the corresponding values in LPS group （ 1.48 ± 0. 23, 1.16 ± 0. 26, 1.84 ± 0. 73, and 3.48 ± 0. 36 pg/mg, all P 〈 0. 05 ）. The levels of MDA, MPO, and cell apoptosis rate in lung and intestine of LPS ＋ CO group were 1.02 ± 0. 23 nmol/mg, 1.74 ± 0. 17 nmol/mg, 7.18 ± 1.62 U/mg, 6. 30 ±0. 97 U/mg, 1.60% ±0. 34%, and 30. 56% ±6. 33%, respectively, significantly lower than the corresponding values in LPS group （ 1.27 ± 0. 33 nmol/mg, 2. 75 ± 0. 39 nmol/mg, 8. 16 ± 1.49 U/mg, 7. 72 ± 1.07 U/mg, 3.18% ±0. 51%, and 41.52% -＋3.36%, all P 〈0.05）. In addition, injury of lung and intestine induced by LPS was attenuated at presence of CO inhalation. Conclusion CO inhalation protects rat lung and intestine from LP

Protection of carbon monoxide intraperitoneal administration from rat intestine injury induced by lipopolysaccharide

Background Treatment with inhaled carbon monoxide （CO） has been shown to ameliorate intestinal injury in experimental animals induced by lipopolysaccharide （LPS） or ischemia-reperfusion. We hypothesized that CO intraperitoneal administration （i.p.） might provide similar protection to inhaled gas. This study aimed to investigate the effects of continuous 2 L/min of 250 ppm CO i.p. on rat intestine injury induced by LPS and to try to develop a more practical means of delivering the gas. Methods A total of 72 male Sprague-Dawley rats were randomly assigned to 4 groups： control group, CO i.p. group, LPS group and LPS＋CO i.p. group. One hour after intravenously received 5 mg/kg LPS, the rats in LPS group and LPS＋CO i.p. group were exposed to room air and 2 L/min of 250 ppm CO i.p., respectively, and the rats of control group and CO i.p. group intravenously received an equal volume of 0.9% NaCI and 1 hour later, were exposed to room air and 2 L/min of 250 ppm CO i.p., respectively. One, 3 and 6 hour of each group after treated with room air or CO i.p., the animals （n=-6 for each time point） were sacrificed and intestinal tissues were collected for determinating the levels of platelet activator factor （PAF） and intercellular adhesion molecule-1 （ICAM-1） with enzyme-lined immunosorbent assays. The maleic dialdehyde （MDA） content and the myeloperoxidase （MPO） activity were determined with a chemical method. The phosphorylated p38 mitogen activated protein kinase （maPK） expression was assayed with Western blotting and the cell apoptotic rate with flow cytometery. The arterial oxygenation was measured by blood gas analysis, and the pathology determined by light microscope. Results After treatment with 2 L/min of 250 ppm CO i.p., the increase of PAF, ICAM-1, MDA, MPO, and cell apoptotic rate induced by LPS was markedly reduced （P 〈0.05 or 0.01）, and accompanied by ameliorating intestine injury. We report the discovery of a peculiar X-ray transient,EP240408a,by Einstein Probe （EP） and follow-up studies made with EP,Swift,NICER,GROND,ATCA and other ground-based multiwavelength *** new transient was first detected with Wide-field X-ray Telescope （WXT） on board EP on April 8th,2024,manifested in an intense yet brief X-ray flare lasting for *** flare reached a peak flux of 3.9×10-9erg cm-2s-1in 0.5-4 ke V,～300 times brighter than the underlying X-ray emission detected throughout the *** and more precise follow-up observations by EP/FXT,Swift and NICER confirmed the finding of this new *** X-ray spectrum is non-thermal in 0.5-10 ke V,with a power-law photon index varying *** X-ray light curve shows a plateau lasting for ～4 d,followed by a steep decay till becoming undetectable ～10 d after the initial *** on its temporal property and constraints from previous EP observations,an unusual timescale in the range of 7-23 d is found for EP240408a,which is intermediate between the commonly found fast and long-term *** counterparts have been found in optical and near-infrared,with the earliest observation at 17 h after the initial X-ray detection,suggestive of intrinsically weak emission in these *** demonstrate that the remarkable properties of EP240408a are inconsistent with any of the transient types known so far,by comparison with,in particular,jetted tidal disruption events,gamma-ray bursts,X-ray binaries and fast blue optical *** nature of EP240408a thus remains an *** suggest that EP240408a may represent a new type of transients with intermediate timescales of the order of～10 *** detection and follow-ups of more of such objects are essential for revealing their origin.

