Aim:To compare the effects of AMP579 and adenosine on L-type Ca2+current(ICa-L)in rat ventricular myocytes and explore the mechanism by which AMP579acts on ***:ICaLwas recorded by patch-clamp technique in ***:Adenosine(10nmol/L to 50μmol/L)showed no effect onbasal ICaL,but it inhibited the ICaLinduced by isoproterenol 10 nmol/L in a concen-tration-dependent manner with the IC50of 13.06 μmol/*** to adenosine,AMP579 also showed an inhibitory effect on the ICaLinduced by ***579 and adenosine(both in 10 μmol/L)suppressed isoproterenol-inducedICaLby 11.1% and 5.2%, *** addition,AMP579 had a direct inhibitoryeffect on basal ICaLin a concentration-dependent manner with IC50(1.17 μmol/L).PD 116948(30μmol/L),an adenosine A1receptor blocker,showed no action on theinhibitory effect of AMP579 on basal ***,GF109203X(0.4μmol/L),aspecial protein kinase C(PKC)blocker,could abolish the inhibitory effect of AMP579on basal I*** the inhibitory effect of AMP579 on basal ICaLwas inducedthrough activating PKC,but not linked to adenosine ***:AMP579 shows a stronger inhibitory effect than adenosine on the ICaLinducedby ***579 also has a strong inhibitory effect on basal ICaLin ratventricular *** of PKC is involved in the inhibitory effect ofAMP579 on basal ICaLat downstream-mechanism.

Aim: To analyze the kayrotypic patterns of 33 cases of intersexuality in Chinese from Taiyuan, China in order tofurther clarify its mechanism of development and the interrelationship between karyotype and phenotypic sex. Meth-ods: High-resolution GTG-banding chromosome technique was used to analyze the karyotype patterns. Results:In these patients, 57.58% were male pseudohermaphrodites (46, XY), 18.18%, female pseudohermaphrodites (46,XX), 12.12%, true hemaphrodites, and 12.12%, other karyotypes, Although testes can be seen in 88.8% of kary-otypes with Y chromosome, 73.68% of the patients were of female social sex. In 42.82% of patients the social sex isin conformity with their karyotypes. There were 2 cases of male pseudohennaphrodites, where the sex chromosome wasnormal, but abnormalities were found in chromosomes 9, 13, or 14. Conclusion: Sex chromosomes determine thedirection of gonadal and sex differentiation, while the development of the normal gonad and extemal genitalia shouldhave the participation of many autosomal chromosomes as well.

In an experimental rat’s renovascular hypertension model, we studied the genesis of anti- cardiac β1-adrenoceptor and M2-muscarinic receptor autoantibodies in relation to the changes in immunological function during the development of renal hypertension. The biological activities of these autoantibodies were also examined. It was shown that after two weeks of operation both the frequency of occurrence and the titre of autoantibodies to cardiac β1-adrenoceptor and M2-muscarinic receptor were significantly increased as compared with the control of pre-treatment. The increased autoantibodies lasted for several weeks and then automatically decreased gradually to the pre-clipping level at 10 weeks. Meanwhile the ratio of CD4 /CD8 + + was also undergone an initial increase followed by gradual recovery and correlated well with the changes in antibody titre. The biological effects of these autoantibodies displayed an “agonistic-like” activities on the beating frequency of cultured neonatal cardiomyocyte. It is suggested that autoimmune mechanisms are involved in the pathogenesis of renal hypertension and the cardiac receptor autoantibodies might be one of the mechanisms leading to cardiac dysfunction.

To clarify whether apoptosis is involved in the injury processes induced by autoantibodyagainst cardiac β1-adrenoceptor,we investigated the biological and apoptotic effects of antibodies on culturedneonatal rat *** rats were immunized with peptides corresponding to the second extra-cellular loop of the β1-adrenoceptor to induce the production of anti-β1-adrenoceptor antibodies in the ***（Ig）G in the sera was detected using synthetic antigen enzyme-linked immunosorbentassay and purified using the diethylaminoethyl cellulose ion exchange *** of cardiomyo-cytes was evaluated using agarose gel electrophoresis and flow *** results showed that thepositive serum IgG greatly increased the beating rates of cardiomyocytes and showed an"agonist-like"***,positive serum IgG induced cardiomyocyte apoptosis after treatment with β1-adrenoceptor overstimulation for *** effects of monoclonal antibody against β-adrenoceptor werealso found to be similar to those of positive serum *** was suggested that the autoantibody could inducecardiomyocyte apoptosis by excessive stimulation of β1-adrenoceptor.

Liver injury induced by various pathogenic factors (such as hepatitis virus, ethanol, drugs and hepatotoxicants, etc.)through their respective special pathogenesis is referred to as 'primary liver injury' (PLI). Liver injury resultedfrom endotoxin (lipopolysaccharide, LPS) and the activation of Kupffer cells by LPS while intestinal endotoxemia (IETM) occurred during the occurrence and development of hepatitis is named the 'secondary liver injury' (SLI).The latter which has lost their own specificities of primary pathogenic factors is ascribed to IETM. The 'secondary liver injury' is of important action and impact on development and prognosis of hepatitis. More severe IETM commonly results in excessive inflammatory responses, with serious hepatic necrosis,further severe hepatitis and even induces acute liver *** milder IETM successively precipitates a cascade,including repeated and persistent hepatocytic impairment accompanied by infiltration of inflammatory cells, hepatic fibrosis, cirrhosis and hepatocarcinoma. Generally, the milder IETM ends with chronic hepatic failure. If PLI caused by various pathogenic factors through their independent specific mechanismis regarded as 'the first hit' on liver, then SLI mediated by different chemical mediators from KCs activated by IETM in the course of hepatitis is 'the second hit' on liver. Thus, fusing and overlapping of the primary and scondary liver injuries determine and influeuce the complexity of the illness and outcome of the patient with hepatitis. For this reason, the viewpoint of 'SLI' induced by the 'second hit' on liver inflicted by IFTM suggests that medical professionals should attach great importance to both 'PLI'and 'SLI' caused by IETM. That is, try to adjust the function of KSs and eliminate endotoxemia of the patient.

Although the aggregated amyloid b-protein (Ab) in senile plaques is one of the key neuropathological features of Alzheimer’s disease (AD), soluble forms of Ab also interfere with synaptic plasticity at the early stage of AD. The suppressive action of acute application of Ab on hippocampal long-term potentiation (LTP) has been reported widely, whereas the mechanism underlying the effects of Ab is still mostly unknown. The present study, using the whole-cell patch clamp technique, investigated the effects of Ab fragments (Ab25–35 and Ab31–35) on the LTP induction-related postsynaptic ligand-gated channel currents in isolated hippocampal CA1 neurons. The results showed a rapid but opposite action of both peptides on excitatory and inhibitory receptor currents. Glutamate application-induced currents weresuppressed by Ab25–35in a dose-dependent manner,andfurtherN-methyl-D-aspartate(NMDA)receptor-mediated currents were selectively inhibited. In contrast, pretreatment with Ab fragments potentiated c-aminobutyric acid (GABA)-induced whole-cell currents. As a control, Ab35–31, the reversed sequence of Ab31–35, showed no effect on the currents induced by glutamate, NMDA or GABA. These results may partly explain the impaired effects of Ab on hippocampal LTP, and suggest that the functional down-regulation of NMDA receptors and up-regulation of GABAA receptors may play an important role in remodeling the hippocampal synaptic plasticity in early AD.
