Objective This study aimed to compare 9 perfluoroalkyl sulfonic acids(PFSA)with carbon chain lengths(C4–C12)to inhibit human placental 3β-hydroxysteroid dehydrogenase 1(3β-HSD1),aromatase,and rat 3β-HSD4 *** Human and rat placental 3β-HSDs activities were determined by converting pregnenolone to progesterone and progesterone secretion in JEG-3 cells was determined using HPLC/MS–MS,and human aromatase activity was determined by *** PFSA inhibited human 3β-HSD1 structure-dependently in the order:perfluorooctanesulfonic acid(PFOS,half-maximum inhibitory concentration,IC50:9.03±4.83μmol/L)>perfluorodecanesulfonic acid(PFDS,42.52±8.99μmol/L)>perfluoroheptanesulfonic acid(PFHpS,112.6±29.39μmol/L)>perfluorobutanesulfonic acid(PFBS)=perfluoropentanesulfonic acid(PFPS)=perfluorohexanesulfonic acid(PFHxS)=perfluorododecanesulfonic acid(PFDoS)(ineffective at 100μmol/L).6:2FTS(1H,1H,2H,2H-perfluorooctanesulfonic acid)and 8:2FTS(1H,1H,2H,2H-perfluorodecanesulfonic acid)did not inhibit human 3β-*** and PFHpS are mixed inhibitors,whereas PFDS is a competitive ***,1–10μmol/L PFOS and PFDS significantly reduced progesterone biosynthesis in JEG-3 *** analysis revealed that PFSA binds to the steroid-binding site of human 3β-HSD1 in a carbon chain length-dependent *** 100μmol/L PFSA solutions did not affect rat 3β-HSD4 and human placental aromatase *** Carbon chain length determines inhibitory potency of PFSA on human placental 3β-HSD1 in a V-shaped transition at PFOS(C8),with inhibitory potency of PFOS>PFDS>PFHpS>PFBS=PFPS=PFHxS=PFDoS=6:2FTS=8:2FTS.

目的分析心肺超声参数及斑点追踪成像技术在肺炎相关脓毒症患者预后中的评估价值。方法选取42例重症医学科(ICU)收治或ICU医师急会诊的肺炎相关脓毒症患者,于起病初期监测心肺超声并储存图像,后期使用EchoPAC程序对超声心动图进行脱机分析,获取左心室长轴整体纵向应变(GLS),收集临床监护数据及实验室检查等指标,根据预后结果分为预后不良组13例和预后良好组29例,应用受试者工作特征(ROC)曲线评价各指标的诊断价值。结果临床参数:预后不良组SOFA评分、呼吸频率、机械通气占比、血流动力学不稳定占比、乳酸水平均高于预后良好组,平均动脉压水平低于预后良好组(P<0.05);超声参数:预后不良组左室流出道速度时间积分(VTI)和最大峰流速(Vmax)、心输出量(CO)、左室射血分数(LVEF)、左室整体GLS(GLS-Avg)、三腔心、四腔心和二腔心GLS(GLS-A3C、GLS-A4C、GLS-A2C)低于预后良好组,左室舒张末期内径(LVEDD)、左室充盈压(E/e’)、前胸壁肺超声评分高于预后良好组(P<0.05)。SOFA评分、VTI、Vmax、CO、LVEDD、E/e’、LVEF、GLS-Avg、前胸壁肺超声评分、乳酸的ROC曲线下面积分别为0.897、0.797、0.789、0.788、0.725、0.699、0.794、0.707、0.688、0.736。结论早期应用心肺超声及斑点追踪成像参数对肺炎相关脓毒症患者预后有较好的评估价值。

Reduction mechanisms of pyrite cinder-carbon composite pellets

The non-isothermal reduction mechanisms of pyrite cinder-carbon composite pellets were studied at laboratory scale under argon （Ar） atmosphere. The composite pellets as well as the specimens of separate layers containing pyrite cinder and coal were tested. The degree of reduction was measured by mass loss. The microstmctures of the reduced composite pellets were characterized by scanning electron mi- croscopy （SEM）. It is found that the reduction processes of the composite pellets may be divided into four stages： reduction via CO and H2 from volatiles in coal at 673-973 K, reduction via H2 and C produced by cracking of hydrocarbon at 973-1123 K, direct reduction by carbon via gaseous intermediates at 1123-1323 K, and direct reduction by carbon at above 1323 K. Corresponding to the four stages, the apparent activation energies （E） for the reduction of the composite pellets are 86.26, 78.54, 72.01, and 203.65 ***-1, respectively.

The efficiency and safety evaluation of hemoglobin hydrolysate as a non-heme iron fortifier

Hemoglobin hydrolysate is derived from the enzymatic degradation of *** work aimed to evaluate whether hemoglobin hydrolysate promotes the absorption of non-heme iron and the safety of absorbed iron in mice by analyzing the iron binding content,iron circulation,and liver *** found that hemoglobin hydrolysate promoted the absorption of non-heme iron with high efficiency in duodenum by spontaneously binding non-heme iron during digestion,and increased hepatic iron content by up-regulating divalent metal transporter 1,zinc transporter 14,but hepatic iron content only increased at 3 *** iron entered the blood through ferroportin without restriction at 3 weeks,and excessive iron entered the liver and then affected the hepatocyte membranes permeability and lipid synthesis through oxidative *** the prolongation of dietary intervention,the up-regulated hepcidin acted on the ferroportin to restrict excess iron from entering the blood,and then the hepatic homeostasis *** addition,hemoglobin hydrolysate enhanced the hepatic antioxidant *** together,hemoglobin hydrolysate has a strong ability to promote the absorption of non-heme iron in vivo,and the absorbed iron is relatively safe due to the regulation of hepcidin.

