Twelve lactating Friesian Cows with body weight of 450 to 550 kg and in the second to fifth lactating season were used after 8 weeks of cal- ving in a complete switch-back design. The cows were fed a basal ration of 35% concentrate feed mix-ture with 40% berseem and 25% rice straw （ on a dry matter （DM） basis） without supplementation （ G1 ） or supplemented with 5 and 10 g zinc methionine per head per day （G2 and G3, respectively）. Nutrient digestibility coefficients and nutritive values were significantly increased （P 〈0.05） by zinc methionine supplementation. Cows in G2 had the highest feed intake of total digestible nutrients （TDN） and digestible crude protein （DCP）, followed by Ga and G1, respectively （ P 〈 0.05 ）. The ruminal total volatile fatty acid concentration increased and ammonia nitrogen decreased significantly （ P 〈 0.05 ） by zinc methio-nine supplementation. Cows in G3 showed the highest plasma total protein, albumin, and globulin concen- trations （ P 〈 0.05 ）. Zinc methionine supplementa- tion led to significant decreases （ P 〈 0.05 ） in the activities of aspartate aminotransferase and alanine aminotransferase. A significant increase （ P 〈 0.05 ） was observed in the milk yield for G2 （2.54 kg/d or 18. 1% ） and G3 （ 1.95 kg/d or 14.2% ） compared with the control group. The corresponding increase in 4% fat-corrected milk （FCM） was 2.85 and 2.03 kg/d （21.63% and 15.74% ） for G2 and G3, respectively. Cows in G2 had the highest fat, total protein, and total solids contents, and the yield of all milk constitu- ents and animals in G3 showed the highest lactose, solids-not-fat, and ash contents. Zinc methionine sup- plementation improved feed conversion, reducing the quantities of DM, TDN, and DCP required to pro- duce 1 kg of 4% FCM （P〈0.05）. The average dai- ly feed cost per kilogram of 4% FCM decreased and the average income from milk production increased with zinc methionine supplementation （ P 〈 0.05 ）. Mounting evidence points to critical roles for DNA modifications, including 5-methylcytosine （5mC） and its oxidized forms, in the development, plasticity and disorders of the mammalian nervous system. The novel DNA base 5- hydroxymethylcytosine （5hmC） is known to be capable of initiating passive or active DNA demethylation, but whether and how extensively 5hmC functions in shaping the post-mitotic neuronal DNA methylome is unclear. here we report the genome-wide distribution of 5hmC in dentate granule neurons from adult mouse hippocampus in vivo. 5hmC in the neuronal genome is highly enriched in gene bodies, especially in exons, and correlates with gene expression. Direct genome-wide comparison of 5hmC distribution between embryonic stem cells and neurons reveals extensive differences, reflecting the functional disparity between these two cell types. Importantly, integrative analysis of 5hmC, overall DNA methylation and gene expression profiles of dentate granule neurons in vivo reveals the genome-wide antagonism between these two states of cytosine modifications, supporting a role for 5hmC in shaping the neuronal DNA methylome by promoting active DNA demethylation.

目的:观察外源性肾上腺髓质素(adrenomedullin,ADM)对大鼠肾脏创伤后外周血Th1/Th2的调节作用,并了解给药时间对此作用的影响。方法:实验于2005年在兰州大学实验中心进行。将104只Wistar大鼠随机分为4大组:①正常对照组(8只);不打击。②创伤组(32只);自由落体生物撞击仪撞击大鼠脊肋区,间接打击双侧肾脏造成肾脏Ⅰ级损伤。③创伤前注射ADM组(32只);打击同创伤组,打击前10min腹腔注射肾上腺髓质素0.1nmol/kg(终体积0.5mL)。④创伤后注射ADM组(32只):打击同创伤组,打击后10min腹腔注射肾上腺髓质素0.1nmol/kg(终体积0.5mL)。后3组分别在打击后1,6,12,24h各处死8只。采用酶联免疫斑点法测定Th1与Th2阳性率。结果:82只大鼠进入结果分析。①创伤组:Th1细胞在创伤后6h显著增加并达峰值,高于创伤后1h及正常对照组[(0.32±0.07)%,(0.13±0.06)%,(0.16±0.07)%,Ph2各时间点比较均无差异;Th1/Th2在创伤后6h达高峰,创伤后24h降至正常对照组水平。②创伤前注射ADM组:Th1在创伤后6h低应答,在创伤后12h增高并达峰值,高于创伤后6h及正常对照组[(0.32±0.08)%,(0.08±0.06)%,(0.16±0.07)%,Ph2在创伤后6-12h的变化趋势与Th1相同;Th1/Th2创伤后6h达最低,低于该时点创伤组(4.58±3.81,23.44±6.19,Ph达高峰。③创伤后注射ADM组:Th1自创伤后1h逐渐上升,至24h达高峰,高于创伤后1,6h[(0.32±0.10)%,(0.10±0.04)%,(0.17±0.07)%,Ph2细胞各时间点比较均无差异;Th1/Th2于创伤后1,6h接近正常对照组,然后逐渐上升,至创伤后24h达最高,高于正常对照组及该时点创伤组(22.49±12.64,5.33±4.55,4.78±3.15,Ph为著),外源性肾上腺髓质素通过抑制Th1优势应答,具有校正Th1/Th2比例失衡的免疫调节作用。②急性创伤后最初6h内,创伤后给予肾上腺髓质素调节Th1/Th2平衡作用明显优于创伤前给药。

