Rigidity(or stiffness)of materials and extracellular matrix has proven to be one of the most significant extracellular physicochemical cues that can control diverse cell behaviors,such as contractility,motility,and spreading,and the resultant pathophysiological phenomena.Many 2D materials engineered with tunable rigidity have enabled researchers to elucidate the roles of matrix biophysical cues in diverse cellular events,including migration,lineage specification,and mechanical memory.Moreover,the recent findings accumulated under 3D environments with viscoelastic and remodeling properties pointed to the importance of dynamically changing rigidity in cell fate control,tissue repair,and disease progression.Thus,here we aim to highlight the works related with material/matrix-rigidity-mediated cell and tissue behaviors,with a brief outlook into the studies on the effects of material/matrix rigidity on cell behaviors in 2D systems,further discussion of the events and considerations in tissue-mimicking 3D conditions,and then examination of the in vivo findings that concern material/matrix rigidity.The current discussion will help understand the material/matrix-rigidity-mediated biological phenomena and further leverage the concepts to find therapeutic targets and to design implantable materials for the treatment of damaged and diseased tissues.

Structural insights on anti-biofilm mechanism of heated slightly acidic electrolyzed water technology against multi-resistant Staphylococcus aureus biofilm on food contact surface

Slightly acidic electrolyzed water(SAEW)has proven to be an efficient and novel sanitizer in food and agriculture field.This study assessed the efficacy of SAEW(30 mg/L)at 40℃on the inactivation of foodbome pathogens and detachment of multi-resistant Staphylococcus aureus(MRSA)biofilm.Furthermore.the underlying mechanism of MRS A biofilm under heated SAEW at 40℃treatment on metabolic profiles was investigated.The results showed that the heated SAEW at 40℃significantly effectively against foodbome pathogens of 1.96-7.56(lg(CFU/g))reduction in pork,chicken,spinach,and lettuce.The heated SAEW at 40℃treatment significantly reduced MRS A biofilm cells by 2.41(lg(CFU/cm^(2))).The synergistic effect of SAEW treatment showed intense anti-biofilm activity in decreasing cell density and impairing biofilm cell membranes.Global metabolic response of MRSA biofilms,treated by SAEW at 40℃,revealed the alterations of intracellular metabolites,including amino acids,organic acid,fatty acid,and lipid.Moreover,signaling pathways involved in amino acid metabolism,energy metabolism,nucleotide synthesis,carbohydrate metabolites,and lipid biosynthesis were functionally disrupted by the SAEW at 40℃treatment.As per our knowledge,this is the first research to uncover the potential mechanism of heated SAEW treatment against MRSA biofilm on food contact surface.

Tetradentate Pt(II) complex as a singlet exciton sensitizing host for highly efficient green fluorescent organic light-emitting diodes

Efficient green fluorescent organic light-emitting diodes were designed with a phosphorescent metal complex as a singlet exciton sensitizing host (SESH) for achieving high efficiency and low efficiency roll-off through exciton and polaron management. Tetradentate Pt(II) complex (Pt-Trz)possessing a tetradentate ligand with a bulky blocking group not only suppresses the severe formation of excimer or dimer itself but also transfers radiative excitons to fluorescent dopant effectively. The SESH assisted fluorescence (SESHF) device with Pt-Trz as the host exhibits the maximum external quantum efficiency of 13.3%and a lower efficiency roll-off compared with exciplex host device which is a general approach for harvesting triplet excitons. Numerical exciton dynamics modeling unravels that the SESHF device effectively utilizes triplet excitons in the host by intersystem crossing and resonance energy transfer from the triplet state of the host to the singlet state of the terminal emitter. Bimolecular quenching rates of the SESHF device are reduced by 11 times for triplet-triplet annihilation and 1.8 times for tripletpolaron annihilation compared with the exciplex device, which demonstrates that Pt-Trz could be a platform to be used as an efficient host for fluorophores.

