PLA_(2) mediates the innate immune response in Asian corn borer, Ostrinia furnacalis

The eicosanoid signaling pathway mediates insect immune reactions to a wide range of stimuli. This pathway begins with the biosynthesis of arachidonic acid (AA) from the hydrolysis of phospholipids catalyzed by phospholipase A_(2) (PLA_(2)). We report here that the PLA_(2) inhibitor, dexamethasone (DEX), impaired the innate immune response including nodulation, encapsulation, and melanization in Ostrinia furnacalis larvae, while AA partially reversed these effects of DEX. We cloned a full-length complementary DNA encoding a PLA_(2), designated as OfsPLA_(2), from O. furnacalis. The open reading frame of OfsPLA_(2) encodes a 195-amino acid residue protein with a 22-residue signal peptide. Sequence alignment analyses indicated that O. furnacalis PLA_(2) might be a Group III secretory PLA_(2). The highest transcript levels of OfsPLA_(2) were detected in the fat body, and its transcript levels increased dramatically after infection with Escherichia coli, Micrococcus luteus, or Beauveria bassiana. Recombinant OfsPLA_(2) significantly induced prophenoloxidase (PPO) activation in larval hemolymph in the presence of Ca^(2+) and encapsulation of agarose beads. Injection of recombinant OfsPLA_(2) into larvae resulted in increased transcript levels of attacin, defencin, and moricin-3 genes. Our results demonstrate the involvement of the eicosanoid signaling pathway in the innate immune response of O. furnacalis larvae and provide new information about the roles of O. furnacalis secretory PLA_(2) in activating PPO and antimicrobial peptide production.

The effects of annealing and irradiation on the evolution of Cu clusters in a-Fe are investigated using object kinetic Monte Carlo *** our model,vacancies act as carriers for chemical species via thermally activated diffusion jumps,thus playing an important role in solute *** the end of the Cu cluster evolution,the simulations of the average radius and number density of the clusters are consistent with the experimental data,which indicates that the proposed simulation model is applicable and *** the simulation of the annealing process,it is found that the evolution of the cluster size roughly follows the 1/2 time power law with the increase in radius during the growth phase and the 1/3 time power law during the coarsening *** addition,the main difference between neutron and ion irradiation is the growth and evolution process of the copper-vacancy *** aggregation of vacancy clusters under ion irradiation suppresses the migration and coarsening of the clusters,which ultimately leads to a smaller average radius of the copper *** proposed simulation model can supplement experimental analyses and provide a detailed evolution mechanism of vacancy-enhanced precipitation,thereby providing a foundation for other elemental precipitation research.

Detailed in-furnace measurements with reburning of superfine pulverized coal in high CO_(2)concentration

This paper presents detailed measurements of gaseous species during the reburning process with high CO_(2)concentration in a bench-scale *** pulverized coal,with the average particle size below or around 20μm,is used as the reburning *** data of flue gas concentration(NOx,HCN,NH_(3),CH_(4),O_(2),CO,CO_(2)etc.)is obtained in order to reveal the intrinsic connection between NOx emissions and other influential gaseous species at different positions of the *** finding concludes that the advantage of superfine pulverized coal with regard to NO reduction is more efficient in homogeneous stage rather than heterogeneous ***,the evolution of HCN and CH_(4)agrees well with each stage of NO reduction,which indicates that these gaseous species are favorable for NO ***,oxygen consumption rate for superfine pulverized coal is relatively faster,conducive to strengthen both homogeneous and heterogeneous NO reduction under CO_(2)reburning condition.

Optimized Simulation Design of Double Glass Curtain Wall sha.ing System in Hot Summer and Cold Winter Zone

The glass curtain wall is widely favored by the owners for its good appearance modeling efthct. In using process, however, excessive energy consumption, low level indoor eomtort and other problems of glass curtain wall are often exposed. Aiming at office buildings in hot Summer and cold Winter zone, taking the optimization of thermal comfort of double glass curtain wall in the summer and the reduetion of building energy consumption throughout the year as the breakthrough point, using the method of energy simulation analysis, through changing the size of internal sha.ing component in the simulated room, this paper analyzes and summarizes the variation law of its energy consumption value, to explore the relatively reasonable design plan of sha.ing systems of the building with glass curtain wall.