目的:观察呼吸道合胞病毒感染肺上皮细胞A549后Toll样受体3表达的变化,探讨Toll样受体3介导呼吸道合胞病毒感染肺上皮细胞的炎性反应及其介导的信号转导通路。方法:实验于2006-05/12在安徽医科大学基础医学院分子生物学实验室完成。用呼吸道合胞病毒感染体外培养的A549细胞,于感染后4,8,16,24h收集细胞和细胞培养上清。用Trizol提取细胞总RNA,RT-PCR法检测TOLL样受体3mRNA、肿瘤坏死因子αmRNA表达的变化;提取细胞总蛋白和核蛋白,免疫印迹法分别检测TOLL样受体3蛋白、核内活性核因子κB的变化;用ELISA检测细胞培养上清肿瘤坏死因子α的表达。以未感染病毒的细胞为正常对照组。结果:①呼吸道合胞病毒感染A549细胞后,Toll样受体3和肿瘤坏死因子α的mRNA和蛋白的表达量均升高且有时间依赖性,Toll样受体3mRNA24h表达量是基础表达量的5倍多,肿瘤坏死因子αmRNA24h表达量是基础表达量的3倍多。②A549细胞中核内活性核因子κB有基础表达,经由呼吸道合胞病毒感染,核内活性核因子κB随感染时间的延长升高;同时呼吸道合胞病毒感染能促进Toll样受体3蛋白的表达,高于正常对照组(Ph分泌达到高峰。结论:呼吸道合胞病毒感染A549细胞后上调Toll样受体3表达,其诱导的炎性反应与Toll样受体3介导的信号转导途径有关。

Laparoscopic pancreaticoduodenectomy: a descriptive and comparative review

Laparoscopic pancreaticoduodenectomy（LPD） is an extremely challenging surgery. First described in 1994, it has been slow to gain in popularity. Recently, however, we have seen an increase in the number of centers performing this operation, including our own institution, as well as an increase in the quantity of published data. The purpose of this review is to describe the current status of LPD as described in the literature. We performed a literature search in the Pub Med database using Me sh terms ＂laparoscopy＂ and ＂pancreaticoduodenectomy＂. We then identified articles in the English language with over 20 patients that focused on LPD only. Review articles were excluded and only one article per institution was used for descriptive analysis in order to avoid overlap. There were a total of eight articles meeting review criteria, consisting of 492 patients. On descriptive analysis we found that percent of LPD due to high-grade malignancy averaged 47% over all articles. Average operative time was 452 minutes, blood loss 369 cc＇s, pancreatic leak rate 15%, delayed gastric emptying 8.6%, length of hospital stay 9.4 days, and short term mortality 2.3%. Comparison studies between open pancreaticoduodenectomy（OPD） and LPD suggested decreased blood loss, longer operative time, similar post-operative complication rate, decreased pain, and shorter hospital length of stay for LPD. There was also increased number of lymph nodes harvested and similar margin free resections with LPD in the majority of studies. LPD is a safe surgery, providing many of the advantages typically associated with laparoscopic procedures. We expect this operation to continue to gain in popularity as well as be offered in increasingly more complex cases. In future studies, it will be beneficial to look further at the oncologic outcome data of LPD including survival.

Thermal and electron transport through organic molecules attached to three-dimensional gold electrodes in two different configurations, namely para and meta with thiol-terminated junctions is studied theoretically in the linear response regime using Green＇s function formalism. We used thiol-terminated（–sh bond） benzene units and found a positive thermopower because the highest occupied molecular orbital（hOMO） is near the Fermi energy level. We investigated the influence of molecular length and molecular junction geometry on the thermoelectric properties. Our results show that the thermoelectric properties are highly sensitive to the coupling geometry and the molecular length. In addition, we observed that the interference effects and increasing molecular length can increase the thermoelectric efficiency of device in a specific configuration.
