Identification of necroptosis-related lncRNAs for prognosis prediction and screening of potential drugs in patients with colorectal cancer

BACKGROUND Tumor recurrence and metastasis lead to a poor prognosis in colorectal cancer(CRC).Necroptosis is closely related to the tumor microenvironment(TME)and affects tumor recurrence and metastasis.We aimed to stratify CRC patients according to necroptosis-related long noncoding RNAs(lncRNAs),which can be used to not only evaluate prognosis and improve precision medicine in clinical practice but also screen potential immunotherapy drugs.AIM To stratify CRC patients according to necroptosis-related lncRNAs(NRLs),which can be used to not only evaluate prognosis and improve precision medicine in clinical practice but also screen potential immunotherapy drugs.METHODS LncRNA expression profiles were collected from The Cancer Genome Atlas.NRLs were identified by coexpression analysis.Cox regression analysis identified a NRL signature.Then,the value of this signature was comprehensively and multidimensionally evaluated,and its reliability for CRC prognosis prediction was assessed with clinical CRC data and compared with that of six other lncRNA were also performed according to the risk score(RS)of the signature.RESULTS An 8-lncRNA signature significantly associated with overall survival(OS)was constructed,and its reliability was validated with clinical CRC data.Most of the areas under the receiver operating characteristic curves(AUCs)values for 1-,3-and 5-year OS for this signature were higher than those for the other six lncRNA signatures.OS,disease-specific survival and the progression-free interval were all significantly poorer in the high-risk group.The RS of the signature showed good concordance with the predicted prognosis,with AUCs for 1-,3-and 5-year OS of 0.79,0.81 and 0.77,respectively.Additionally,the calibration plots for this signature combined with clinical factors showed that this combination could effectively improve the ability to predict OS.The RS was correlated with tumor stage,lymph node metastasis and distant metastasis.Most of the enriched Kyoto Encyclopedia of Genes and Genomes and Gene Ontol

KISS1 methylation and expression as predictors of disease progression in colorectal cancer patients

AIM:To examine the effect of aberrant methylation of the KISS1 promoter on the development of colorectal cancer(CRC)and to investigate reversing aberrant methylation of the KISS1 promoter as a potential therapeutic target.METHODS:KISS1 promoter methylation,mRNA expression and protein expression were detected by methylation-specific polymerase chain reaction(PCR),real-time quantitative PCR and Western blotting,respectively,in 126 CRC tissues and 142 normal colorectal tissues.Human CRC cells with KISS1 promoter hypermethylation and poor KISS1 expression were treated in vitro with 5-aza-2’-deoxycytidine(5-Aza-CdR).After treatment,KISS1 promoter methylation,KISS1 mRNA and protein expression and cell migration and invasion were evaluated.RESULTS:Hypermethylation of KISS1 occurred frequently in CRC samples(83.1%,105/126),but was infrequent in normal colorectal tissues(6.34%,9/142).Moreover,KISS1 methylation was associated with tumor differentiation,the depth of invasion,lymph node metastasis and distant metastasis(P<0.001).KISS1methylation was also associated with low KISS1 expression(P<0.001).Furthermore,we observed re-expression of the KISS1 gene and decreased cell migration after 5-Aza-CdR treatment in a CRC cell line.CONCLUSION:These data suggest that KISS1 is down-regulated in cancer tissues via promoter hypermethylation and therefore may represent a candidate target for treating metastatic CRC.

