The ventral part of the anteromedial thalamic nucleus(AMv)is in a position to convey information to the cortico-hippocampal-amygdalar circuit involved in the processing of fear memory.Corticotropin-releasing-factor(CRF)neurons are closely associated with the regulation of stress and fear.However,few studies have focused on the role of thalamic CRF neurons in fear memory.In the present study,using a conditioned fear paradigm in CRF transgenic mice,we found that the c-Fos protein in the AMv CRF neurons was significantly increased after cued fear expression.Chemogenetic activation of AMv CRF neurons enhanced cued fear expression,whereas inhibition had the opposite effect on the cued fear response.Moreover,chemogenetic manipulation of AMv CRF neurons did not affect fear acquisition or contextual fear expression.In addition,anterograde tracing of projections revealed that AMv CRF neurons project to wide areas of the cerebral cortex and the limbic system.These results uncover a critical role of AMv CRF neurons in the regulation of conditioned fear memory.

Developing the methodologies that allow for safe and effective delivery of therapeutic drugs to target sites is a very important research area in cancer therapy.In this study,polyethylene glycol(PEG)-coated magnetic polymeric liposome(MPL)nanoparticles(NPs)assembled from octadecyl quatemized carboxymethyl chitosan(OQC),PEGylated OQC,cholesterol,and magnetic NPs,and functionalized with epithelial growth factor receptor(EGFR)peptide,were successfully prepared for in-vivo liver targeting.The two-step liver targeting strategy,based on both magnetic force and EGFR peptide conjugation,was evaluated in a subcutaneous hepatocellular carcinoma model of nude mouse.The results showed that EGFR-conjugated MPLs not only accumulated in the liver by magnetic force,but could also diffuse into tumor cells as a result of EGFR targeting.In addition,paclitaxel(PTX)was incorporated into small EGFR-conjugated MPLs(102.0土0.7 nm),resulting in spherical particles with high drug encapsulation efficiency(>90%).The use of the magnetic targeting for enhancing the transport of PTX-loaded EGFR-conjugated MPLs to the tumor site was further confirmed by detecting PTX levels.In conclusion,PTX-loaded EGFR-conjugated MPLs could potentially be used as an effective drug delivery system for targeted liver cancer therapy.

Effect on microstructure and plastic deformation behavior of a Zr-based amorphous alloy by cooling rate control

In this study,Zr_(41.2)Ti_(13.8)Cu_(12.5)Ni_(10)Be_(22.5) amorphous alloys samples with the same diameter(8 mm)were prepared by using self-designed molds(viz.refractory steel,pure graphite,and copper molds)with different cooling capacities.Moreover,by eliminating the size effect,the effect of the cooling rate on the microstructure and compression deformation behavior of Zr_(41.2)Ti_(13.8)Cu_(12.5)Ni_(10)Be_(22.5) amorphous alloys was investigated.Differentiation of the cooling curves revealed that the instantaneous cooling rates of the alloy melt at the glass transition temperature(Tg)are 45,52,and 64 K·s^(-1) for refractory steel,pure graphite,and copper molds,respectively.X-ray diffraction,differential scanning calorimetry,and highresolution transmission electron microscopy analysis revealed that with the decrease in the cooling rate,trace icosahedral-like atomic clusters and nanocrystals appear in local areas of the amorphous alloy and that the amount of free volume decreases with the increase in the amount of icosahedra-like atomic clusters and nanocrystals.Compression test results revealed that the elastic strain,yield strength,and compressive strength of the amorphous alloy marginally change with the decrease in the cooling rate,while the plastic strain gradually increases.By fitting,the effective size of the vein-like pattern was linearly related to the enthalpy released during structural relaxation and plastic strain,indicating that at a low cooling rate,the trace nanocrystals in the amorphous alloy could not effectively improve its plasticity and that the amount of free volume mainly affects its plasticity.

