Metagenomic insights into microorganisms and antibiotic resistance genes of waste antibiotic fermentation residues along production,storage and treatment processes

Antibiotic fermentation residue(AFR)is nutrient-rich solid waste generated from fermentative antibiotic production *** is demonstrated that AFR contains high-concentration of remaining antibiotics,and thus may promote antibiotic resistance development in receiving environment or feeding farmed ***,the dominate microorganisms and antibiotic resistance genes(ARGs)in AFRs have not been adequately explored,hampering understanding on the potential antibiotic resistance risk development caused by ***,seven kinds of representative AFRs along their production,storage,and treatment processes were collected,and multiple methods including amplicon sequencing,metagenomic sequencing,and bioinformatic approaches were adopted to explore the biological characteristics of *** expected,antibiotic fermentation producer was found as the predominant species in raw AFRs,which were collected at the outlet of fermentation ***,except for producer species,more environment-derived species persisted in stored AFRs,which were temporarily stored at a semi-open *** genus,classified as Firmicutes phylum and Bacilli class,became predominant bacterial taxa in stored AFRs,which might attribute to its tolerance to high concentration of *** from metagenomic sequencing together with assembly and binning approaches sh.wed that these newly-colonizing species(e.g.,Lactobacillus genus)tended to carry ARGs conferring resistance to the remaining ***,after thermal treatment,remaining antibiotic could be efficiently removed from AFRs,and microorganisms together with DNA could be strongly *** sum,the main risk from the AFRs was the remaining antibiotic,while environment-derived bacteria which tolerate extreme environment,survived in ARFs with high content antibiotics,and may carry ***,hydrothermal or other harmless treatment technologies are recommended to remove antibiotic content and inactivate bacteria befor

High-Temperature Stability of Mg-1Al-12Y Alloy Containing LPSO Phase and Mechanism of Its Portevin-Le Chatelier(PLC)Effect

In this study,the high-temperature stability and the generation mechanism of the Portevin-Le Chatelier(PLC)effect in solid-solution Mg-1Al-12Y alloy with different heat treatment processes were investigated by adjusting the content of long-period stacking ordered(LPSO)*** was found that the content of LPSO phases in the alloys differed the most after heat treatment at 530℃for 16 h and 24 h,with values of 13.56%and 3.93%***,high-temperature tensile experiments were conducted on these two alloys at temperatures of 150℃,200℃,250℃,and 300℃.The results sh.wed that both alloys exhibited the PLC effect at temperatures ranging from 150 to 250℃.However,at a temperature 300℃,only the alloy with a greater concentration of LPSO phases exhibited the PLC effect,whereas the alloy with a lower proportion of LPSO phases did not exhibit this ***,both alloys exhibited remarkable high-temperature stability,with the alloy containing a greater percentage of LPSO phases also demonstrating superior *** underlying mechanism for this phenomenon lies in the exceptional high-temperature stability exhibited by the second phase within the ***,the LPSO phase effectively obstructs the movement of dislocations,and it also undergoing kinking to facilitate plastic deformation of the *** results indicate that the PLC effect can be suppressed by reducing dislocation pile-up at grain boundaries,which leads to a decrease in alloy plasticity but an increase in *** presence of the PLC effect in the WA121 alloy is attributed to the abundant dispersed second phase within the alloy,which initially hinders the movement of dislocations,leading to an increase in stress,and subsequently releases the dislocations,allowing them to continue their movement and thereby reducing in stress.

Mesenchymal stromal cells modulate unfolded protein response and preserve β-cell mass in type 1 diabetes

Introduction:Transplantation of mesenchymal stromal cells(MSCs)is a promising therapy for type 1 diabetes(T1D).However,whether the infused MSCs affect the endoplasmic reticulum stress or subsequent unfolded protein response inβcells remains ***:To investigate this,we induced early-onset T1D in non-obese diabetic mice using ***,T1D mice were randomly assigned to receive either MSCs or phosphate-buffered *** observed the in vivo homing of MSCs and assessed their effectiveness by analyzing blood glucose levels,body weight,histopathology,pancreatic protein expression,and serum levels of cytokines,proinsulin,and ***:Infused MSCs were found in the lungs,liver,spleen,and pancreas of T1D *** exhibited various effects,including reducing blood glucose levels,regulating immunity,inhibiting inﬂammation,increasingβ-cell areas,and reducing the expression of key proteins in the unfolded protein response *** serum proinsulin and C-peptide levels were signiﬁcantly higher in the MSCs treatment group than in the T1D model ***,there was no signiﬁcant difference in the biomarker ofβ-cell endoplasmic reticulum stress,the ratio of fasting serum proinsulin to C-peptide,between the two ***:Ourﬁndings reveal that MSCs infusion does not alleviate endoplasmic reticulum stress inβcells directly but modulates the unfolded protein response pathway to preserveβ-cell mass and function in T1D mice.

