Performance evaluation of NeuMoDx 96 system for hepatitis B and C viral load

BACKGROUND Hepatitis B virus(HBV)and hepatitis C virus(HCV)viral load(VL)estimation is essential for the management of both HBV and HCV *** to a longer turnaround time for VL estimation,many patients drop out from the cascade of *** achieve the global goals of reducing morbidity and mortality due to HBV/HCV and moving towards their elimination by 2030,molecular diagnostic platforms with faster and random(*** sample)access are *** To evaluate the performance of the recently launched NeuMoDx 96 random access system with the conventional COBAS^(■)AmpliPrep/COBAS TaqMan system for HBV and HCV VL *** Archived once-thawed plasma samples were retrieved and tested on both *** between the assays was determined by linear regression and Bland-Altman *** study included samples from 186 patients,99 for HBV of which 49 were true infected HBV cases(hepatitis B surface antigen,antihepatitis B core antibody,and HBV DNA-positive)and 87 for HCV assay in which 39 were true positives for HCV infection(anti-HCV and HCV RNA-positive).RESULTS The median VL detected by NeuMoDx for HBV was 2.9(interquartile range[IQR]:2.0-4.3)log_(10)IU/mL and by COBAS it was 3.70(IQR:2.28-4.56)log_(10)IU/mL,with excellent correlation(R2=0.98).In HCV,the median VL detected by NeuMoDx was 4.9(IQR:4.2-5.4)log_(10)IU/mL and by COBAS it was 5.10(IQR:4.07-5.80)log_(10)IU/mL with good correlation(R2=0.96).CONCLUSION The overall concordance between both the systems was 100%for both HBV and HCV VL ***,no genotype-specific bias for HBV/HCV VL quantification was seen in both the *** findings reveal that NeuMoDx HBV and HCV quantitative assays have shown overall good clinical performance and provide faster results with 100%sensitivity and specificity compared to the COBAS AmpliPrep/COBAS TaqMan system.

A Retrospective Renal Study from a Lupus Vasculitis Clinic

Aim: In July 2009, a combined Renal Rheumatology Lupus Vasculitis (RRLV) clinic, the first of its kind for adult patients in Australia, was started at Royal Brisbane & Women’s Hospital. This is an audit of progression of renal disease to assess if patients attending this clinic had comparable results to published studies of similar cohorts with lupus Nephritis (LN) and vasculitis. Methods: We conducted a retrospective audit of all the patients who attended this clinic from July 2009 to October 2013. There were 33 patients followed up in the vasculitis group and 36 in the LN group. Patients with other connective tissue disorders were excluded from the analysis as the numbers were insignificant. Results: The mean estimated glomerular filtration rate of vasculitis and LN patients improved from 32.06 to 45.82 ml/min/1.73m2 and 62.42 to 65.53 ml/min/1.73m2 respectively. The mean urine protein/creatinine ratio of vasculitis and LN patients improved from 420 to 85 and 406 to 70 respectively. No patients died in either group. One vasculitis and two LN patients required maintenance dialysis. Three LN patients underwent renal transplantation. Conclusion: The results show excellent patient and renal survival and support the concept of a combined renal rheumatology clinic in managing renal disease from systemic connective tissue disorders.

This work emphasis on the competence of(100)-oriented PMN–PT buffer layered(0.68PbMg_(1/3)Nb_(2/3)O_(3)–0.32PbTiO_(3)with Pb(Zr_(0.3)Ti_(0.7))O_(3)/PbO_(x)buffer layer)and(001)-oriented PMN–PT(0.67PbMg_(1/3)Nb_(2/3)O_(3)–0.33PbTiO_(3))for low grade thermal energy harvesting using Olsen *** analysis(based on well-reported experiments in literature)reveals that these films show colossal energy harnessing *** the films are found to have maximum harnessable energy densities(PMN–PT buffer layered:8 MJ/m^(3);PMN–PT:6.5 MJ/m^(3))in identical ambient conditions of 30–150℃and 0–600 kV/*** energy harnessing plausibility is found to be nearly five times higher than the previously reported values to date.

