Efficient and multiplex gene upregulation in plants through CRISPR-Cas-mediated knockin of enhancers

Geneupregulation through genome editing is important for plant research and *** insertion of short transcriptional enhancers(STEs)into gene promoters may offer a universal solution akin to transgene-mediated overexpression while avoiding the drawbacks associated with ***,we introduce an“in locus activation”technique in rice that leverages well-characterized STEs for refined,heritable,and multiplexed gene *** address the scarcity of potent enhancers,we developed a large-scale mining approach and discovered a suite of STEs that are capable of enhancing gene expression in rice *** in locus integration of these STEs into eight rice genes resulted in substantial tran-scriptional upregulation in the edited plants,with up to 869.1-fold increases in their transcript ***-ploying a variety of STEs,we achieved delicate control of gene expression,enabling the fine-tuning of key phenotypic traits such as plant *** approach also enabled efficient multiplexed gene upregu-lation,with up to four genes activated simultaneously,significantly enhancing the nicotinamide mononucleotide metabolic ***,heritability studies from the To to T3 generations confirmed the stable and heritable nature of STE-driven gene ***,our work demon-strates that coupled with STE mining,leveraging genome editing for in locus activation and gene upregu-lation holds great promise to be widely adopted in fundamental plant research and crop breeding.

It is widely recognized that the development of ZABs is impeded by the kinetic bottleneck of oxygen evolution reaction(OER)and oxygen reduction reaction(ORR).The application of conformational entropy strategy to oxides often involves introducing multiple elements with different properties,thereby providing outstanding bifunctional catalytic activity for OER/***,the possible underlying catalytic pathways and potential interactions between various components are still poorly *** paper presents an excellent medium-entropy perovskite oxide,Sr(FeCoNiMo)_(1/4)O_(3−δ)(lower overpotential of 301 mV at 10 mA cm^(−2)).Zinc-air batteries employing it as a cathode catalyst demonstrate excellent round-trip efficiency(62%).By combining theoretical calculation with experiments,we aim to establish the link between the electronic structure of perovskite oxides with different elemental compositions and their OER *** reveals that the conformational entropy strategy can simultaneously shift the O 2p-band center and metal d-band center of perovskite oxide towards the vicinity of the Fermi energy level,thereby triggering a more favorable lattice oxygen-participated mechanism(LOM)during the OER *** outcomes of this work provide crucial insights into the role of conformational entropy strategies in oxygen catalysis and offer potential avenues for constructing efficient and stable electrocatalysts.

Topological study about failure behavior and energy absorption of honeycomb structures under various strain rates

High-speed impact threats and terrorist actions on the battlefield require the development of more effective protective materials and structures,and various protective structure is designed according their energy-absorbing *** this research,the deformation behavior,microscopic failure modes and energy absorption characteristics of re-entrant hexagonal structure,regular hexagonal structure and regular quadrilateral structure are studied under different strain rates *** re-entrant hexagonal structure forms a“X”-shaped deformation zone,the regular quadrilateral and regular hexagonal structure form an“I”-shaped deformation *** microscopic appearance of the section is a mixed fracture *** effects of the topological shape,cell angle,and cell height on the impact behavior of the structure were *** the cell height is fixed and the cell angle is changed,the energy absorption of the structure increase and then decrease as the relative density *** mechanical properties of the structure are optimal when the relative density is about 18.6%and the cell angle is22.5°.When the cell angle is fixed and the cell height is changed,as the relative density increases,the energy absorption of the structure gradually *** regular quadrilateral structure and the reentrant hexagonal structure experienced clear strain rate effects under dynamic impact conditions;the regular hexagonal structure did not exhibit obvious strain rate *** results presented herein provide a basis for further rational design and selection of shock-resistant protective structures that perform well in high-speed impact environments.

