The effects of single versus combined therapy using LIM-kinase 2 inhibitor and type 5 phosphodiesterase inhibitor on erectile function in a rat model of cavernous nerve injury-induced erectile dysfunction

We aimed to determine whether combination of LIM-kinase 2 inhibitor(LIMK2i)and phosphodiesterase type-5 inhibitor(PDE5i)could restore erectile function through suppressing cavernous fibrosis and improving caver nous apoptosis in a rat model of cavernous nerve crush injury(CNCI).Seventy 12-week-old Sprague-Dawley rats were equally distributed into five groups as follows:(1)sham surgery(Group S),(2)CNCI(Group I),(3)CNCI treated with daily intraperitoneal administration of 10.0 mg kg^-1 LIMK2i(Group I+L),(4)daily oral administration of 20.0 mg kg-1 udenafil,PDE5i(Group I+U),and(5)combined administration of 10.0 mg kg^-1 LIMK2i and 20.0 mg kg^-1 udenafil(Group I+L+U).Rats in Groups I+L,I+U,and I+L+U were treated with respective regimens for 2 weeks after CNCI.At 2 weeks after surgery,erectile response was assessed using electrostimulation.Penile tissues were processed for histological studies and western blot.Group I showed lower intracavernous pressure(ICP)/mean arterial pressure(MAP),lower area under the curve(AUC)/MAP,decreased immunohistochemical staining for alpha-smooth muscle(SM)actin,higher apoptotic index,lower SM/collagen ratio,increased phospho-LIMK2-positive fibroblasts,decreased protein kinase B/endothelial nitric oxide synthase(Akt/eNOS)phosphorylation,increased LIMK2/cofilin phosphorylation,and increased protein expression of fibronectin,compared to Group S.In all three treatment groups,erectile responses,protein expression of fibronectin,and SM/collagen ratio were improved.Group I+L+U showed greater improvement in erectile response than Group I+L.SM content and apoptotic index in Groups I+U and I+L+U were improved compared to those in Group I.However,Group I+L did not show a significant improvement in SM content or apoptotic index.The number of phospho-LIMK2-positive fibroblasts was normalized in Groups I+L and I+L+U,but not in Group I+U.Akt/eNOS phosphorylation was improved in Groups I+U and I+L+U,but not in Group I+L.LIMK2/cofilin phosphorylation was improved in Groups I+L and I+L+U,but not in Group

We aimed to evaluate the current nationwide trend,efficacy,safety,and quality of life(QoL)profiles of hormone treatment in real-world practice settings for prostate cancer(PCa)patients in Korea.A total of 292 men with any biopsy-proven PCa(TanyNanyMany)from 12 institutions in Korea were included in this multi-institutional,observational study of prospectively collected data.All luteinizing hormone-releasing hormone(LHRH)agonists were allowed to be investigational drugs.Efficacy was defined as(1)the rate of castration(serum testosterone≤50 ng dl^-1)at 4-week visit and(2)breakthrough(serum testosterone>50 ng dl^-1 after castration).Safety assessments included routine examinations for potential adverse events,laboratory tests,blood pressure,body weight,and bone min era I density(BMD,at baseli ne and at the last follow-up visit).QoL was assessed using the Expa nded Prostate Cancer Index Composite-26(EPIC-26).The most comm on initial therapeutic regimen was LHRH agonist with an ti-androge n(78.0%),and the most commonly used LHRH agonist for combination and mono therapy was leuprolide(64.0%for combi natio n and 58.0%for monotherapy).The castration and breakthrough rates were 78.4%and 6.6%,respectively.The laboratory results related to dyslipidemia worsened after 4 weeks of hormone treatment.In addition,the mean BMD T-score was significantly lower at the last follow-up(mean:-1.950)compared to baseline(mean:-0.195).The mean total EPIC-26 score decreased from 84.8(standard deviation[s.d.]:12.2)to 78.3(s.d.:8.1),with significant deterioration only in the urinary domain(mean:23.5 at baseli ne and 21.9 at the 4-week visit).These findi ngs dem on strate the n ationwide trend of current practice setti ngs in hormone treatment for PCa in Korea.

The complex of Fas-associated factor 1 with Hsp70 stabilizes the adherens junction integrity by suppressing RhoA activation

Fas-associated factor 1 (FAF1) is a scaffolding protein that plays multiple functions, and dysregulation of FAF1 is associated withmany types of diseases such as cancers. FAF1 contains multiple ubiquitin-related domains (UBA, UBL1, UBL2, UAS, and UBX), eachdomain interacting with a specific partner. In particular, the interaction of UBL1 with heat shock protein 70 (Hsp70) is associatedwith tumor formation, although the molecular understanding remains unknown. In this study, the structural analysis revealed thatHis160 of FAF1 is important for its interaction with Hsp70. The association of Hsp70 with FAF1 is required for the interaction withIQGAP1. FAF1 negatively regulates RhoA activation by FAF1–Hsp70 complex formation, which then interacts with IQGAP1. Thesesteps play a key role in maintaining the stability of cell-to-cell junction. We conclude that FAF1 plays a critical role in the structureand function of adherens junction during tissue homeostasis and morphogenesis by suppressing RhoA activation, which induces theactivation of Rho-associated protein kinase, phosphorylation of myosin light chain, formation of actin stress fiber, and disruptionof adherens junction. In addition, depletion of FAF1 increased collective invasion in a 3D spheroid cell culture. These results provideinsightinto how the FAF1–Hsp70 complex acts as a novelregulator ofthe adherens junction integrity. The complex can be a potentialtherapeutic target to inhibit tumorigenesis and metastasis.

