Nomograms for predicting pathological response to neoadjuvant treatments in patients with rectal cancer

BACKGROUND In recent decades, neoadjuvant therapy(NT) has been the standardized treatment for locally advanced rectal cancer(LARC). Approximately 8%-35% of patients with LARC who received NT were reported to have achieved a complete pathological response(pCR). If the pathological response(PR) can be accurately predicted, these patients may not need surgery. In addition, no response after NT implies that the tumor is destructive, resistant to both chemotherapy and radiotherapy, and prone to having a high metastatic potential. Therefore,developing accurate models to predict PR has great clinical significance and can help achieve individualized treatment in LARC patients.AIM To establish nomograms for predicting PR to different NT regimens based on pretreatment parameters for patients with LARC.METHODS Rectal cancer patients were identified from the database of The Sixth Affiliated Hospital, Sun Yat-sen University from January 2012 to December 2016. Logistic regression and nomograms were developed to predict the probability of pCR and good downstaging to ypT0-2N0M0(ypTNM 0-I), respectively, based on pretreatment parameters for all LARC patients. Nomograms were also developed for three NT regimens(capecitabine/deGramont-RT, mFOLFOX6, and m FOLFOX6-RT) to predict pCR probability.RESULTS Four hundred and three patients were included in this study; 72(17.9%) had pCR at the final pathology report, and 177(43.9%) achieved good downstaging to ypT0-2N0M0(ypTNM 0-I). The nomogram for predicting pCR probability showed that NT regimens, tumor differentiation, mesorectal fascia(MRF) status,and tumor length significantly influenced pCR probability. When predicting the probability of good downstaging, tumor differentiation, MRF status, and clinical T stage were the significant factors. Nomograms were developed based on NT regimens. For the capecitabine/de Gramont-RT group, the multivariate analysis showed that the neutrophil-lymphocyte ratio(NLR) was the only significant factor, thus we could not develop a nomogram for this regim

BACKGROUND Carcinoembryonic antigen(CEA)is a commonly used biomarker in colorectal cancer.However,controversy exists regarding the insufficient prognostic value of preoperative serum CEA alone in rectal cancer.Here,we combined preoperative serum CEA and the maximum tumor diameter to correct the CEA level,which may better reflect the malignancy of rectal cancer.AIM To assess the prognostic impact of preoperative CEA/tumor size in rectal cancer.METHODS We retrospectively reviewed 696 stage I to III rectal cancer patients who underwent curative tumor resection from 2007 to 2012.These patients were randomly divided into two cohorts for cross-validation:training cohort and validation cohort.The training cohort was used to generate an optimal cutoff point and the validation cohort was used to further validate the model.Maximally selected rank statistics were used to identify the optimum cutoff for CEA/tumor size.The Kaplan-Meier method and log-rank test were used to plot the survival curve and to compare the survival data.Univariate and multivariate Cox regression analyses were used to determine the prognostic value of CEA/tumor size.The primary and secondary outcomes were overall survival(OS)and disease-free survival(DFS),respectively.RESULTS In all,556 patients who satisfied both the inclusion and exclusion criteria were included and randomly divided into the training cohort(2/3 of 556,n=371)and the validation cohort(1/3 of 556,n=185).The cutoff was 2.429 ng/mL per cm.Comparison of the baseline data showed that high CEA/tumor size was correlated with older age,high TNM stage,the presence of perineural invasion,high CEA,and high carbohydrate antigen 19-9(CA 19-9).Kaplan-Meier curves showed a manifest reduction in 5-year OS(training cohort:56.7%vs 81.1%,P<0.001;validation cohort:58.8%vs 85.6%,P<0.001)and DFS(training cohort:52.5%vs 71.9%,P=0.02;validation cohort:50.3%vs 79.3%,P=0.002)in the high CEA/tumor size group compared with the low CEA/tumor size group.Univariate and multivariate analyses identified CEA/tumor siz

Enhanced cyclic stability of partially disordered spinel cathodes through direct fluorination with gaseous fluorine

Spinel-type cathodes are considered an optimal substitute for conventional layered oxide cathodes owing to their use of inexpensive and earth-abundant manganese as the redox-active element.Moreover,the introduction of cation disorder can effectively suppress the detrimental two-phase reaction to realize high capacities in a wide voltage range.However,the continuous capacity decay during cycles has hindered the widespread application of these cathode materials.Inorganic fluorides exhibit excellent electrochemical stability at high voltage;therefore,in this study,the direct F2 gas reaction with a partially disordered spinel cathode(Li1.6Mn1.6O3.7F0.3,LMOF1.6) was initially applied to investigate the impacts of fluorination on the surface structure and electrochemical performances.The inorganic fluorinated layer,mainly containing LiF,was distributed uniformly on the surface of LMOF1.6nanoparticles after fluorination for an appropriate time without the turbulence caused by the valency of manganese cation,which improved the capacity retention and rate capability by the suppression of structural damage,parasitic reaction,and cation dissolution.The LMOF1.6cathode fluorinated for 0.5 h exhibited a capacity of283.6 mAh·g-1at 50 mA·g-1and an enhanced capacity retention of 29.6% after 50 cycles in the voltage range of1.5-4.8 V,as compared to the pristine LMOF1.6 with only27.9% capacity retention.

In this work,we mainly consider the Cauchy problem for the reverse space-time nonlocal Hirota equation with the initial data rapidly decaying in the solitonless sector.Start from the Lax pair,we first construct the basis Riemann-Hilbert problem for the reverse space-time nonlocal Hirota equation.Furthermore,using the approach of Deift-Zhou nonlinear steepest descent,the explicit long-time asymptotics for the reverse space-time nonlocal Hirota is derived.For the reverse space-time nonlocal Hirota equation,since the symmetries of its scattering matrix are different with the local Hirota equation,the v(λ_(i))(i=0,1)would like to be imaginary,which results in theδ_(λi)^(0)contains an increasing t(±Imv(λ_(i)))/2,and then the asymptotic behavior for nonlocal Hirota equation becomes differently.