MGB probe assay for rapid detection of mtDNA11778 mutation in the Chinese LHON patients by real-time PCR

Objective： Leber＇s hereditary optic neuropathy （LHON） is a maternally inherited degeneration of the optic nerve caused by point mutations of mitochondrial DNA （mtDNA）. Many unsolved questions regarding the penetrance and pathophysiological mechanism of LHON demand efficient and reliable mutation testing. This study aims to develop a minor groove binder （MGB） probe assay for rapid detection of mtDNA11778 mutation and heteroplasmy in Chinese LHON patients by real-time polymerase chain reaction （PCR）. Methods： Forty-eight patients suspected of having LHON and their maternal relatives underwent a molecular genetic evaluation, with 20 normal individuals as a control group at the same time. A real-time PCR involving two MGB probes was used to detect the mtDNA 1 1778 mutation and heteroplasmy. A linear standard curve was obtained by pUCmLHONG and pUCmLHONA clones. Results： All 48 LHON patients and their maternal relatives were positive for rntDNA11778 mutation in our assay, 27 heteroplasmic and 21 homoplasmic. Eighteen cases did not show an occurrence of the disease, while 9 developed the disease among the 27 heteroplasmic mutation cases. Eleven did not show an occurrence of the disease, while 10 cases developed the disease among 21 homoplasmic mutation cases. There was a significant difference in the incidence between the heteroplasmic and the homoplasmic mutation types. The time needed for running a real-time PCR assay was only 80 min. Conclusion： This real-time PCR assay is a rapid, reliable method for mtDNA mutation detection as well as heteroplasmy quantification. Detecting this ratio is very important for predicting phenotypic expression of unaffected carriers.

Efficient catalytic hydrogen generation by intermetallic platinum-lead nanostructures with highly tunable porous feature

The water-gas shift(WGS) reaction is an essential industrial reaction for upgrading hydrogen(H2) by removing carbon monoxide(CO), while highly efficient platinum(Pt)-based catalysts for WGS with simultaneously high activity and stability are still yet to be developed due to the poisoning issue during the reaction. Herein, we report on the porous PtPb peanut nanocrystals(porous PtPb PNCs) and porous PtPb octahedron nanocrystals(porous PtPb ONCs) with controllable ratios of Pt/Pb as extremely active and stable catalysts towards WGS reaction. It exhibits the composition-dependent activity with porous PtPb PNCs-40/ZnO being the most active for WGS to H_2, 16.9 times higher than that of the commercial Pt/C. The porous PtPb PNCs-40/ZnO also display outstanding durability with barely activity decay and negligible structure and composition changes after ten successive reaction cycles. X-ray photoelectron spectroscopy(XPS) results reveal that the suitable binding energy of Pt 4f_(7/2) and the high ratio of Pt(0)to Pt(II) in porous PtPb PNCs/ZnO and porous PtPb ONCs/ZnO are crucial for the enhanced WGS *** CO stripping results indicate the optimized CO adsorption strength on the Pt surface ensure the excellent WGS activity and the outstanding durability. The present work demonstrates an important advance in tuning the porous metal nanomaterials as highly efficient and durable catalysts for catalysis,energy conversion and beyond.

Circuit model based design and analysis for a four-structure-switchable wireless power transfer system

The four-coil wireless power transfer(WPT)technology can effectively improve the transfer *** high efficiency,however,cannot be obtained along the whole transfer distance due to the phenomenon of frequency splitting in the over coupled *** at this limitation,this paper presents a switchable WPT system to improve the overall efficiency by changing the number of working *** switching conditions for the designed system are determined based on the analysis of the transfer efficiencies of four structures,which is deduced through modeling the equivalent *** simulation results well comply with the experimental results and both of them indicate that the switchable system can greatly improve the overall transfer efficiency along the whole transfer *** overall efficiency of the experimental system can reach above 70%at9.97 MHz without additional complexity,which is higher than any single structure system.

Surface engineering at the interface of core/shell nanoparticles promotes hydrogen peroxide generation