An in-situ Raman spectroscopic study on potential-pO^(2-) phase diagram of lanthanum in LiCl-KCl eutectic melt

The oxidative reaction between lanthanum trichloride and oxide ion was studied in the molten LiCl-KCl eutectic at 773 K by in-situ Raman spectroscopy using an yttria-stabilized zirconia electrode in conjunction with electrochemical methods.The formation of lanthanum oxychloride,LaOCl(s),was confirmed by studying the vibrational properties of crystalline LaOCl and X-ray diffraction analysis.The solubility product of LaOCl in the LiCl-KCl molten eutectic at 773 K is found to be pKsp(LaOCl)=7.569±0.100,by combining the results of Raman spectroscopy and potentiometric titration with the addition of barium oxide.The use of Raman spectroscopic quantitation of dissolved lanthanum ions in the system allows faster and more accurate determination of the stable phase of lanthanum as well as the solubility product of LaOCl compared with that measured by potentiometric titration only.Based on experimentally obtained pKsp and standard equilibrium potentials,the potentialpO2- phase diagram of lanthanum in the molten LiCl-KCl eutectic was reported.

Rational design of ultrafine FeSe_(2) nanocrystals emb e dde d within hollow mesoporous carbon bowls for potassium-ion batteries with long-term cycling stability and high volumetric capacity

Hollow structures are commonly used to alleviate the mechanical stress on electrode materials and to provide more active sites in potassium-ion batteries(KIBs).Nevertheless,the excessive internal voids within these structures significantly reduce the packing density of particles,resulting in a relatively low volumetric energy density of the fabricated electrodes,which is undesirable for practical use.We designed a hollow mesoporous carbon bowl embedded with ultrafine bis(selanylidene)iron(FeSe 2)nanocrystals(FeSe 2@HMCB)via a controllable impregnation method and subsequent selenization pro-cess for high-performance KIBs.The as-obtained FeSe 2@HMCB can inherit the advantages of conventional hollow carbon-based composites,such as alleviation of volume variation in active materials,abundant ion storage sites,and high electrical conductivity.Simultaneously,the bowl structure has a higher pack-ing density than the conventional hollow structure,resulting in a significant increase in the volumet-ric energy density of the fabricated electrodes.Because of these advantages,the FeSe 2@HMCB exhibits a high,stable reversible capacity of 326 mA h g^(-1) even after 1000 cycles at 0.5 A g^(-1),and excellent rate capacities(182 mA h g^(-1) at 3.0 A g^(-1)).Compared with the hollow structured counterpart,the vol-umetric capacity(mA h cm−3)of FeSe 2@HMCB increased by 60%.Furthermore,a full cell consisting of FeSe 2@HMCB//Prussian blue(PB)exhibits excellent electrochemical performance(99 mA h g^(-1) after 100 cycles at 0.1 A g^(-1)).

MOF‑Derived CoSe2@N‑Doped Carbon Matrix Confined in Hollow Mesoporous Carbon Nanospheres as High‑Performance Anodes for Potassium‑Ion Batteries

In this work,a novel vacuum-assisted strategy is proposed to homogenously form Metal-organic frameworks within hollow mesoporous carbon nanospheres(HMCSs)via a solid-state reaction.The method is applied to synthesize an ultrafine CoSe2 nanocrystal@N-doped carbon matrix confined within HMCSs(denoted as CoSe2@NC/HMCS)for use as advanced anodes in highperformance potassium-ion batteries(KIBs).The approach involves a solvent-free thermal treatment to form a Co-based zeolitic imidazolate framework(ZIF-67)within the HMCS templates under vacuum conditions and the subsequent selenization.Thermal treatment under vacuum facilitates the infiltration of the cobalt precursor and organic linker into the HMCS and simultaneously transforms them into stable ZIF-67 particles without any solvents.During the subsequent selenization process,the“dual confinement system”,composed of both the N-doped carbon matrix derived from the organic linker and the small-sized pores of HMCS,can effectively suppress the overgrowth of CoSe2 nanocrystals.Thus,the resulting uniquely structured composite exhibits a stable cycling performance(442 mAh g^−1 at 0.1 A g^−1 after 120 cycles)and excellent rate capability(263 mAh g^−1 at 2.0 A g^−1)as the anode material for KIBs.