Background:Although laparoscopic technology has achieved rapid development in the surgical field,it has not been applied to liver transplantation,primarily because of difficulties associated with laparoscopic vascular anastomosis.In this study,we introduced a new magnetic-assisted vascular anastomosis technique and explored its application in laparoscopic liver transplantation in pigs.Methods:Two sets of magnetic vascular anastomosis rings(MVARs)with different diameters were developed.One set was used for anastomosis of the suprahepatic vena cava(SHVC)and the other set was used for anastomosis of the infrahepatic vena cava(IHVC)and portal vein(PV).Six laparoscopic orthotopic liver transplantations were performed in pigs.Donor liver was obtained via open surgery.Hepatectomy was performed in the recipients through laparoscopic surgery.Anastomosis of the SHVC was performed using hand-assisted magnetic anastomosis,and the anastomosis of the IHVC and PV was performed by magnetic anastomosis with or without hand assistance.Results:Liver transplants were successfully performed in five of the six cases.Postoperative ultrasonographic examination showed that the portal inflow was smooth.However,PV bending and blood flow obstruction occurred in one case because the MVARs were attached to each other.The durations of loading of MVAR in the laparoscope group and manual assistance group for IHVC and PV were 13±5 vs.5±1 min(P<0.01)and 10±2 vs.4±1 min(P<0.05),respectively.The durations of MVAR anastomosis in the laparoscope group and manual assistance group for IHVC and PV were 5±1 vs.1±1 min(P<0.01),and 5±1 vs.1±1 min(P<0.01),respectively.The anhepatic phase was 43±4 min in the laparoscope group and 23±2 min in the manual assistance group(P<0.01).Conclusions:Our study showed that magnetic-assisted laparoscopic liver transplantation can be successfully carried out in pigs.

Zinc Oxide(ZnO)has been extensively applied as electron transport material(ETM)in perovskite solar cells(PSCs)since the emergence of PSCs.However,some chemisorbed oxygen species on the surface of ZnO can cause the degradation of CH3NH3+(MA^(+))based perovskite.To avoid the destructive effect of ZnO,a facile solution strategy was proposed to produce a ZnS shell around the ZnO nanorods arrays(ZnO-NRs),i.e.ZnO@ZnS core-shell nanorods(ZnO-NRs@ZnS).The ZnO-NRs@ZnS cascade structure can not only facilitate carrier transport,but also enhance the stability of ZnO based PSCs.A power conversion efficiency(PCE)of 20.6%was finally yielded,which is the-state-of-the-art efficiency for PSCs with one-dimensional(1 D)ZnO electron transport materials(ETMs).Moreover,over 90%of the initial efficiency was retained for the unencapsulated device with ZnO-NRs@ZnS ETMs at 85℃for 500 h,demonstrating excellent stability.This work provides a simple and efficient avenue to simultaneously enhance the photovoltaic(PV)performance and stability of 1 D ZnO nanostructure-based PSCs.

Gastrointestinal(GI)cancer is one of the leading causes of cancer-related deaths worldwide.According to the Global Cancer Statistics,colorectal cancer is the second leading cause of cancer-related mortality,closely followed by gastric cancer(GC).Environmental,dietary,and lifestyle factors including cigarette smoking,alcohol intake,and genetics are the most important risk factors for GI cancer.Furthermore,infections caused by Helicobacter pylori are a major cause of GC initiation.Despite improvements in conventional therapies,including surgery,chemotherapy,and radiotherapy,the length or quality of life of patients with advanced GI cancer is still poor because of delayed diagnosis,recurrence and side effect.Resveratrol(3,4,5-trihydroxy-trans-stilbene;Res),a natural polyphenolic compound,reportedly has various pharmacologic functions including anti-oxidant,anti-inflammatory,anti-cancer,and cardioprotective functions.Many studies have demonstrated that Res also exerts a chemopreventive effect on GI cancer.Research investigating the anti-cancer mechanism of Res for the prevention and treatment of GI cancer has implicated multiple pathways including oxidative stress,cell proliferation,and apoptosis.Therefore,this paper provides a review of the function and molecular mechanisms of Res in the prevention and treatment of GI cancer.