Therapeutic options are quite limited in clinics for the successful repair of infected/degenerated tissues.Although the prevalent treatment is the complete removal of the whole infected tissue,this leads to a loss of tissue function and serious complications.Herein the dental pulp infection,as one of the most common dental problems,was selected as a clinically relevant case to regenerate using a multifunctional nanotherapeutic approach.For this,a mesoporous bioactive glass nano-delivery system incorporating silicate,calcium,and copper as well as loading epidermal growth factor(EGF)was designed to provide antibacterial/pro-angiogenic and osteo/odontogenic multiple therapeutic effects.Amine-functionalized Cu-doped bioactive glass nanospheres(Cu-BGn)were prepared to be 50–60 nm in size,mesoporous,positive-charged and bone-bioactive.The Cu-BGn could release bioactive ions(copper,calcium and silicate ions)with therapeutically-effective doses.The Cu-BGn treatment to human umbilical vein endothelial cells(HUVEC)led to significant enhancement of the migration,tubule formation and expression of angiogenic gene(e.***.vascular endothelial growth factor,VEGF).Furthermore,the EGF-loaded Cu-BGn(EGF@Cu-BGn)showed pro-angiogenic effects with antibacterial activity against E.faecalis,a pathogen commonly involved in the pulp infection.Of note,under the co-culture condition of HUVEC with E.faecalis,the secretion of VEGF was up-regulated.In addition,the osteo/odontogenic stimulation of the EGF@Cu-BGn was evidenced with human dental pulp stem cells.The local administration of the EGF@Cu-BGn in a rat molar tooth defect infected with E.faecalis revealed significant in vivo regenerative capacity,highlighting the nanotherapeutic uses of the multifunctional nanoparticles for regenerating infected/damaged hard tissues.

High-index-contrast photonic structures:a versatile platform for photon manipulation

In optics,the refractive index of a material and its spatial distribution determine the characteristics of light propagation.Therefore,exploring both low-and high-index materials/structures is an important consideration in this regard.Hollow cavities,which are defined as low-index bases,exhibit a variety of unusual or even unexplored optical characteristics and are used in numerous functionalities including diffraction gratings,localised optical antennas and low-loss resonators.In this report,we discuss the fabrication of hollow cavities of various sizes(O.2-5μm in diameter)that are supported by conformal dielectric/metal shells,as well as their specific applications in the ultraviolet(photodetectors),visible(light-emitting diodes,solar cells and metalenses),near-infrared(thermophotovoltaics)and mid-infrared(radiative coolers)regions.Our findings demonstrate that hollow cavities tailored to specific spectra and applications can serve as versatile optical platforms to address the limitations of current optoelectronic devices.Furthermore,hollow cavity embedded structures are highly elastic and can minimise the thermal stress caused by high temperatures.As such,future applications will likely include high-temperature devices such as thermophotovoltaics and concentrator photovoltaics.

Photocatalytic decomposition of mobile-source related pollutants using a continuous-flow reactor

This study evaluated the application of a continuous-flow photocatalytic reactor for the control of two mobile-derived pollutants, methyl-tertiary butyl ether （MTBE） and naphthalene, present at in-vehicle levels. Variables tested for this study included the hydraulic diameter （HD）, stream flow rate （SFR）, relative humidity （RH）, and feeding type （FT）. The fixed parameters included contaminant concentration, ultraviolet light source, and the weight of TiO2. In all experimental conditions the adsorption process reached equilibrium within 30 to 180 min for the target compounds, and the outlet concentrations of the photocatalytic oxidation （PCO） reactor while operating reached a steady state within 60 to 180 min. The degradation of the target compounds was dependent on RH, HD, FT, or SFR. The PCO system exhibited high degradation （up to nearly 100% for certain conditions） and mineralization efficiencies of target compounds, suggesting that this system can effiectively be employed to improve indoor air quality. Moreover, it was confirmed that trichloroethylene at urban-ambient level also could enhance the degradation efficiency of naphthalene when applying the PCO technology inside vehicles.

Dear Editor,Macular hole(MH)formation is uncommon in patients with diabetic macular edema(DME).Few cases of MH associated with DME treatment have been described,including a report of four out of eight eyes with MH and diabetic retinopathy(DR)having DME[1].However,no case of resolution of MH after DME treatment has been reported.We report three consecutive cases of MH closure achieved by intravitreal injection for DME treatment.

