In vivo protective effect of late embryogenesis abundant protein(ApSK3 dehydrin)on Agapanthus praecox to promote post-cryopreservation survival

Dehydrins(DHNs),as members of the late embryogenesis abundant protein family,play critical roles in the protection of seeds from dehydration and plant adaptation to multiple abiotic stresses.Vitrification is a basic method in plant cryopreservation and is characterized by forming a glassy state to prevent lethal ice crystals produced during cryogenic storage.In this study,ApSK3 type DHN was genetically transformed into embryogenic calluses(EC)of Agapanthus praecox by overexpression(OE)and RNA interference(RNAi)techniques to evaluate the in vivo protective effect of DHNs during cryopreservation.The cell viability showed a completely opposite trend in OE and RNAi cell lines,the cell relative death ratio was decreased by 20.0%in ApSK3-OE EC and significantly increased by 66.15%in ApSK3-RNAi cells after cryopreservation.Overexpression of ApSK3 increased the content of non-enzymatic antioxidants(AsA and GSH)and up-regulated the expression of CAT,SOD,POD,and GPX genes,while ApSK3-RNAi cells decreased antioxidant enzyme activities and FeSOD,POD,and APX genes expression during cryopreservation.These findings suggest that ApSK3 affects ROS metabolism through chelating metal ions(Cu^(2+)and Fe^(3+)),alleviates H_(2)O_(2)and OH·excessive generation,activates the antioxidant system,and improves cellular REDOX balance and membrane lipid peroxidation damage of plant cells during cryopreservation.DHNs can effectively improve cell stress tolerance and have great potential for in vivo or in vitro applications in plant cryopreservation.

The primary chemical components of Astragalus membranaceus include polysaccharides,saponins,flavonoids,and amino acids.Recent studies have shown that Astragalus membranaceus has multiple functions,including improving immune function and exerting antioxidative,anti-radiation,anti-tumor,antibacterial,antiviral,and hormone-like effects.Astragalus membranaceus and its extracts are widely used in clinical practice because they have obvious therapeutic effects against various autoimmune diseases and relatively less adverse reaction.Multiple sclerosis(MS)is an autoimmune disease of central nervous system(CNS),which mainly caused by immune disorder that leads to inflammatory demyelination,inflammatory cell infiltration,and axonal degeneration in the CNS.In this review,the authors analyzed the clinical manifestations of MS and experimental autoimmune encephalomyelitis(EAE)and focused on the efficacy of Astragalus membranaceus and its chemical components in the treatment of MS/EAE.

Metabolomic characteristics of spontaneously hypertensive rats under chronic stress and the treatment effect of Danzhi Xiaoyao Powder, a traditional Chinese medicine formula

Objective: Numerous studies have demonstrated the close relationship between chronic stress and blood pressure(BP). Hypertensive subjects exhibit exaggerated reactions to stress, especially higher BP. The mechanisms by which stress affects pre-existing hypertension still need to be explored. Danzhi Xiaoyao Powder(DP), a historical traditional Chinese medicine formula, is a promising treatment for BP control in hypertensive patients under stress. The present study investigated the metabolomic disruption caused by chronic stress and the treatment effect and mechanism of DP.Methods: Spontaneously hypertensive rats(SHRs) were subjected to chronic restraint stress(CRS) for 4 weeks. BP was measured via the tail-cuff method, and anxiety-like behavior was quantified using the elevated-plus-maze test. Meanwhile, DP was administered intragastrically, and its effects were observed. Global metabolomic analysis was performed using ultra-high-performance liquid chromatography-quadrupole time-of-flight mass spectrometry, followed by multivariate statistical analysis to detect differential metabolites and pathways.Results: DP alleviated the CRS-induced increase in BP and anxiety-like behavior. Systematic metabolic differences were found among the three study groups. A total of 29 differential plasma metabolites were identified in both positive-and negative-ion modes. These metabolites were involved in triglyceride metabolism, amino acid(phenylalanine, tryptophan, and glycine) metabolism, and steroid hormone pathways.Conclusion: These findings expose the metabolomic disturbances induced by chronic stress in SHRs and suggest an innovative treatment for this disorder.