A Regional Economy's Resistance to the COVID-19 Shock: Sales Revenues of Micro-, Small-, and Medium-Sized Enterprises in South Korea

The coronavirus disease 2019(COVID-19) is a global pandemic that has heavily impacted not only the health sector, but also the economic sector in general.Many countries have projected a negative economic impact, and the effect on micro-, small-, and medium-sized enterprises(MSMEs) is predicted to be significantly large.This study estimated the regional resistance of MSME sales revenues and identified the regional economic factors that affect resistance by analyzing South Korea, a country with one of the lowest economic impact projections from COVID-19. Resistance was estimated by comparing sales revenues and changes in resistance observed during the early COVID-19 period to those recorded in the same weeks(weeks 6 to 9) of 2019. The factors that affect regional resistance were determined by conducting robust regression and spatial regression analyses. The results show that the number of confirmed COVID-19 cases, a direct risk factor, is negatively related to regional resilience, while diversity is positively related to regional resistance. To improve the regional resistance against uncertain events, this study recommends increased diversity among regional industrial structures to reduce the duration of the early shock of an unexpected adverse event.

Evaluation of early-age strains and stresses in roller-compacted concrete pavement

In the structural design of a roller-compacted concrete pavement(RCCP), it is crucial to estimate strain and stress developments in the RCCP slab realistically. Since the RCCP mix uses less cement and lower amount of water, shrinkage strain and concrete temperature during the hardening stage are expected to be reduced as compared to those of conventional concrete mixture, resulting in a reduction of the concrete early-age deformation and stress developments in the RCCP slab. In this paper, early-age concrete strain and stress developments in RCCP slab subjected to environmental loads were evaluated. A full-scale test section of RCCP under real climatic conditions was monitored. The early-age total strains,stress-independent strains, shrinkage strains, and coefficient of thermal expansion(CTE) of the RCCP were measured and analyzed. Using the results of measured strains, in-situ CTE and shrinkage strain, and temperature, the early-age concrete stress development is computed by incorporating a viscoelastic property of the early-age concrete. The results revealed that the shrinkage strain of the RCCP is quite low as compared to that of conventional concrete. The early-age stress developments in the RCCP slab are strongly governed by the thermal-induced stresses. Shrinkage-induced stresses were quite small and might be negligible in a preliminary estimation of early-age stress developments in the RCCP slab.

Synergistically coupling of Ni–Fe LDH arrays with hollow Co–Mo sulfide nanotriangles for highly efficient overall water splitting

Developing bifunctional catalysts that can catalyze both oxygen evolution reaction(OER)and hydrogen evolution reaction(HER)is pivotal to commercializing large-scale water splitting.Herein,a novel hollow nanotriangle composed of NiFe LDH-CoMoS_(x) heterojunction(H-CMSx@NiFe LDH)is proposed as a highly efficient bifunctional electrocatalyst for both OER and HER.To fabricate a heterojunction system,ultra-thin nickel–iron layered double hydroxide(NiFe LDH)nanosheets are uniformly electrodeposited onto a metal–organic framework-derived hollow CoMoS_(x) nanotriangle.The strong coupling of CoMoS_(x) and NiFe LDH catalysts forms the intimate heterojunction interfaces to facilitate interfacial charge transfer,which is favorable to enhance the bifunctional catalytic activity.Moreover,the large void of CoMoS_(x) nanotriangles and interconnected ultra-thin NiFe LDH nanosheets result in good electrolyte penetration and gas release.Therefore,the as-prepared H-CMSx@NiFe LDH on nickel foam(NF)exhibits an impressive catalytic activity and durability for OER and HER activities,delivering a current density of 100 mA·cm^(−2) at the small overpotentials of 214 and 299 mV in OER and HER,respectively.Meanwhile,H-CMSx@NiFe LDH/NF proves to be an effective electrode for an alkaline electrolyzer,as a voltage of only 1.99 V is enough to achieve a current density voltage of only 1.99 V is enough to achieve a current density of 400 mA·cm^(−2) with no degradation in performance over 50 h.

Novel N-doped multichannel carbon nanofiber architecture with porous CoS nanoprisms for high-performance potassium-ion batteries

Potassium-ion batteries(PIBs) have gained significant attention as an alternative to lithium-ion batteries(LIBs) due to the abundance of potassium(K) and low cost.Nevertheless,the difficulty in finding appropriate electrode materials that can efficiently store the larger K ions has hindered their practical application.Herein,we report a novel anode material,N-doped multichannel carbon nanofibers embedded with porous CoS nanoprisms(CSP@NMCNF),for high-performance PIBs.The CSP@NMCNF was synthesized using a two-step strategy comprising of the electrospinning of Co acetate hydroxide nanoprism/binary polymer blend and a subsequent heat treatment.The porous CoS nanoprisms with an anisotropic morphology were well aligned along the length axis of the N-doped multichannel carbon nanofibers,thus ensuring their structural stability during the repeated charge-discharge process.In addition,numerous pores facilitated the transport of electrons and ions.Accordingly,the CSP@NMCNF anode exhibited excellent electrochemical performance,delivering a high specific capacity of 368 mAh·g-1at 0.5 A·g-1after 200 cycles and excellent rate capability(232 mAh·g-1at 2.0 A·g-1).
