Targeting the inflammasome and adenosine type-3 receptors improves outcome of antibiotic therapy in murine anthrax

AIM:To establish whether activation of adenosine type-3 receptors(A3Rs)and inhibition of interleukin- 1β-induced inflammation is beneficial in combination with antibiotic therapy to increase survival of mice challenged with anthrax spores. METHODS:DBA/2 mice were challenged with Bacillus anthracis spores of the toxigenic Sterne strain 43F2. Survival of animals was monitored for 15 *** treatment(50 mg/kg,once daily,intraperitoneally) was initiated at day+1 simultaneously with the ad- ministration of inhibitors,and continued for 10 *** doses(2.5 mg/kg and 12.5 mg/kg)of acetyl-tyrosylvalyl-alanyl-aspartyl-chloromethylketone(YVAD)and three doses(0.05,0.15 and 0.3 mg/kg)of 1-[2-Chloro- 6-[[(3-iodophenyl)methyl]amino]-9H-purin-9-yl]-1- deoxy-N-methyl-β-D-ribofuranuronamide(Cl-IB-MECA) were *** received YVAD on days 1-4,and Cl-IB-MECA on days 1-10 once daily,subcutaneously. Human lung epithelial cells in culture were challenged with spores or edema toxin and the effects of IB-MECAon phosphorylation of AKT and generation of cAMP were tested. RESULTS:We showed that the outcome of antibiotic treatment in a murine anthrax model could be substantially improved by co-administration of the caspase-1/4 inhibitor YVAD and the A3R agonist *** treatment with these substances and ciprofloxacin resulted in up to 90%synergistic *** untreated mice died,and antibiotic alone protected only 30% of *** conclude that both substances target the aberrant host signaling that underpins anthrax mortality. CONCLUSION:Our findings suggest new possibilities for combination therapy of anthrax with antibiotics,A3R agonists and caspase-1 inhibitors.

In this paper we investigate the gazeau–Klauder coherent states using a newly introduced diagonal ordering operation technique, in order to examine some of the properties of these coherent states. The results coincide with those obtained from other purely algebraic methods, but the calculations are greatly simplified. We apply the general theory to two cases of gazeau–Klauder coherent states： pseudoharmonic as well as the Morse oscillators.

Current self-limitation in a transverse nanosecond discharge with a slotted cathode

A high-voltage transverse pulsed nanosecond discharge with a slotted hollow cathode was found to be a source of high-energy （few kV） ribbon electron beams. Conditions for the formation and extinction of electron beams were experimentally studied in discharges in helium at pressures of 1-100 Torr. It was found that interaction of fast electrons with a non-uniform electric field near the slotted cathode led to limitation of the magnitude of the discharge current. A physical model was developed to describe the discharge current self-limitation that was in satisfactory agreement with the experimental results. Some technical solutions that are expected to increase the upper current limits in transverse nanosecond discharge are discussed.

The present paper is devoted to a theoretical analysis of sliding friction under the influence of oscillations perpendicular to the sliding *** contrast to previous works we analyze the influence of the stiffness of the tribological contact in detail and also consider the case of large oscillation amplitudes at which the contact is lost during a part of the oscillation period,so that the sample starts to 'jump'.It is shown that the macroscopic coefficient of friction is a function of only two dimensionless parameters-a dimensionless sliding velocity and dimensionless oscillation *** function in turn depends on the shape of the contacting *** the present paper,analysis is carried out for two shapes:a flat cylindrical punch and a parabolic *** we consider 'stiff systems',where the contact stiffness is small compared with the stiffness of the *** role of the system stiffness will be studied in more detail in a separate paper.

