An efficient and reproducible procedure for clonal multiplication through in vitro culture of Rauwolfia serpentina (L.) Benth. (Sarpagandha) is standardized. The different explants like leaf and stem were used for callus induction and regeneration of the complete plantlets. High-frequency callusing was induced in leaf and stem explant of Rauwolfia serpentina on modified Murashige and Skoog (MS) medium supplemented with 2.5 mg/l 2,4-D. Maximum regeneration of shoots from callus (90%) was observed in MS medium supplemented with 0.2 mg/l NAA and 1.5 mg/l BA. However direct regeneration (96%) was recorded best in MS medium supplemented with BAp 2.5 mg/l. The maximum number of shoots per explant (6.5) was also highest in this phyto-hormone combination. Higher induction of root (100%) was observed in MS medium supplemented with NAA 0.5 mg/l. The rooted plantlets were successfully established in the field.

Benefits of Simple Exchange Transfusion in Sickle Cell Disease (HbSS) with Vaso-Occlusive Crisis Not Responding to Standard Therapy

Sickle cell disease is an autosomal recessive genetic disease. Vaso occlusive crisis (VOC) is frequently seen in such patients. painful VOC is usually recurrent, of variable severity due to many factors and its management poses important challenge in the clinical practice. Few patients do not respond to standard therapies and continue to suffer severe pain for prolonged period or land to serious life threatening situation. The red cell exchange by aphaeresis is presumed to be one efficient alternative in this situation which can reduce the level of HbS below 40% - 50%. However, it is costly and not available everywhere. Both circumstances are common in our state where incidence of sickle cell disease is quite high. in such situations simple red cell exchange i.e. removing 1 unit (350 ml) of blood manually (by phlebotomy) and replacement with one unit normal red cell is effective. All of our four cases of SCA with severe acute VOC, are not responding to standard therapy but responded efficiently to this simple red cell exchange transfusion. Our present observation may pave the way of one simple, affordable, and effective measure to reduce the pain of severe acute VOC not responding to standard therapy. Moderate reduction of HbS by 8% - 14% by simple red cell exchange transfusion was associated with relief of pain of acute VOC;a new observation was reported in all our 4 cases which need to be validated by larger controlled studies.

ATpase 6/8 gene （842 bp） of mitochondrial DNA was sequenced in Labeo rohita samples （n = 253） collected from nine rivers belonging to four river basins; indus, Ganges, Brahmaputra and Mahanadi. Analysis revealed 44 haplotypes with high haplotype diversity （Hd） 0.694 and low nucleotide diversity （π） 0.001. The within population variation was larger （83.44%） than among population differences （16.56%）. The mean Fsv value （0.166; p 〈 0.05） for overall populations revealed moderate level of genetic structuring in the wild L. rohita populations. The haplotype network presented a single clade for wild L. rohita population, from different rivers. Negative values for Fu＇s index （Fs）, mismatch distribution analysis indicated period of expansion in L. rohita population. The time after recent expansion was estimated for each population, between 0.042 to 0.167 mya. The pattern of isolation by Distance （iBD） was not significant （r = -0.113, p 〈 0.287）, when all the sampling locations were compared （Mantel test）, however, when an outlier （indus, Brahmaputra and Mahanadi） was removed from the whole population set, a clear positive correlation between pairwise Fsv and geographic distance （Km） was seen. The analysis of data demonstrated that ATpase6/8 gene polymorphism is a potential marker to understand genetic population structure of wild L. rohita existing in different rivers. The study identified population substructure in wild L. rohita with common ancestral origin [Current Zoology 60 （4）： 460--471, 2014].

The present work describes microstructure, texture, mechanical and ballistic properties correlation of a hot rolled and peak aged AA-7017 alloy plate at surface and centre. Both the microstructures and textures are different on the surface and centre of the plate. The surface of the plate shows recrystallized grains and a weak over all texture. The centre of the plate displays elongated grains and a sharp texture. Tensile properties, hardness and impact toughness are evaluated at surface and centre of the plate. it is observed that strength and hardness is high at centre, whereas ductility and impact toughness is more at the surface. Ballistic properties of the plate at centre and surface are measured by impacting against two different 7.62 mm deformable projectiles. The plates impacted on the surface shows better ballistic resistance. Ballistic performance of the plate at surface and centre has been correlated with the microstructure, texture and mechanical properties.

The present experiment was carried out to evaluate the effects of vitamin E and C supplementation on the antioxidant status of broiler breeder hens during summer. Two hundred seventy broiler breeder birds (coloured) were divided into nine groups with three replicates having 10 birds in each, and were supplemented two levels of antioxidant vitamins E (250 mg or 500 mg/kg) or C (200 mg or 400 mg/kg) alone or in combinations for a period of 8 weeks. All the vitamin supplemented groups showed significantly lowered malondialdehyde (MDA) level, higher activities of superoxide dismutase (SOD), catalase (CAT) enzymes and higher Ferric Reducing Antioxidant power (FRAp) activities in erythrocytes than control. However, supplementation with combination of vitamins proved to be better than individual supplementation. it could be concluded that the combination of vitamins C and E at lower level (@ 200 mg/kg and 250 mg/kg, respectively) had higher erythrocytic antioxidant status in coloured broiler breeder hens as compared to individual vitamin supplementation during hot and humid stress.

Rural Alabama currently lacks resources to significantly impact the trend of increasing rates of diabetes. Therefore, this pilot study was designed to evaluate the effect of an education intervention on knowledge, motivation, and attitudes of participants with Type ii diabetes mellitus. The aim was to improve lifestyle and overall control of diabetes for a rural, vulnerable population. Ten participants (7 with diabetes and 3 with pre-diabetes) attended three class sessions over a two-month period with presentations focused on general diabetes information, diet, exercise, and proper management through glucose measurement and medications. A pre- and post-survey design evaluated participants’ behaviors, knowledge, motivation and attitude towards making lifestyle changes. Responses were compared pre- to post-intervention. While knowledge levels related to diabetes, diet, and exercise increased, the changes were not statistically significant. Similarly, participants expressed increased motivation towards weight loss, healthful eating, exercise and positive lifestyle choices. However, these changes were not statistically significant. This pilot study demonstrates an increase in personal knowledge, motivation, and attitude for a small group of patients with diabetes or pre-diabetes. These group classes are inexpensive and will be easy to replicate with larger groups. They demonstrate the potential to motivate patients towards positive change.
