Anti-lipopolysaccharide factors(ALFs)exhibit a potent antimicrobial activity against a broad range of bacteria,filamentous fungi,and viruses.In previous reports,seven groups of ALFs(groups A–G)were identified in penaeid shrimp.Among them,group D showed negative net charges and weak antimicrobial activity.Whether this group has antiviral function is not clear.In this study,the ALF sequences of penaeid shrimp were analyzed,and eight groups of ALF family(groups A–H)were identified.The four ALFs including MjALF-C2,MjALF-D1,MjALF-D2,and MjALF-E2 from kuruma shrimp Marsupenaeus japonicus were expressed recombinantly in Escherichia coli,and the antiviral activity was screened via injection of purified recombinant ALFs into shrimp following white spot syndrome virus(WSSV)infection.Results showed that the expression of Vp28(WSSV envelope protein)decreased significantly in the MjALF-D2-injected shrimp only.Therefore,MjALF-D2 was chosen for further study.Expression pattern analysis showed that MjAlf-D2 was upregulated in shrimp challenged by WSSV.The WSSV replication was detected in RNA,genomic DNA,and protein levels using VP28 and Ie1(immediate-early gene of WSSV)as indicators in MjALF-D2-injected shrimp following WSSV infection.Results showed that WSSV replication was significantly inhibited compared with that in the rTRX-or PBS-injected control groups.After knockdown of MjAlf-D2 in shrimp by RNA interference,the WSSV replication increased significantly in the shrimp.All these results suggested that MjALF-D2 has an antiviral function in shrimp immunity,and the recombinant ALF-D2 has a potential application for viral disease control in shrimp aquaculture.

An efficient and eco-friendly carbon nitride nanosheet(NM-g-C3N4)-catalyzed decarboxylative coupling reaction of imidazo-fused heterocycles(i.e.,imidazo[1,2-a]pyridines,benzo[d]imidazo[2,1-b]thiazole)with N-phenylglycines in dimethyl carbonate(DMC)has been developed.The toxic solvents,external oxidants,and restricted reaction conditions could be effectively avoided in this powerful and sustainable protocol.Remarkably,NM-g-C3N4 could be straightforwardly recovered by simple centrifugation and recycled and reused at least 7 times without an obvious decrease in catalytic activity.

What is already known about this topic?Oncomelania hupensis(O.hupensis)is the only intermediate host of Schistosoma japonica.The distribution of O.hupensis is affected by a series of climate,geographical,sand ocioeconomic factors,which reflect the risk areas of schistosomiasis.What is added by this report?There were certain geographical aggregations in the potential high distribution areas of O.hupensis,which were mainly distributed in Poyang Lake area,Dongting Lake area,and the middle and the lower reaches of the Yangtze River.What are the implications for public health practice?Monitoring and forecasting the distribution of O.hupensis is conducive to improving the early warning capabilities of the potential risk of schistosomiasis transmission and progressively promoting the elimination of schistosomiasis nationwide.

BACKGROUND Acute gastrointestinal bleeding is an emergency condition that can lead to significant morbidity and mortality.Embolization is considered the preferred therapy in the treatment of lower gastrointestinal bleeding when it is unrealistic to perform the surgery or vasopressin infusion in this population.Treatment of acute lower gastrointestinal(GI)bleeding(any site below the ligament of Treitz)using this technique has not reached a consensus,because of the belief that the risk of intestinal infarction in this condition is extremely high.The purpose of the study is to evaluate the effectiveness and safety of this technique in a retrospective group of patients who underwent embolization for acute lower GI bleeding.AIM To evaluate the efficacy and safety of super-selective arterial embolization in the management of acute lower GI bleeding.METHODS A series of 31 consecutive patients with angiographically demonstrated small intestinal or colonic bleeding was retrospectively reviewed.The success rate and complication rate of super-selective embolization were recorded.RESULTS Five out of thirty-one patients(16.1%)could not achieve sufficiently selective catheterization to permit embolization.Initial control of bleeding was achieved in 26 patients(100%),and relapsed GI bleeding occurred in 1 of them at 1 wk after the operation.No clinically apparent bowel infarctions were observed in patients undergoing embolization.CONCLUSION Super-selective embolization is a safe therapeutic method for acute lower GI bleeding,and it is suitable and effective for many patients suffering this disease.Importantly,careful technique and suitable embolic agent are essential to the successful operation.

