Rac1 regulates the release of Weibel-Palade Bodies in human aortic endothelial cells

Background The release of Weibel-Palade Bodies (WPB) is a form of endothelial cell activation But the signal transduction pathway leading to WPB release is not yet defined We hypothesized that small G-protein rac1 and reactive oxygen species (ROs) mediate the ligand induced release of Weibel-Palade Bodies Methods We tested this hypothesis by using wild-type and mutant adenoviral rac1 expression vectors, and by manipulating the production and destruction of superoxide and hydrogen peroxide in human aortic endothelial cells (HAEC) KH*2/5DResults Thrombin (1 0 Unit, 30 min) induced the increase of WPB release by 3 7-fold in HAEC, and that H 2O 2 (0 1 mmol/L, 30 min) induced by 4 5-fold These results correlated with thrombin-stimulated activation of rac-GTP binding activity by 3 5-fold, and increase of ROs production by 3 4-fold The dominant negative adenoviral rac-N17 gene transfer dramatically inhibited the release of WPB by 64 2% (control) and 77 3% (thrombin-stimulation), and decreased ROs production by 65 5% (control) and 83 6% (thrombin-stimulation) compared with non-infected cells, respectively Anti-oxidants, catalase and N-acetyl-cysteine significantly decreased the release of WPB by 34% and 79% in control cells, and further decreased by 63 6% and 46 7% in rac-N17 transferred cells compared with non-infected cells We also confirmed that rac1 was located upstream of ROs in the WPB release pathway KH*2/5DConclusions small G-protein rac1 medicates ligand-induced release of Weibel-Palade Bodies in human aortic endothelial cells, and the signal pathway of WPB release is a rac1-dependent ROs regulating mechanism

BACKGROUND Pancreaticoduodenectomy(PD)is a technically complex operation,with a re-latively high risk for *** ability to rescue patients from post-PD complications is as a recognized quality *** protocols were instituted at our low volume facility in the year *** To document the rate of rescue from post-PD complications with tailored protocols in place as a measure of *** A retrospective audit was performed to collect data from patients who experienced major post-PD complications at a low volume pancreatic surgery unit in Trinidad and Tobago between January 1,2013 and June 30,***-dardized definitions from the International study Group of Pancreatic surgery were used to define post-PD complications,and the modified Clavien-Dindo classification was used to classify post-PD *** Over the study period,113 patients at a mean age of 57.5 years(standard deviation[sD]±9.23;range:30-90;median:56)underwent PDs at this *** complications were recorded in 33(29.2%)patients at a mean age of 53.8 years(sD:±7.9).Twenty-nine(87.9%)patients who experienced major morbidity were salvaged after aggre-ssive treatment of their ***(3.5%)died from bleeding pseudoaneurysm(1),septic shock secondary to a bile leak(1),anastomotic leak(1),and myocardial infarction(1).There was a significantly greater salvage rate in patients with American society of Anesthesiologists scores≤2(93.3%vs 25%;P=0.0024).CONCLUsION This paper adds to the growing body of evidence that volume alone should not be used as a marker of quality for patients requiring *** low volumes at our facility,we demonstrated that 87.9%of patients were rescued from major *** attributed this to several factors including development of rescue protocols,the competence of the pancreatic surgery teams and continuous,and adaptive learning by the entire institution,cul-minating in the development of tailored peri-pancreatectomy protoc

A database of modeled gridded dry deposition velocities for 45 gaseous species and three particle size ranges across North America

The dry deposition process refers to the flux loss of an atmospheric pollutant due to uptake of the pollutant by the earth’s *** deposition flux of a chemical species is typically calculated as the product of its surface-layer concentration and its dry deposition velocity(V_(d)).Field measurement based V_(d) data are very scarce or do not exist for many chemical species considered in chemistry transport *** the present study,gaseous and particulate dry deposition schemes were applied to generate a database of hourly V_(d) for 45 gaseous species and three particle size ranges for two years(2016–2017)at a 15 kmby 15 km horizontal resolution across North *** V_(d) of the 45 gaseous species ranged fromsec across the whole domain,with chemical species-dependentmedian(mean)values being in the range of 0.018–1.37 cm/sec(0.05–1.43 cm/sec).The spatial distributions of the two-year average V_(d) showed values higher than 1–3 cm/sec for those soluble and reactive species over certain land *** species have the highest V_(d) over water surfaces,while insoluble but reactive species have the highest V_(d) over *** V_(d) of PM_(2.5) across the whole domain ranged from 0.039 to 0.75 cm/sec with median(mean)value of 0.18(0.20)cm s^(−1),while the mean V_(d) for PM_(2.5)–10 is twice that of PM_(2.5).Uncertainties in the modeled V_(d) are typically on the order of a factor of 2.0 or larger,which needs to be considered when applying the dataset in other studies.