Adsorptive interaction between typical VOCs and various topological zeolites:Mixture effect and mechanism

Adsorption is one of the most feasible and effective methods to alleviate the volatile organic compounds(VOCs)pollution.However,the mixture effect and mechanism for competitive adsorption of VOCs on zeolites are barely addressed.In this study,toluene,acetone,and ethyl acetate as prevalent VOCs species were removed by four potential zeolites(13X,USY,Beta,ZSM-5)in both single-and multi-component systems.The structure-property relationship between adsorbate-adsorbent pairs was revealed by X-ray diffraction,scanning electron microscopy,energy dispersive spectrometer,X-ray fluorescence,N_(2) adsorption and density function theory calculation.The molecular polarity and volatility of VOCs species played key roles in adsorption and the dynamic uptakes were generally listed as follows:ethyl acetate>toluene>acetone.As for the above VOCs mixtures,13X zeolite selectively adsorbed oxygenated VOCs rather than toluene.In contrast,USY exhibited a preference to trap toluene.Ethyl acetate could be readily enriched by ZSM-5 and Beta selectively.The possible explanations and implications are discussed based on the subtle change in electron density.The results obtained are vital for understanding the mixture effect of VOCs adsorption and may guide the selection of proper adsorbent for real applications.

Natural products have been an unprecedented starting point for drug and agrochemical discovery.They represent the only validated source of chemical diversity capable of delivering a sustainable pipeline for novel drug candidates.As of 1990,80%of clinically used drugs were natural products or natural product-inspired or semi-synthetic derivatives.From 1994 to 2007,almost half of the new chemical entities approved by FDA were based on natural products[1].By the end of 2013,natural products or their inspired compounds comprised about 40%of the FDA approved drugs[2].Streptomyces are by far the most prolific natural product producing organisms and as such this genus has been the research subjects for lead compound discovery for decades.

An Information Entropy-Based Methodology to Construct the Avulsion Threshold in the Tail Reach of the Estuarine Alluvial Plain

Channel avulsion is a natural phenomenon that occurs abruptly on alluvial river deltas,which can affect the channel stability.The causes for avulsion could be generally categorized as topography-and flood-driven factors.However,previous studies on avulsion thresholds usually focused on topography-driven factors due to the centurial or millennial avulsion timescales of the world’s most deltas,but neglected the impacts of flood-driven factors.In the current study,a novel demarcation equation including the two driven factors was proposed,with the decadal timescale of avulsion being considered in the Yellow River Estuary(YRE).In order to quantify the contributions of different factors in each category,an entropy-based methodology was used to calculate the contributing weights of these factors.The factor with the highest weight in each category was then used to construct the demarcation equation,based on avulsion datasets associated with the YRE.An avulsion threshold was deduced according to the demarcation equation.This avulsion threshold was then applied to conduct the risk assessment of avulsion in the YRE.The results show that:two dominant factors cover respectively geomorphic coefficient representing the topography-driven factor and fluvial erosion intensity representing the flood-driven factor,which were thus employed to define a two dimensional mathematical space in which the demarcation equation can be obtained;the avulsion threshold derived from the equation was also applied in the risk assessment of avulsion;and the avulsion threshold proposed in this study is more accurate,as compared with the existing thresholds.

Precision of the Chinese Space Station Telescope(CSST)stellar radial velocities

The Chinese Space Station Telescope(CSST)spectroscopic survey plans to deliver high-quality low-resolution(R>200)slitless spectra for hundreds of millions of targets down to a limiting magnitude of about 21 mag,covering a large survey area(17500 deg^(2))and a wide wavelength range(255-1000 nm by three bands GU,GV,and GI).In this work,we use empirical spectra of the Next Generation Spectral Library to simulate the CSST stellar spectra at R=250,and investigate their capabilities in measuring radial velocities.We find that velocity uncertainties depend strongly on effective temperature,weakly on metallicity for only FGK stars,and hardly on surface gravity.It is possible to deliver stellar radial velocities to a precision of about 3 kms^(-1)for AFGKM stars,and about 10 km s^(-1)for OB stars,at signal-to-noise ratio(SNR)of 100.Velocity uncertainties using single GU/GV/GI band spectra are also explored.Given the same SNR,the GU band performs best,the GV band the second best,and then the GI band.The effects of spectral normalization and the imperfect template on velocity measurements are investigated and found to be very weak.The uncertainties caused by wavelength calibration are considered and found to be moderate.Given the possible precision of radial velocities,the CSST spectroscopic survey can enable interesting science such as searching for hyper-velocity stars.Limitations of our results are also discussed.