Objective:To assess the hepatoprotective effct of Ipomoea carnea(***)extract against antitubercular drug-induced liver toxicity in experimental ***:*** extracts(125,250 and 500 mg/kg,*** weight)were administered daily for 35 d in experimental *** toxicity was induced by combination of three antitubercular drugs(isoniazid 7.5 mg/kg,rifampicin 10 mg/kg and pyrazinamide 35 mg/kg)given orally as suspension for 35 d in *** groups received *** extracts along with antitubercular *** hepatoprotective activity was assessed using various biochemical parameters like aspartate aminotransferase,alanine aminotransferase,alkaline phosphatise and total ***,in-vivo antioxidant activities as lipid peroxidation,reduced glutathione,superoxide dismutase and catalase were measured in rat liver homogenate along with ATPase and *** biochemical observations were supplemented by histopathological ***:Obtained results demonstrated that treatment with *** extracts significantly(P<0.05-P<0.001)and dose-dependently prevented drug induced increase in serum levels of hepatic ***,*** extracts significantly(up to P<0.001)reduced the lipid peroxidation in the liver tissue and restored activities of defence antioxidant enzymes,reduced glutathione,superoxide dismutase and catalase towards normal *** of the liver tissue showed that *** extracts attenuated the hepatocellular necrosis,massive fatty changes and led to reduction in inflammatory cells ***:The results of this study strongly indicate the protective effect of *** extracts against liver injury,which may be attributed to its hepatoprotective activity,and there by scientifically support its traditional use.

Current evidence of concept analyses recommending nanotechnology for biomedical uses abounds in recent research. The area of biotechnology interfaces with nanostructures, reconfigures their composition, and alters their characteristics;which influences the dispersion of the particles, the biotransformation they cause, and their potential toxic effect. It is vital to link the idea of the lifecycle of nanostructures to the biological impacts and use methodologies to identify, estimate, and track the gradual bioprocessing of nanostructures in vivo, from a body-wide level to a nanoscopic size. This is necessary because understanding how nanostructures processing, degradation, persistence, and recycling predict potential exposure risks. The safe implementation of nanotechnology-based products in biomedical applications necessitates an extensive understanding of the recycling and transformations of nanomaterials in a living organism. Long-term fate in the body is crucial, as it governs potential environmental risks to human health. Strategies may be used to manage the long-term outcome of nanostructures in an organism since, in addition to composition, their design also affects how long they last and how easily they degrade. The lifespan of nanoparticles, a flexible and biocompatible category of nanostructures that have made it into clinical trials, is the subject of this article. Strategies may be used to manage the long-term outcome of nanoparticles in an organism since, in addition to composition, their design also affects how long they last and how easily they degrade. This review explained the safety of nanoscale materials, biotransformation, and the multifunctional recycling mechanism of nanostructures.

Propofol vs midazolam sedation for elective endoscopy in patients with cirrhosis:A systematic review and meta-analysis of randomized controlled trials

Precision phenotyping of contrasting potato(Solanum tuberosum L.) varieties in a novel aeroponics system for improving nitrogen use efficiency: In search of key traits and genes

With increasing population, degrading soil health, limited arable land area, and high cost of nitrogen(N) fertilizers, improving nitrogen use efficiency(NUE) of potato is an inevitable approach to save the environment and achieve sufficient tuber yields with less N fertilizer supply. Recently, we have developed an aeroponics system to study NUE in potato using genomics, physiology, and breeding approaches. This study aims on precision phenotyping of plants of two distinct potato varieties(Kufri gaurav, N efficient;Kufri Jyoti, N inefficient) in the novel aeroponics system. Plants were grown in aeroponics under controlled conditions with low N(0.75 mmol L^-1NO3^-) and high N(7.5 mmol L^-1NO3^-) levels. Plant biomass, root traits, total chlorophyll content, and plant N were increased with increasing N supply, whereas higher NUE parameters namely NUE, agronomic NUE(Ag NUE), N uptake efficiency(NUp E), harvest index(HI), and N harvest index(NHI) were observed at low N. An NUE efficient cv. Kufri gaurav showed higher tuber dry weight, fresh tuber yield, tuber number per plant, early start of tuber harvesting, root traits, stolon traits, NUE parameters, and higher amino acid(aspartic acid and asparagine) content at low N supply. Higher expression of nitrate reductase(NR), nitrite reductase(NIR), and asparagine synthetase(AS) genes was observed in the leaf tissues of Kufri gaurav at high N. Thus, aeroponics-based precision phenotyping enables identification of NUE efficient genotypes based on key traits and genes involved in improving NUE in potato. Further, this study suggests that the potential of aeroponics can be utilized to investigate N biology in potato under different N regimes.
