Treatment of pediatric average-risk medulloblastoma using craniospinal irradiation less than 2500 cGy and chemotherapy: single center experience in Korea

Background:Although craniospinal irradiation (CSI) of 2340 cGy plus tumor booster with chemotherapy have been established as a standard treatment of childhood average-risk (AvR) medulloblastoma (MBL) in Western counties,there are a few recent reports in outcomes ofAvR MBL using this strategy in Korean and other Asian *** investigated the outcome of the Korean children with AvR MBL who were treated with CSI <2500 cGy and ***:Between January 2001 and December 2010,clinical characteristics and outcomes of 42 patients who were diagnosed with AvR MBL postoperatively and treated with radiation including CSI <2500 cGy and chemotherapy in Seoul National University Children's Hospital were ***:Their median age was 9 years (range:3-18.8),and 29 were *** subtypes were classic type in 28 patients,nodular/desmoplastic in 7,and large cell/anaplastic (LCA) in *** the patients received adjuvant radiotherapy (CSI with median 2340 cGy and booster) and multiagent chemotherapy as the first-fine *** a median follow-up of 54 months,12 patients experienced relapse or progression of the *** 3-and 5-year disease-free survival (DFS) rates were 78.0%±6.5% and 75.0%±6.9%,respectively,and overall survival (OS) rates were 85.3%±5.6% and 76.8%±6.9%,*** LCA subtype was associated with poorer DFS (P=0.023) and OS (P=0.008),compared with non-LCA ***:The outcomes of children and adolescents with AvR MBL treated with radiation including CSI <2500 cGy and chemotherapy,are compatible to those in Western countries;however,the LCA subtype has a poor outcome with this strategy.

A Predictive Model for Pathologic Complete Response in Breast Cancer Patients Treated with Neoadjuvant Chemotherapy Using Machine Learning

Background: In patients with breast cancer after Neoadjuvant Chemotherapy (NAC), pathological Complete Response (pCR) was associated with better long-term outcomes. We here attempted to predict pCR using machine learning. Patients and Methods: From 2008 to 2017, 1308 breast cancer patients underwent NAC before surgery, of whom 377 patients underwent Cancer SCANTM for gene data. Of 377, 238 were analyzed here, with 139 excluded due to incomplete medical data. Results: The pCR (-) vs. (+) group had 200 vs. 38 patients. In our predictive model with gene data, the Area Under the Curve (AUC) of the Receiver Operating Characteristic (ROC) curve was 0.909 and accuracy was 0.875. In another model without gene data, the AUC of ROC curve was 0.743 and accuracy was 0.800. Medulloblastoma（MBL）,the most common malignant pediatric brain tumor,is incurable in about one-third of patients and can lead to long-term disabilities despite current multimodal *** purpose of this study was to demonstrate in vitro biological effects of neurotrophins-3（NT-3） on MBL cells and to evaluate the growth-inhibitory effect of neurotrophin-3（NT-3）-secreting stem cells on tumor *** confirmed by western blotting that D283-MED cells express tyrosine kinase C,a specific receptor for *** the biological effects of NT-3 on MBL cells,we evaluated autophagy,apoptosis,senescence,and differentiation of tumor cells with *** NT-3 induced a concentrationdependent increase in apoptosis in the tumor cell line（P ＜ 0.001）.The high concentrations of NT-3 increased the expression of class III β-tubulin（P ＜ 0.001） and decreased the expression of Nestin（P ＜ 0.05）.NT-3-secreting stem cells were produced by nucleofecting ***-NT3 into human adipose tissue-derived mesenchymal stem cells（hAT-MSCs） and were confirmed by fluorescence microscopy and ***-layered co-culture experiments with the NT-3-secreting hAT-MSCs and D283-MED MBL cells were performed,and NT-3-induced cell death was studied by 3-（4,5-dimethylathiazol-2-yl）-2,5-dephenyltetrazolium bromide（MTT） ***,the high concentrations of NT-3-secreting hAT-MSCs significantly（P ＜ 0.05） increased the death of D283-MED cells in *** present study demonstrated that both apoptotic cell death and neuronal differentiation of tumor cells were the mechanisms of growth-inhibitory effect of NT-3-secreting hAT-MSCs on MBL cell line.