A star-based method for precise wavelength calibration of the Chinese Space Station Telescope(CSST) slitless spectroscopic survey

The Chinese Space Station Telescope(CSST)spectroscopic survey aims to deliver high-quality low-resolution(R>200)slitless spectra for hundreds of millions of targets down to a limiting magnitude of about 21 mag,distributed within a large survey area(17500 deg2)and covering a wide wavelength range(255-1000 nm by three bands GU,GV,and GI).As slitless spectroscopy precludes the usage of wavelength calibration lamps,wavelength calibration is one of the most challenging issues in the reduction of slitless spectra,yet it plays a key role in measuring precise radial velocities of stars and redshifts of galaxies.In this work,we propose a star-based method that can monitor and correct for possible errors in the CSST wavelength calibration using normal scientific observations,taking advantage of the facts that(ⅰ)there are about ten million stars with reliable radial velocities now available thanks to spectroscopic surveys like LAMOST,(ⅱ)the large field of view of CSST enables efficient observations of such stars in a short period of time,and(ⅲ)radial velocities of such stars can be reliably measured using only a narrow segment of CSST spectra.We demonstrate that it is possible to achieve a wavelength calibration precision of a few km s^(-1) for the GU band,and about 10 to 20 kms^(-1) for the GV and GI bands,with only a few hundred velocity standard stars.Implementations of the method to other surveys are also discussed.

Anxiety symptoms in patients with Mayer-Rokitansky-Küster-Hauser syndrome:a cross-sectional study

Background:As a congenital malformation that results in infertility and an inability to have vaginal intercourse,Mayer-Rokitansky-Küster-Hauser(MRKH)syndrome places a considerable psychological burden on patients,which results in anxiety symptoms.However,only single case studies or a few small to medium-sized cross-sectional studies were identified to focus on anxiety symptoms in MRKH patients.Thus,the aim of this study was to explore the status of anxiety symptoms and the related factors in patients with MRKH syndrome.Methods:This cross-sectional study involving 141 patients with MRKH syndrome and 178 healthy women was conducted from January 2018 to December 2018.All participants were required to complete a demographic questionnaire and the Generalized Anxiety Disorder 7-item scale(GAD-7),Patient Health Questionnaire-9,Eysenck Personality Questionnaire-Revised,Short Scale for Chinese,and Chinese Version of the Female Sexual Function Index.The main outcome was the anxiety symptoms measured by the GAD-7.Main outcome was compared between the MRKH syndrome group and the healthy control group.Then,we explored the related factors by comparing patients with and without anxiety symptoms.Results:Of the respondents,24.1%experienced moderate to severe anxiety symptoms.Patients with MRKH syndrome manifested more severe anxiety symptoms than healthy women.Negative self-evaluation of femininity(odds ratio[OR]2.706,95%confidence interval[CI]1.010-7.247),neurotic personality traits(OR 1.100,95%CI 1.029-1.175),and coexisting depressive symptoms(OR 4.422,95%CI 1.498-13.049)were more prevalent in anxious patients.Conclusion:The findings stress the importance of anxiety symptom screening in MRKH patients and identify patients at risk of anxiety symptoms,providing a possible basis for future intervention.

Evolutionary Pattern and Effect of Administrative Division Adjustment During Urbanization of China:Empirical Analysis on Multiple Scales

The adjustment of administrative divisions is one of the important factors guiding China's urbanization, which has profound economic and social effects for regional development. In this paper, we comprehensively describe the process of the adjustment of administrative divisions at provincial and municipal levels in China and summarize the effects on the basic structure and patterns of the spatial development. We quantitatively assess the effects on fields such as urbanization and social economy through the use of multidimensional scaling. The results show that: 1) Upgrading county to municipality(or city-governed district) is the main way of adjusting the administrative divisions. It is also an important factor in the spatial differentiation of interprovincial urbanization. China's population urbanization can be divided into four patterns including interprovincial migration, provincial migration, natural growth, and growth caused by the adjustment of administrative divisions, which is also the main reason for the increased Chinese urbanization rate at the provincial level. 2) Taking the city of Beijing as an example, we generalize five adjustment patterns made to administrative divisions: the set-up of sub-districts, the set-up of regional offices, the upgrading of townships to sub-districts, the upgrading of townships to towns, and the set-up of towns and the addition of new regional offices. We summarize the municipal urban spatial structure, including the sub-district office area in the central urban area, the regional office area in the new urban area, the mixed area of villages, towns, and sub-district offices in the suburb area, and the township area in the outer suburb area. 3) The adjustment of administrative divisions triggers a significant circulative accumulation effect, resulting in the spatial locking of population and industrial agglomeration.