The Arabidopsis R-SNARE protein YKT61 is essential for gametophyte development

Gametophyte development is a pre-requisite for plant reproduction and seed yield;therefore,studies of gametophyte development help us understand fundamental biological questions and have potential applications in *** biogenesis and dynamics of endomembrane compartments are critical for cell survival,and their regulatory mechanisms are just beginning to be ***,we report that the Arabidopsis thaliana SNARE(soluble N-ethylmaleimide sensitive factor attachment protein receptor)protein YKT61 is essential for both male and female *** using clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9)-based genome editing,we demonstrated that male and female gametophytes carrying YKT61 loss-offunction alleles do not ***,loss of YKT61 function resulted in the arrest of male gametophytic development at pollen mitosis I and the degeneration of female gametophytes.A three-base-pair deletion in YKT61 in the ykt61-3 mutant resulted in a single-amino acid deletion in the longin domain of YKT61;the resulting mutant protein does not interact with multiple SNAREs and showed substantially reduced membrane association,suggesting that the N-terminal longin domain of YKT61 plays multiple roles in its *** study demonstrates that Arabidopsis YKT61 is essential for male and female gametogenesis and sets an example for functional characterization of essential genes with the combination of Cas9-mediated editing and expression from a Cas9-resistant transgene.

The amyloid-β(Aβ)oligomer,rather than the Aβmonomer,is considered to be the primary initiator of Alzheimer’s *** was hypothesized that p(Aβ3-10)10-MT,the recombinant Aβ3-10 gene vaccine of the Aβoligomer has the potential to treat Alzheimer’s *** this study,we intramuscularly injected the p(Aβ3-10)10-MT vaccine into the left hindlimb of APP/PS1/tau triple-transgenic mice,which are a model for Alzheimer’s *** results showed that the p(Aβ3-10)10-MT vaccine effectively reduced Aβoligomer levels and plaque deposition in the cerebral cortex and hippocampus,decreased the levels tau protein variants,reduced synaptic loss,protected synaptic function,reduced neuron loss,and ameliorated memory impairment without causing any cerebral ***,this novel DNA vaccine,which is safe and highly effective in mouse models of Alzheimer’s disease,holds a lot of promise for the treatment of Alzheimer’s disease in humans.

Preparation of New Cementitious System using Fly Ash and Dehydrated Autoclaved Aerated Concrete

We experimentally studied the interaction between pozzolanic material（fly ash） and dehydrated autoclaved aerated concrete（DAAC）. The DAAC powder was obtained by grinding aerated concrete waste to particles fi ner than 75μm and was then heated to temperatures up to 900 ℃. New cementitious material was prepared by proportioning fly ash and DAAC, named as AF. X-ray diffraction（XRD） was employed to identify the crystalline phases of DAAC before and after rehydration. The hydration process of AF was analyzed by the heat of hydration and non-evaporable water content（Wn）. The experimental results show that the highest reactivity of DAAC can be obtained by calcining the powder at 700 ℃ and the dehydrated products are mainly β-C2 S and CaO. The cumulative heat of hydration and Wn was found to be strongly dependent on the replacement level of fl y ash, increasing the replacement level of fl y ash lowered them in AF. The strength contribution rates on pozzolanic effect of fl y ash in AF are always negative, showing a contrary tendency of that of cement-fl y ash system.

Theoretical study on the effect of H_(2)O on the formation mechanism of NO_(x) precursors during indole pyrolysis

The incineration technology of kitchen waste is one of the effective technologies to achieve the resource utilization of municipal solid *** is an important stage of *** is a rich initial product in the pyrolysis process of kitchen waste,and the presence of H_(2)O has a significant impact on the decomposition of indole to form NO_(x) ***,this study uses density functional theory method to study the effect of H_(2)O on the thermal decomposition of indole to produce NH_(3),HNCO,and *** H_(2)O participates in the reaction,it can provide oxidative groups to generate a new product HNCO,which is different from the previous findings by indole pyrolysis without the presence of H_(2)***,this study theoretically proves that NH_(3) is easier to form than *** is consistent with the phenomenon that NH_(3) release is higher than HCN release in pyrolysis *** addition,compared with the individual pyrolysis of indole,the participation of H_(2)O reduces the energy barriers for the formation of NH_(3) and HCN during indole pyrolysis,thereby promoting the formation of NH_(3) and HCN.
