Pharmacological applications of a novel neoepitope antibody to a modified amyloid precursor protein-derived beta-secretase product

We have previously described a novel artificial NFEVβ-secretase(BACE1)cleavage site,which when introduced into the amyloid-βprecursor protein(APP),significantly enhances APP cleavage by BACE1 in in vitro and cellular *** this study,we describe the identification and characterization of a single chain fragment of variable region(scFv),specific to the EV neo-epitope derived from BACE1 cleavage of the NFEV-containing peptide,and its conversion to *** the scFv displayed on phage and EV-IgG1 show exquisite specificity for binding to the EV neoepitope without cross-reactivity to other NFEV containing peptides or WT-APP KMDA cleavage ***-IgG1 can detect as little as 0.3 nmol/L of the EV ***-IgG1 antibody was purified,conjugated with alkaline phosphatase and utilized in various biological *** the BACE1 enzymatic assay using NFEV substrate,a BACE1 inhibitor MRK-3 inhibited cleavage with an IC50 of 2.4 nmol/L with excellent *** an APP_NFEV stable SH-SY5Y cellular assay,the EC_(50) for inhibition of EV-Aβ peptide secretion with MRK-3 was 236 nmol/L,consistent with values derived using an EV polyclonal *** an APP_NFEV knock-in mouse model,both Aβ_EV40 and Aβ_EV42 peptides in brain homogenate showed excellent gene dosage *** conclusion,the EV neoepitope specific monoclonal antibody is a novel reagent for BACE1 inhibitor discovery for both in vitro,cellular screening assays and in vivo biochemical *** methods described herein are generally applicable to novel synthetic substrates and enzyme targets to enable robust screening platforms for enzyme inhibitors.

A Numerical Study of a TOGA-COARE Squall-Line Using a Coupled Mesoscale Atmosphere-Ocean Model

An atmosphere-ocean coupled mesoscale modeling system is developed and used to investigate the interactions between a squall line and the upper ocean observed over the western Paci?c warm pool during the Tropical Ocean/Global Atmosphere Coupled Ocean and Atmosphere Response Experiment (TOGA-COARE). The modeling system is developed by coupling the Advanced Regional Prediction Sys- tem (ARPS) to the Princeton Ocean Model (POM) through precipitation and two-way exchanges of mo- mentum, heat, and moisture across the air-sea interface. The results indicate that the interaction between the squall-line and the upper ocean produced noticeable di?erences in the sensible and latent heat ?uxes, as compared to the uncoupled cases. Precipitation, which is often ignored in air-sea heat ?ux estimates, played a major role in the coupling between the mesoscale convective system and the ocean. Precipitation a?ected the air-sea interaction through both freshwater ?ux and sensible heat ?ux. The former led to the formation of a thin stable ocean layer underneath and behind the precipitating atmospheric convection. The presence of this stable layer resulted in a more signi?cant convection-induced sea surface temperature (SST) change in and behind the precipitation zone. However, convection-induced SST changes do not seem to play an important role in the intsensi?cation of the existing convective system that resulted in the SST change, as the convection quickly moved away from the region of original SST response.

Correlation between tear fluid and serum vitamin D levels

Background:Vitamin D deficiency is associated with a range of systemic diseases including ocular *** objective of this study is to measure tear vitamin D levels and investigate the correlation between serum and tear vitamin D ***:A total of 48 healthy volunteers without any systemic and ocular disease were recruited for this observational cohort *** was collected using clot activator coated Vacutainer®Plus *** fluid was collected using Schirmer’s *** and tear total 25-hydroxyvitamin D levels were measured by competitive chemiluminescent ELISA and the correlation between the levels were ***:The measured serum 25-hydroxyvitamin D level ranged between 3.3 and 27.5 ng/ml(Mean±SEM,9.4±0.7 ng/ml;Median 8.4 ng/ml).Significantly(p<0.0001)higher level of 25-hydroxyvitamin D was detected in the tears(Mean±SEM,17.0±1.6 ng/ml;Range 3.2–45.8 ng/ml;Median,16.3 ng/ml)compared to *** average of~2 fold(Mean±SEM,1.9±0.2;Range 0.4–5.8;Median,1.7)higher 25-hydroxyvitamin D was observed in tears compared to serum in the *** addition,a positive correlation was observed between serum and tear 25-hydroxyvitamin D levels(r=0.5595;p<0.0001).Conclusions:A higher level of 25-hydroxyvitamin D was observed in the tear fluid compared to that of the *** would be beneficial to consider tear vitamin D levels to study its role with reference to ocular surface diseases.

Sleep spindles detection from human sleep EEG signals using autoregressive (AR) model: a surrogate data approach

A new algorithm for the detection of sleep spindles from human sleep EEG with surrogate data approach is presented. Surrogate data ap-proach is the state of the art technique for nonlinear spectral analysis. In this paper, by developing autoregressive (AR) models on short segment of the EEG is described as a superposition of harmonic oscillating with damping and frequency in time. Sleep spindle events are detected, whenever the damping of one or more frequencies falls below a prede-fined threshold. Based on a surrogate data, a method was proposed to test the hypothesis that the original data were generated by a linear Gaussian process. This method was tested on human sleep EEG signal. The algorithm work well for the detection of sleep spindles and in addition the analysis reveals the alpha and beta band activities in EEG. The rigorous statistical framework proves essential in establishing these results.

Antibacterial activity of four Gracilaria species of red seaweeds collected from Mandapam Coast,Gulf of Mannar Marine Biosphere Reserve,India