AIM To explore the significance of corticotropin-releasing hormone(CRH)-receptor(R)2 in mucosal healing of dextran sulfate sodium(DSS)-induced colitis and the effect of Tong-Xie-Yao-Fang(TXYF) on CRH-R2 expression and regulation.METHODS Ulcerative colitis was induced in mice by administration of 3%(w/v) DSS for 7 d. Once the model was established,mice were administered urocortin-2(30 μg/kg), a peptide which binds exclusively to CRH-R2, or various doses of aqueous TXYF extracts(2.8-11.2 g/kg), a CRH-R2 antagonist Astressin(Ast)2B(20 μg/kg), Ast2B + Ucn2, or Ast2B with various doses of aqueous TXYF extracts for 9 d. Colonic mucosal permeability was then evaluated by measuring the fluorescence intensity in serum. The colitis disease activity index(DAI), histology, body weight loss and colon length were assessed to evaluate the condition of colitis. Terminal deoxynucleotidyl transferase d UTP nick-end labeling was used to detect apoptosis of the intestinal epithelial cells. The expression level of Ki-67 represented the proliferation of colonic epithelial cells and was detected by immunohistochemistry. The expression levels of inflammation cytokines IL-6, TNF-α and CXCL-1 were examined in colon tissues using real-time PCR and ELISA kits.RESULTS Compared with the DSS group, mice treated with the CRH-R2 antagonist Ast2B showed greater loss of body weight, shorter colon lengths(4.90 ± 0.32 vs 6.21 ± 0.34 cm, P < 0.05), and higher DAI(3.61 ± 0.53 vs 2.42 ± 0.32, P < 0.05) and histological scores(11.50 ± 1.05 vs 8.33 ± 1.03, P < 0.05). Additionally, the Ast2B group showed increased intestinal permeability(2.76 ± 0.11 μg/mL vs 1.47 ± 0.11 μg/mL, P < 0.001), improved secretion of inflammatory cytokines in colon tissue, and reduced colonic epithelial cell proliferation(4.97 ± 4.25 vs 22.51 ± 8.22, P < 0.05). Increased apoptosis(1422.39 ± 90.71 vs 983.01 ± 98.17, P < 0.001) was also demonstrated. 目的:以多种中药挥发油作为原料,开发一款高效广谱抑菌的空气抑菌剂。方法:选取丁香、艾叶、香茅3种挥发油,采用Design-Expert 12.0中自带的Optimal(custom)试验设计进行最优混料配方筛选试验。通过采样我校饭堂空间空气浮游菌,先按软件设计配方加样抑菌孵育并计数,再拟合出孵育菌落数与各挥发油配方比例的模型方程,最后计算出最佳抑菌效能配方比例,并采用实验室试验和现场试验验证优选配方。结果:利用286组抑菌孵育数据计算得出,孵育菌落数(Y)与各挥发油配方比例之间的拟合方程为Y=17.60A+53.01B+36.76C-106.11AB-84.87AC-63.93BC+782.79A^(2)BC-1178.89AB^(2)C+413.15ABC2。丁香与艾叶、香茅挥发油抑菌的交互作用对孵育菌落数量有显著影响(p<0.05)。最终选择的丁香、艾叶、香茅挥发油比例为45.2:41.6:13.2,该配方的实验室试验和现场试验验证了其抑菌效能良好。结论:中药抑菌挥发油复配后具有明显的增效作用。该配方的空气抑菌剂抑菌效果明显,气味芳香易于被人们接受,具有良好的市场前景和推广价值。

Reactive incorporation of Ag into porous TiO2 coating and its influence on its microstructure, in vitro antibacterial efficacy and cytocompatibility

Ag-doped TiO2coatings were successfully fabricated on Ti using plasma electrolytic oxidation(PEO) in the electrolytes containing Ag or Ag2O nanoparticles(NPs). The results showed that the different Ag sources resulted in significant variations of morphology, composition and chemical state. The thermal decomposition of Ag2O during PEO process contributed to the formation of Ag NPs, which might reactively incorporate into TiO2, leading to the presence of a Ca, P-rich amorphous layer between entrapped Ag NPs and the peripheral TiO2. By contrast,Ag specie tends to be present as micro-sized clusters consisting of metallic Ag NPs in the PEO coating fabricated via Ag NPs addition. The different microstructures inevitably affected the Ag ionic release behaviour and thus antibacterial capabilities and cytocompatibilities. It is indicated that the application of Ag2O NPs as Ag source is promising to fabricate Ag-doped PEO coating with both long-term antibacterial activity and acceptable cytocompatibility.
