Fast and sensitive LC–MS/MS method for the simultaneous determination of lisinopril and hydrochlorothiazide in human plasma

A sensitive and rapid liquid chromatography-tandem mass spectrometry(LC–MS/MS) method has been developed for the simultaneous determination of lisinopril(LIS) and hydrochlorothiazide(HCTZ) in human plasma using their labeled internal standards(ISs). Sample pre-treatment involved solid phase extraction on Waters Oasis HLB cartridges using 100 μL of plasma, followed by liquid chromatography on Hypersil Gold C_(18)(50 mm×3.0 mm, 5 μm) column. The analytes were eluted within 2.0 min using acetonitrile-5.0 m M ammonium formate, p H 4.5(85:15, v/v) as the mobile phase. The analytes and ISs were analyzed in the negative ionization mode and quantified using multiple reaction monitoring. The method showed excellent linearity over the concentration range of 0.50–250.0 ng/m L for both the analytes. The intra-batch and inter-batch precision(% CV) was ≤5.26% and their extraction recoveries were in the range of 96.6%–103.1%. Matrix effect evaluated in terms of IS-normalized matrix factors ranged from 0.97 to 1.03 for both the analytes. The validated method was successfully applied to determine the plasma concentration of the drugs using 10 mg lisinopril and 12.5 mg hydrochlorothiazide fixed dose formulation in 18 healthy Indian volunteers.

Pakistan is highly exposed to climate-induced disasters, especially floods. Flooding history shows that educational establishments have been disproportionately hard-hit by flooding events. In Pakistan, school safety and preparedness is still a choice, rather than a mandatory requirement for all schools. But schools in Pakistan do have a responsibility to keep safe the students in their care,especially during and after the catastrophic events. This implies the need to maintain the environment in and around school property, so as to minimize the impacts of floods and to have the mechanisms in place to maximize a school's resilience. This study examined the emergency preparedness activities of 20 schools in four districts of Khyber Pakhtunkhwa Province that had recently been severely affected by floods. Through face to face interviews and a structured questionnaire(n = 100) we collected data on the four pillars of emergency preparedness:emergency planning, preparation measures, safe school facilities, and hazard education and training. The study revealed that the majority of the sample schools hadexperienced more than one natural hazard-induced disaster,predominantly flooding, yet despite this had not undertaken adequate emergency preparedness activities. There are particular gaps with regard to plans for students with disabilities, the continuity of school operations after a disaster,the presence of maps to identify evacuation routes, the availability of emergency equipment and resources, disaster preparedness guidelines, and psychological first aid and crisis counseling. The strengths, weaknesses, opportunities,and threats analysis that our researchers carried out indicates that, although schools in the survey have taken many steps towards flood preparedness, many weaknesses still exist and there remain significant opportunities to strengthen the preparedness level of many schools. The motion of mono-disperse spherical steel particles in a vibration driven quasi-two-dimensional （2D） square cell is studied. The cell is horizontally vibrated to eliminate the effect of gravity compaction. The velocity distributions at different particle number densities are studied and found to obey the form exp[-β（｜Vy｜/σy）α], in which Vy and （σy are velocity and its variance in the transverse direction, and α and β are fitting parameters. The value of α is found to decrease with the number density of particles increasing. To investigate the effect of the bottom plate, the molecular dynamics simulation without considering any bottom friction is performed. The accordance between the simulation result and the experimental result shows that the influence of bottom plate friction force on the high energy tail of the velocity distribution can be neglected.

The structural,morphological and optical properties of single-phase polycrystalline La2-xSrxNiMnO6(x=0,0.3 and 0.5),synthesized by solid state reaction were *** samples were characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM)/energy dispersive analysis of X-rays(EDAX),Raman spectroscopy and diffuse reflectance spectroscopy(DRS)to elucidate the role of A-site Sr-doping in double perovskite La2 *** analysis of XRD patterns revealed that all the samples have monoclinic structure with space group P21/*** gradient in the Williamson Hall plots revealed the presence of tensile strain in all the *** morphological studies revealed that average grain size increases along with appreciable decrease in porosity with Sr *** Ni/Mn antisite disorder was introduced in the La2 NiMnO6 by Sr-doping confirmed by an increase in the full width at half maximum(FWHM)and decrease in intensity of the Raman modes at around 540 and 665 cm-1 which correspond to the antisymmetric stretching and symmetric stretching modes,*** results reveal that the band gap in La2 NiMnO6 can be tuned down by Sr-doping to a value of1.37 eV(very close to 1.40 eV,considered as optimum value for better efficiency of a solar cell).Thus,Sr-doped La2 NiMnO6 may be of prime importance for applications in solar cells.

Lead-free magnetoelectric composites(1-x)K0.5Na0.5NbO3–(x)BaFe12O19(x = 30, 40, and 50 wt%) are synthesized using solid state reaction method. X-ray diffraction(XRD) patterns confirm formation of diphase composites. Field emission scanning electron microscopy(FE-SEM) gives information about grain size, connectivity, and microstructure of constituent phases. Dielectric parameters of composite samples are studied as a function of temperature and the transition temperatures corresponding to both the constituent phases are observed in the composite samples. Dielectric constant has been found to decrease with addition of ferrite. Room temperature multiferroic behaviour has been confirmed using P–E and M–H hysteresis loops and magnetoelectric measurement. Polarization is found to decrease;however, magnetization increases with ferrite weight percentage. The highest αME of 4.08 mV/(cm·Oe) is obtained for x = 30 wt% composite and it is realized that ferrite content significantly affects magnetoelectric behaviour.

The stability of a river-bed protection block in a flood event is affected by underscouring of the block. The effects of block spacing on block stability were investigated in different underscour conditions. Three blocks were placed at the bottom in a flume, and the underscour depth of the frontal block and distance between the first and second blocks were changed. Drag and lift forces were measured directly in each experimental condition. Particle Image Velocimetry （PIV） was used to visualize the flow and eva- luate the streamline pattern. Pressure measurements around the block to understand the pressure distribution show that the pressure distribution on the rear and bottom surfaces of the block is significantly affected by changes in block spacing and underscour depth. The experimental results show that for the same underscour gap, lift, and drag coefficients decreased with increasing space between the two blocks. In addition, if the underscour gap is increased with the same horizontal spacing, the lift coefficient increases and drag coefficient decreases. These results demonstrate that wider spacing enhance stability of the frontal block in the underscour condition. Moreover, analysis of block stability with changing underscouring and horizontal spacing shows that bed protection blocks seem to have more stability when the gap between two bed protection blocks was equal to height of the blocks.