Hydrogen peroxide(HO), an environmentally friendly oxidant, has already been widely used in many chemical synthesis and industrials as an alternative to replace traditional oxidants including chlorinated oxidizers and strong acids. However, the conventional synthesis method confronts intense energy cost,tedious separation procedures and high cost, which is not competitive with traditional oxidants. Although direct HOsynthesis from Hand Ois a green and atomically economic reaction, satisfactory activity and desirable selectivity still remain formidable challenges. Herein, for the first time, a class of Pd@NiO-x nanoparticles(NPs)(x = 1, 2, 3 and 4) with a unique core@shell interface structure has been created to achieve high activity, selectivity and stability for the direct HOsynthesis. A precise thermal annealing on Pd@Ni-x NPs revealed that the resulting Pd@Ni O-x NPs exhibited the volcano-like activity toward direct HOsynthesis as a function of annealing temperature and time. By tuning the composition of Pd@Ni O-x NPs and the reaction condition, the efficiency of HO-2 synthesis could be well optimized with 5 wt%Pd@NiO-3/TiOexhibiting the highest productivity(89 mol/(kgh)) and selectivity(91%) to HOas well as excellent stability, making it one of the best catalysts for direct HOsynthesis reported to date.

Molecular Identification of Dried Shellfish Products Sold on the Market Using DNA Barcoding

The dried shellfish products with rich nutrients and low-calorie content are favorite food in China,especially in coastal ***,the species of dried shellfish products in the market are usually unknown,as the taxonomic features were removed during the production *** study described the application of DNA barcoding technique to the identification of 100 dried shellfish(scallop,squid,octopus and cuttlefish)products in *** were authenticated by comparing mitochondrial cytochrome oxidase subunit I(COI)gene and 16S ribosomal RNA(16S rRNA)gene sequences with public reference taxonomic *** results showed that all the 100 products can be identified at species *** four scallop adductor products were processed using the bay scallop,Argopecten irradians,and one was from Portuguese oyster,Crassostrea *** the 27 squid,2 cuttlefish and 6 octopus products were produced by the Jumbo flying squid,Dosidicus *** neighbour-joining tree is in agreement with the results of DNA barcoding *** 64 scallop samples formed one *** cluster,leaving Sca65 clustered with the reference oyster sequence ***(MH997922).All the 35 cephalopod(squid,octopus and cuttlefish)samples formed a *** *** investigation revealed a low variety of dried shellfish products sold on the market,and highlighted the high rate of mislabeling and species *** present work provides a practical method for tracing and authenticating shellfish products.

External blast flow field evolution and response mechanism of single-layer reticulated dome structure

Single-layer reticulated dome structure are commonly high-profile building in the public and can be attractive targets for terrorist bombings,so the public can benefit from enhanced safety with a stronger understanding of the behavior of single-layer reticulated dome structure under *** paper investigates the fluid-structure interaction process and the dynamic response performance of the singlelayer reticulated dome under external blast *** experimental and numerical results shown that structural deformation is remarkably delayed compared with the velocity of blast wave,which advises the dynamic response of large-span reticulated dome structure has a negligible effect on the blast wave propagation under *** failure modes are identified by comparing the plastic development of each ring and the residual spatial geometric of the structure,i.e.,minor vibration,local depression,severe damage,and overall *** plastic deformation energy and the displacement potential energy of the structure are the main consumers of the blast *** addition,the stress performance of the vertex member and the deep plastic ratio of the whole structure can serve as qualitative indicators to distinguish different failure modes.

Sensitivity analysis and probability modelling of the structural response of a single-layer reticulated dome subjected to an external blast loading

The structural response of a single-layer reticulated dome to external explosions is shaped by many variables,and the associated uncertainties imply non-deterministic *** deterministic methods for predicting the consequences of specific explosions do not account for these ***,the impact of the uncertainties associated with these input variables on the structures’response needs to be studied and *** this study,a parametric uncertainty analysis was conducted ***,local and global sensitivity analyses were carried out to identify the drivers of the structural dynamic response.A probabilistic structural response model was established based on sensitive variables and a reasonable sample ***,some deterministic empirical methods for explosion-resistance design,including the plane blast load model of CONWEP,the curved blast load model under the 50%assurance level,and the 20%mass-increased method,were used for evaluating their *** results of the analyses revealed that the structural response of a single-layer reticulated dome to an external blast loading is lognormally ***,the MB0.5 method based on the curved reflector load model yielded results with a relatively stable assurance rate and reliability,but CONWEP did not;thus,the 1.2MB0.5 method can be used for making high-confidence simple *** addition,the results indicated that the structural response is very sensitive to the explosion *** on these results,it is suggested that for explosion proofing,setting up a defensive barrier is more effective than structural strengthening.