Role of adaptation measures in addressing heatwave exposure in China

Heatwave exposure has increased dramatically because of climate warming and population growth,along with their interactive effects.However,effective adaptation measures can reduce these impacts.Nonetheless,the dynamic changes,regional inequality in adaptive capacity and their potential contributions to reducing exposure in the future remain unclear.This study quantifies the impact of adaptive capacity and underscores regional variations in heatwave magnitudes,population exposure and adaptation levels in China.We projected the future adaptive capacity using air-conditioner penetration,factoring in climate cooling requirements and individuals'purchasing power.Utilising population and gross domestic product(GDP)data from four Shared Socioeconomic Pathways(SSP1,SSP2,SSP3 and SSP5)and daily temperature data from four SSP-based emission scenarios(SSP1-2.6,SSP2-4.5,SSP3-7.0 and SSP5-8.5),we estimated heatwave duration,population exposure and avoided impacts through adaptation across China and its sub-regions.Results show a substantial increase in heatwave duration in Southwest and Southern China,especially under the SSP5-8.5 scenario,with a projection of 163.2±36.7 d during 2081-2100.Under the SSP3|SSP3-7.0 scenario,total exposure reaches 156.4±76.8 billion person d per year,which is the highest among all scenarios and 23 times greater than that in 1986-2005 without adaptation.Upon considering adaptation measures,a noteworthy reduction in population exposure is observed,especially in the SSP3|SSP3-7.0 and SSP5JSSP5-8.5 scenarios,with reductions of(62.6±3.9)%and(65.8±5.1)%,respectively,compared with the scenario without adaptation during 2081-2100.Remarkable regional disparities in avoided impacts are also evident,with variations of up to 50%across different regions.The implementation of effective and environmentally friendly adaptation measures can notably address climate change,thereby alleviating the profound threats posed to human well-being.

Developing a novel quantitative parameter for characterizing spatial distribution of fish following exposure to chemicals and wastewater:Behavioral Gini coefficient

While the spatial distribution pattern of fish is increasingly used for toxicological test o chemicals or wastewater,no ideal parameter is available for quantitative assessment of spa tial distribution,especially uneven distribution with multiple hotspots.Here,to develop a quantitative assessment parameter for spatial distribution,the zebrafish were exposed to ethanol,pentylenetetrazole(PTZ),paraquat dichloride(paraquat)and wastewater,followed by a behavioral test in a narrow tank.Behavioral data was acquired and analyzed by id Tracker and MATLAB.By comparing the effects of all treatments on behavior parameters we confirmed that the spatial distribution was more easily altered rather than general loco motor parameters,e.g.0.7-70 mg/L PTZ and 5-20 mg/L paraquat being effective for altering spatial distribution but having little effects on general locomotor parameters.Based on the heatmap,i.e.,the cumulative proportion of grids and that of frequency in grids,we calcu lated the behavioral Gini coefficient(G_(b))for quantitative assessment of fish spatial distri bution.The Gini coefficient ranged from zero to 1,with larger values meaning poorer even ness of spatial distribution.Of note,G_(b)showed smaller coefficient of variations(CV)with3%-19%between replicate tanks in all treatments than the highest frequency(4%-79%),dis playing well robustness.Especially,G_(b)addressed the challenge of the complicated heatmap with multiple hotspots.Overall,the behavioral Gini coefficient we established is an idea parameter to quantitatively assess spatial distribution of fish shoal,which is expected to be applied in toxicity testing for chemicals and wastewater and automatic quality monitoring for surface water and aquaculture water.

Aerial target threat assessment based on gated recurrent unit and self-attention mechanism

Aerial threat assessment is a crucial link in modern air combat, whose result counts a great deal for commanders to make decisions. With the consideration that the existing threat assessment methods have difficulties in dealing with high dimensional time series target data, a threat assessment method based on self-attention mechanism and gated recurrent unit(SAGRU) is proposed. Firstly, a threat feature system including air combat situations and capability features is established. Moreover, a data augmentation process based on fractional Fourier transform(FRFT) is applied to extract more valuable information from time series situation features. Furthermore, aiming to capture key characteristics of battlefield evolution, a bidirectional GRU and SA mechanisms are designed for enhanced features.Subsequently, after the concatenation of the processed air combat situation and capability features, the target threat level will be predicted by fully connected neural layers and the softmax classifier. Finally, in order to validate this model, an air combat dataset generated by a combat simulation system is introduced for model training and testing. The comparison experiments show the proposed model has structural rationality and can perform threat assessment faster and more accurately than the other existing models based on deep learning.