3-N-Butylphthalide mitigates high glucose-induced injury to Schwann cells:association with nitrosation and apoptosis

A high glucose state readily causes peripheral axon atrophy, demyelination, loss of nerve fiber function, and delayed regeneration. However, few studies have examined whether nitration is also critical for diabetic peripheral neuropathy. Therefore, this study investigated the effects of high glucose on proliferation, apoptosis, and 3-nitrotyrosine levels of Schwann cells treated with butylphthalide. In addition, we explored potential protective mechanisms of butylphthalide on peripheral nerves. Schwann cells were cultured in vitro with high glucose then stimulated with the peroxynitrite anion inhibitors uric acid and 3-n-butylphthalide for 48 hours. Cell Counting Kit-8 and flow cytometry were used to investigate the effects of uric acid and 3-n-butylphthalide on proliferation and apoptosis of Schwann cells exposed to a high glucose environment. Effects of uric acid and 3-n-butylphthalide on levels of 3-nitrotyrosine in Schwann cells were detected by enzyme-linked immunosorbent assay. The results indicated that Schwann cells cultured in high glucose showed decreased proliferation, but increased apoptosis and intracellular 3-nitrotyrosine levels. However, intervention with uric acid or 3-n-butylphthalide could increase proliferation of Schwann cells cultured in high glucose, and inhibited apoptosis and intracellular 3-nitrotyrosine levels. According to our data, 3-n-butylphthalide may inhibit cell nitrification and apoptosis, and promote cell proliferation, thereby reducing damage to Schwann cells caused by high glucose.

Mechanical mechanism and support design analysis on bolt-beam-net support in soft rock roadway in Qigou Coal Mine

The deformation and failure mechanical mechanism in soft rock roadway is related to the stability of supported tunnels, which is important to coal mine production and construction. By physical mechanics experiments and X-ray diffraction （XRD） tests, the engineering mechanical properties of soft rock, as well as main mineral composition of the surrounding soft rock of Qigou Coal Mine, were obtained. Based on analysis results, a method using bolt-beam-net combination to support was put forward. Mechanical analysis of the support form was done by using the calculation software FLAC3D. Results show that clay minerals of this mine are kaolinite and illite mixed layer, of which the water absorption is relatively obvious and presented mudding characteristic after absorbing water, with the plasticity index of 0.35, with small expansibility, which is weakly consolidated colloid with strong connected force in unit cell. The rock blocks have the characteristics of moisture absorption softening, and the deformation mechanical mechanism of which is with the coexistence of molecular expansive mechanism, colloid expansive mechanism, and weak layer trend type. The calculation results show that the bolt-beam-net support structure makes the bolt, beam, and roof deform compatibly. The beams make the force in the bolt relatively homogeneous, which restricts the displacement of the tunnel roof as well. Finally, using in situ monitoring, the numerical results were verified.

为解决某矿围岩破碎、巷道锚网支护设施损坏严重、返修频率高等问题,应用爆破振动和围岩与支护相互作用机理分析了支护失效的原因,提出了“掘到哪儿,支到哪儿,喷到哪儿”的喷锚网优化支护工序。即新掘进的巷道及时清理浮石以后,首先采用素喷一层厚度50~100 mm的混凝土,然后进行钢网和锚杆支护,最后再喷一层厚度50~100 mm的混凝土。经过可行性分析和现场工业试验,证明可在不改变原支护工艺和支护材料的基础上,通过调整和优化施工顺序,可以提高支护效果,维护巷道围岩稳定,比现行支护工序节约86142.71元,研究成果可供同类矿山参考借鉴。

Research on goal-setting method of rock drivage based on the balance among period, cost and quality

On the basis of the analysis on the disadvantages of the original goal-setting about rock drivage, this paper defined the ＂life cycle quality＂. With project management theory and the Cobb-Douglas function, ＂quality-cost and quality-price curve model＂ and the ＂total cost-period prediction model＂ were built. Then the goal-setting method of the balance among quality, cost and period of rock drivage was constructed by finding ＂life cycle cost＂ through ＂life cycle quality＂ using ＂quality-cost and quality-price curve model＂ and ensuring period through ＂life cycle cost＂ using ＂total cost-period prediction model＂（hereinafter referred to as the ＂three goals balance method＂）. ＂Value contribution＂ which is the value of the contribution to a mine because of rock drivage, was found in the process of constructing the ＂quality-cost and quality-price curve model＂. An industrial test was done in coal mine A with the research results, staff footage efficiency improved by 24.24%, the period shortened by 14.3%, the ＂life cycle cost＂ dropped by 2.09%, the ＂life cycle quality price＂ improved by 3.29%, and value contribution increased by 25.3%. The result shows that the new goal method setting on the basis of coal mine profit maximization can ensure construc- tion period. At the same time, it can realize cost and quality objectives and the optimization and balance of relationship among them; rewarding excavation teams by ＂value contribution＂ can combine organizational goal with personal goal, it significantly raise the employee＇s work efficiency.