Deciphering gastric inflammation-induced tumorigenesis through multi-omics data and AI methods

Gastric cancer(GC), the fifth most common cancer globally, remains the leading cause of cancer deaths worldwide. Inflammation-induced tumorigenesis is the predominant process in GC development;therefore, systematic research in this area should improve understanding of the biological mechanisms that initiate GC development and promote cancer hallmarks. Here, we summarize biological knowledge regarding gastric inflammation-induced tumorigenesis, and characterize the multi-omics data and systems biology methods for investigating GC development. Of note, we highlight pioneering studies in multi-omics data and state-of-the-art network-based algorithms used for dissecting the features of gastric inflammation-induced tumorigenesis, and we propose translational applications in early GC warning biomarkers and precise treatment strategies. This review offers integrative insights for GC research, with the goal of paving the way to novel paradigms for GC precision oncology and prevention.

Fast Cross-Platform Binary Code Similarity Detection Framework Based on CFGs Taking Advantage of NLP and Inductive GNN

Cross-platform binary code similarity detection aims at detecting whether two or more pieces of binary code are similar or not. Existing approaches that combine control flow graphs(CFGs)-based function representation and graph convolutional network(GCN)-based similarity analysis are the best-performing ones. Due to a large amount of convolutional computation and the loss of structural information, the use of convolution networks will inevitably bring problems such as high overhead and sometimes inaccuracy. To address these issues, we propose a fast cross-platform binary code similarity detection framework that takes advantage of natural language processing(NLP)and inductive graph neural network(GNN) for basic blocks embedding and function representation respectively by simulating extracting structural features and temporal features. GNN’s node-centric and small batch is a suitable training way for large CFGs, it can greatly reduce computational overhead. Various NLP basic block embedding models and GNNs are evaluated. Experimental results show that the scheme with long short term memory(LSTM)for basic blocks embedding and inductive learning-based Graph SAGE(GAE) for function representation outperforms the state-of-the-art works. In our framework, we can take only 45% overhead. Improve efficiency significantly with a small performance trade-off.

Highly efficient capture of thorium ion by citric acid-modified chitosan gels from aqueous solution

A novel citric acid-modified chitosan gel(CSCA)was synthesized through a simple one-step process and was used to extract thorium ions from wastewater.The CSCA samples with varying chemical compositions were analyzed using SEM with mapping EDS,FT-IR,and static water contact angle measurements,and their adsorption behaviors were studied in detail.The results showed that the adsorption performance of CSCA improves with the increase of CA content in the sample.CSCA possesses an impressive capacity for thorium adsorption of 279.8 mg/g.Furthermore,it showed an ultra-fast adsorption rate and reached equilibrium within 30 min.In terms of recyclability,the CSCA still retained more than 86%of its initial adsorption capacity after 6 cycles of reuse.Density functional theory(DFT)analysis reveals that the good selectivity of this material towards thorium ions should be attributed to the high density of adsorption sites and strong interaction between carboxyl groups and thorium ions.This work could be beneficial in the design and synthesis of new polymer materials for extracting thorium.

How the enhanced East Asian summer monsoon regulates total gross primary production in eastern China

Recognizing the relationship between gross primary production(GPP)and precipitation in eastern China,the East Asian Summer Monsoon(EASM)plays a crucial role in shaping GPP.Despite confirmation of the strong link between EASM and GPP,there remains a notable research gap in understanding the specific impact of the EASM on GPP in different regions of eastern China.Here we used simulations from Trends in Net Land-Atmosphere Carbon Exchanges(TRENDY)models from 1951 to 2010 and divided eastern China into five subregions for the study.We also used the New East Asian Summer Monsoon Index(NEWI)as a quantitative metric to distinguish between periods of strong and weak EASM.Building on this,this study aims to investigate the response of GPP in different subregions of eastern China.Regionally,under strengthened EASM years(1954,1957,1965,1969,1977,1980,1983,1987,1993 and 1998),East China experienced the most pronounced increase in GPP at 12±21(mean±1 sigma)gC m^(-2) mon^(-1) compared to the weak EASM years(1958,1961,1972,1973,1978,1981,1985,1994,1997 and 2004).In contrast,Southwest China showed a decline in GPP at-4±10 gC m^(-2) mon^(-1).Moreover,GPP also increased in Northeast and North China when EASM strengthened,while South China showed a decline in GPP.This indicated that GPP changed with monsoon intensity.According to the mechanism analysis,during strong EASM,there was intense moisture convergence through alterations in the atmospheric circulation field over East China and abundant precipitation,which further contributed to the increase in GPP.Downward solar radiation in Southwest China decreased with EASM enhancement,which suppressed GPP and hindered vegetation growth.Overall,the results highlight the importance of accurately predicting the impact of different EASM intensities of regional carbon fluxes.

The Extension Dimension of Subcategories and Recollements of Abelian Categories

We investigate the behavior of the extension dimension of subcategories of abelian categories under recollements.LetΛ',Λ,Λ"be art in algebras such that(modΛ',mod A,modΛ")is a recollement,and let D'and D"be subcategories of modΛand modΛ"respectively.For any n,m≥0,under some conditions,we get dimΩ^(k)(D)≤dimΩ^(n)(D')+dimΩ^(m)(D")+1,where k=max{m,n}and D is the subcategory of modΛglued by D'and D";moreover,we give a sufficient condition such that the converse inequality holds true.As applications,some results for Igusa-Todorov subcategories and syzygy finite sub categories are obtained.
