Molecular evolution of hepatitis A virus in a human diploid cell line

aiM： To investigate the hoLspoLs, direction, and the time course of evolution of hepatitis A virus in the process of consecutive cell culture passage in human KMB17 diploid cells.METHODS： Wild type hepatitis A virus H2w was serially propagated in KMB17 cells until passage 30, and the fulllength genomes of H2w and its six chosen progenies were determined by directly sequencing RT-PCR products amplified from viral genomic RNA. Alignment comparison of sequences from H2w with its six progenies and phylogenetic analysis of the whole VP1 region from H2w, progenies of H2w, and other cell culture adapted hepatitis A virus were then carried out to obtain data on the molecular evolution of hepatitis A virus in the process of consecutive passage in KMB17 cells.RESULTS： Most of the mutations occurred by passage 5 and several hotspots related to adaptation of the virus during cell growth were observed. After that stage, few additional mutations occurred through the remaining duration of passage in KMB17 cells except for mutation in the virulence determinants, which occurred in the vicinity of passage 15. The phylogenetic analysis of the whole VP1 region suggested that the progenies of H2w evolved closely to other cell culture adapted hepatitis A virus, i.e. MBB, L-A-l, other than its progenitor H2w.CONCLUSION： Hepatitis A virus served as a useful model for studying molecular evolution of viruses in a given environment. The information obtained in this study may provide assistance in cultivating the next generation of a seed virus for live hepatitis A vaccine production.

From risk control to resilience: developments and trends of urban roads designed as surface flood passages to cope with extreme storms

Urban roads can be designated as surface flood passages to transport excess runoff during extreme storms, thereby preventing local flooding, which is known as the major drainage system. However, this practice poses significant risks, including human loss and property damage, due to the high flow rate and velocity carried by roads. Moreover, urban roads with low flood-resilience may significantly hamper the transportation function during severe storms, leading to dysfunction of the city. Therefore, there is an urgent need to transform risk-oriented flood passages into resilient urban road-based flood passages. This paper presents a systematic review of existing methodologies in designing a road network-based flood passage system, along with the discussion of new technologies to enhance system resilience. The study also addresses current knowledge gaps and future directions. The results indicate that flood management measures based on the urban road network should integrate accessibility assessment, lifeline and emergency planning to ensure human well-being outcomes. Furthermore, the special needs and features of vulnerable groups must be taken into serious consideration during the planning stage. In addition, a data-driven approach is recommended to facilitate real-time management and evaluate future works.

H2 strain attenuated live hepatitis A vaccines:protective efficacy in a hepatitis A outbreak

aiM:To investigate the protective efficacy of H2 strain attenuated live hepatitis A vaccines (H2-strain vaccines) in hepatitis A (HA) ***:With the permission of their parents, 5551 pre-school and grade 1-3 primary school children were inoculated with 1 dose (10(6.5) TCID(50)) of H2 strain vaccines in a nonrandomized, controlled trial conducted in Fucheng County, Hebei Province in May *** 6485 children in the same grades and compatible in gender and age were enrolled as controls. Epidemiological and serological survey was conducted to evaluate the protective efficacy of the vaccines. ELISA was used to detect serum IgM ***:HA outbreak started in early May 1998, peaked in the middle of the same month, and lasted about 80 days. Overall 302 HA cases were found, 192(63.58%) were 5-9 years old. One vaccinee and 25 control cases were found to have hepatitis A, which account for 0.28% (1/356) and 5.92% (25/422) of all vaccinees and controls in the 14 villages, respectively. The protective efficacy of vaccines was 95.27% (95% CI: 85.83%-104.72%). In subjects tested for anti-HAV IgM from 13 villages, 1(0.40%) overt and 11(4.06%) asymptomatic HAV cases were found in 271 vaccinees but 21(6.69%) of overt and asymptomatic ones were found in 314 ***:H2 strain vaccines were excellent in preventing overt hepatitis A,but not so effective in preventing asymptomatic hepatitis A virus infection.A booster dose might be needed to get permanent reliable immunity.

GNN-based temporal knowledge reasoning for UAV mission planning systems

Digital twin system framework and information model for industry chain based on industrial Internet

The integration of industrial Internet,cloud computing,and big data technology is changing the business and management mode of the industry ***,the industry chain is characterized by a wide range of fields,complex environment,and many factors,which creates a challenge for efficient integration and leveraging of industrial big *** at the integration of physical space and virtual space of the current industry chain,we propose an industry chain digital twin(DT)system framework for the industrial *** addition,an industry chain information model based on a knowledge graph(KG)is proposed to integrate complex and heterogeneous industry chain data and extract industrial ***,the ontology of the industry chain is established,and an entity alignment method based on scientific and technological achievements is ***,the bidirectional encoder representations from Transformers(BERT)based multi-head selection model is proposed for joint entity–relation extraction of industry chain ***,a relation completion model based on a relational graph convolutional network(R-GCN)and a graph sample and aggregate network(GraphSAGE)is proposed which considers both semantic information and graph structure information of *** results show that the performances of the proposed joint entity–relation extraction model and relation completion model are significantly better than those of the ***,an industry chain information model is established based on the data of 18 industry chains in the field of basic machinery,which proves the feasibility of the proposed method.