seasonal Behaviour of Mesoscale Eddy Trajectories in the North Indian Ocean Based on satellite Altimetry

In the north Indian Ocean (NIO), maps of sea level anomaly from satellite altimetry were analysed from January-1995 to December-2000. The study attempted to trace the trajectories of the individual mesoscale anomalies manually and to understand seasonal changes in terms of phase speed. Mesoscale anomalies are detected as concentric circular shapes and diameters of ~90 km to 600 km and the minimum 30 days life cycle. Relatively higher eddy kinetic energy was noticed in the northwestern region of the NIO. Individual mesoscale anomalies, namely positive (warm, anticyclonic eddies) and negative (cold, cyclonic eddies) showing travelling direction westward in the NIO basins. In autumn, the number of negative anomalies detected is more than positive anomalies and vice versa during summer. The westward propagating positive (negative) anomalies in the Arabian sea start appearing in winter (spring) along (away from) the west coast of India and west of 65°E;individual anomalies move to the west in spring/summer/autumn and collide along somalia’s & Arabian coast. A group of positive (negative) anomalies trajectories appears as a tail at the southern tip of India are located west of the Laccadive ridge in winter (summer to autumn) associated with LH (LL). The Bay of Bengal (BB) trajectories show southwestward in northern BB, westward in central BB and northwestward in southern BB;individual anomalies are appearing along the west coast of Andaman & Nicobar ridge. The zonal phase speed decreases away from the equator, and the magnitude varies longitudinally in each season in the form of a wave-like pattern propagating westward from autumn to summer;the life cycle of the wave is almost 365 days (a year). The theoretical phase speed of the first mode of the baroclinic Rossby waves is quite similar to that of averaged zonal speed. Therefore mesoscale anomalies (eddies) are embedded into the large waves like phenomenon (Rossby waves), responsible for creating high variability and EKE

Alkaline earth metal （Mg,Ca,sr and Ba）-doped Mo-V-sb-O x catalysts,prepared by a dry-up method,have been investigated for their catalytic performance in the oxidation of propane under different reaction *** catalysts have been characterized by N2 adsorption-desorption,temperature-programmed desorption （TPD） of NH3,sEM and *** of water vapor on the catalytic performance,particularly on the selectivities to acetic acid and acrylic acid,has also been *** selectivity to acrylic acid was improved significantly by the doping of alkaline earth metals to Mo-V-sb-O x *** surface acidic sites of the catalyst decreased with the doping of the catalyst with alkaline earth metals,which ultimately was found to be beneficial for obtaining high selectivity to acrylic *** catalytic activity and product selectivities were found to be influenced by the reaction temperature,C3H8/O2 ratio and space velocity.A significant improvement in the selectivity to acrylic acid has also been observed by the addition of water vapor in the feed of propane and oxygen in the oxidation of propane.

A novel method of water remediation of organic pollutants and industrial wastes by solution- route processed CZTs nanocrystals

