Performance investigation of LAMBDA and bootstrapping methods for PPP narrow-lane ambiguity resolution

Precise point positioning with ambiguity resolution(PPP-AR)is a powerful tool for geodetic and time-constrained applications that require high precision.The performance of PPP-AR highly depends on the reliability of the correct integer carrier-phase ambiguity estimation.In this study,the performance of narrow-lane ambiguity resolution of PPP using the Least-squares AMBiguity Decorrelation(LAMBDA)and bootstrapping methods is extensively investigated using real data from 55 IGS stations over one-month in 2020.Static PPP with 24-,12-,8-,4-,2-,1-and½-h sessions using two different cutoff angles(7°and 30°)was conducted with three PPP modes:i.***.ambiguity-float and two kinds of ambiguity-fixed PPP using the LAMBDA and bootstrapping methods for narrow-lane AR,respectively.The results show that the LAMBDA method can produce more reliable results for 2 hour and shorter observation sessions com-pared with the bootstrapping method using a 7°cutoff angle.For a 30°cutoff angle,the LAMBDA method outperforms the bootstrapping method for observation sessions of 4 h and less.For long observation times,the bootstrapping method produced much more accurate coordinates compared with the LAMBDA method without considering the wrong fixes cases.The results also show that occurrences of fixing the wrong integer ambiguities using the bootstrapping method are higher than that of the LAMBDA method.

Metagenomics and volatile profile of Turkish artisanal Tulum cheese microbiota

Turkish artisanal Tulum cheese is a traditional raw milk cheese and starter cultures are not used for ripening process.In this study,16S rRNA amplicon-based sequencing approach was used for determining bacterial diversity in Tulum cheeses.The volatile organic compound(VOC)profile which contributes to sensory properties of the cheeses was determined via solid-phase microextraction combined with gas chromatography-mass spectrometry(GC-MS/SPME)and correlated with the microbiota.In Tulum cheese samples,Lactobacillaceae(30,71%)phylum widely varied but Streptococcaceae(59,86%)was dominant.It mainly belongs to the four dominant genera of lactic acid bacteria(LAB)and includes Lactobacillus,Lactococcus,Streptococcus and Pediococcus.The bacterial diversity of TC5 sample was much higher than that of other samples.Moreover,several species of non-starter LAB were frequently detected.A total of 36 volatile compounds were identified including hydrocarbons(3),acids(13),alcohols(5),ketones(3),aldehydes(3),esters(8)and miscellaneous compound(1).Pentadecanoic acid,octadecanoic acid,tetradecanoic acid,octanoic acid,hexanoic acid were determined to be dominant in all cheese samples and to be related to microbial flora.Also,the sensory profiles of the samples were examined.The most popular TC6 did not contain 2-butanone and hexanal from the volatile organic compound(VOC)values,while in the most unpopular TC3,2-butanone(7.73%)and hexanal(5.95%)were found to be the highest.The outcome of this study will contribute to literature on bacterial diversity and aromatic profile of Tulum cheese which should be investigated in order to improve quality and safety.

Evaluation of hepatitis B surface antigen and hepatitis B virus-DNA results in postmortem plasma specimens

Objective:To assess the presence of hepatitis B surface antigen,one of the serologic markers of hepatitis B virus(HBV)infection,in postmortem blood samples from autopsy cases using ELISA,and to compare the results with those obtained by PCR,which is the gold standard method in assessing HBV infection.Methods:The HBV test results of the blood samples from 880 autopsy cases determined in our laboratory,were retrospectively studied.Results:When compared with the gold standard method PCR,the sensitivity and specificity of postmortem ELISA were 100%and 84.1%,respectively.Conclusions:The increasingly used molecular diagnostic methods,such as PCR,should be used in cases where serological tests remain insufficient.We think that prospective studies on the comparison of ELISA and PCR assessment of postmortem blood samples with larger material should be carried out.

Background: Genetically engineered animals are essential for gaining a proper understanding of the disease mechanisms of cystic fibrosis(CF). The rat is a relevant laboratory model for CF because of its zootechnical capacity, size, and airway characteristics, including the presence of submucosal glands.Methods: We describe the generation of a CF rat model(F508 del) homozygous for the p.Phe508 del mutation in the transmembrane conductance regulator(Cftr) gene. This model was compared to new Cftr-/-rats(CFTR KO). Target organs in CF were examined by histological staining of tissue sections and tooth enamel was quantified by micro-computed tomography. The activity of CFTR was evaluated by nasal potential difference(NPD) and short-circuit current measurements. The effect of VX-809 and VX-770 was analyzed on nasal epithelial primary cell cultures from F508 del rats.Results: Both newborn F508 del and Knock out(KO) animals developed intestinal obstruction that could be partly compensated by special diet combined with an osmotic laxative. The two rat models exhibited CF phenotypic anomalies such as vas deferens agenesis and tooth enamel defects. Histology of the intestine, pancreas, liver, and lungs was normal. Absence of CFTR function in KO rats was confirmed ex vivo by short-circuit current measurements on colon mucosae and in vivo by NPD, whereas residual CFTR activity was observed in F508 del rats. Exposure of F508 del CFTR nasal primary cultures to a combination of VX-809 and VX-770 improved CFTR-mediated Cl-transport.Conclusions: The F508 del rats reproduce the phenotypes observed in CFTR KO animals and represent a novel resource to advance the development of CF therapeutics.

Patients with cystic fibrosis are particularly at risk of infection with non-tuberculous mycobacteria (NTM). Prevalence of these infections increases with ag e to around 15%. The main species involved are M. abscessus and M. avium, the l atter not found in children under 15. Diagnosis relies on clinical, radiological and above all bacteriological criteria defined by the ATS. Identification of th e causal species of NTM is essential and requires genetic techniques, some of wh ich are currently evaluated. Treatment depends on the mycobacterial species. For M. avium, combined therapy with rifampicin, clarythromycin and ethambutol must be extended 12 months after negativation. M. abscessus infection is particularly resistant to therapy. Usual treatment is a one month course of intravenous imip enem or cefoxitin plus amikacin followed by oral clarithromicin plus ethambutol for at least 12 months after negativation. In case of local lesions, surgery is an option.