New patterns of localized excitations in (2+1)-dimensions:The fifth-order asymmetric Nizhnik-Novikov-Veselov equation

By applying the mastersymmetry of degree one to the time-independent symmetry K_(1), the fifth-order asymmetric Nizhnik-Novikov-Veselov system is derived. The variable separation solution is obtained by using the truncated Painlevé expansion with a special seed solution. New patterns of localized excitations, such as dromioff, instanton moving on a curved line, and tempo-spatial breather, are constructed. Additionally, fission or fusion solitary wave solutions are presented,graphically illustrated by several interesting examples.

Income inequality is widespread in human society and has important implications for human ***’s perception of the environment bridges income inequality and individual behavioral *** research suggests that social income inequality is usually biased,either overestimated or ***,such phenomena have not been fully explored with quantitive prove,especially in the working environment based on performance data of actual *** fact,the correct perception of people is the basis of a fair environment for their production ***,the perception bias may weaken the adaptability and competitiveness of a company in the *** paper first confirms the prevalence of individual perception bias of income inequality within the working environment based on actual production *** results show that people tend to underestimate income inequity around them,and this underestimation grows with the real unfairness of the working ***,this paper proposes a network generation-based framework with a three-layer structure to correct perception bias using a cooperative network reengineering *** the framework,a homophily-based generative network model is proposed as the key *** simulation results show that our proposed framework effectively reduces individuals’perception bias of income inequality.

Provenance of the Lower Jurassic pyroclastic sediments in the Zigui Basin:Implication for crustal thickening in the eastern Qinling orogenic belt

The Late Triassic witnessed significant collisional orogenic events in the Qinling orogenic belt,accompanied by magma underplating and tectonic *** processes potentially resulted in substantial crustal thickening and uplift of the Qinling ***,due to the absence of igneous rock records from this period in the eastern section of the Qinling orogen,the changes in crustal thickness during this orogenic process have not been thoroughly investigated.A series of foreland basins emerged during the Early Mesozoic to the south of the East Qinling orogenic *** basins have preserved clastic sedimentary rocks derived from the uplift and erosion of the orogenic *** sedimentary records serve as crucial records to reconstruct the evolutionary history of the Qinling *** further clarify the collisional orogenic process of the Qinling orogenic belt,this study conducted in situ volcanic lithic fragment geochemistry,detrital zircon U-Pb chronology and trace element composition analysis on the sandstones of the Lower Jurassic Tongzhuyuan Formation in the Zigui *** results suggest that the sandstones,which exhibit a significant abundance of volcanic lithic fragments,has a characteristic detrital zircon age group of 250–200 Ma,indicating a major provenance from the Triassic volcanic *** with regional correlation and paleocurrent analysis,the detrital zircon U-Pb age data show that the source area of volcanic rocks should be in the Qinling orogenic belt to the north of the *** interpretation is further supported by the Triassic granitic rocks exposed in the western part of the orogenic belt,representing the magmatism during the Triassic collisional orogenesis in the Qinling *** on the co-varying relationships between present-day crust thickness with the chemical compositions of granite rocks and zircons,the La/Yb ratio of volcanic lithic fragments in the Tongzhuyuan Formation and the Eu/Eu*ratio of detrital zircon

Research on the Asymmetric Effect of Fixed-Asset Investment on China's Primary Energy Production in the Context of Energy Transformation

Under China’s“Dual Carbon”target(DCT),“clean replacement”on the energy supply side and“electric energy replacement”on the energy consumption side are the ways to achieve energy ***,energy projects have a long construction period,complex technology categories,and investment risks that greatly affect the development of energy *** judging the effect of investment changes on primary energy production is of great practical significance to the realization of the *** on this,NARDL and TVP-SV-VAR models are innovatively used to reveal the nonlinear effect of fixed-asset investment on China’s primary energy *** results show that the marginal effect of investment growth on coal production is about 1.44 times that of investment ***,the marginal effect of oil and gas investment growth is about 1.21times that of investment *** to the influence of resource constraints,China’s traditional fossil energy still has varying degrees of path dependence on the investment-driven development *** non-fossil energy,investment in hydropower and nuclear power has an inverse correlation with the change in *** marginal efficiency and diseconomies of scale have hindered the development of the hydropower and nuclear power *** addition,the asymmetric effect is not yet significant for the short development time and technical constraints of wind and solar *** the impulse response results,the impact curves of investment in wind and solar power are generally positive,and investment has different degrees of time-delay and time-varying effects on various energy production,which verifies the heterogeneity of investment adjustment mechanisms in different energy industries.