CZTs(Cu_(2)Znsns_(4)),a P-type semiconductor with a direct bandgap(1.2-1.7eV),earth-abundant,non-toxic,and has a large absorption coefficient makes it extremely useful in optoelectronics and light-harvesting *** this work,CZTs is prepared by an ingenious,cost-effective colloidal route using the‘hotinjection’method with the usage of different *** XRD and Raman spectroscopy shows the single-phase highly crystalline CZTs nanoparticles with kesterite *** TEM results show that the size of CZTs nanoparticles is about 2-5 nm and monodispersity is confirmed by DLs(Dynamic Light scattering).FTIR confirms the presence of different ligands used in CZTs *** Uv-vis absorption shows the direct bandgap of *** contact angle study shows the hydrophobic nature of as-synthesized CZTs nanoparticles which were further ligand exchanged with L-cysteine hydrochloride to make it hydrophilic to study the photocatalytic degradation activity of organic pollutants and industrial waste in the *** photocatalysis experiments were performed under two conditions:(i)under bare sunlight(Intensity~900 W/m^(2))(ii)focussing the sample under the sunlight via converging lens(1800 W/m^(2)).The photocatalytic efficiencies were then compared and the best photocatalytic efficiency achieved under sunlight was 98.4%for organic pollutants and 75%for industrial waste via converging lens while the corresponding efficiencies with bare sunlight were 98.1% and 73% *** the best of the author’s knowledge,a rapid and highly efficient photocatalysis of CZTs NPs employing a converging lens for water-remediation without the usage of noble&transition-metals has been reported for the first time.

Valorisation of fruit waste for enhanced exopolysaccharide production by Xanthomonas campestries using statistical optimisation of medium and process

The aim of this research work was to enhance the fermentative production of exopolysaccharide(EPs)with mixed fruit waste as a key *** campestries was isolated from spoiled orange and its EPs production efficiency was determined using EPs *** to optimisation,the production was 3.4 g/L,upon optimizing the medium the production improved to 9.2 g/*** medium formulation was studied by sequential addition of medium components and replacement of sugar with mixed fruit *** formulated medium,the production enhanced to 11.1 g/*** further optimisation,the response surface methodology was used,which resulted in the enhancement of production to 14.5 g/*** scale-up study was undertaken in a 10 L shake flask and *** 10 L flask EPs,production was 14.45 g/L in 100 h,whereas in 10 L fermenter it was 21.10 g/L in 60h *** optimisation studies gave nearly 500%(6.2fold)enhanced EPs production and dwindle in fermentation *** of sucrose with fruit waste extract made the process environment-friendly;omission of L-cystine and use of 50%reduced amount of sodium acetate in the medium lowered the production cost of *** developed process resulted in the utilization of fruit waste and was also useful in solving the fruit waste disposal problem.

Advanced Endoscopic Palliation of Unresectable Periampullary Carcinoma: A systematic Review

Background and Aims: Periampullary malignancies often present at an advanced unresectable stage requiring palliation of symptoms for the better quality of life. Though they require multispecialty support;role of endoscopic interventions in palliation is paramount because of the advancement in technology and increase experience of Gastroenterologist. Methods: We did extensive review of articles regarding endoscopic advances in the management of Unresectable Periampullary Malignancies. several systematic reviews, Meta analysis, and Randomized controlled trials published over the last 2 decades were thoroughly searched on PUBMED and GOOGLE sCHOLAR. Results: Advanced Endoscopic procedures have been emerging as a superior modality than conventional measures because of minimal invasiveness and greater clinical and technical success. Conclusion: Periampullary malignancies often present at an advanced unresectable stage requiring palliation of symptoms for the better quality of life. several endoscopic interventions have already been established as the standard of care in palliation of symptoms of unresectable periampullary malignancies, we can say with confidence that with growing advances in EUs, advancement in technologies and increasing experiences, the role of gastroenterologist will be pivotal in these groups of patients.

Genetic Diversity and Population structure in Landraces and Improved Rice Varieties from India

A set of 50 rice genotypes comprising landraces, local selections, and improved varieties were characterized using simple sequence repeat（ssR） and inter simple sequence repeat（IssR） markers to study genetic diversity and population structure. Following unweighted pair group method with arithmetic mean based clustering using binary data of polymorphic markers, the genotypes were grouped into 5 clusters and 11 sub-clusters, whereas population structure analysis separated 50 rice genotypes into 5 sub-populations. Grouping of rice genotypes showed better resemblance with the pedigree information of the genotypes. Both genetic diversity and population structure analysis separated majority of the improved varieties from landraces and local selections. some of the ssR markers amplified unique alleles which were specific to a particular genotype and could distinguish them from the rest. The results indicate that these rice genotypes exhibit a higher genetic diversity and can be very useful in rice improvement program.