Peptidyl-prolyl isomerase-like 1(PPIL1)is associated with the human spliceosome complex.However,its function in pre-mRNA splicing remains unclear.In this study,we show that Arabidopsis thaliana CYCLOPHILIN 18-2(AtCYP18-2),a PPIL1 homolog,plays an essential role in heat tolerance by regulating pre-mRNA splicing.Under heat stress conditions,AtCYP18-2 expression was upregulated in mature plants and GFP-tagged AtCYP18-2 redistributed to nuclear and cytoplasmic puncta.We determined that AtCYP18-2 interacts with several spliceosome complex B^(ACT)components in nuclear puncta and is primarily associated with the small nuclear RNAs U5 and U6 in response to heat stress.The AtCYP18-2 loss-of-function allele cyp18-2 engineered by CRISPR/Cas9-mediated gene editing exhibited a hypersensitive phenotype to heat stress relative to the wild type.Moreover,global transcriptome profiling showed that the cyp18-2 mutation affects alternative splicing of heat stress–responsive genes under heat stress conditions,particularly intron retention(IR).The abundance of most intron-containing transcripts of a subset of genes essential for thermotolerance decreased in cyp18-2 compared to the wild type.Furthermore,the intron-containing transcripts of two heat stress-related genes,HEAT SHOCK PROTEIN 101(HSP101)and HEAT SHOCK FACTOR A2(HSFA2),produced functional proteins.HSP101-IR-GFP localization was responsive to heat stress,and HSFA2-Ⅲ-IR interacted with HSF1 and HSP90.1 in plant cells.Our findings reveal that CYP18-2 functions as a splicing factor within the B~(ACT)spliceosome complex and is crucial for ensuring the production of adequate levels of alternatively spliced transcripts to enhance thermotolerance.

The TNFα/TNFR2 axis mediates natural killer cell proliferation by promoting aerobic glycolysis

Natural killer(NK)cells are predominant innate lymphocytes that initiate the early immune response during infection.NK cells undergo a metabolic switch to fuel augmented proliferation and activation following infection.Tumor necrosis factor-alpha(TNFα)is a well-known inflammatory cytokine that enhances NK cell function;however,the mechanism underlying NK cell proliferation in response to TNFαis not well established.Here,we demonstrated that upon infection/inflammation,NK cells upregulate the expression of TNF receptor 2(TNFR2),which is associated with increased proliferation,metabolic activity,and effector function.Notably,IL-18 can induce TNFR2 expression in NK cells,augmenting their sensitivity toward TNFα.Mechanistically,TNFα-TNFR2 signaling upregulates the expression of CD25(IL-2Rα)and nutrient transporters in NK cells,leading to a metabolic switch toward aerobic glycolysis.Transcriptomic analysis revealed significantly reduced expression levels of genes involved in cellular metabolism and proliferation in NK cells from TNFR2 KO mice.Accordingly,our data affirmed that genetic ablation of TNFR2 curtails CD25 upregulation and TNFα-induced glycolysis,leading to impaired NK cell proliferation and antiviral function during MCMV infection in vivo.Collectively,our results delineate the crucial role of the TNFα-TNFR2 axis in NK cell proliferation,glycolysis,and effector function.

joint member angle limits of composite beams and CFT square columns

This work aims to understand the relationship between the member angle limit and the energy ratio for 24 test samples of composite beams and CFT square steel tube columns. This work also compares the formula applicability for member angle limit with the previous test result to provide the basic data for the design of composite beam-CFT column. The evaluation of the member angle limit was performed with concrete compressive strength (fC =22.16 MPa, 30.49 MPa), breadth-to-thickness ratio (B/t=25.0, 33.3, 43.5), and axial capacity ratio (N/N0=0, 0.3, 0.4, 0.5) as the main variables of the test samples. For the relationship between the member angle limit (Ru) and the energy ratio (ES/EC ), the test result shows that the energy ratio becomes lower as the axial capacity ratio and the breadth-to-thickness ratio increase. The energy ratio is lower for the Type B test sample compared with that of Types A and C. For the formula suggested by SATO, the test samples are distributed evenly for comparison between test values and the member angle limit; however, other formulas indicate a deviation. Specifically, for the comparison between R u,cal and R u (Test), Maeda's formula shows severe deviation.