Enhanced cyclic stability of partially disordered spinel cathodes through direct fluorination with gaseous fluorine

Spinel-type cathodes are considered an optimal substitute for conventional layered oxide cathodes owing to their use of inexpensive and earth-abundant manganese as the redox-active element.Moreover,the introduction of cation disorder can effectively suppress the detrimental two-phase reaction to realize high capacities in a wide voltage range.However,the continuous capacity decay during cycles has hindered the widespread application of these cathode materials.Inorganic fluorides exhibit excellent electrochemical stability at high voltage;therefore,in this study,the direct F2 gas reaction with a partially disordered spinel cathode(Li1.6Mn1.6O3.7F0.3,LMOF1.6) was initially applied to investigate the impacts of fluorination on the surface structure and electrochemical performances.The inorganic fluorinated layer,mainly containing LiF,was distributed uniformly on the surface of LMOF1.6nanoparticles after fluorination for an appropriate time without the turbulence caused by the valency of manganese cation,which improved the capacity retention and rate capability by the suppression of structural damage,parasitic reaction,and cation dissolution.The LMOF1.6cathode fluorinated for 0.5 h exhibited a capacity of283.6 mAh·g-1at 50 mA·g-1and an enhanced capacity retention of 29.6% after 50 cycles in the voltage range of1.5-4.8 V,as compared to the pristine LMOF1.6 with only27.9% capacity retention.

Transcriptomic analysis reveals the effects of maternal exposure to bisphenol AF on hypothalamic development in male neonatal mice

Fragmented data suggest that bisphenol AF(BPAF),a chemical widely used in a variety of products,might have potential impacts on the hypothalamus.Here,we employed male neonatal mice following maternal exposure to explore the effects of low-dose BPAF on hypothalamic development by RNA-sequencing.We found that maternal exposure to approximately 50μg/(kg·day)BPAF from postanal day(PND)0 to PND 15 altered the hypothalamic transcriptome,primarily involving the pathways and genes associated with extracellular matrix(ECM)and intercellular adhesion,neuroendocrine regulation,and neurological processes.Further RNA analysis confirmed the changes in the expression levels of concerned genes.Importantly,we further revealed that low-dose BPAF posed a stimulatory impact on pro-opiomelanocortin(POMC)neurons in the arcuate nucleus of the hypothalamus and induced the browning of inguinal white adipose tissue.All findings indicate that developmental exposure to low-dose BPAF could interfere with hypothalamic development and thereby lead to alterations in the metabolism.Interestingly,5000μg/(kg·day)BPAF caused slighter,non-significant or even inverse alterations than the low dose of 50μg/(kg·day),displaying a dose-independent effect.Further observations suggest that the the dose-independent effects of BPAF might be associated with oxidative stress and inflammatory responses caused by the high dose.Overall,our study highlights a risk of low-dose BPAF to human neuroendocrine regulation and metabolism.

Intensive management enhances mycorrhizal respiration but decreases free-living microbial respiration by affecting microbial abundance and community structure in Moso bamboo forest soils