Recently,high/medium-entropy alloys(HEAs/MEA s)have been considered attractive catalysts due to their unique physicochemical ***,the synthesis of nano-sized HEAs/MEAs catalysts with desirable morphology presents significant ***,we report the synthesis of NiCoFeCu MEA nanoparticles encapsulated in nitrogen-doped carbon nanotubes(NCTs)via a straightforward one-step pyrolysis *** unique structure of NiCoFeCu/NCTs and the nano-sized MEA catalysts contributes to the improved hydrogen desorption kinetics of MgH_(2).The onset dehydrogenation temperature of the MgH_(2)-NiCoFeCu/NCTs composite decreased to 173.4℃,a reduction of 117.4℃compared to pure MgH_(2).The MgH_(2)-NiCoFeCu/NCTs composite could release 6.50 wt%H_(2)within 30 min at 325℃.Furthermore,an activation energy of 116.3 kJ·mol^(-1)for the MgH_(2)-NiCoFeCu/NCTs composite has been obtained,much lower than pure milled MgH_(2),demonstrating an enhanced hydrogen desorption ***,the exceptional dispersion capability of the carbon material contributes to outstanding cyclic stability without any loss of capacity even after 10 cycles of de/hydrogenation at300℃.

N-Linked glycosylation of hemagglutinin(HA) has been demonstrated to regulate the virulence and receptor-binding specificity of avian influenza virus(AIV).In this study,we characterized the variation trend of naturally isolated H9 N2 viruses for the potential N-linked glycosylation sites in HA proteins,and explored any important role of some glycosylation *** genes of 19 H9 N2 subtype AIV strains since 2001 were sequenced and analyzed for the potential glycosylation *** results showed that the viruses varied by losing one potential glycosylation site at residues 200 to 202,and having an additional one at residues 295 to 297 over the past few *** molecular and single mutation analysis revealed that the N200 Q mutation lost an N-linked glycosylation at positions 200 to 202 of the HA protein and affected the human-derived receptor *** further found that this N-linked glycosylation increased viral productivity in the lung of the infected *** findings provide a novel insight on understanding the determinants of host adaption and virulence of H9 N2 viruses in mammals.

The ent-kaurenoids(e.g.,oridonin and enmein)from the Isodon genus(Lamiaceae)are one class of diterpenoids with rich structural diversity and intriguing pharmaceutical *** contrast to the well-established gibberellin pathway,oxidative modifications diversifying the ent-kaurene skeleton in Isodon have remained undetermined for half a *** we report a chromosome-level genome assembly of ***,a well-recognized oridonin producer long favored by Asian people as a traditional herb with antitumor *** shoot apex was confirmed to be the actual region actively producing ent-kaurene *** comparative genomics and phylogenetic analyses,we discovered a cluster of tandem-duplicated CYP706V oxygenase-encoding genes located on an ancient genomic block widely distributed in eudicots,whereas almost exclusively emerged in Isodon *** the shoot apex,IrCYP706V2 and IrCYP706V7 oxidized the ent-kaurene core in the initial stage of oridonin *** of CYP706Vs in other Lamiaceae plants offered an explanation for the specific kaurenoid production in Isodon ***,we found that the Isodon genomes encode multiple diterpenoid synthases that are potentially involved in generating diterpenoid *** findings provided new insights into the evolution of the lineage-specific diterpenoid pathway and laid a foundation for improving production of bioactive ent-kaurene-type diterpenoids by molecular breeding and synthetic biology approaches.

A newly developed rust layer on Q370qENH weathering steel(sample C)was studied under dry–wet cycle test of 3.5 wt.%NaCl solution and salt spray test,compared with the bare weathering steel(sample A)and the weathering steel with additive contents(CuSO_(4)0.2,FeSO_(4)0.1,NaHSO_(3)0.02,NaCl 0.02,Na_(2)HPO_(4)0.01,and water balance,in mass%,termed as sample B).Corrosion mechanism of Q370qENH weathering steel after different surface treatments was investigated by means of surface potential scanning,mass change,polarization curve,and X-ray *** results of X-ray diffraction indicate that rust of bare weathering steel has rare Fe_(3)O_(4),and a dense oxide layer mainly composed of Fe_(3)O_(4) is formed on rust of both sample B and sample *** surface potential of sample A gradually increases to−0.2 V in the dry–wet cycle test,while the surface potential of sample A maintains at about−0.6 V in the salt spray *** surface potential of sample B and sample C is higher than that of sample A in the early stage and remains stable during the short-term accelerated corrosion ***,the mass change of sample C gradually stabilizes with time in the dry–wet cycle and salt spray *** corrosion current of the sample A is lower than that of sample C in the initial stage of short-term accelerated corrosion ***,the corrosion current of sample C is smaller than that of the sample A in the *** main components of the deep rust of sample A areγ-FeOOH andα-FeOOH,while those of the deep rust of sample B and sample C are Fe_(3)O_(4).