Nitrogen and Carbon Isotope Data of Olenekian to Anisian Deposits from Kamenushka/South Primorye, Far-Eastern Russia and Their Palaeoenvironmental Significance

The Kamenushka Formation, exposed in the northern part of South Primorye （Kamennshka-1 and Kamenushka-2 sections）, is one of the few localities in the world with richly fossiliferous Lower-Upper Olenekian sedimentary successions. Lower to Middle Triassic ammonoid-, brachiopod- and conodont-bearing silty-clayey deposits of the Kamenushka-1 and Kamenushka-2 sections have been isotope-geochemically investigated in detail. As a result, these sections, together with the previously investigated Abrek Section, exposed in the southern part of South Primorye, provide almost complete ^15Non- and ^13Corg- records for the Lower Triassic of this region. Nine N- isotope intervals and the five negative C-isotope excursions, reflecting, apparently, unstable climatic and hydrological conditions, have been distinguished in the Lower Triassic of South Primorye. On the basis of the new C-isotope data the Mesohedenstroemia bosphorensis Zone （upper part）, Shimanskyites shimanskyi and Neocolumbites insignis zones of South Primorye are correlated now with the Lower Smithian part of the Yinkeng Formation, the Upper Smithian part of the Helongshan Formation and the Middle Spathian part of the Nanlinghu Formation in South China, respectively, as has been observed in the Abrek, Kamenushka-2, West Pingdingshan and Majiashan sections.

The influence of out-of-plane oscillations on friction is a well-known phenomenon that has been studied extensively with various experimental methods,e.g.,pin-on-disk ***,existing theoretical models have yet achieved only qualitative correspondence with *** we argue that this may be due to the system dynamics (mass and tangential stiffness) of the pin or other system components being *** paper builds on the results of a previous study [19] by taking the stiffness and resulting dynamics of the system into *** main governing parameters determining macroscopic friction,including a dimensionless oscillation amplitude,a dimensionless sliding velocity and the relation between three characteristic frequencies (that of externally excited oscillation and two natural oscillation frequencies associated with the contact stiffness and the system stiffness) are *** the limiting cases of a very soft system and a very stiff system,our results reproduce the results of previous *** between these two limiting cases there is also a resonant case,which is studied here for the first *** resonant case is notable in that it lacks a critical sliding velocity,above which oscillations no longer reduce *** obtained for the resonant case are qualitatively supported by experiments.

Non-adhesive and adhesive contacts of an elastic quarter-or eighth-space with freely sliding sides

The contact of an elastic quarter-or eighth-space is studied under the condition that the movement of the side surface of the quarter-space is constrained:It can slide freely along the plane of the side surface but its normal movement is blocked(for example,by a rigid wall).The solution of this contact problem can be easily achieved by additionally applying a mirrored load to an elastic ***-adhesive contact and the Johnson–Kendall–Roberts(JKR)-type adhesive contact between a rigid sphere and an elastic quarter-space under such a boundary condition is numerically simulated using the fast Fourier transform(FFT)-assisted boundary element method(BEM).Contacts of an elastic eighth-space are investigated using the same *** on the position of the sphere relative to the side edge,different contact behavior is *** the case of adhesive contact,the force of adhesion first increases with increasing the distance from the edge of the quarter-space,achieves a maximum,and decreases further to the JKR-value in large distance from the *** enhancement of the force of adhesion compared to the half-space-contact is associated with the pinning of the contact area at the *** provide the maps of the force of adhesion and their analytical approximations,as well as pressure distributions in the contact plane and inside the quarter-/eighth-space.

Method of reduction of dimensionality in contact and friction mechanics:A linkage between micro and macro scales

Active control of friction by transverse oscillations

The present paper is devoted to a theoretical analysis of sliding friction under the influence of in-plane oscillations perpendicular to the sliding direction. Contrary to previous studies of this mode of active control of friction, we consider the influence of the stiffness of the tribological contact in detail and show that the contact stiffness plays a central role for small oscillation amplitudes. In the present paper we consider the case of a displacement-controlled system, where the contact stiffness is small compared to the stiffness of the measuring system. It is shown that in this case the macroscopic coefficient of friction is a function of two dimensionless parameters— a dimensionless sliding velocity and dimensionless oscillation amplitude. In the limit of very large oscillation amplitudes, known solutions previously reported in the literature are reproduced. The region of small amplitudes is described for the first time in this paper.
