Comparative study of haemagglutination inhibition, Agar gel precipitation test, Serum neutralization and Enzyme linked immunosorbent assay for detection to avian influenza viruses

The sensitivity, specificity and reproducibility of the serological tests for detection of avian in-fluenza viruses were carried-out by using Ham- agglutination inhibition (HI), Agar gel precipita-tion test (AGPT), and Enzyme linked immu-nosorbent assay (ELISA) and Serum neutraliza-tion test. The geometric mean titre (GMT) of hae- magglutination inhibition antibodies recor- ded as log2 indicated that the post vaccination titres in the field were on higher side i.e., 7.9 for H7 and 5.9 for H9. The correlation between HI titre and AGPT affirmed that for the AGPT test need high antibody titre for positive reaction. The pooled sera were also used to correlate the se-rum neutralization test and enzyme linked im- muno-sorbent assay. The serial two fold dilute- ons were tested for the serum neutralization ac- tivity and concluded that the HI titre log2 4 pro-vided 100% protection, than 52% and 45% pro-tection in 1:2 and 1:4 dilution was recorded, respectively. Similarly, the ELISA test showed positive results up to 1:16 HI titre, i.e. log2 4 and confirmed the linear relation between these two serological tests. In HI test, the concentration of antigen can influence the result. It also needs careful preparation of concentration of eryth-rocyte suspension. Agar Gel immuno-diffusion is basically a qualitative test as it can not de-termine the quantity of antigen or antibody with the help of this test. It lacks the level of sensi-tivity as offered by other test. If serum neutrali-zation test is performed on a pooled serum sam- ples, then it could lead to a false conclusion on antibodies status. ELISA is most sensitive, spe-cific and accurate as compare to all other sero-logical tests.

Performance, Immune, and Carcass Characteristics of Broiler Chickens as Affected by Thyme and Licorice or Enzyme Supplemented Diets

Numbers of 300 day-old broiler chickens through a CRD design with 5 treatments, 3 replicates and 20 chicks in each pen were used to evaluate the effect of thyme (T), licorice (L), thyme + licorice (TL), and enzyme supplemented (E) diets on performance, immune and carcass characteristics. According to the results, performance traits, immune indices, and carcass traits in herbal medicine and enzyme supplemented diets were improved significantly than control diet (P < 0.05). Weight gain and FCR in T and E groups were significantly higher and lower than other groups respectively (P < 0.05). Internal organs such as abdominal fat and liver weight as indicators of lipogenesis rate were decreased in T, L, and TL diets than control or E diet significantly (P < 0.05). Immune organs such as burse and spleen weight as indicators of immune situation were increased in TL diet than other treatments significantly (P < 0.05). These findings indicated that thyme and licorice singly or in combination as organic herbal medicine can affect performance, carcass and immune characteristics. Also an improved immune organ such as burse or spleen in this study indicates that this herbal medicine can promote the immune situation and efficacy of health and livability.

Background： Reducing labor pain and anxiety is one of the most important goals of maternity care. Objective： This study aimed to assess the effects of aromatherapy with Rosa damascena on pain and anxiety in the first stage of labor among nulliparous women. Design, setting, participants and interventions： This was a randomized clinical trial of 110 nulliparous women. The eligible participants were randomly assigned to two groups of aromatherapy and control in an Iranian maternity hospital. The participants received 0.08 mL of Rosa damascena essence in the aro- matherapy group and 0.08 mL of normal saline in the control group, every 30 min. Pain was measured 3 times, once each at three stages of cervical dilation （4-5, 6-7, and 8-10 cm）. Anxiety was measured twice, once each at two stages of cervical dilation （4-7 and 8-10 cm）. The tools for data collection were the Spielberger anxiety questionnaire, numerical pain rating scale, demographic and obstetric questionnaire, and an observational checklist. Data analyses included the t-test, Mann-Whitney U test and Chi-square test. Main outcome measures： Severity of labor pain and severity of anxiety were used as primary outcome measures. Labor and delivery characteristics （including number of contractions, duration of contractions in second stage, Bishop score, augmentation by oxytocin, Apgar score, and mode of delivery）, demographic characteristics, and fertility information were used as secondary outcome measures. Results： Pain severity in the group receiving aromatherapy with R. damoscena was significantly lower than in the control group after treatment at each pain assessment （cervical dilation of 4-5, 6-7, and 8-10 cm; P 〈 0.05）. Anxiety levels were also significantly lower in the treatment group than in the control group after treatment at each time of measurement （cervical dilation of 4-7 and 8-10 cm; P 〈 0.05）. For the development of 19-plex Y STR system and polymorphism studies in local ethnic populations sixteen markers of non-recombining regions (NRY) of Y chromosome, which show high power of discrimination among individuals, were selected in this study. Blood samples (600) were collected from the males of three most common castes of Pakistani population (Arain, Awan and Rajput) with different parent lineages. Three markers (DYS385a/b, DYS389I/II and YCAIIa/b) among 16 Y STRs are double-targeted regions of the Y chromosome and thus provide two polymorphic peaks for each respective primer set. These 16 Y-STRs were developed into Megaplex system for simultaneous amplification of all markers within the population. The overall power of discrimination observed in focused populations was 60.5%, 66.5% and 55% in Rajput, Awan and Arain casts respectively. This discrimination power will be helpful in human identification for forensic casework studies including sexual assaults and paternity testing.