An association study of HFEgene mutation with idiopathic male infertility in the Chinese Han population

Mutations in the haemochromatosis gene （HFE） influence iron status in the general population of Northern Europe, and excess iron is associated with the impairment of spermatogenesis. The aim of this study is to investigate the association between three mutations （C282Y, H63D and S65C） in the HFEgene with idiopathic male infertility in the Chinese Han population. Two groups of Chinese men were recruited： 444 infertile men （including 169 with idiopathic azoospermia） and 423 controls with proven fertility. The HFEgene was detected using polymerase chain reaction-restriction fragment length polymorphism （PCR-RFLP） technique. The experimental results demonstrated that no C282Y or $65C mutations were detected. Idiopathic male infertility was not significantly associated with heterozygous H63D mutation （odds ratio=O.801, 95% confidence interval=0.452-1.421, X2=0.577, P=0.448）. The H63D mutation frequency did not correlate significantly with the serum luteinizing hormone （LH）, follicle-stimulating hormone （FSH） and testosterone （T） levels in infertile men （P=0.896, P=0.404 and P=O.05, respectively）. Our data suggest that the HFEH63D mutation is not associated with idiopathic male reproductive dysfunction.

血红素氧合酶(hemeoxygenase,HO)降解血红素的主要代谢产物一氧化碳(carbonmonoxide,CO)具有抗氧化、抗炎症和抑制细胞凋亡作用,而脂多糖(lipopolysaccharide,LPS)诱导的肺组织过氧化、炎症性损伤及大量肺泡上皮和血管内皮细胞凋亡正是导致肺损伤(lunginjury,LI)的关键。由此我们猜想,CO有可能通过上述机制对LI起保护作用。通过静脉注入LPS(5mg/kg体重)诱导大鼠LI,观察吸入室内空气或2.5×10-4(V/V)CO3h后,肺氧化酶学、炎症细胞因子、细胞凋亡、HO-1表达及组织形态学变化。结果显示,静脉注入LPS诱导LI后,CO吸入组大鼠肺肿瘤坏死因子α(tumornecrosisfactor-α,TNF-α)、白细胞介素6(interlukin-6,IL-6)、丙二醛(maleicdialdehyde,MDA)、髓过氧化物酶(myeloperoxidase,MPO)和细胞凋亡分别为(0.91±0.25)pg/mg蛋白、(0.64±0.05)pg/mg蛋白、(1.02±0.23)nmol/mg蛋白、(7.18±1.62)U/mg蛋白、(1.60±0.34)%,均显著低于LI组的(1.48±0.23)pg/mg蛋白、(1.16±0.26)pg/mg蛋白、(1.27±0.33)nmol/mg蛋白、(8.16±1.49)U/mg蛋白、(3.18±0.51)%(P<0.05)。CO吸入组HO-1、白细胞介素10(interlukin-10,IL-10)表达和超氧化物歧化酶(superoxidedismutase,SOD)活性分别为(5.43±0.92)、(0.26±0.07)pg/mg蛋白、(60.09±10.21)U/mg蛋白,它们均显著高于LI组的(3.08±0.82)、(0.15±0.03)pg/mg蛋白、(50.98±6.88)U/mg蛋白(P<0.05)。与LI组相比,CO吸入组肺损伤减轻。研究结果表明,低浓度CO吸入通过抗氧化、抗炎症、抑制细胞凋亡、上调HO-1表达而减轻LPS诱导的肺损伤。

目的:观察低浓度一氧化碳(CO)吸入和腹腔给予对脂多糖(LPS)诱导大鼠小肠损伤的作用及作用过程中丝裂原活化蛋白激酶p38(p38 maPK)磷酸化水平的变化。方法:6组SD大鼠静脉注入5 mg/kg体质量LPS或等容量生理盐水;1 h后,对照及LPS注入组吸入室内空气,CO吸入及LPS注入+CO吸入组吸入体积分数为2.5×10-4CO,CO腹腔及LPS注入+CO腹腔组腹腔通入体积分数为2.5×10-4CO。观察1、3、6 h后放血处死,取回盲部上小肠,酶联免疫吸附法测定血小板活化因子(PAF)及细胞间黏附分子-1(ICAM-1)水平;光镜观察组织形态学变化;蛋白印迹法测定p38 maPK磷酸化水平。结果:LPS注入组PAF、ICAM-1及p38 maPK磷酸化水平显著高于相应时间点的对照、CO吸入及CO腹腔组(P均maPK磷酸化水平进一步增高(P均maPK信号转导通路可能参与了这一过程。