Transcriptional control by HNF-1:Emerging evidence showing its role in lipid metabolism and lipid metabolism disorders

The present review focuses on the roles and underlying mechanisms of action of hepatic nuclear factor-1(HNF-1)in lipid metabolism and the development of lipid metabolism disorders.HNF-1 is a transcriptional regulator that can form homodimers,and the HNF-1αand HNF-1βisomers can form heterodimers.Both homo-and heterodimers recognize and bind to specific cis-acting elements in gene promoters to transactivate transcription and to coordinate the expression of target lipid-related genes,thereby influencing the homeostasis of lipid metabolism.HNF-1 was shown to restrain lipid anabolism,including synthesis,absorption,and storage,by inhibiting the expression of lipogenesis-related genes,such as peroxisome proliferator-activated receptorγ(PPARγ)and sterol regulatory element-binding protein-1/2(SREBP-1/2).Moreover,HNF-1 enhances the expression of various genes,such as proprotein convertase subtilisin/kexin type 9(PCSK9),glutathione peroxidase 1(GPx1),and suppressor of cytokine signaling-3(SOCS-3)and negatively regulates signal transducer and activator of transcription(STAT)to facilitate lipid catabolism in hepatocytes.HNF-1 reduces hepatocellular lipid decomposition,which alleviates the progression of nonalcoholic fatty liver disease(NAFLD).HNF-1 impairs preadipocyte differentiation to reduce the number of adipocytes,stunting the development of obesity.Furthermore,HNF-1 reduces free cholesterol levels in the plasma to inhibit aortic lipid deposition and lipid plaque formation,relieving dyslipidemia and preventing the development of atherosclerotic cardiovascular disease(ASCVD).In summary,HNF-1 transcriptionally regulates lipid-related genes to manipulate intracorporeal balance of lipid metabolism and to suppress the development of lipid metabolism disorders.

Background:Oncomelania hupensis is only intermediate snail host of Schistosomajaponicum,and distribution of 0.hupensis is an important indicator for the surveillance of schistosomiasis.This study explored the feasibility of a random forest algorithm weighted by spatial distance for risk prediction of schistosomiasis distribution in the Yangtze River Basin in China,with the aim to produce an improved precision reference for the national schistosomiasis control programme by reducing the number of snail survey sites without losing predictive accuracy.Methods:The snail presence and absence records were collected from Anhui,Hunan,Hubei,Jiangxi and Jiangsu provinces in 2018.A machine learning of random forest algorithm based on a set of environmental and climatic variables was developed to predict the breeding sites of the 0.hupensis intermediated snail host of S.japonicum.Different spatial sizes of a hexagonal grid system were compared to estimate the need for required snail sampling sites.The predictive accuracy related to geographic distances between snail sampling sites was estimated by calculating Kappa and the area under the curve(AUC).Results:The highest accuracy(AUC=0.889 and Kappa=0.618)was achieved at the 5 km distance weight.The five factors with the strongest correlation to 0.hupensis infestation probability were:(1)distance to lake(48.9%),(2)distance to river(36.6%),(3)isothermality(29.5%),(4)mean daily difference in temperature(28.1%),and(5)altitude(26.0%).The risk map showed that areas characterized by snail infestation were mainly located along the Yangtze River,with the highest probability in the dividing,slow-flowing river arms in the middle and lower reaches of the Yangtze River in Anhui,followed by areas near the shores of China's two main lakes,the Dongting Lake in Hunan and Hubei and the Poyang Lake in Jiangxi.Conelusions:Applying the machine learning of random forest algorithm made it feasible to precisely predict snail infestation probability,an approach that could improve the sensitivity of the

What is already known about this topic?Over 90%of Oncomelania snails,the only intermediate host of Schistosoma japonicum,are distributed in the middle and low reaches of Yangtze River.Flooding can extend the distribution of Oncomelania snails and hence accelerate the transmission of schistosomiasis.What is added by this report?Although the dispersal of Oncomelania snails was negligible in north Poyang Lake after flooding in 2020,2 samples of cattle feces with Schistosoma egg and 2 infected snails samples were indeed found.All four risk sites were distributed in Lushan County.Cattle feces were observed in the six out of seven field sites in Lushan County.What are the implications for public health practice?The present national control strategy focusing on control of infection source should be reinforced in Lushan and other schistosomiasis endemic areas.Overlaps of infected snails and cattle feces with Schistosoma egg were not observed,which called for intensive surveillance in Lushan County.

China has accumulated multiple practices and experiences in building and enhancing malaria surveillance and response system. As China’s engagement into global health has gathered stronger momentum than ever, China together with the Swiss Tropical and Public Health Institute and WHO has organised five sessions of the International Forum on Surveillance-Response System Leading to Tropical Diseases Elimination during 2012–2020, in which malaria elimination has always been one of the hottest topics. In this study, the roles of international network on the surveillance and response system were explored to achieve a global malaria-free goal. China’s approach to malaria elimination has demonstrated significance of global collaboration on taking joint prevention and control, and building a worldwide institutional-based network.