Electrochemical and electronic properties of nitrogen doped fullerene and its derivatives for lithium-ion battery applications

We have studied the redox potentials and electronic properties of C60 and C59N using density functional theory method. It is found that doping C60 with one nitrogen atom results in a slight increase in redox potential. Next, we have also studied C59N functionalized with various redox-active oxygen containing functional groups and strongly electron withdrawing functional groups. It is found that the intrinsic electronic structure of the molecule is the major determinant of the redox potential. Our DFT calculations show that the electron affinity to redox potential of functionalized C59N is correlated with the LUMO of the systems very well. This is the first systematic study on the redox properties and electronic structures of N-doped C60 systems.

Microbiota-based treatments in alcoholic liver disease

Gut microbiota plays a key role in the pathogenesis of alcoholic liver disease(ALD). Consumption of alcohol leads to increased gut permeability, small intestinal bacterial overgrowth, and enteric dysbiosis. These factors contribute to the increased translocation of microbial products to the liver via the portal tract. Subsequently, bacterial endotoxins such as lipopolysaccharide, in association with the Toll-like receptor 4 signaling pathway, induce a gamut of damaging immune responses in the hepatic milieu. Because of the close association between deleterious inflammation and ALD-induced microbiota imbalance, therapeutic approaches that seek to reestablish gut homeostasis should be considered in the treatment of alcoholic patients. To this end, a number of preliminary studies on probiotics have confirmed their effectiveness in suppressing proinflammatory cytokines and improving liver function in the context of ALD. In addition, there have been few studies linking the administration of prebiotics and antibiotics with reduction of alcoholinduced liver damage. Because these preliminary results are promising, large-scale randomized studies are warranted to elucidate the impact of these microbiotabased treatments on the gut flora and associated immune responses, in addition to exploring questions about optimal delivery. Finally, fecal microbiota transplant has been shown to be an effective method of modulating gut microbiota and deserve further investigation as a potential therapeutic option for ALD.

Influence of sulfides on the tribological properties of composites produced by pulse electric current sintering

Self-lubricating A1203-15wt% ZrO2 composites with sulfides, such as molybdenum disulfide （MoS2） and tungsten disulfide （WSz） serving as solid lubricants, were fabricated by using the pulse electric current sintering （PECS） technique. The coefficient of friction （COF） of the A1203-15wt% ZrO2 composite without/with sulfides was in the range of 0.37-0.48 and 0.27-0.49, respectively. As the amoant of sul- fides increased, the COF and the wear rate decreased. The reduction in COF and wear rate of the sulfide-containing composite is caused by a reduction in shear stresses between the specimen and the tribological medium due to the formation of a lubricating film resulting from the lamellar structure of sulfides located on the worn surface.

Transcriptional coactivators regulate the rate of gene expression in the *** receptor coactivator 6(NCOA6),a coactivator,has been implicated in embryonic development,metabolism,and cancer pathogenesis,but its role in innate immunity and inflammatory diseases remains ***,we demonstrated that NCOA6 was expressed in monocytes and macrophages and that its level was increased under proinflammatory ***,nuclear NCOA6 was found to translocate to the cytoplasm in activated monocytes and then become incorporated into the inflammasome with NLRP3 and ASC,forming cytoplasmic ***,NCOA6 associated with the ATP hydrolysis motifs in the NACHT domain of NLRP3,promoting the oligomerization of NLRP3 and ASC and thereby instigating the production of IL-1βand active *** note,Ncoa6 deficiency markedly inhibited NLRP3 hyperactivation caused by the Nlrp3^(R258W) gain-of-function mutation in *** ablation of Ncoa6 substantially attenuated the severity of two NLRP3-dependent diseases,folic-induced acute tubular necrosis and crystal-induced arthritis,in *** with these findings,NCOA6 was highly expressed in macrophages derived from gout patients,and NCOA6-positive macrophages were significantly enriched in gout macrophages according to the transcriptome profiling ***,NCOA6 is a critical regulator of NLRP3 inflammasome activation and is therefore a promising target for NLRP3-dependent diseases,including gout.
