This study presents the assumptions and strategies for the practical implementation of the dynamic mode decomposition approach in the wide-area monitoring system of the Italian transmission system operator,Terna.The procedure setup aims to detect poorly damped interarea oscillations of power systems.Dynamic mode decomposition is a data-driven technique that has gained increasing attention in different fields;the proposed implementation can both characterize the oscillatory modes and identify the most influenced areas.This study presents the results of its practical implementation and operational experience in power system monitoring.It focuses on the main characteristics and solutions identified to reliably monitor the interarea electromechanical modes of the interconnected European power system.Moreover,conditions to issue an appropriate alarm in case of critical operating conditions are described.The effectiveness of the proposed approach is validated by its application in three case studies:a critical oscillatory event and a short-circuit event that occurred in the Italian power system in the previous years,and a 15-min time interval of normal grid operation recorded in March 2021.

As a dynamic ex situ conservation strategy,a clonal seed orchard was started in a nursery in Pomaio(POM)in central Italy in 1993 for an assisted migration experiment of Abies nebrodensis(Lojac.)Mattei.Two artifi cial ex situ populations were planted with this gene pool:a seedling arboretum in Pieve Santo Stefano(PSS)and a small dendrological collection in Papiano(PAP),both originating from the Sicilian relict population.Here,using AFLP markers,we estimated the relatedness among the relocated genotypes of the three collections to check whether the three collections had suffi cient genetic variability to be considered as additional sources of variability to the original gene pool for the assisted migration strategy.High individual genetic variability was found in the collections;each plant had a diff erent genotype and was confi rmed to belong to its population of origin.PAP and PSS trees were shown to be only from the original population of A.nebrodensis species and were derived from a limited set of maternal fertile genotypes.Based on the Sicilian fi r population inventory,nursery production in Sicily,and structure clustering analysis,close genetic relationships among POM,PAP and several PSS trees(1–35)were evident.Similarly,the PSS group(36–78)was genetically close to tree 1 of POM and in a lesser proportion to plants 7 and 9 of POM.The sampling of seedlings used to form batches in the nursery might have infl uenced the structure of the resultant plantations.All genotypes will be useful for enriching the original gene pool.

Since the coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has rapidly spread worldwide,there is still limited knowledge about this condition and its natural history.Children have been relatively spared during COVID-19 pandemic but a novel syndrome known as multisystem inflammatory syndrome(MIS-C)has emerged,following a SARS-CoV-2 infection in children and adolescents.This syndrome can lead to shock and multiple organ failure requiring intensive care.Although COVID-19 clinical research focuses on respiratory symptoms,extrapulmonary involvement such as gastrointestinal(GI)and hepatic manifestations should also be considered.In fact,GI and hepatic involvement play an important role among the most common presenting symptoms of both pediatric and adult COVID-19 and MIS-C.This involvement can not only be one of the most common presenting clinical features but also one of the sequelae of these syndromes.Abdominal ultrasonography monitoring could be very useful to identify a potential involvement of the GI tract and liver.Moreover,long-term follow-up is needed and would be essential to define the long-term outcomes of these patients.

Purpose:Assessing the quality of the ocular surface by in vivo scanning laser confocal microscopy(IVCM)in primary open angle glaucoma(POAG)patients treated by Xen 45 Gel Stent,medical therapy and trabeculectomy.Methods:Retrospective,single-center,single-masked,comparative study including 60 eyes of 30 patients(mean age 61.16±10 years)affected by POAG.Eyes were divided into 3 groups:Group 1 eyes underwent the Xen 45 Gel Stent procedure,Group 2 eyes were under medical therapy,Group 3 eyes were surgically treated by trabeculectomy.All patients underwent HRT II IVCM analysis of cornea,limbus,conjunctiva,sub-tenionian space and sclera.Results:The Xen 45 Gel stent,if properly positioned in the sub-conjunctival space preserves goblet cells and limits ocular surface inflammation.Regular corneal epithelial cells with micro-cysts,and normo-reflective sub-epithelial nerve plexus are documented by IVCM.In sub Tenon’s implants an alternative lamellar intra-scleral filtration is detectable.Combined surgical procedures show a noticeable number of inflammatory cells with rare micro-cysts.Post-trabeculectomy inflammatory reaction is more evident than Xen 45 Gel Stent associated surgical procedures,but less than medical therapy where a conspicuous presence of Langerhans cells,peri-neural infiltrates,marked loss of goblet cells and fibrosis is visible.Conclusion:Ocular surface inflammation was more notable in topical therapy than after trabeculectomy,which itself causes more inflammation than XEN Gel stents.