Intensive management is known to markedly alter soil carbon(C)storage and turnover in Moso bamboo forests compared with extensive management.However,the effects of intensive management on soil respiration(RS)components remain unclear.This study aimed to evaluate the changes in different RScomponents(root,mycorrhizal,and free-living microorganism respiration)in Moso bamboo forests under extensive and intensive management practices.A1-year in-situ microcosm experiment was conducted to quantify the RScomponents in Moso bamboo forests under the two management practices using mesh screens of varying sizes.The results showed that the total RSand its components exhibited similar seasonal variability between the two management practices.Compared with extensive management,intensive management significantly increased cumulative respiration from mycorrhizal fungi by 36.73%,while decreased cumulative respiration from free-living soil microorganisms by 8.97%.Moreover,the abundance of arbuscular mycorrhizal fungi(AMF)increased by 43.38%,but bacterial and fungal abundances decreased by 21.65%and 33.30%,respectively,under intensive management.Both management practices significantly changed the bacterial community composition,which could be mainly explained by soil pH and available potassium.Mycorrhizal fungi and intensive management affected the interrelationships between bacterial members.Structural equation modeling indicated that intensive management changed the cumulative RSby elevating AMF abundance and lowering bacterial abundance.We concluded that intensive management reduced the microbial respiration-derived C loss,but increased mycorrhizal respiration-derived C loss.

A highly reduced Mo_(74) polyoxometalate featuring high proton conductivity accessed by building block strategy

Highly reduced polyoxometalates(POMs) are predicted to be used as rather high energy density materials;however,it still suffers from the limited cluster species and reduction ratio.Here we demonstrate that it is possible to employ the building block strategy to generate a highly reduced polyoxomolybdate(C_(2)H_(8)N)_(14)(NH_(4))_(4)H_(14)[Mo_(48)-ⅤMo_(26)ⅥO_(202)(OH)_(12)(SO_(4))_(6)]·46H_(2)O(Mo_(74)).The fundamental Mo-based{Mo_x}(x=4,5,and 6) building blocks,which are templated by tetra-coordinated anions{MoO_(4)}or{SO_(4)},not only lay foundation for the formation of Mo_(74) featuring an unprecedented reduction ratio of 65%,but also give rise to SBBs-mediated(secondary building blocks) supramolecular dense packing interactions among the isolated Mo_(74) clusters that are favorable for proton conduction.Remarkably,high proton conductivity(2.04×10^(-2)S cm^(-1)) had been realized at 50℃ and 90% relative humidity,revealing one of the well-known POMs-based crystalline proton conducting materials.This result highlights that this building block approach possesses great potential in producing highly reduced POM systems that can achieve controllable reduced ratio and desirable properties.

Late Triassic Intracontinental Deformation of South Tianshan,Central Asia:Evidence from Syn-tectonic Sedimentation and Detrital Zircon Provenances of the Kuqa Depression

The Tianshan range,a Paleozoic orogenic belt in Central Asia,has undergone multiple phases of tectonic activities characterized by the N-S compression after the early Mesozoic,including the far-field effects of the Cenozoic Indian-Asian collision.However,there are limited reports on the tectonic deformation and initiation of Triassic intracontinental deformation in the Tianshan range.Understanding this structural context is crucial for interpreting the early intracontinental deformation history of the Eurasian continent during the early Mesozoic.Growth strata and syn-tectonic sediments provide a rich source of information on tectonic activities and have been extensively used in the studies of orogenic belts.Based on detail fieldwork conducted in this study,the middle-late Triassic Kelamayi Formation of the northern Kuqa Depression in the southern Tianshan fold-thrust belt has been identified as the typical syn-tectonic growth strata.The youngest detrital zircon component in two lithic sandstone samples from the bottom and top of the Kelamayi growth strata yielded U-Pb ages of 223.4±3.1 and 215.5±2.9 Ma,respectively,indicating that the maximum depositional age of the bottom and top of the Kelamayi growth strata is 226-220 and 218-212 Ma.The geochronological distribution of detrital samples from the Early-Middle Triassic and Late Triassic revealed abrupt changes,suggesting a new source supply resulting from tectonic activation in the Tianshan range.The coupling relationship between the syn-tectonic sedimentation of the Kelamayi Formation and the South Tianshan fold-thrust system provides robust evidence that the Triassic intracontinental deformation of the South Tianshan range began at approximately 226-220 Ma(during the Late Triassic)and ended at approximately 218-212 Ma.These findings provide crucial constraints for understanding the intraplate deformation in the Tianshan range during the Triassic.