ORPA:a fast and efficient phylogenetic analysis method for constructing genome-wide alignments of organelle genomes

Creating a multi-gene alignment matrix for phylogenetic analysis using organelle genomes involves aligning single-gene datasets manually,a process that can be time-consuming and prone to *** HomBlocks pipeline has been created to eliminate the inaccuracies arising from manual *** processing of a large number of sequences,however,remains a time-consuming *** conquer this challenge,we develop a speedy and efficient method called Organelle Genomes for Phylogenetic Analysis(ORPA).ORPA can quickly generate multiple sequence alignments for whole-genome comparisons by parsing the result files of NCBI BLAST,completing the task just in 1 *** increasing data volume,the efficiency of ORPA is even more pronounced,over 300 times faster than HomBlocks in aligning 60 high-plant chloroplast *** phylogenetic tree outputs from ORPA are equivalent to HomBlocks,indicating its outstanding *** to its speed and accuracy,ORPA can identify species-level evolutionary conflicts,providing valuable insights into evolutionary cognition.

Traditional Chinese Medicine Integrated Responsive Microneedles for Systemic Sclerosis Treatment

Traditional Chinese medicine,such as Tripterygium wilfordii and Paeonia lactiflora,has potential values in treating systemic sclerosis(SSc)and other autoimmune diseases,while their toxic side effect elimination and precise tropical drug delivery are still ***,we present multiple traditional Chinese medicine integrated photoresponsive black phosphorus(BP)microneedles(MNs)with the desired features for the SSc *** employing a template-assisted layer-by-layer curing method,such MNs with triptolide(TP)/paeoniflorin(Pae)needle tips and BP-hydrogel needle bottoms could be well *** combined administration of TP and Pae can not only provide anti-inflammatory,detoxification,and immunomodulatory effects to treat skin lesions in the early stage of SSc but also remarkably reduce the toxicity of single drug ***,the additive BPs possess good biocompatibility and near-infrared(NIR)responsiveness,imparting the MN photothermal-controlled drug release *** on these features,we have demonstrated that the traditional Chinese medicine integrated responsive MNs could effectively improve skin fibrosis and telangiectasia,reduce collagen deposition,and reduce epidermal thickness in the SSc mouse *** results indicated that the proposed Chinese medicine integrated responsive MNs had enormous potential in clinical therapy of SSc and other diseases.

Ghost Module Based Residual Mixture of Self-Attention and Convolution for Online Signature Verification

Online Signature Verification (OSV), as a personal identification technology, is widely used in various ***, it faces challenges, such as incomplete feature extraction, low accuracy, and computational heaviness. Toaddress these issues, we propose a novel approach for online signature verification, using a one-dimensionalGhost-ACmix Residual Network (1D-ACGRNet), which is a Ghost-ACmix Residual Network that combines convolutionwith a self-attention mechanism and performs improvement by using Ghost method. The Ghost-ACmix Residualstructure is introduced to leverage both self-attention and convolution mechanisms for capturing global featureinformation and extracting local information, effectively complementing whole and local signature features andmitigating the problem of insufficient feature extraction. Then, the Ghost-based Convolution and Self-Attention(ACG) block is proposed to simplify the common parts between convolution and self-attention using the Ghostmodule and employ feature transformation to obtain intermediate features, thus reducing computational ***, feature selection is performed using the random forestmethod, and the data is dimensionally reducedusing Principal Component Analysis (PCA). Finally, tests are implemented on the MCYT-100 datasets and theSVC-2004 Task2 datasets, and the equal error rates (EERs) for small-sample training using five genuine andforged signatures are 3.07% and 4.17%, respectively. The EERs for training with ten genuine and forged signaturesare 0.91% and 2.12% on the respective datasets. The experimental results illustrate that the proposed approacheffectively enhances the accuracy of online signature verification.