We applied a multi-taxa approach integrating the co-occurrence of plants,ground beetles,spiders and springtails with soil parameters(temperatures and chemical characteristics)in order to describe the primary succession along two glacier forelands in the Maritime Alps(Italy),a hotspot of Mediterranean biodiversity.We compared these successions to those from Central Alps:Maritime glacier forelands markedly differ for their higher values of species richness and species turnover.Contrary to our expectation,Maritime glacier forelands follow a‘replacement change model’,like continental succession of Inner Alps and differently from other peripheral successions.We propose that the temperatures along these Mediterranean glacier forelands are warmer than those along other Alpine glacier forelands,which promote the faster species turnover.Furthermore,we found that early and mid successional stages of the investigated glaciers are richer in cold-adapted and endemic species than the later ones:we confirmed that the‘replacement change’model disadvantages pioneer,cold-adapted species.Given the overall correspondence among coldadapted and endemic species,the most threatened in this climate phase,our results raise new concerns about the extinction risk of these species.We also describe supraglacial habitat of Maritime glaciers demonstrating that supraglacial debris represents an environment decoupled from the regional climate and may have an important role as refugium for coldadapted and hygrophilous plant and animal species,whose survival can be threatened by climate change and by a rapid ecological succession in the adjacent forelands.

SARS-CoV-2 virus is responsible for the current worldwide coronavirus disease 2019 (COVID-19) pandemic, infecting millions of people and causing hundreds of thousands of deaths. Understanding the antibody response to SARS-CoV-2 is crucial for the development of vaccines, therapeutics and public health interventions. However, lack of consistency in methods used to monitor antibody response to SARS-CoV-2 leaves some uncertainty in our fine understanding of the human antibody response mounted following SARS-CoV-2 infection. We developed a peptide-based enzyme-linked immunosorbent assay (ELISA) by selecting 7 synthetic peptides from the spike, membrane, and nucleocapsid protein sequences of SARS-CoV-2, which effectively detects the antibody response mounted by all COVID-19 convalescent tested. Strikingly, the assay shows a profound difference in antibody response among individual subjects, which may have a significant impact on disease severity. Together, our results define an efficient and specific serological assay to consistently measure the antibody response following SARS-CoV-2 infection, as well as help the design of vaccine and therapeuticals for prevention and treatment of COVID-19.

BACKGROUND Treatment of infantile-onset inflammatory bowel disease(IO-IBD)is often challenging due to its aggressive disease course and failure of standard therapies with a need for biologics.Secondary loss of response is frequently caused by the production of anti-drug antibodies,a well-known problem in IBD patients on biologic treatment.We present a case of IO-IBD treated with therapeutic drug monitoring(TDM)-guided high-dose anti-tumor necrosis factor therapy,in which dose escalation monitoring was used as a strategy to overcome anti-drug antibodies.CASE SUMMARY A 5-mo-old boy presented with a history of persistent hematochezia from the 10th d of life,as well as relapsing perianal abscess and growth failure.Hypoalbuminemia,anemia,and elevated inflammatory markers were also present.Endoscopic assessment revealed skip lesions with deep colic ulcerations,inflammatory anal sub-stenosis,and deep fissures with persistent abscess.A diagnosis of IO-IBD Crohn-like was made.The patient was initially treated with oral steroids and fistulotomy.After the perianal abscess healed,adalimumab(ADA)was administered with concomitant gradual tapering of steroids.Clinical and biochemical steroid-free remission was achieved with good trough levels.After 3 mo,antibodies to ADA(ATA)were found with undetectable trough levels;therefore,we optimized the therapy schedule,first administering 10 mg weekly and subsequently up to 20 mg weekly(2.8 mg/kg/dose).After 2 mo of high-dose treatment,ATA disappeared,with concomitant high trough levels and stable clinical and biochemical remission of the disease.CONCLUSION TDM-guided high-dose ADA treatment as a monotherapy overcame ATA production.This strategy could be a good alternative to combination therapy,especially in very young patients.

Obstetric traumatic separation of the distal humeral epiphysis is a very uncommon injury, which presents a diagnostic challenge. These case serials reviewed the functional outcomes of 5 patients who had sustained a fracture-separation of the distal humeral epiphysis at birth. The diagnosis was made at a mean time of 40.8 h after delivery. All the patients were treated with gentle close manipulation, reduction under fluoroscopy and above-elbow cast application. After discharge, the patients were followed up for a mean of 30 months. Clinico-radiological results were excellent in four patients. One case necessitated closed reduction and percutaneous K-wire fixation at one week follow-up due to failed reduction. Cubitusvarus deformity was the only complication noted in 1 case. Good functional outcome can be expected in newborns with fracture-separation of the distal humeral epiphysis wherein the physis is anatomically reduced.
